The use of optogenetic tools offers an excellent method for spatially and temporally regulated gene and protein expression in cell therapeutic approaches. This could be useful as a concomitant therapeutic measure, especially in small body compartments such as the inner ear, for example, during cochlea implantation, to enhance neuronal cell survival and function. Here, we used the blue light activatable CRY2/CIB system to induce transcription of brain-derived neurotrophic factor (BDNF) in human cells. Transfection with three plasmids, encoding for the optogenetic system and the target, as well

People with mood disorders often have disruptions in their circadian rhythms. Recent molecular genetics has linked circadian clock genes to mood disorders. Our objective was to study two core circadian clock genes, CRY1 and CRY2 as well as TTC1 that interacts with CRY2, in relation to depressive and anxiety disorders. Of these three genes, 48 single-nucleotide polymorphisms (SNPs) whose selection was based on the linkage disequilibrium and potential functionality were genotyped in 5910 individuals from a nationwide population-based sample. The diagnoses of major depressive disorder, dysthymia

BACKGROUND: Hypoxia-inducible factor 1 (HIF-1) affects the circadian clock in obstructive sleep apnea (OSA) and may have a bidirectional relationship with circadian mechanisms. This study examined the link between circadian clock and HIF-1 in OSA patients versus controls.
METHODS: 70 participants underwent polysomnography (PSG), and were assigned into OSA (apnea-hypopnea index (AHI) ≥ 5, n = 54) or control (AHI < 5, n = 16) groups. BMAL1 (brain and muscle ARNT like 1), CLOCK (circadian locomotor output cycles kaput), PER1 (period 1), CRY1 (cryptochrome 1), HIF-1α, and HIF-1β gene expressions

The light input pathways and the molecular clock are tightly linked, with light serving as the most potent zeitgeber that entrains the clock to the external environment. Our present study focuses on the Drosophila melanogaster populations that have evolved with a precise circadian clock as a correlated response to selection for adult emergence in a narrow window of time over 335 generations. The results of our study showed that flies from populations selected for the timing of adult emergence sleep more during the night phase compared to controls. This sleep was even more enhanced when the lig

Ultraviolet-sensitive photoreceptors have been shown to be important for a variety of visual tasks performed by bees, such as orientation, color and polarization vision, yet little is known about their spatial distribution in the compound eye or optic lobe. We cloned and sequenced a UV opsin mRNA transcript from Bombus impatiens head-specific cDNA and, using western blot analysis, detected an eye protein band of approximately 41 kDa, corresponding to the predicted molecular mass of the encoded opsin. We then characterized UV opsin expression in the retina, ocelli and brain using immunocytochem

In the mammalian circadian clock, the transcription factor CLOCK/BMAL1 cycles between an active state recruiting co-activators like MLL1 and repressed states associated with PER1/2 and CRY1/2 clock proteins. The MLL1 complex component WDR5 was also found in a repressive PER complex. Here we show that WDR5 directly binds to the CRY binding domain (CBD) regions of PER1 and PER2. PER2 binds WDR5 via a WBM motif within the PER2/CRY interface, imposing competition between PER2/WDR5-, PER2/CRY- and RbBP5/WDR5 complexes. PER1 binds WDR5 predominantly via a WIN motif outside the CBD, enabling formatio

Obesity, characterized by excessive fat accumulation, disrupts metabolism, leading to insulin resistance, hyperglycemia, and dyslipidemia. Circadian rhythm genes such as BMAL1 and CRY1 regulate energy balance, lipid metabolism, and glucose homeostasis, with enzymes, for example, DGAT1 and DGAT2, playing critical roles in triglyceride synthesis. Melatonin, a circadian regulator, influences metabolic processes, including glucose and insulin regulation. This study examined melatonin's therapeutic effects in obese Wistar rats. A total of 48 male rats were fed either a standard diet (C group) or a

Ultraviolet-C (UVC) electromagnetic radiation is the most damaging type of the UV radiation and causes many cellular and physiological responses. UVC has been using for sterilization and disinfection, and the risk of exposure to the UVC is increasing. Here, we determined the effect of the UVC on the cellular circadian clock system. UVC irradiation synchronized the biological clock system and induced time-dependent expression of clock genes including Clock, Cry1, and Per1. The rhythmic expression of clock genes is also followed by time-dependent mRNA degradation or non-canonical translation ini

Circadian clock genes, such as BMAL1 (brain and muscle aryl-hydrocarbon receptor nuclear translocator-like 1), CLOCK (circadian locomotor output cycles kaput), PER1/2/3 (period 1/2/3), and CRY1/2 (cryptochrome 1/2), can play crucial roles in regulating cardiovascular and renal functions, potentially via influencing salt sensitivity and blood pressure regulation. This thematic review elucidates the complex interaction between salt sensitivity, genetic variations in circadian rhythm genes, and systemic hypertension. We have particularly focused on the profound impact of obstructive sleep apnea (

The mineralocorticoid receptor (MR) is a ligand-activated transcription factor that regulates cardiorenal physiology and disease. Ligand-dependent MR transactivation involves a conformational change in the MR and recruitment of coregulatory proteins to form a unique DNA-binding complex at the hormone response element in target gene promoters. Differences in the recruitment of coregulatory proteins can promote tissue-, ligand- or gene-specific transcriptional outputs. The goal of this study was to evaluate the circadian protein TIMELESS as a selective regulator of MR transactivation. TIMELESS h

Dusk feeding is practised probably to satisfy the energy requirement during night. However, little is known on the changes with clock gene expressions during this feeding behaviour. In our present study, the linkage of clock gene expressions and feeding behaviour in dusk feeding was investigated in laying hens under two lighting programs: the conventional lighting program (Control) with a light period from 05:00 AM to 21:00 PM and a dark period from 21:00 PM to 05:00 AM; or the shifted lighting program group (SLP) with a light period from 02:00 AM to 18:00 PM and a dark period from 18:00 PM to

Dysregulation of the circadian clock machinery is a critical mechanism in the pathogenesis of fibrosis. This study aimed to investigate whether the antifibrotic effect of melatonin is associated with attenuation of circadian clock pathway disturbances in mice treated with carbon tetrachloride (CCl4) and in human hepatic stellate cells line LX2. Mice received CCl4 5 μL/g body weight i.p. twice a week for 4 or 6 weeks. Melatonin was given at 5 or 10 mg/kg/day i.p., beginning 2 weeks after the start of CCl4 administration. Treatment with CCl4 resulted in fibrosis evidenced by the staining of α-sm

OBJECTIVE: In mice, a lack of cryptochrome results in up-regulation of aldosterone production due to high expression of the 3β-hydroxysteroid dehydrogenases (HSD3β) gene. The HSD3β pathway might play a pivotal role in aldosterone synthesis. This study aimed to determine the association of HSD3β and HSD3β2 gene variations with primary aldosteronism in a Taiwanese population.
METHOD: In this case-control cohort, 688 consecutive ethnically matched unrelated individuals including 362 primary aldosteronism and 326 essential hypertension cases were recruited. Nineteen tag single-nucleotide polymorp

Melatonin and its autonomic regulation serve important physiological functions. We recently demonstrated that stimulation of beta-adrenergic receptors only increases nighttime arylalkylamine N-acetyltransferase (Aa-Nat, the rate-limiting enzyme in melatonin synthesis) mRNA levels in mouse pineal gland in vitro, which suggests that pineal clocks may gate Aa-Nat gene expression. In the present study, our data reveal that cAMP analog increased Aa-Nat at any time of day but only in the presence of ionomycin. Using Fura-2AM in ratiometric calcium measurements, we show that isoproterenol stimulation

BACKGROUND: The prevalence of insomnia and mental illness varies by sex. The mRNA expression level of the CRY gene in patients with insomnia is significantly decreased, and the expression level of the CRY2 gene is closely related to depression. However, sex differences in the link between insomnia and symptoms of anxiety, depression, and stress, as well as whether CRY2 gene DNA methylation can regulate this association, are not yet fully understood.
OBJECTIVE: Investigate sex-specific differences in the associations between insomnia symptoms and symptoms of anxiety, depression, and stress amo

Phase transitions driven by intrinsically disordered protein regions (IDRs) have emerged as a ubiquitous mechanism for assembling liquid-like RNA/protein (RNP) bodies and other membrane-less organelles. However, a lack of tools to control intracellular phase transitions limits our ability to understand their role in cell physiology and disease. Here, we introduce an optogenetic platform that uses light to activate IDR-mediated phase transitions in living cells. We use this "optoDroplet" system to study condensed phases driven by the IDRs of various RNP body proteins, including FUS, DDX4, and H

When chicks are exposed to constant light (CL) during growth, their corneas become flatter and lighter in weight, and their anterior segments become shallower than those of chicks exposed to cyclical periods of light and dark. These effects have been correlated with CL suppression of cyclical changes in melatonin levels. The question of whether light directly influences corneal growth (e.g. via cryptochromes in the cornea) or acts remotely via the suppression of the melatonin rhythm has not yet been answered. Retinoic acid (RA), an ubiquitous morphogen, also causes non-functional flattening du

Metabolic dysfunction-associated steatotic liver disease (MASLD) is characterized by profound disturbances in hepatic lipid metabolism. However, the role of N6-methyladenosine (m6A)-mediated dysregulation of specific stress-responsive proteins in governing metabolically critical gene expression, particularly the clock system, in this context remains largely unexplored. Notably, there is no prior experimental evidence linking Hypoxia Up-Regulated 1 (HYOU1) to MASLD pathology. Combining bioinformatics with validation, we identify HYOU1 as aberrantly upregulated in MASLD. Elevated HYOU1 promotes

BACKGROUND: Transgenic mice expressing the C414A-mutant cryptochrome 1 (CRY1 C414A) develop diabetes with progressive β-cell senescence and the formation of mucinous intra-islet ductal lesions. These lesions exhibit features resembling metaplasia; however, their molecular basis and the potential involvement of endocrine cells remain unclear. This study aimed to define the metaplasia-like program in intra-islet ducts and to characterize endocrine cell phenotypes in this context.
METHODS: Gene expression microarrays were analyzed using islets from aged CRY1 C414A transgenic and wild-type mice.
