Biostimulation of indigenous communities for the successful dechlorination of tetrachloroethene (perchloroethylene)-contaminated groundwater.

Chlorinated ethenes are of environmental concern with most reports of successful microbial-mediated remediation being associated with major dechlorinating groups such as Dehalococcoides (Dhc) species. However, limited information is available on the community dynamics and dechlorinating activities of indigenous non-Dhc groups. Here, we present evidence of dechlorination of tetrachloroethene (perchloroethylene, PCE) in groundwater samples by indigenous microbial communities. 100 % PCE conversion to ethene was observed in acetate-stimulated 24 week-microcosms (controls; 15 %). Microbial communit

Effects of different electron donor feeding patterns on TCE reductive dechlorination performance.

This study investigates how the feeding pattern of e(-) donors might affect the efficiency of enhanced in situ bioremediation in TCE-contaminated aquifers. A series of lab-scale batch experiments were conducted using butyrate or hydrogen gas (H2) as e(-) donor and a TCE-dechlorinating microbial consortium dominated by Dehalococcoides spp. The results of these experiments demonstrate that butyrate is similarly efficient for TCE dechlorination whether it is injected once or in doses. Moreover, the present work indicates that the addition of butyrate in great excess cannot be avoided, since it mo

Anaerobic microbial reductive dechlorination of tetrachloroethene to predominately trans-1,2-dichloroethene.

While most sites and all characterized PCE and TCE dechlorinating anaerobic bacteria produce cis-DCE as the major DCE isomer, significant amounts of trans-DCE are found in the environment. We have obtained microcosms from some sites and enrichment cultures that produce more trans-DCE than cis-DCE. These cultures reductively dechlorinated PCE and TCE to trans-DCE and cis-DCE simultaneously and in a ratio of 3(+/-0.5):1 that was stable through serial transfers with a variety of electron donors and occurred in both methanogenic and nonmethanogenic enrichments. Two sediment-free, nonmethanogenic e

Phylogenetic analysis of bacterial populations in an anaerobic microbial consortium capable of degrading saturation concentrations of tetrachloroethylene.

An anaerobic microbial consortium able to biodegrade saturation levels of perchloroethylene (PCE) in a column containing a source zone of PCE was examined phylogenetically to determine microbial community structure and spatial variation in relation to the PCE source. The consortium was comprised of at least 34 members with 7 organisms sharing affiliations with known respiratory or cometabolic dechlorinators. Seven other organisms had their closest phylogenetic relative detected in other environments containing chlorinated compounds. Based on denaturing gradient gel electrophoresis, significant

Reductive dechlorination of tetrachloroethene to ethene by a two-component enzyme pathway.

Two membrane-bound, reductive dehalogenases that constitute a novel pathway for complete dechlorination of tetrachloroethene (perchloroethylene [PCE]) to ethene were partially purified from an anaerobic microbial enrichment culture containing Dehalococcoides ethenogenes 195. When titanium (III) citrate and methyl viologen were used as reductants, PCE-reductive dehalogenase (PCE-RDase) (51 kDa) dechlorinated PCE to trichloroethene (TCE) at a rate of 20 micromol/min/mg of protein. TCE-reductive dehalogenase (TCE-RDase) (61 kDa) dechlorinated TCE to ethene. TCE, cis-1,2-dichloroethene, and 1,1-di

The humic acid analogue antraquinone-2,6-disulfonate (AQDS) serves as an electron shuttle in the electricity-driven microbial dechlorination of trichloroethene to cis-dichloroethene.

Quinone moieties in humic substances have previously been shown to serve as extracellular electron acceptors in different metabolic pathways. Here we show that the humic acid analogue antraquinone-2,6-disulfonate (AQDS) can also serve as an electron donor in the microbial reductive dechlorination of TCE to cis-DCE. In a bioelectrochemical system (BES), equipped with a glassy carbon electrode (cathode) polarized at -250mV vs. SHE, electrically reduced AQDS served as the shuttle of electrons between the electrode surface and the dechlorinating bacteria. Interestingly, AQDS selectively stimulated

Chlorinated ethene plume evolution after source thermal remediation: Determination of degradation rates and mechanisms.

The extent, mechanism(s), and rate of chlorinated ethene degradation in a large tetrachloroethene (PCE) plume were investigated in an extensive sampling campaign. Multiple lines of evidence for this degradation were explored, including compound-specific isotope analysis (CSIA), dual C-Cl isotope analysis, and quantitative real-time polymerase chain reaction (qPCR) analysis targeting the genera Dehalococcoides and Dehalogenimonas and the genes vcrA, bvcA, and cerA. A decade prior to this sampling campaign, the plume source was thermally remediated by steam injection. This released dissolved org

Bioelectrochemical system accelerates reductive dechlorination through extracellular electron transfer networks.

Bioelectrochemical system is considered as a promising approach for enhanced bio-dechlorination. However, the mechanism of extracellular electron transfer in the dechlorinating consortium is still a controversial issue. In this study, bioelectrochemical systems were established with cathode potential settings at -0.30 V (vs. SHE) for trichloroethylene reduction. The average dechlorination rate (102.0 μM Cl·d-1) of biocathode was 1.36 times higher than that of open circuit (74.7 μM Cl·d-1). Electrochemical characterization via cyclic voltammetry illustrated that electrostimulation promoted elec

Model-based evidence for the relevance of microbial community variability to the efficiency of the anaerobic reductive dechlorination of TCE.

The composition of mixed dechlorinating communities varies considerably in field and laboratory conditions. Dechlorinators thrive alongside with distinctive populations that help or hinder dechlorination. The variability of the composition of dechlorinating communities inevitably precludes a firm consensus regarding the optimal strategies for biostimulation. This lack of consensus motivated a model-based approach for the investigation of how the variability of the composition of a microbial community impacts the electron donor supply strategies for accelerating chloroethene removal. To this en

Use of silicate minerals for long-term pH control during reductive dechlorination of high tetrachloroethene concentrations in continuous flow-through columns.

The long-term buffering potential of three silicate minerals (diopside, fayalite and forsterite) present as fine particles in porous quartz sand medium was evaluated in flow-through column experiments over a period of 6.5 months. The columns were operated with PCE concentrations close to saturation and inoculated with the organohalide-respiring consortium SDC-9™, which is able to completely dechlorinate PCE to ethene at high concentrations. In the absence of pH buffering agents, fermentation and organohalide respiration drove the pH close to 6.1, leading to severe inhibition of PCE dechlorinat

Bioaugmentation for treatment of dense non-aqueous phase liquid in fractured sandstone blocks.

Laboratory experiments were performed in discretely fractured sandstone blocks to evaluate the use of bioaugmentation to treat residual dense non-aqueous phase liquid (DNAPL) tetrachloroethene (PCE). Significant dechlorination of PCE and growth of Dehalococcoides spp. (DHC) occurred within the fractures. DNAPL dissolution was enhanced during bioaugmentation by up to a factor of approximately 3.5, with dissolved PCE concentrations at or near aqueous solubility. The extent of dechlorination and DNAPL dissolution enhancement were dependent upon the fracture characteristics, residence time in the

Interaction of perchlorate and trichloroethene bioreductions in mixed anaerobic culture.

This work evaluated the interaction of perchlorate and trichloroethene (TCE), two common co-contaminants in groundwater, during bioreduction in serum bottles containing synthetic mineral salts media and microbial consortia. TCE at concentrations up to 0.3mM did not significantly affect perchlorate reduction; however, perchlorate concentrations higher than 0.1mM made the reduction of TCE significantly slower. Perchlorate primarily inhibited the reduction of vinyl chloride (VC, a daughter product of TCE) to ethene. Mechanistic analysis showed that the inhibition was mainly because perchlorate re

1,1-dichloroethene as a predominant intermediate of microbial trichloroethene reduction.

A microbial culture derived from a landfill site in Dover, DE consistently reduced trichloroethene (TCE) to ethene through 1,1-dichloroethene (DCE) as a dominant intermediate in the presence of ampicillin. A constant 1,1-DCE-to-cis-DCE ratio of 2.4 +/- 0.3 was observed for more than two years, while trans-DCE was never detected. Without ampicillin, however, TCE was reduced to ethene almost exclusively through cis-DCE, suggesting that the culture contained at leasttwo TCE-dechlorinating populations. Two subcultures, which were established using 1,1-DCE or vinyl chloride as an electron acceptor,

Stratification of chlorinated ethenes natural attenuation in an alluvial aquifer assessed by hydrochemical and biomolecular tools.

Biomolecular and hydrochemical tools were used to evaluate natural attenuation of chlorinated ethenes in a Quaternary alluvial aquifer located close to a historical source of large-scale tetrachloroethylene (PCE) contamination. Distinct stratification of redox zones was observed, despite the aquifer's small thickness (2.8 m). The uppermost zone of the target aquifer was characterised by oxygen- and nitrate-reducing conditions, with mixed iron- to sulphate-reducing conditions dominant in the lower zone, along with indications of methanogenesis. Natural attenuation of PCE was strongly influenced

Exploiting the intrinsic microbial degradative potential for field-based in situ dechlorination of trichloroethene contaminated groundwater.

Bioremediation of trichloroethene (TCE) polluted groundwater is challenging, with limited next generation sequencing (NGS) derived information available on microbial community dynamics associated with dechlorination. Understanding these dynamics is important for designing and improving TCE bioremediation. In this study, biostimulation (BS), biostimulation-bioaugmentation (BS-BA) and monitored natural attenuation (MNA) approaches were applied to contaminated groundwater wells resulted in ≥ 95% dechlorination within 7 months. Vinyl chloride's final concentrations in stimulated wells were between

Monitoring bioaugmenation with single-well push-pull tests in sediment systems contaminated with trichloroethene.

Bioaugmentation to enhance the rate and extent of reductive dechlorination of chlorinated ethenes was investigated in intermediate ( approximately 1 m) scale physical aquifer models (PAMs) designed to simulate the groundwater flow field near an injection well. Push-pull tests were used to quantify the reductive dechlorination of injected trichloroethene (TCE) and trichlorofluoroethene (TCFE) in prepared sediment packs with and without an added dechlorinating culture containing Dehalococcoides spp. Distribution of the added culture throughout the sediment pack was confirmed by microscopic obser

Large-scale production of bacterial consortia for remediation of chlorinated solvent-contaminated groundwater.

Chlorinated solvents such as perchloroethylene (PCE) and trichloroethylene (TCE) continue to be significant groundwater contaminants throughout the USA. In many cases efficient bioremediation of aquifers contaminated with these chemicals requires the addition of exogenous microorganisms, specifically members of the genus Dehalococcoides (DHC). This process is referred to as bioaugmentation. In this study a fed-batch fermentation process was developed for producing large volumes (to 3,200 L) of DHC-containing consortia suitable for treating contaminated aquifers. Three consortia enriched from t

A Polyhydroxybutyrate (PHB)-Biochar Reactor for the Adsorption and Biodegradation of Trichloroethylene: Design and Startup Phase.

In this work, polyhydroxy butyrate (PHB) and biochar from pine wood (PWB) are used in a mini-pilot scale biological reactor (11.3 L of geometric volume) for trichloroethylene (TCE) removal (80 mgTCE/day and 6 L/day of flow rate). The PHB-biochar reactor was realized with two sequential reactive areas to simulate a multi-reactive permeable barrier. The PHB acts as an electron donor source in the first "fermentative" area. First, the thermogravimetric (TGA) and differential scanning calorimetry (DSC) analyses were performed. The PHB-powder and pellets have different purity (96% and 93% w/w) and

Microbiome reengineering by four environmental factors for the rapid biodegradation of trichloroethylene.

Trichloroethylene (TCE) was once a widely applied industrial solvent, but is now an infamous contaminant in groundwater. Although anaerobic reductive dechlorination is considered a greener remediation approach, the accumulation of toxic intermediates, such as vinyl chloride (VC), and a longer remediation period are highly concerning. Biostimulation and bioaugmentation have been developed to solve these problems. The former method may not be effective, and the latter may introduce foreign genes. Here, we propose a new approach by applying environmental stresses to reshape the indigenous microbi
