Coal hydrogenation and environmental health.

Planning of coal hydrogenation processes, such as liquifaction and gasification, requires consideration of public health implications. Commercial plants will require coal quantities greater than or equal to 20,000 tons/day and the large size of these plants calls for careful consideration of the potential health hazards from the wastes and products of such processes. Analysis of pollution potential can roughly be divided into three categories: raw material structure and constituents, process design, and mode of plant operation. Identifiable pollutants include hydrogen cyanide, phenols, cresols

Sample preparation and chromatographic methods for the determination of protein-bound uremic retention solutes in human biological samples: An overview.

Protein-bound uremic retention solutes, such as indole-3-acetic acid, indoxyl sulfate, p-cresol and p-cresol sulfate, are associated with the development of several pathologies, namely renal, cardiovascular, and bone toxicities, due to their potential accumulation in the human body, thus requiring analytical methods for monitoring and evaluation. The present review addresses conventional and advanced sample treatment procedures for sample handling and the chromatographic analytical methods developed for quantification of these compounds in different biological fluids, with particular focus on

Development of Rapid and Economical Colorimetric Screening Method for p-Phenylenediamine in Variety of Biological Matrices and its Application to Eleven Fatal Cases of p-Phenylenediamine Poisoning.

A rapid colorimetric method for detection of p-phenylenediamine (PPD) in various biological samples is developed. The o-cresol test for acetaminophen detection has been modified to detect PPD in blood, urine, gastric contents, and liver. After precipitating protein with trichloroacetic acid solution (2 mL, 10% w/v), biological specimens were required to convert PPD metabolites to PPD by acid hydrolysis. Finally, o-cresol solution (1 mL, 1% w/v), hydrogen peroxide (200 μL, 3%v/v), and concentrated ammonium hydroxide (0.5 mL) were added in the biological samples. The presence of PPD was indicate

Response of Lucilia sericata (Diptera: Calliphoridae) to Screwworm Oviposition Attractant.

The sheep blowfly, Lucilia sericata Meigen (Diptera: Calliphoridae), causes sheep myiasis in various parts of the world. Female flies are attracted to sheep following various olfactory cues emanating from the sheep's body, and oviposit on suitable substrates on sheep ultimately causing myiasis. Earlier workers attempted to reduce fly population in the field, with some success, using traps baited with various attractants. This research was conducted to determine if L. sericata would respond to a recently developed synthetic attractant that has attracted gravid screwworms, Cochliomyia hominivora

Flow-through sol-gel optical biosensor for the colorimetric determination of acetazolamide.

An optical biosensor based on immobilised carbonic anhydrase and its application to the determination of the anti-glaucoma agent acetazolamide by enzyme inhibition measurements, is described. The enzyme and a pH indicator dye, cresol red, were physically immobilised in overlapped sol-gel thin films, in a dual-layer format. Carbonic anhydrase catalyses the dehydration of HCO3-, which in turn causes a change of pH in the microenvironment of the sensor. By following the colour transition of cresol red, the enzymatic reaction as well as its inhibition by acetazolamide can be monitored. The sensor

Patulin biosynthesis: the metabolism of m-hydroxybenzyl alcohol and m-hydroxybenzaldehyde by particulate preparations from Penicillium patulum.

The ring hydroxylation of m-hydroxybenzyl alcohol to gentisyl alcohol by a particulate preparation from Penicillium patulum has been characterised. The activity was shown to be closely associated with, but not necessarily identical to, m-cresol 2-hydroxylase activity of the 105 000 X g microsomal fraction. As with both the m-cresol hydroxylases of this system, m-hydroxybenzyl alcohol hydroxylase required oxygen and NADPH for activity. A Km value for m-hydroxybenzyl alcohol of 15 muM was measured. Inhibition of the hydroxylase activity and its reversal by light, as well as the action of cytochr

Thrombolome and Its Emerging Role in Chronic Kidney Diseases.

Patients with chronic kidney disease (CKD) are at an increased risk of thromboembolic complications, including myocardial infarction, stroke, deep vein thrombosis, and pulmonary embolism. These complications lead to increased mortality. Evidence points to the key role of CKD-associated dysbiosis and its effect via the generation of gut microbial metabolites in inducing the prothrombotic phenotype. This phenomenon is known as thrombolome, a panel of intestinal bacteria-derived uremic toxins that enhance thrombosis via increased tissue factor expression, platelet hyperactivity, microparticles re

Treatment of women who have undergone chemically induced abortions.

The pathophysiology of abortion induced by the transcervical introduction of compounds containing mixtures of soap, cresol and phenol is complex. A plan is necessary for the treatment of such critically ill women. The complex, multisystem damage produced by those compounds requires an extremely sophisticated treatment approach to avoid the many pitfalls that beset those cases.

Decoding structural determinants of aryl hydrocarbon receptor antagonism by monoterpenoids.

Monocyclic monoterpenoids carvones have been recently identified as atypical negative allosteric modulators of aryl hydrocarbon receptor (AhR). In the current work, we performed AhR antagonist activity screening of 100 natural and synthetic monoterpenoids, and their analogues. Using SAR approach, structural determinants of AhR antagonist activity were assigned, including CO presence/position, planarity, and C3/C5-alkylation. Applying pyramidal selection criteria, including absence of residual agonist activity, no cytotoxicity, strong antagonist potency, and pan-antagonism against diverse AhR a

Functional characterization of the RYR1 mutation p.Arg4737Trp associated with susceptibility to malignant hyperthermia.

Aside from the in vitro contracture test, genetic screening for causative RYR1 mutations is the established procedure to diagnose susceptibility to malignant hyperthermia (MH). However, currently only 34 out of more than 300 known RYR1 mutations have been confirmed to be causative for MH by experimental studies addressing their functional impact on intracellular calcium homeostasis. The RYR1 mutation p.Arg4737Trp has been recently detected in a German MH family. To evaluate the effects of that mutation on intracellular calcium handling, the response after stimulation with the RYR1 agonist 4-ch

Inhibitory effects of toxic compounds on nitrification process for cokes wastewater treatment.

Cokes wastewater is one of the most toxic industrial effluents since it contains high concentrations of toxic compounds such as phenols, cyanides and thiocyanate. Although activated sludge process has been adapted to treat this wastewater, nitrification process has been occasionally upset by serious inhibitory effects of toxic compounds. In this study, therefore, we examined inhibitory effects of ammonia, thiocyanate, free cyanide, ferric cyanide, phenol and p-cresol on nitrification in an activated sludge system, and then correlated their threshold concentrations with the full-scale pre-denit

Physicochemical and microbiological stability of insulin eye drops in an artificial tear vehicle used in the treatment of refractory neurotrophic keratopathy.

Neurotrophic keratopathy (NK) is a degenerative corneal disease with a loss of corneal sensitivity and impairment of corneal healing. Low dose insulin eyedrops have been shown to be a simple and effective treatment for refractory NK when the response to the usual treatment is incomplete. At present, there are no commercially available forms, and there is no data regarding the stability of these products as prepared by compounding pharmacies. In this work, we studied the physicochemical and microbiological stability of an insulin ophthalmic formulation obtained by mixing insulin lispro in artif

Functional characterization of 2 known ryanodine receptor mutations causing malignant hyperthermia.

BACKGROUND: Malignant hyperthermia (MH) is a potentially lethal pharmacogenetic disorder. More than 300 variants in the ryanodine receptor 1 (RYR1) have been associated with MH; however, only 31 have been identified as causative. To confirm a mutation in RYR1 as being causative for MH, segregation of the potential mutation in at least 2 unrelated families with MH susceptibility must be demonstrated and functional assays must show abnormal calcium release compared with wild-type RYR1. METHODS: We used "Hot-spot" DNA screening to identify mutations in RYR1 in 3 New Zealand families. B-lymphobla

A novel application of the Margin of Exposure approach: segregation of tobacco smoke toxicants.

This paper presents a rationale for utilising a Margin of Exposure (MOE) approach to the segregation of tobacco smoke toxicants for risk assessment and management purposes. Future regulatory frameworks and product modifications aimed at tobacco harm reduction could utilise data that segregate toxicants using associations with specific diseases caused by cigarette smoking together with an indication of their relative contribution to that disease. Compounds with MOEs >10,000 accompanied by appropriate narrative are considered "low priority for risk management actions". This paper applies the MOE

Redox-gated polyvinylferrocene interfaces for cathodic peroxymonosulfate activation.

Electrochemical advanced oxidation can intensify contaminant removal while avoiding consumable metal catalysts, but cathodic activation of anionic peroxymonosulfate (PMS, HSO5-) is limited by electrostatic exclusion from negatively polarized interfaces. Here, we constructed a polyvinylferrocene-modified biochar (PVF-BC) cathode operated by a two-stage potential program. At 0 V, reaction of surface-bound ferrocene (Fc) with PMS partially formed ferrocenium (Fc+), whereas subsequent operation at -1.2 V accelerated cathodic PMS reduction. Molecular tracking of the in-situ bioaugmentation process of Pseudomonas sp. BO3-4 using key degradation genes.

In-situ bioaugmentation is often constrained by unclear environmental factors and the difficulty of tracking microbial activity, limiting the efficiency of groundwater remediation. This study developed a functional gene-based approach to track microbial degradation activity during field-scale bioaugmentation of toluene-contaminated groundwater. Based on transcriptomic analysis, todC1 (dioxygenase) and todE (ring-cleavage enzyme) were selected as molecular markers due to their critical roles in toluene degradation and high upregulation (>700-fold) in Pseudomonas sp. BO3-4. Following bioaugmenta
