DNA nanostructures show great potential as drug carriers in biomedicine, however, biological barriers and stability issues limit their applications, particularly for aggressive melanoma shielded by the skin barrier. This study presents a hyaluronic acid-based microneedle patch integrated with DNA nanoflowers to effectively penetrate the skin barrier. The DNA nanoflowers (Z/H@DFs) are self-assembled from PD-L1 antisense oligonucleotide (ASO), AS1411 G-quadruplex units, Hemin, and ZnPc. In a B16F10 melanoma mouse model, the Z/H@DFs-based microneedle patches (Z/H@DFMNs) significantly inhibited tu

Neutrophils release neutrophil extracellular traps (NETs) to neutralize infections, a process that also contributes to immunothrombosis. While beneficial in localized infections, excessive NET formation can lead to widespread coagulopathy and organ failure. While the roles of NET-associated proteins such as histones in immunothrombosis are well characterized, NET-derived DNAs are much less known. To address this issue, we report herein the direct interaction between thrombin and DNA scaffolds and further, the identification of short tandem repeats of single-stranded (ATTCC)n in NETs that selec

The pathogenesis of the neurologic abnormalities associated with the acquired immune deficiency syndrome (AIDS) is poorly understood. Although human immunodeficiency virus type 1 (HIV-1) transcripts have been detected in endothelial cells and macrophages of the central nervous system in patients with AIDS, infection of neuronal cells by HIV-1 has not been established. The purpose of this study was to localize HIV-1 transcripts in the central nervous system. 3H and digoxigenin-UTP-labeled riboprobes generated from a 942-bp fragment of DNA from the 5' end of the HIV-1 gag sequence were used for

The cAMP response element-binding protein (CREB) is believed to play a pivotal role in dopamine (DA) receptor-mediated nuclear signaling and neuroplasticity. Here we demonstrate that the significance of CREB for gene expression depends on the experimental paradigm. We compared the role of CREB in two different but related models: l-DOPA administration to unilaterally 6-hydroxydopamine lesioned rats, and cocaine administration to neurologically intact animals. Antisense technology was used to produce a local knockdown of CREB in the lateral caudate-putamen, a region that mediates the dyskinetic

BACKGROUND AND PURPOSE: Appetite suppression induced by amphetamine has been attributed to its inhibition of neuropeptide Y (NPY) neurons and activation of pro-opiomelanocortin (POMC) neurons in the hypothalamus. This study examined whether STAT3 was involved in these actions of amphetamine.
EXPERIMENTAL APPROACH: Rats were given amphetamine daily for 4 days. Changes in the expression of NPY, POMC, melanocortin MC3 receptors, PI3K and STAT3 in the hypothalamus were assessed by RT-PCR and Western blotting. Antisense oligonucleotides to STAT3 were also used.
KEY RESULTS: Expression of NPY decr

In the present study, mitral valve tissues from three mitral stenosis patients with RHD by valve replacement and two healthy donors were harvested and conducted DNA methylation signature on PRKCA by MeDIP-qPCR. The presence of hypomethylated CpG islands at promoter and 5' terminal of PRKCA was observed in RHD accompanied with highly expressed PRKCA and down-regulated antisense long non-coding RNA (lncRNA) PRKCA-AS1 compared to health control. Furthermore, the enrichments of DNMT1/3A/3B on PRKCA were detected by ChIP-qPCR assay in vivo and in human cardiomyocyte AC16 and RL-14 cells exposed to

The inhibitor of apoptosis proteins (IAPs) constitutes a family of highly conserved apoptosis suppressor proteins that were originally identified in baculoviruses. Although IAP homologs have recently been demonstrated to suppress apoptosis in mammalian cells, their expression and role in human ovarian epithelial cancer and chemotherapy resistance are unknown. In the present study we used cisplatin-sensitive and -resistant human ovarian surface epithelial (hOSE) cancer cell lines and adenoviral antisense and sense complementary DNA expression to examine the role of IAP in the regulation of apop

Antisense oligonucleotides are small pieces of modified DNA or RNA, which offer therapeutic potential for many diseases. We report on the synthesis of 7',5'-α-bc-DNA phosphoramidite building blocks, bearing the A, G, T and MeC nucleobases. Solid-phase synthesis was performed to construct five oligodeoxyribonucleotides containing modified thymidine residues, as well as five fully modified oligonucleotides. Incorporations of the modification inside natural duplexes resulted in strong destabilizing effects. However, fully modified strands formed very stable duplexes with parallel RNA complements.

The G4C2 repeat expansion in the C9orf72 gene is the most common genetic cause of Amyotrophic Lateral Sclerosis and Frontotemporal Dementia. Many studies suggest that dipeptide repeat proteins produced from this repeat are toxic, yet, the contribution of repeat RNA toxicity is under investigated and even less is known regarding the pathogenicity of antisense repeat RNA. Recently, two clinical trials targeting G4C2 (sense) repeat RNA via antisense oligonucleotide failed despite a robust decrease in sense-encoded dipeptide repeat proteins demonstrating target engagement. Here, in this brief repo

The COVID-19 pandemic brought to light the demand for nucleic-acid sensing platforms that are rapid, resilient, and scalable for integration at the point-of-care. In this study, we described a green-engineered fluorescence biosensing platform using a Ag-TiO2-CeO2 ternary nanocomposite fabricated with Morinda citrifolia leaf extract. The biosynthetic approach facilitated the production of a nanocomposite that is biocompatible, exhibits high surface area, and contains oxygen vacancy related to surface sites that promote favorable interactions with nucleic acids. The nanocomposite was functionali

Antisense therapy is widely used as an alternative therapeutic option for various diseases. RNA interference might be effective in infections, through the degradation of messenger RNA and, therefore, translation process. Hence, proteins essential for microorganisms and viruses' proliferation and metabolism are inhibited, leading to their elimination. The present study aimed to evaluate the use of oligonucleotide in patients infected by Epstein-Barr (EBV) or Herpes Simplex Viruses 1/2 or with Lyme Disease caused by Borrelia burgdorferi. Blood samples were collected from 115 patients and the dif

Diffuse large B-cell lymphoma (DLBCL) is one of the malignancies with a high mortality rate. The molecular mechanisms involved in transformation of DLBCL remain unclear. Therefore, it is critically important to investigate the biological mechanisms of DLBCL. Accumulating evidence indicates that long non-coding RNAs (lncRNAs) serve key functions in tumorigenesis, cancer progression and metastasis. Compared with follicular lymphoma (FL), a total of 123 upregulated lncRNAs and 192 downregulated lncRNAs in DLBCL were identified. Subsequently, a specific DLBCL-associated competing endogenous RNA (c

DNA nanostructures are programmable, biocompatible platforms for biomedicine; however, their mechanical properties are coupled to sequence and structural dimensions, limiting control over stiffness. Here, we report a paradigm shift from static design to mechanical engineering of DNA nanostructures. Using a nanoemulsion interfacial-confined coordination assembly strategy, hollow DNA nanocapsules (HDCs) were constructed with mechanical properties decoupled from chemical identity, without hard templates. By tuning the nanoemulsion-to-DNA-solution ratio, HDCs with sizes (∼160 nm) and surface charg

In this issue of Cell, Kornblihtt and colleagues report a strategy to improve antisense oligonucleotide spinal muscular atrophy therapy. They discover that the oligonucleotide drug nusinersen, which induces exon inclusion, also promotes repressive chromatin modifications, which in turn work against exon inclusion. Notably, co-administration of histone deacetylase inhibitors counteracted this effect to augment exon inclusion.

Herein, we examine the complexation and biological delivery of a short single-stranded antisense oligonucleotide (ASO) payload with four polymer derivatives that form two architectural variants (polyplexes and micelleplexes): a homopolymer poly(2-dimethylaminoethyl methacrylate) (D), a diblock polymer poly(ethylene glycol)methylether methacrylate-block-poly(2-dimethylaminoethyl methacrylate) (ObD), and two micelle-forming variants, poly(2-dimethylaminoethyl methacrylate)-block-poly(n-butyl methacrylate) (DB) and poly(ethylene glycol)methylether methacrylate-block-poly(2-dimethylaminoethyl meth

TREATMENT PERSPECTIVES FOR PATIENTS NOT ELIGIBLE FOR PROTEIN THERAPIES FOR CYSTIC FIBROSIS. The vast majority of patients with cystic fibrosis can benefit from proteic therapy. 10% of patients remain without therapeutic access. RNA editing strategies such as antisense oligonucleotides for specific splice mutations or RNA editing are now in the clinical phase. Gene therapy, DNA editing or readthrough therapies face numerous challenges in terms of vectorization and efficiency. Strong interactions between academic laboratories, harmaceutical industries, associations and health authorities may be

DNA topoisomerase IIα (TOP2α; 170 kDa, TOP2α/170) is an essential enzyme for proper chromosome dysjunction by producing transient DNA double-stranded breaks and is an important target for DNA damage-stabilizing anticancer agents, such as etoposide. Therapeutic effects of TOP2α poisons can be limited due to acquired drug resistance. We previously demonstrated decreased TOP2α/170 levels in an etoposide-resistant human leukemia K562 subline, designated K/VP.5, accompanied by increased expression of a C-terminal truncated TOP2α isoform (90 kDa; TOP2α/90), which heterodimerized with TOP2α/170 and w

To explore whether mouse short interspersed nuclear element B1 antisense RNA (B1 antisense RNA, B1 asRNA) could improve the proliferation activity and killing tumor function of spleen lymphocytes from aged mice and to investigate the underlying mechanisms, we transfected B1 asRNA into spleen lymphocytes isolated from 12-month-old mice. We found that B1 asRNA substantially increased the proliferative rate of lymphocytes, the number of EdU-positive cells and the number of S phase cells. B1 asRNA decreased the apoptosis of lymphocytes and regulated the mRNA expression levels of senescence-related

Orexin (hypocretin)-containing neurons of the hypothalamus project to brainstem sites that are involved in the neural control of REM sleep, including the locus coeruleus, the dorsal raphe nucleus, the cholinergic zone of the mesopontine tegmentum, and the pontine reticular formation (PRF). Orexin knockout mice exhibit narcolepsy/cataplexy, and a mutant and defective gene for the orexin type II receptor is present in dogs with an inherited form of narcolepsy/cataplexy. However, the physiological systems mediating these effects have not been described. We reasoned that, since the effector neuron
