Interacting influences of pregnancy and corneal injury on rabbit lacrimal gland immunoarchitecture and function.

PURPOSE: Previous reports indicated that pregnancy and corneal injury (CI) trigger alterations of lacrimal gland (LG) growth factor expression and redistributions of lymphocytes from periductal foci to acini. The purpose of this study was to test our hypothesis that pregnancy would exacerbate the changes induced by CI. METHODS: Corneas were injured with scalpel blades, and, 2 weeks later, LGs were collected for immunocytochemistry and Western blot analysis. Lacrimal fluid was collected under basal- and pilocarpine-stimulated conditions for protein determination and Western blot analyses. RES

[An obstetrical injury as the etiology of chronic corneal edema. Clinical study with scanning microscopy].

The case of a young woman with an unilateral corneal edema with regular folds of the descemet's membrane is reported. These lesions originate from an obstetrical traumatism by forceps and are associated with cutaneous scars of the temples. Scanning electron microscopy of the cornea shows multiple endothelial fissures; they are responsible for the late corneal decompensation.

Plasma fibrin membranes loaded with bone marrow mesenchymal stem cells and corneal epithelial cells promote corneal injury healing via attenuating inflammation and fibrosis after corneal burns.

The shortage of corneal donors has prompted the development of tissue-engineered corneal grafts as an alternative solution. Currently, amniotic membranes with good biocompatibility are widely used as scaffolds for loading stem cells in the treatment of corneal injury. However, this approach has its limitations. In this study, BMSCs were induced to differentiate into corneal epithelial cells via direct contact co-culture, and platelet-poor plasma was used to prepare fibrin gels, which were compressed to remove excess liquid and then lyophilized to obtain plasma fibrin membranes (PFMs). A tissue

Corneal Nerve Injury and Regeneration: Mechanisms, Clinical Consequences, and Emerging Therapeutic Strategies.

The cornea is among the most densely innervated tissues in the body and depends on intact sensory and autonomic innervation to maintain epithelial integrity, tear film homeostasis, and ocular surface defense. Corneal nerve injury may occur after surgery, infection, inflammation, trauma, or systemic disease and can lead to reduced corneal sensitivity, impaired epithelial healing, dry eye, neuropathic pain, and neurotrophic keratopathy. Effective management requires both treatment of the underlying cause and restoration of a microenvironment permissive for reinnervation. This review examines the

Treatment of nitrogen mustard-induced corneal injury with alpha-melanocyte stimulating hormone.

Nitrogen mustard (NM) exposure leads to severe corneal damage, resulting in persistent corneal inflammation, epithelial damage, endothelial dysfunction, and vision impairment. Effective therapeutic strategies to mitigate these effects remain limited. This study evaluates the protective effects of alpha-melanocyte-stimulating hormone (α-MSH) in a murine model of NM-induced corneal injury. C57BL/6 mice were exposed to NM and treated with systemic α-MSH for 28 days. Clinical assessments, histological analysis, anterior segment optical coherence tomography (AS-OCT), and immunohistochemistry were p

Alkaline Phosphatase-Triggered Spatiotemporal Repair of Corneal Injury with TB500 Peptide Hydrogel.

Corneal injury remains a significant clinical challenge due to the limited regenerative capacity of the cornea and the difficulties associated with maintaining drug retention at the injury site. This study presents a novel spatiotemporal repair strategy for corneal wounds, utilizing an alkaline phosphatase (ALP)-triggered, lesion-responsive peptide hydrogel that incorporates TB500, a biologically active peptide with the amino acid sequence LKKTETQ, which has not been previously explored for corneal disease treatment. The hydrogel is designed through enzyme-instructed self-assembly (EISA) of a

Metabolomics for identifying pathways involved in vesicating agent lewisite-induced corneal injury.

Lewisite (LEW) is an arsenical vesicant that can be a potentially dangerous chemical warfare agent (CWA). Eyes are particularly susceptible to vesicant induced injuries and ocular LEW exposure can act swiftly, causing burning of eyes, edema, inflammation, cell death and even blindness. In our previous studies, we developed a LEW exposure-induced corneal injury model in rabbit and showed increased inflammation, neovascularization, cell death, and structural damage to rabbit corneas upon LEW exposure. In the present study, we further assessed the metabolomic changes to delineate the possible mec

Corneal burn injury in picosecond laser treatment: A potential hazard in cosmetic therapy.

Key PointsWithout appropriate protection, cosmetic laser treatments can cause severe ocular side effects and permanent visual impairment. We present this case to emphasize the importance of ocular protection during the entire treatment period, especially treatments of lesions over the eyelids. According to our knowledge and database search, this is the first reported ocular injury caused by picolaser.

Phase-specific functions of macrophages determine injury-mediated corneal hem- and lymphangiogenesis.

Macrophages are critical mediators of injury-associated corneal hemangiogenesis (HA) and lymphangiogenesis (LA). Yet, molecular regulators of the hem- and lymphangiogenic potential of corneal wound macrophages are poorly understood. Using two different mouse models of acute (perforating corneal incision injury) and chronic (corneal suture placement model) corneal injury, here we identified distinct functions of early- versus late-phase corneal wound macrophages in corneal HA and LA. Whereas early-phase wound macrophages are essential for initiation and progression of injury-mediated corneal HA

The Role of Microglia and Peripheral Monocytes in Retinal Damage after Corneal Chemical Injury.

Eyes that have experienced alkali burn to the surface are excessively susceptible to subsequent severe glaucoma and retinal ganglion cell loss, despite maximal efforts to prevent or slow down the disease. Recently, we have shown, in mice and rabbits, that such retinal damage is neither mediated by the alkali itself reaching the retina nor by intraocular pressure elevation. Rather, it is caused by the up-regulation of tumor necrosis factor-α (TNF-α), which rapidly diffuses posteriorly, causing retinal ganglion cell apoptosis and CD45+ cell activation. Herein, we investigated the involvement of

CD18 and ICAM-1-dependent corneal neovascularization and inflammation after limbal injury.

Extensive limbal injury is a leading cause of irreversible blindness. The destruction of corneal limbal stem cells often results in corneal neovascularization and an optically inferior epithelium. Previous work has shown that the neovascularization after limbal injury is vascular endothelial growth factor (VEGF)-dependent, with much of the VEGF emanating from the inflammatory cells that invade the cornea. Using a relevant mouse model of limbal injury, we examined the role of CD18 and intercellular adhesion molecule-1 (ICAM-1) in limbal injury-induced neovascularization. The results show that C
