BACKGROUND: Fleas (Insecta: Siphonaptera) are small flightless parasites of birds and mammals; their blood-feeding can transmit many serious pathogens (i.e., the etiological agents of bubonic plague, endemic and murine typhus). The lack of flea genome assemblies has hindered research, especially comparisons to other disease vectors. Accordingly, we sequenced the genome of the cat flea, Ctenocephalides felis, an insect with substantial human health and veterinary importance across the globe.
RESULTS: By combining Illumina and PacBio sequencing of DNA derived from multiple inbred female fleas w

The authors describe techniques for handling and rearing large numbers of body lice (Pediculus humanus humanus L.), oriental rat fleas (Xenopsylla cheopis (Rothschild)), and cat fleas (Ctenocephalides felis felis (Bouché)).Body lice may be fed on man or on domestic rabbits. In the latter case, the lice are kept on woollen patches in glass dishes at 30 degrees C and 60% relative humidity. The patches are placed on the clipped belly of a rabbit once a day and the lice allowed to feed. Eggs are deposited on the patches, and the adult lice are transferred to new patches every two or three days. The

Xenopsylla cheopis, also called oriental rat flea, is an ectoparasite as well as disease vector for murine typhus and bubonic plague. In the study, the whole mitochondrial genome of X. cheopis was sequenced and assembled, which is the second report of mitochondrial genome in the family Pulicidae and the sixth mitochondrial genome in the order Siphonaptera (fleas). The mitochondrial genome is 18,902 bp in length, consisting of 40% A, 44% T, 6% G, and 10% C. Phylogenetic analysis of all available mitochondrial genomes from Siphonaptera indicated that X. cheopis clustered with Ctenocephalides fel

BACKGROUND: Bartonella henselae is transmitted amongst cats by Ctenocephalides felis and is associated with multiple clinical syndromes in cats and people. In a previous study, monthly spot-on administration of 10% imidacloprid/1% moxidectin was shown to block transmission of B. henselae amongst cats experimentally exposed to infected C. felis. The purpose of this study was to determine whether application of a flea and tick collar containing 10% imidacloprid and 4.5% flumethrin would lessen C. felis transmission of B. henselae amongst cats for 8 months.
METHODS: Specific pathogen free cats (

A total of 380 scrub hares (Lepus saxatilis) from 5 localities, ranging from the north-eastern Transvaal to the eastern and the south-western Cape Province of South Africa, were examined for fleas and lice. Ctenocephalides felis damarensis was almost exclusively responsible for the flea infestations on hares at 4 of the 5 sites. At 4 localities the prevalence and abundance of this flea reached peaks between August and October and declined thereafter to their lowest levels between February and April. Its prevalence and abundance in the north-eastern Transvaal were not correlated with the breedi

Fleas, lice and mites were collected from 24 and 120 scrub hares at Pafuri and Skukuza, Northern and Eastern Transvaal, respectively, in the Kruger National Park, and from 34 scrub hares in the Hluhluwe region, north-eastern KwaZulu-Natal. Ctenocephalides felis damarensis, the only flea recovered, reached peak burdens on the hares at each locality during late winter or spring. Juvenile hares harboured significantly fewer fleas than did adult animals. The lice Haemodipsus lyriocephalus and Haemodipsus setoni were collected from hares at each locality, with H. setoni generally being the most abu

Ectoparasites are capable of transmitting infectious diseases and, therefore, are of zoonotic concern. Cats submitted to a spay/neuter program in the city of Rio de Janeiro were examined to determine the distribution of ectoparasites in cats from a city with a tropical climate. Independent of gender, breed, or age, 292 cats were combed and subjected to otoscopic examination. Ectoparasites were collected, and blood samples were taken to determine packed cell volume. The majority of the 292 cats were female (71%), and most of them were categorized as domestic short hair cats (92%). Different spe

Photoperiod affected the production of cat flea eggs by cats infested with cat flea, Ctenocephalides felis (Bouché). Cat flea eggs have a smooth chorion and do not adhere to the host. About 60% of fluorescing eggs placed in the pelage were recovered in collecting trays underneath the host in 2 h. An average of 87.7 eggs per cat was laid during the last 8 h of the scotophase compared with 49.9 and 59.1 eggs during the other 8-h periods. Significant variation in the number of eggs collected daily existed among the six cats. The activity budgets were similar for all the cats with the maximum slee

Bartonella spp. and haemoplasmas are pathogens of veterinary and medical interest with ectoparasites mainly involved in their transmission. This study aimed at molecular detection of Bartonella spp. and haemoplasmas in cats (n = 93) and dogs (n = 96), and their related fleas (n = 189) from countries in East and Southeast Asia. Ctenocephalides felis was the dominant flea species infesting both cats (97.85%) and dogs (75%) followed by Ctenocephalides orientis in dogs (18.75%) and rarely in cats (5.2%). Bartonella spp. DNA was only detected in blood samples of flea-infested cats (21.51%) (p < .00

The flea-borne rickettsioses murine typhus (Rickettsia typhi) and flea-borne spotted fever (FBSF) (Rickettsia felis) are febrile diseases distributed among humans worldwide. Murine typhus has been known to be endemic to Kenya since the 1950s, but FBSF was only recently documented in northeastern (2010) and western (2012) Kenya. To characterize the potential exposure of humans in Kenya to flea-borne rickettsioses, a total of 330 fleas (134 pools) including 5 species (Xenopsylla cheopis, Ctenocephalides felis, Ctenocephalides canis, Pulex irritans, and Echidnophaga gallinacea) were collected fro

Rickettsia typhi is the causal agent of murine typhus; a worldwide zoonotic and vector-borne infectious disease, commonly associated with the presence of domestic and wild rodents. Human cases of murine typhus in the state of Yucatán are frequent. However, there is no evidence of the presence of Rickettsia typhi in mammals or vectors in Yucatán. The presence of Rickettsia in rodents and their ectoparasites was evaluated in a small municipality of Yucatán using the conventional polymerase chain reaction technique and sequencing. The study only identified the presence of Rickettsia typhi in bloo

Murine typhus, an acute febrile illness caused by Rickettsia typhi, is distributed worldwide. Mainly transmitted by the fleas of rodents, it is associated with cities and ports where urban rats (Rattus rattus and Rattus norvegicus) are abundant. In the United States, cases are concentrated in suburban areas of Texas and California. Contrary to the classic rat-flea-rat cycle, the most important reservoirs of infection in these areas are opossums and cats. The cat flea, Ctenocephalides felis, has been identified as the principal vector. In Texas, murine typhus cases occur in spring and summer, w

An investigation was conducted into a severe flea infestation on dairy calves. Inspection of the dairy revealed a high infestation of Ctenocephalides felis felis on four calves and thousands of fleas in the stable where cows and calves were brought in for milking. A 3-month-old female calf was highly infested with fleas. The animal showed an evident lethargy, weight loss, as well as pale mucous membranes and dehydration. Hematological analysis revealed anemia, with 10% packed cell volume and a 1.72 x 10(6) microl(-1) erythrocyte count. Blood transfusions were performed and the flea infestation

In recent years, discovery proteomics has emerged as a pivotal tool in biological research, especially when studying the intricate relationships among multiple organisms. To delve deeper into these interactions, we pioneered a bottom-up proteomics workflow. Using nanoLC-MS/MS and a label-free quantification method, this work specifically examines the differential protein expression in fleas (Ctenocephalides felis felis) that have been experimentally infected with Bartonella henselae, the causative agent of cat scratch disease (CSD). Our detailed methodology, from protein cleanup to data analys
