Genetic ablation of CD36 induces age-related corneal neovascularization.

PURPOSE: Corneal avascularity is tightly regulated by a balance between angiogenic and antiangiogenic factors (angiogenic privilege). In the current study, we tested the hypothesis that the CD36+/+ antiangiogenic receptor contributes toward the maintenance of corneal avascularity. METHODS: Corneas of CD36 wild-type (CD36) and knockout (CD36) mice aged 4, 16, 52, and 78 weeks were histologically evaluated for corneal haze and neovascularization (NV). Quantitative real-time polymerase chain reaction was performed on corneal tissue from CD36+/+ and CD36-/- mice aged 4 and 52 weeks to examine the

Role of macrophage migration inhibitory factor in corneal neovascularization.

PURPOSE: To determine the role of macrophage migration inhibitory factor (MIF) in inflammatory corneal neovascularization. METHODS: Corneal neovascularization was induced by suturing 10-0 nylon 1 mm away from limbal vessel or limbal scraping after 0.15 M NaOH application in BALB/c mice. MIF expression was evaluated by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR), Western blot analysis, and immunohistochemistry. To investigate the function of MIF in inflammatory corneal neovascularization, the neovascularized area and number of infiltrating F4/80-positive cells (mo

Role of protein tyrosine phosphorylation in rat corneal neovascularization.

BACKGROUND: Recent studies have suggested that tyrosine kinase pathways that are activated by angiogenic growth factors may play a role in corneal neovascularization. METHODS: Corneal neovascularization was induced in rat corneas by chemical cauterization. At 6, 24, 48, 96, and 168 h after chemical cauterization the rat corneas without the corneal epithelium were prepared for gel electrophoresis. Total protein profiles of the corneal samples were examined by staining gels with Coomassie brilliant blue. Tyrosine-phosphorylated proteins, three angiogenic growth factors (basic fibroblast growth

Corneal neovascularization induced by stimulated lymphocytes in inbred mice.

The intrastromal implantation of Concanavalin A-stimulated allogeneic lymphocytes induced corneal neovascularization (CNV) in inbred C57BL/6 and BALB/c mice. Control syngeneic stimulated, allogeneic non-stimulated, and allogeneic stimulated irradiated lymphocytes were not angiogenic. CNV induced by allogeneic stimulated lymphocytes in BALB/c recipients was significantly greater than in C57BL/6 recipients. This response reflected host-versus-graft reactivity, since parental recipients responded to F1 hybrid donors, while F1 hybrids did not respond to parental donors. The ability of stimulated l

Corneal argon laser photocoagulation for neovascularization in penetrating keratoplasty.

Corneal argon laser photocoagulation (CALP) was used in 13 patients to treat deep stromal vascular ingrowth. Eight patients had undergone successful penetrating keratoplasty but had developed deep stromal vessels into the graft associated with signs of graft rejection, which did not improve with steroid treatment alone (group 1). After CALP, there was marked regression of the neovascularization with reversal of graft rejection in all eyes. Three additional patients with vascularized corneas, referred for penetrating keratoplasty, underwent CALP preoperatively with obliteration of the vessels (

[Regression of corneal neovascularization by laser treatment? (author's transl)].

The corneal neovascularization produced by NaOH burns was examined in two groups of pigmented rabbits following laser treatment. The treatment was carried out with energy level 500 mW, spot diameter 200 micron, and exposure time 1 s throughout. In the first group, a single newly formed vessel was coagulated in each case. Subsequent fluorescein angiography invariably showed an incomplete occlusion of the vessel. In the second group, we coagulated a section of the neovascularization network at its origin in the corneal limbus. After 48 h, fluorescein perfusion was once again observed, but the ve

A Mouse Model for Corneal Neovascularization by Alkali Burn.

Corneal neovascularization (CoNV), a pathological form of angiogenesis, involves the growth of blood and lymph vessels into the avascular cornea from the limbus and adversely affects transparency and vision. Alkali burn is one of the most common forms of ocular trauma that leads to CoNV. In this protocol, CoNV is experimentally induced using sodium hydroxide solution in a controlled manner to ensure reproducibility. The alkali burn model is useful for understanding the pathology of CoNV and can be extended to study angiogenesis in general because of the avascularity, transparency, and accessib

The functional role of decorin in corneal neovascularization in vivo.

Our earlier decorin (Dcn) gene overexpression studies found that the targeted Dcn gene transfer into the cornea inhibited corneal angiogenesis in vivo using a rabbit model. In this study, we tested the hypothesis that anti-angiogenic effects of decorin in the cornea are mediated by alterations in a normal physiologic balance of pro- and anti-angiogenic factors using decorin deficient (Dcn-/-) and wild type (Dcn+/+) mice. Corneal neovascularization (CNV) in Dcn-/- and Dcn+/+ mice was produced with a standard chemical injury technique. The clinical progression of CNV in mice was monitored with s

Intrastromal injection of bevacizumab in patients with corneal neovascularization.

Corneal neovascularization (NV) not only reduces visual acuity, but it also causes loss of the cornea's immune privilege, strongly contributing to a worse prognosis in penetrating keratoplasty. Several mediators participate in corneal angiogenesis, and the role of vascular endothelial growth factor (VEGF) has been extensively proven. Anti-VEGF agents have been shown to be effective in slowing the growth of corneal neovessels. Bevacizumab, an anti-VEGF agent, has been successfully used in the treatment of corneal neovascularization. In this paper, we report a series of patients who underwent in

Indomethacin v. dexamethasone in the suppression of corneal neovascularization.

A 7-0 silk suture was placed in one of the corneas of each of 18 albino rabbits as a vasogenic stimulus. Two drops of normal saline, a 10.3 mg/ml suspension of indomethacin or a 0.1% suspension of dexamethasone, allocated in double-masked fashion, were then instilled in the 18 eyes three times per day. There was a statistically significant difference (p less than 0.01) in the rate of neovascularization between the 6 control corneas and the 12 treated corneas but no significant difference in the rate or the quality of neovascularization between the 6 indomethacin-treated corneas and the 6 dexam

A synergistic antioxidant nanozyme platform for corneal neovascularization treatment.

Corneal neovascularization (CNV) is recognized as one of the most common blinding eye diseases, afflicting millions of individuals globally each year. Substantial evidence has demonstrated that oxidative stress and inflammation constitute pivotal pathogenic factors in the initiation and progression of CNV. In this study, we prepared a synergistic nanozyme platform for the antioxidant therapy of CNV. Specifically, we co-assembled manganese(iii) meso-tetra(4-carboxyphenyl)porphine (Mn-TCPP) with octahydrated zirconium(iv) chloride, followed by surface modification with platinum nanoparticles (Pt

TLR4 signal inhibition alleviates alkali-burn induced corneal neovascularization.

Toll-like receptor 4 (TLR4), recognized as a fundamental mediator of inflammatory signaling, plays a crucial role in orchestrating the inflammatory response. Previous studies suggested that TLR4 knockout (KO) notably reduced corneal vascular areas induced by silver nitrate burn based on the morphological observation. The current study seeks to elucidate the influence of TLR4 signaling on corneal neovascularization (CNV) and to examine the underlying mechanisms. The model of alkali burn (AB)-induced CNV was built using TLR4 KO and wildtype (WT) mice. CNV was detected using a slit lamp. Corneal

Sunitinib-eluting thin films for Inhibition of corneal neovascularization.

Corneal disease is a leading cause of blindness globally, and is commonly associated with corneal neovascularization, which results in the loss of optical clarity and visual impairment. A wide variety of factors contribute to abnormal growth of new blood vessels into the cornea, including contact lens wear, infectious keratitis, corneal graft rejection, inflammatory disorders and trauma. In each case, proangiogenic factors such as vascular endothelial, platelet-derived, and fibroblast growth factors fuel new blood vessel growth and proliferation. Topical corticosteroids are a front-line treatm
