Injectable Hydrogel as Intracanal Medication for Root Canal Disinfection.

Due to the complex anatomical structures of the root canal, thorough intracanal disinfection has always been challenging in endodontic treatment. Existing intracanal medicaments exhibit limitations such as low permeability and suboptimal antibacterial performance. Thus, an intracanal medicament that combines excellent operating performance with potent antibacterial properties is required. Therefore, we designed an injectable hydrogel loaded with modified triple antibiotic drugs (mTAD) through a Schiff base reaction of carboxymethyl chitosan (CMCS) and polyethylene glycol aldehyde (OHC-PEG-CHO)

Influence of smear layer on depth of penetration of three endodontic sealers: an SEM study.

The purpose of this study was to examine the effect of the smear layer on the penetration depth of Sealapex, Roth 811, and CRCS root canal sealers into the dentinal tubules. Forty recently extracted, human single-rooted teeth were used. The crowns were cut and removed at the cemento-enamel junction level and the root canals were prepared to a #60 K-file. The teeth were then randomly divided into two groups of 20, group A and group B. The smear layer was removed from all teeth in group A with EDTA and NaOCl. The smear layer remained in all teeth in group B. Two roots in each group were used as

Altered tissue contribution to interfacial bond strength with acid conditioned dentin.

PURPOSE: To measure the shear bond strength of resin composite to dentin using three fourth generation bonding agents in combination with three different acid treatments. One such treatment was followed by sodium hypochlorite. The purpose was to determine the contribution which the collagen rich, demineralized zone made to bond strength. MATERIALS AND METHODS: Three groups, representing (1) All Bond 2/BisFil, (2) Optibond Dual Cure/XRV and (3) Scotchbond Multi-Purpose/Z100, contained four sub-groups each comprised of 10 caries-free molar teeth whose dentin was exposed in a transverse mid-coro

Ultrastructure of intraradicular dentin after irrigation with BioPure MTAD. II. The consequence of obturation with an epoxy resin-based sealer.

In addition to an initial report on the creation of thick demineralized collagen matrices in root dentin that were irrigated with BioPure MTAD or EDTA as final rinses for 5 minutes, this study examined the effect of a 2-minute irrigation time of these final irrigants on the extent of demineralization of intact intraradicular dentin and on interfacial nanoleakage after obturation with AH Plus and gutta-percha using warm vertical compaction. Transmission electron microscopy confirmed that the reduction in irrigation time did not compromise the smear layer removing capacity of these two irrigants

Susceptibility of Nd:YAG laser-irradiated root surfaces in replanted teeth to external inflammatory resorption.

Nd:YAG laser-induced modification of the root surface may inhibit development of external inflammatory resorption in replanted teeth. This study tested this hypothesis in vivo. The pulp chambers of six mandibular premolars in each of two dogs were accessed, inoculated with plaque, and sealed (Groups 1, 2). Two additional premolars in each dog were endodontically treated without inoculation (Groups 3, 4). After 2 weeks, teeth were hemisected and extracted. Each root had a 2 x 3 mm surface area denuded of cementum on the buccal and lingual surface. In Groups 1 (n = 12 roots) and 3 (n = 4), the d

Experimental intra-oral caries models in fluoride research.

The use of experimental intra-oral caries models has increased in fluoride research. This paper focuses on the pre-clinical intra-oral models, the in situ and in vivo models, the various types, their benefits and disadvantages. Both preparation and sterilization of the hard tissue substrates can affect the substrates and therefore the results. Care needs to be taken that dentine samples are not exposed to drying and consequently shrinking during preparation and evaluation. Sterilization by gamma-radiation is at present the least tissue-damaging method. The most realistic experimental model is

Calcium ion diffusion from calcium hydroxide-containing materials in endodontically-treated teeth: an in vitro study.

The aim of this in vitro study was to determine the amount and duration of diffusion of calcium ions from both a calcium hydroxide-containing root canal sealer and an intracanal medicament, through the apical foramen and the dentinal tubules of endodontically-treated teeth. The root canals of 88 freshly extracted single-rooted teeth were prepared using the modified double flared, balanced force technique with patency filing. The teeth were divided into four test groups of 20 teeth each and a control group of eight teeth. One group was dressed with a non-setting calcium hydroxide while the othe

Pulpal hemodynamics and interstitial fluid pressure: balance of transmicrovascular fluid transport.

The net rate of fluid movement across the microvascular endothelium is governed by hydrostatic and protein osmotic pressures operating on each side of the vessel wall. These basic forces regulating the pulpal transmicrovascular fluid exchange are the same as in other tissues. However, the pulp is special in regard to its enclosement between rigid dentin walls, implying low interstitial compliance. In addition, the pulp has a relatively low perfusion pressure. In the low compliant pulpal interstitium, even a modest rise in net filtration will favor a relatively large increase in interstitial fl

Impact of claudin-10 deficiency on amelogenesis: Lesson from a HELIX tooth.

In epithelia, claudin proteins are important components of the tight junctions as they determine the permeability and specificity to ions of the paracellular pathway. Mutations in CLDN10 cause the rare autosomal recessive HELIX syndrome (Hypohidrosis, Electrolyte imbalance, Lacrimal gland dysfunction, Ichthyosis, and Xerostomia), in which patients display severe enamel wear. Here, we assess whether this enamel wear is caused by an innate fragility directly related to claudin-10 deficiency in addition to xerostomia. A third molar collected from a female HELIX patient was analyzed by a combinati

Fucoidan-derived carbon dots against Enterococcus faecalis biofilm and infected dentinal tubules for the treatment of persistent endodontic infections.

Enterococcus faecalis (E. faecalis) biofilm-associated persistent endodontic infections (PEIs) are one of the most common tooth lesions, causing chronic periapical periodontitis, root resorption, and even tooth loss. Clinical root canal disinfectants have the risk of damaging soft tissues (e.g., mucosa and tongue) and teeth in the oral cavity, unsatisfactory to the therapy of PEIs. Nanomaterials with remarkable antibacterial properties and good biocompatibility have been developed as a promising strategy for removing pathogenic bacteria and related biofilm. Herein, carbon dots (CDs) derived fr

The influence of endodontic treatment upon periodontal wound healing.

The interrelationship between periodontal and endodontic disease has aroused much speculation, confusion, and controversy. Pulpal and periodontal problems are responsible for more than 50% of tooth mortality today. Diagnosis is often difficult since these diseases have been studied primarily as separate entities. The toxic substances of the pulp may initiate periodontal defects through canal ramifications and patent dentinal tubules, thus impairing wound healing in regenerative procedures. Although no studies exist addressing the direct effect of pulpal infection on the outcome of guided tissu

The effect of tubular penetration of root canal sealers on dye microleakage.

An in vitro study of a possible correlation between penetration of dentinal tubules by four root canal sealers and microleakage of external fluids into the canal was done using a dye leakage method and scanning electron microscopy. The root canals of 45 teeth were instrumented and the smear layer removed prior to obturation of root canals with gutta-percha and one of four sealers: Diaket, Endomethasone, CRCS or Ketac-Endo. The extent of leakage was scored after immersion in India ink for 72 h. The same specimens were also used for scanning electron microscopic evaluation. There was a statistic

Polydopamine@Zinc oxide coated macroporous membrane for antibacterial protection and early pulp repair in pulpitis.

Continuous decompression and drainage are a vital surgical strategy for managing severe tissue infections. In vital pulp therapy (VPT) for irreversible pulpitis, there is a clinical demand for advanced biomaterials capable of effectively sealing the pulp cavity, alleviating pulpal hypertension, preventing bacterial infiltration, and resolving acute pulp inflammation in bacteria-rich environments. In this study, we developed a polydopamine@zinc oxide nanoparticle (ZnO-NP)-coated polytetrafluoroethylene (PTFE) membrane with tunable Zn content ranging from 2.51 to 7.45 wt%. The polydopamine enhan

Dental pulp response to bacterial cell wall material.

Lipopolysaccharides (LPS) from Bacteroides oralis and Veillonella parvula and cell wall material from Lactobacillus casei were studied for their capacity to induce leukocyte migration in the dental pulp and in an implanted wound chamber. Three adult monkeys were challenged using lyophilized material sealed into buccal Class V cavities prepared in dentin. Pulp tissue responses were observed histologically eight and 72 hours after initiation of the experiment. Subjacent to cut dentinal tubules, bacterial materials induced polymorphonuclear leukocyte (PMN's) infiltration in the pulp tissue of the

Tissue pressure and blood flow in pulpal inflammation.

The initial vascular reactions during inflammation are vasodilation and increased vessel permeability. Both these basic reactions cause increased pulpal fluid volume. In the dental pulp the inflammatory vascular reactions take place in a rigid enclosed dentin chamber, which to some extent makes the pulp vulnerable. Due to this lack of distensibility any gain in pulpal volume will necessarily increase the pulpal tissue pressure. If the tissue pressure rises to the same level as the blood pressure it will compress the pulpal vessels, thus counteracting a beneficial blood flow increase during inf
