Development and clinical application of a strategy for preimplantation genetic diagnosis of single gene disorders combined with HLA matching.

Preimplantation HLA matching has recently emerged as a tool for couples desiring to conceive a potential donor progeny for transplantation in a sibling with a life-threatening disorder. In this paper we describe a strategy optimized for preimplantation genetic diagnosis (PGD) of haemoglobinopathies combined with HLA matching. This procedure involves a minisequencing-based genotyping of HLA regions A, B, C and DRB combined with mutation analysis of the gene regions involved by mutation. Analysis of at least eight polymorphic short tandem repeat (STR) markers scattered through the HLA complex ha

Genetic linkage analysis of nerve growth factor (beta) in familial Alzheimer's disease.

Recent studies have not shown linkage of late-onset (mean age, greater than 60 years) familial Alzheimer's disease (FAD) to the chromosome 21 locus reported linked to early-onset FAD. Beta nerve growth factor (beta-NGF) has been considered a candidate gene in the pathogenesis and therapy of FAD, based on its localization in the cortex and hippocampus and its ability to enhance the growth and survival of cholinergic neurons. A 1.5-kb fragment of the beta-NGF gene was used to detect a BglII restriction fragment length polymorphism, which was then used for linkage analysis. A total of 30 families

Meiotic nondisjunction in the mouse: methodology for genetic testing and comparison with other methods.

Since trisomies produce adverse effects relatively late in development or even postnatally, they are an important component of the array of genetic damages that might be caused by environmental agents. Whole-chromosome aneuploidy (as opposed to breakage-derived aneuploidy) might come about secondarily from crossover depression, or could follow damage to the meiotic spindle or to kinetochores. For simplicity, the event-by whichever of the mechanisms-is referred to as meiotic nondisjunction (ND). A genetic method has been devised which is based on the facts that ND involving the sex chromosomes

Molecular topography of the secondary constriction region (qh) of human chromosome 9 with an unusual euchromatic band.

Heterochromatin confined to pericentromeric (c) and secondary constriction (qh) regions plays a major role in morphological variation of chromosome 9, because of its size and affinity for pericentric inversion. Consequently, pairing at pachytene may lead to some disturbances between homologous chromosomes having such extreme variations and may result in abnormalities involving bands adjacent to the qh region. We encountered such a case, where a G-positive band has originated de novo, suggesting a maternal origin from the chromosome 9 that has had a complete pericentric inversion. In previously

An index marker map of chromosome 9 provides strong evidence for positive interference.

An index marker map of chromosome 9 has been constructed using the Centre d'Etude du Polymorphisme Humain reference pedigrees. The map comprises 26 markers, with a maximum intermarker interval of 13.1 cM and only two intervals > 10 cM. Placement of all but one marker into the map was achieved with > 10,000:1 odds. The sex-equal length is 151 cM, with male length of 121 cM and female length of 185 cM. The map extends to within 2%-3% of physical length at the telomeres, and its coverage therefore is expected to be within 20-30 cM of full map length. The markers are all of the GT/CA repeat type a

An unequal crossover event in RCCX modules of the human MHC resulting in the formation of a TNXB/TNXA hybrid and deletion of the CYP21A.

The central region of the human major histocompatibility complex contains tandemly arranged genes of RP, C4, CYP21, and TNX. The C4 gene region is prone to rearrangements that generates duplications, conversions, and deletions. Diversity in gene number and size causes reorganization and may lead to genetic disorders. The RP, C4, CYP21, and TNX genes form a genetic unit called RCCX. We describe molecular studies on RCCX haplotypes revealing a unique recombination giving rise to a TNXB/TNXA hybrid gene, CYP21A deletion and CYP21B duplication on one chromosome of the propositus. His other chromos

Genomic impacts of chromosomal inversions in parapatric Drosophila species.

Chromosomal inversions impact genetic variation and facilitate speciation in part by reducing recombination in heterokaryotypes. We generated multiple whole-genome shotgun sequences of the parapatric species pair Drosophila pseudoobscura and Drosophila persimilis and their sympatric outgroup (Drosophila miranda) and compared the average pairwise differences for neutral sites within, just outside and far outside of the three large inversions. Divergence between D. pseudoobscura and D. persimilis is high inside the inversions and in the suppressed recombination regions extending 2.5 Mb outside o

Network-informed multi-trait genomic analysis decodes the shared genetic architecture and therapeutic landscape of pelvic floor disorders.

Pelvic floor disorders, including pelvic organ prolapse and urinary incontinence, represent a common health burden with substantial clinical comorbidity, but their shared genetic architecture remains incompletely understood. We performed a multi-trait genomic analysis of six pelvic-floor-related phenotypes using publicly available GWAS summary statistics from FinnGen R12 and the GWAS Catalog, with Pan-UKBB summary statistics used only for cross-ancestry validation. Linkage disequilibrium score regression and Genomic Structural Equation Modeling identified two broad latent genetic dimensions: a

Association between osteoporosis and rotator cuff tears: evidence from causal inference and colocalization analyses.

Osteoporosis is a known risk factor for rotator cuff tears (RCTs), but the causal correlation and underlying mechanisms remain unclear. This study aims to evaluate the impact of osteoporosis on RCT risk and investigate their genetic associations. Using data from the UK Biobank (n = 457 871), cross-sectional analyses demonstrated that osteoporosis was significantly associated with an increased risk of RCTs (adjusted OR [95% CI] = 1.38 [1.25-1.52]). A longitudinal analysis of a subset of patients (n = 268 117) over 11 years revealed that osteoporosis increased the risk of RCTs (adjusted HR [95%

Principles of genetics applied to medicine.

Genes and environment interact continuously throughout life, from the moment of conception, to regulate and affect all aspects of human structure and function. The normal rules of mendelian inheritance, ie, the transmission of genes over the generations, are usually best studied by observation of abnormal mutations, especially in plants and animals under precise breeding conditions. The study of such didactic models as the guinea pig crosses discussed here is a prerequisite to an understanding of mendelian genetics in man and the terms used in other articles to appear in this series.

The role of multiple drug resistance proteins in fetal and/or placental protection against teratogen-induced orofacial clefting.

Previous studies have shown a role for multiple drug resistance proteins in protecting the fetus from a limited number of teratogens. We have expanded the number of proteins and teratogens examined by comparing the influence of the mdr1a and mdr2 proteins on teratogen-induced orofacial clefting using their respective knockouts in crosses with the A/J, high susceptibility strain. Western blots identified the presence of mdr1a and possibly mdr2 in the placenta and fetus. The mdr1a knockout, on its unique genetic background showed lower, similar, and higher incidences of clefting compared to A/J

Genome-wide assessment of shared genetic landscape of idiopathic pulmonary fibrosis and its comorbidities.

Idiopathic pulmonary fibrosis (IPF) is a progressive interstitial lung disease accompanied by both local and systemic comorbidities. Genetic factors play a role in the development of IPF and certain associated comorbidities. Nevertheless, it is uncertain whether there are shared genetic factors underlying IPF and these comorbidities. To bridge this knowledge gap, we conducted a systematic investigation into the shared genetic architecture between IPF and ten prevalent heritable comorbidities (i.e., body mass index [BMI], coronary artery disease [CAD], chronic obstructive pulmonary disease [COP

Predictive power of individual genetic and environmental factor scores.

This study explores the use of an individual's genetic (IGFS) and environmental factor score (IEFS), constructed using genetic model fitting of a multivariate strength phenotype. Maximal isometric and dynamic strength measures, one maximal repetition load (1RM) and muscle cross-sectional area (MCSA) were measured in 25 monozygotic and 16 dizygotic twin pairs. The use of IGFS and IEFS in predicting the sensitivity to environmental stress was evaluated by the association of the scores with strength training gains after a 10-week high resistance strength training programme. Results show a high co

Holding All the Cards-How Fanconi Anemia Proteins Deal with Replication Stress and Preserve Genomic Stability.

Fanconi anemia (FA) is a hereditary chromosomal instability disorder often displaying congenital abnormalities and characterized by a predisposition to progressive bone marrow failure (BMF) and cancer. Over the last 25 years since the discovery of the first linkage of genetic mutations to FA, its molecular genetic landscape has expanded tremendously as it became apparent that FA is a disease characterized by a defect in a specific DNA repair pathway responsible for the correction of covalent cross-links between the two complementary strands of the DNA double helix. This pathway has become incr

Sister chromatid exchanges in protein-energy malnutrition.

The frequency of sister chromatid exchanges in children suffering from severe protein-energy malnutrition was investigated by the fluorescent-plus-Giemsa method. Children suffering from kwashiorkor had significantly higher mean SCEs per circulating lymphocyte than did normal children. A small but statistically significant decrease in these levels was observed following nutritional rehabilitation.

The HLA types of two families with hereditary spherocytosis (HS) suggest that the HS gene(s) may be linked with the HLA locus. If sustantiated by further family studies, this linkage would place HS on chromosome 6.

Cytologic effects of adriamycin on human peripheral lymphocytes.

Preliminary data show adriamycin (AM) induces chromosomal structural changes and division delay in human peripheral lymphocytes. Cells treated for one hour with 0.01, 0.03, 0.05 and 0.10 mug/ml AM show chromatid lesions mainly breaks and deletions. Chromosome type lesions and chromatid exchanges were less frequent even in cells with the highest percentage of aberrations. Dicentric chromosomes were very rare and were scored only in cells treated with higher concentrations (0.10 mug/ml) and recovered 24 hour post-treatment. Based solely on cytologic evidence G1 and S phase cells appear to be mor

A Novel Environmental Azole Resistance Mutation in Aspergillus fumigatus and a Possible Role of Sexual Reproduction in Its Emergence.

This study investigated the dynamics of Aspergillus fumigatus azole-resistant phenotypes in two compost heaps with contrasting azole exposures: azole free and azole exposed. After heat shock, to which sexual but not asexual spores are highly resistant, the azole-free compost yielded 98% (49/50) wild-type and 2% (1/50) azole-resistant isolates, whereas the azole-containing compost yielded 9% (4/45) wild-type and 91% (41/45) resistant isolates. From the latter compost, 80% (36/45) of the isolates contained the TR46/Y121F/T289A genotype, 2% (1/45) harbored the TR46/Y121F/M172I/T289A/G448S genotyp

Cryptococcus neoformans Mating and Genetic Crosses.

The Cryptococcus pathogenic species complex is a group of opportunistic human fungal pathogens that cause cryptococcal meningoencephalitis, an infection associated with unacceptably high mortality rates. The public health relevance of these pathogens has galvanized extensive research over the past several decades and led to characterization of their sexual cycles. This research has allowed several Cryptococcus species to develop into model fungal organisms for both pathogenesis and basic science studies. Many of these studies require observation of the meiotic process and its associated mating
