High-quality genome and transcriptome analyses under temperature stress reveal heat shock protein-centric adaptive enhancement mechanisms in Taiwanese loach.

Within the family Cobitidae, the Paramisgurnus dabryanus demonstrates notable environmental resilience and accelerated growth. This is particularly true for the subspecies Taiwanese loach (P. dabryanus ssp. Taiwan). This study presents a chromosome-level genome for the Taiwanese loach, offering a valuable genomic resource for Cobitoidei. We also identified expansions in gene families associated with environmental adaptation, such as heat shock proteins (HSPs), in Taiwanese loaches. Transcriptomic analysis revealed that HSPs may exert a central regulatory role under temperature stress. Correspo

Assessment of the fish tumors or other deformities beneficial use impairment and associated risks at two Lake Michigan Areas of Concern.

The U.S. Great Lakes Areas of Concern (AOCs) have been designated due to significant environmental degradation, with multiple Beneficial Use Impairments (BUIs) requiring remediation before delisting. The “fish tumors or other deformities” BUI remains at the Sheboygan River and the Lower Green Bay and Fox River AOCs, necessitating evaluation in white sucker (Catostomus commersonii), a sentinel species. This study documented the prevalence of skin and liver neoplastic and preneoplastic lesions at these AOCs, assessed temporal trends at the Sheboygan River AOC from 2012 to 2021, and investigated

Aneuploid progenies of triploid hybrids between diploid and tetraploid loach Misgurnus anguillicaudatus in China.

Triploid Chinese loach, Misgurnus anguillicaudatus, hybrids between tetraploids from Hubei Province and diploids from Liaoning Province were mated with either diploid wild-type or triploid hybrids to analyze viability and ploidy of the resultant progenies. Both triploid males and females generated fertile gametes, but progenies from the crosses using gametes of triploid hybrids did not survive beyond the larval stages. In crosses between wild-type diploid females and triploid hybrid males, embryos ranging from 2.2n to 2.6n were predominant with a mode of either 2.4n (chromosome numbers 59, 60,

Aberrant Meiotic Configurations Cause Sterility in Clone-Origin Triploid and Inter-Group Hybrid Males of the Dojo Loach, Misgurnus anguillicaudatus.

Gonochoristic wild-type dojo loaches (Misgurnus anguillicaudatus) are diploid (2n = 50) and reproduce bisexually. However, sympatric clonal diploids generate unreduced diploid isogenic eggs that develop gynogenetically. Clone-origin triploidy arises following the incorporation of a haploid wild-type sperm nucleus into the diploid egg. Triploid females produce fertile haploid eggs by meiotic hybridogenesis, while triploid males are sterile. Clonal loaches arose from past hybridization event(s) between genetically diverse groups, A and B. Artificial hybrid females between the 2 groups produce un

Intersex and other reproductive disruption of fish in wastewater effluent dominated Colorado streams.

Intersex white suckers (Catostomus commersoni) were collected in Boulder Creek and the South Platte River downstream of wastewater treatment plant (WWTP) effluent but not at reference sites. Eighty-three percent of the 60 white suckers collected downstream of the Boulder WWTP in spring and fall were female compared to 45% upstream (n=33). Only female (n=16) and intersex white suckers (n=4) were collected in the South Platte River downstream of the Denver WWTP in the spring. Gonad deformities and delayed follicular maturation were noted in some white suckers in Boulder Creek and the South Platt

Investigation of genetic diversity in the loach Misgurnus anguillicaudatus revealed by whole-genome resequencing.

BACKGROUND: The loach (Misgurnus anguillicaudatus) is a significant freshwater economic fish in China, renowned for its tender meat, delicious taste, and high nutritional value. It is widely distributed across the country, except for the western plateau. However, the loach is currently at risk of population decline and degradation of wildlife resources. Research on genetic diversity provides a crucial foundation for the conservation and development of these fish resources. To enhance the protection and utilization of wild loach germplasm resources, we analyzed the genetic structure and diversi

Gonadotropin subunits of the characiform Astyanax altiparanae: Molecular characterization, spatiotemporal expression and their possible role on female reproductive dysfunction in captivity.

To better understand the endocrine control of reproduction in Characiformes and the reproductive dysfunctions that commonly occur in migratory fish of this order when kept in captivity, we chose Astyanax altiparanae, which has asynchronous ovarian development and multiple spawning events, as model species. From A. altiparanae pituitary total RNA, we cloned the full-length cDNAs coding for the follicle-stimulating hormone β subunit (fshb), the luteinizing hormone β subunit (lhb), and the common gonadotropin α subunit (gpha). All three sequences showed the highest degree of amino acid identity w

New circumscription of freshwater fish parasites Monobothrium diesing, 1863 and Promonobothrium mackiewicz, 1968 (Cestoda: Caryophyllidea) using morphological and molecular evidence.

Two genera of caryophyllidean cestodes, Monobothrium Diesing, 1863 and Promonobothrium Mackiewicz, 1968 , from cypriniform fishes (cyprinids and suckers) in the Holarctic Region, are revised using morphological and molecular evidence. Monobothrium, which includes morphologically distinct species that occur in European cyprinids (Cyprinidae) and North American suckers (Catostomidae), is separated into 3 genera. Monobothrium becomes monotypic and is represented by its type species, Monobothrium wageneri Nybelin, 1922 , which occurs in European tench, Tinca tinca (Cyprinidae). Monobothrium auricu

Neural pathway for aggressive display in Betta splendens: midbrain and hindbrain control of gill-cover erection behavior.

Horseradish peroxidase (HRP) was used to identify parts of the presumptive neural pathway for gill cover erection, a behavioral display pattern performed by Siamese fighting fish (Betta splendens) during aggressive interactions. Motor, motor integration and sensory areas were identified in the medulla and mesencephalon. Motor neurons of the dilator operculi muscle, the effector muscle for gill cover erection, are located in the lateral and medial parts of the caudal trigeminal motor nucleus. Iontophoretic injections of HRP into the lateral trigeminal motor nucleus resulted in labeled cell bodi

T cell receptor (TCR) α and β genes of loach (Misgurnus anguillicaudatus): Molecular cloning and expression analysis in response to bacterial, parasitic and fungal challenges.

In vertebrates, the T cell receptor (TCR) plays a crucial role in immune system. To date, the roles of fish TCRs in response to pathogen infection are still poorly understood. In the present study, we firstly cloned and identified the TCRα and TCRβ from dojo loach (Misgurnus anguillicaudatus) by RACE approaches. The full-length cDNAs of Ma-TCRα and Ma-TCRβ include an open reading frame (ORF) of 723 and 879 bp encoding a polypeptide of 241 and 293 amino acids, respectively. Structural analysis indicated that Ma-TCRα and Ma-TCRβ had a signal peptide, IgV domain, IgC domain, a connecting peptide

Immunoglobulin (Ig) heavy chain gene locus and immune responses upon parasitic, bacterial and fungal infection in loach, Misgurnus anguillicaudatus.

Teleost fish are the most primitive bony vertebrates that contain immunoglobulin (Ig). Although teleost Ig is known to be important during tetrapod evolution and comparative immunology, little is known about the genomic organization of the immunoglobulin heavy-chain (IgH) locus. Here, three Ig isotype classes, IgM, IgD and IgT, were firstly identified in dojo loach (Misgurnus anguillicaudatus), and the IgH locus covering τ, μ and δ genes was also illustrated. Variable (V) gene segments lie upstream of two tandem diversity (D), joining (J) and constant (C) clusters and the genomic organization

Molecular structure, expression, and function analysis of BAFF gene in Chinese sucker, Myxocyprinus asiaticus.

B cell activating factor (BAFF), belonging to the tumor necrosis factor superfamily (TNFSF), is a critical cytokine for B cell survival and immunoglobulin secretion. Here, the BAFF gene of Chinese sucker (Myxocyprinus asiaticus) (MaBAFF) was cloned using RT-PCR and RACE (rapid amplification of cDNA end) techniques. The open reading frame (ORF) of MaBAFF encodes a 272-amino acid protein containing a transmembrane domain, a TNF family signature, and a putative furin protease cleavage site as seen in BAFFs from other species. Tissue expression profiles of MaBAFF determined by absolute and relativ

FISH Identifies Chromosome Differentiation Between Contemporary Genomes of Wild Types and the Ancestral Genome of Unisexual Clones of Dojo Loach, Misgurnus anguillicaudatus.

In dojo loach (Misgurnus anguillicaudatus), although most wild types are gonochoristic diploids that are genetically differentiated into 2 groups, A and B, clonal lineages appear in certain localities. Clonal loaches have been considered to have hybrid origins between the 2 groups by a series of genetic studies. In this study, using FISH with a newly developed probe (ManDra-A), we identified 26 (1 pair of metacentric and 12 pairs of telocentric chromosomes) of 50 diploid chromosomes in contemporary wild-type group A loach. In contrast, ManDra-A signals were not detected on metacentric chromoso

The New Data on the Serotonin and FMRFamide Localization in the Nervous System of Opisthorchis felineus Metacercaria.

BACKGROUND: Trematoda Opisthorchis felineus Rivolta, 1884 is the causative agent of dangerous parasite disease-opisthorchiasis, widespread in the Russian Federation. The details of the neuroanatomical localization of the serotoninergic and FMRFamidergic neurotransmitter elements as well as their functional roles remain not studied enough in both adult and larval forms of O. felineus. The studies in this area are important in term of the development of a new pharmacological strategy of the struggle with the causative agent of opisthorchiasis affecting the neuronal signal substances and the func

Differences in optokinetic and vestibular ocular reflex performance in teleosts and their relationship to different life styles.

Horizontal eye movements in response to vestibular and optokinetic stimulation were investigated in 20 species of marine and fresh water teleosts. Most species performed spontaneous saccadic eye movements in light and in darkness. Successive saccades occurred either at irregular intervals and in different directions (few species), or in bursts of smaller saccades in one direction followed by saccades in the opposite direction (majority of species). Thus, in the latter group the lines of sight shifted cyclically from one side to the other about every 10-15 s. Eye movement responses in goldfish

The complex trophic anatomy of silver carp, Hypophthalmichthys molitrix, highlighting a novel type of epibranchial organ.

Hypophthalmichthys molitrix, silver carp, is an invasive Asian carp that has become increasingly widespread and ecologically destructive within the upper Mississippi River Basin. Its complex trophic anatomy may help explain the apparent efficiency with which they consume phytoplankton, outcompeting native filter feeders. This cypriniform species is characterized by trophic synapomorphies that include a palatal organ, loss of upper pharyngeal jaws, and a hypertrophied lower pharyngeal jaw. However, in silver carp these structures have become greatly modified and diverge from the more basal cond

Cdk1 heterozygosity triggers premeiotic endoreplication and unreduced sperm formation via the Cdk2/Ccne1-Fzr1a pathway in loach, a teleost.

Polyploidy represents a major driver of eukaryotic genome evolution. In vertebrates, premeiotic endoreplication is considered as an important route to polyploidization, yet the molecular basis governing this process remains elusive. Here, we reported that heterozygous mutation of cdk1 in the loach, Misgurnus anguillicaudatus (Teleostei, Cobitidae), triggered premeiotic endoreplication in spermatogonia, resulting in the production of unreduced diploid sperms. Through integrated proteomic and phosphoproteomic profiling, we uncovered that Cdk1 heterozygosity led to widespread dysregulation of the
