Desmoglein 2-Dependent Arrhythmogenic Cardiomyopathy Is Caused by a Loss of Adhesive Function.

BACKGROUND: The desmosomal cadherin desmoglein 2 (Dsg2) localizes to the intercalated disc coupling adjacent cardiomyocytes. Desmoglein 2 gene (DSG2) mutations cause arrhythmogenic cardiomyopathy (AC) in human and transgenic mice. AC is characterized by arrhythmia, cardiodilation, cardiomyocyte necrosis with replacement fibrosis, interstitial fibrosis, and intercalated disc dissociation. The genetic DSG2 constellations encountered are compatible with loss of adhesion and altered signaling. To further elucidate pathomechanisms, we examined whether heart-specific Dsg2 depletion triggers cardiomy

Desmoglein 3, its pathogenecity and a possibility for therapeutic target in pemphigus vulgaris.

INTRODUCTION: Desmoglein 3 (Dsg3) is one of desmosomal cadherins and functions in epidermal keratinocyte adhesion. IgG anti-Dsg3 autoantibodies are detected in pemphigus vulgaris, an autoimmune bullous disease showing blisters and erosions on the skin and oral mucosa. Other types of pemphigus also show anti-Dsg3 antibodies. Genetic disease of Dsg3 has not been reported. AREAS COVERED: Many in vitro and in vivo studies have indicated pathogenic role of anti-Dsg3 antibodies. Blisters in pemphigus vulgaris are thought to be developed by loss of keratinocyte adhesions by binding of anti-Dsg3 anti

Utility of desmoglein ELISA in the clinical correlation and disease monitoring of pemphigus vulgaris.

BACKGROUND: Pemphigus vulgaris (PV) is a relatively common autoimmune blistering disease in India. Recently, enzyme-linked immunosorbent assay (ELISA) test against desmoglein1 (Dsg 1) and desmoglein3 (Dsg 3) has been developed, which is found to be highly sensitive and specific. OBJECTIVE: To assess the utility of ELISA for monitoring the disease of PV. METHODS: Serum samples from 63 active PV patients were tested for anti-Dsg1 and anti-Dsg3 using ELISA. They were tested for anti-Dsg1 and anti-Dsg3 reactivity before treatment. The test was performed during remission period and after relapse

Desmoglein 2 is a receptor for adenovirus serotypes 3, 7, 11 and 14.

We have identified desmoglein-2 (DSG-2) as the primary high-affinity receptor used by adenoviruses Ad3, Ad7, Ad11 and Ad14. These serotypes represent key human pathogens causing respiratory and urinary tract infections. In epithelial cells, adenovirus binding of DSG-2 triggers events reminiscent of epithelial-to-mesenchymal transition, leading to transient opening of intercellular junctions. This opening improves access to receptors, for example, CD46 and Her2/neu, that are trapped in intercellular junctions. In addition to complete virions, dodecahedral particles (PtDds), formed by excess amo

Staphylococcal scalded skin syndrome: loss of desmoglein 1 in patient skin.

Staphylococcal scalded skin syndrome (SSSS) is a blistering disease of the skin caused by an infection with certain strains of Staphylococcus aureus. In vitro studies have suggested that exfoliative toxins secreted by these bacteria cleave the desmosomal adhesion molecule desmoglein 1 leading to loss of cell-cell contact in the superficial epidermis. In this study we investigated the fate of desmoglein 1 in biopsies of patients with SSSS to see whether the ectodomain of desmoglein 1 is cleaved. Our data largely confirm previous in vitro data. The different biopsies demonstrated the loss of the

Pemphigus vulgaris IgG directly inhibit desmoglein 3-mediated transinteraction.

The autoimmune blistering skin disease pemphigus is caused by autoantibodies against keratinocyte surface Ags. In pemphigus vulgaris (PV), autoantibodies are primarily directed against desmosomal cadherins desmoglein (Dsg) 3 and Dsg 1, whereas pemphigus foliaceus (PF) patients only have Abs against Dsg 1. At present, it is unclear whether Dsg autoantibodies contribute to pemphigus pathogenesis by direct inhibition of Dsg transinteraction. Using atomic force microscopy, we provide evidence that PV-IgG directly interfere with homophilic Dsg 3 but, similar to PF-IgG, not with homophilic Dsg 1 tra

Pemphigus vegetans Neumann type with anti-desmoglein and anti-periplakin autoantibodies.

Pemphigus vegetans is a rare variant of pemphigus vulgaris characterized by vegetating lesions in the folds and mouth and by the presence of autoantibodies against desmoglein 3. We describe two Caucasian patients with pemphigus vegetans, one of them presented antibodies to desmoglein 3 and 1 and the other one to desmoglein 3. Both patients also had circulating antibodies against a 190 kDa protein co-migrating with periplakin. Anti-periplakin reactivity is usually detected in paraneoplastic pemphigus, while it has never been reported in pemphigus vegetans. Our observation enlarges the spectrum

Pemphigoid nodularis with IgA autoantibodies against the intracellular domain of desmoglein 1.

Pemphigoid nodularis is a rare variant of bullous pemphigoid. We report a 49-year-old Japanese male with clinical and histopathological features of pemphigoid nodularis including circulating and in vivo-bound IgG antibasement membrane zone antibodies and IgA anti-intercellular antibodies. Although the precise molecular target of the IgG autoantibodies could not be determined, intriguingly, immunoblotting showed that the IgA in the patient's serum reacted with the intracellular domain of desmoglein 1, the target antigen in cases of pemphigus foliaceus. However, the IgA did not react with the ex

Herpetiform pemphigus showing reactivity with pemphigus vulgaris antigen (desmoglein 3).

We report a patient with herpetiform pemphigus (HP) who showed reactivity only with pemphigus vulgaris (PV) antigen but not with pemphigus foliaceus (PF) antigen. Direct and indirect immunofluorescence revealed keratinocyte cell surface staining in the lower layers of the epidermis, where desmoglein 3 (Dsg3) is expressed. Immunoblot analysis, using ethylenediamine tetra-acetic acid-separated human epidermal extracts, revealed that the patient's serum recognized only a 130-kDa polypeptide which co-migrated with Dsg3. By antigen-specific immunoadsorption studies, using desmoglein 1 (Dsg1) and Ds

A non-desmoglein component of bovine epidermis reactive with pemphigus foliaceus sera.

A component from bovine epidermis has been purified and found to react with sera from patients with pemphigus foliaceus (PF). This component appears to have molecular weight characteristics similar to those of a previously reported component of human and bovine epidermis which was reactive with PF sera, but only detectable by radiolabeling. It is composed of an 80 kD and a 45 kD polypeptide which are thought to be covalently linked. By chromatographic data, the native form appears to be a noncovalently joined dimer of the 80 and 45 kD polypeptides. Amino acid analysis and sequence data indicat

Desmoglein-1 coordinates cell proliferation and actin dynamics to promote epidermal morphogenesis.

Skin development depends on coordinated biochemical and mechanical signaling to form a multilayered epidermal barrier. Our previous work demonstrated that the stratified tissue-specific desmosomal cadherin Desmoglein-1 (Dsg1) promotes epidermal differentiation in human in vitro models. Dsg1 first appears in the developing mouse epidermis around E14, coinciding with a developmental transition in stratification and the onset of terminal differentiation, raising the question of how Dsg1 contributes to epidermal morphogenesis in vivo . We generated a wild-type (WT) single-cell RNA sequencing (scRN

Correlation of Clinical and Histopathological Features With Anti-Desmoglein Antibody Profile in Pemphigus Patients.

BACKGROUND: The desmoglein compensation hypothesis (DCH) proposes that clinical and histopathological features of pemphigus correlate with differential expression of desmoglein 1 (Dsg1) and desmoglein 3 (Dsg3) in skin and mucosa, and corresponding autoantibody profiles. While DCH explains many classical patterns, increasing exceptions have challenged its universality. AIM: This study aimed to correlate clinical phenotype and histopathological split level with anti-Dsg1 and anti-Dsg3 antibody profiles in pemphigus vulgaris (PV) and pemphigus foliaceus (PF), and evaluate consistency with DCH.

Desmoglein-driven dynamic signaling in pemphigus vulgaris: a systematic review of pathogenic pathways.

Epithelial tissue integrity is maintained through specialized intercellular junctions known to coordinate homeostatic processes. In this context, outside-in signaling and mechanotransduction through desmosomal cadherins, the building blocks of desmosomes and main stress bearers in epithelial tissue, are only starting to emerge. To better understand the dual function of desmosomal cadherins in structural integrity and cellular signaling, we here performed a systematic, unbiased review on pathogenic signaling effectors identified in models and patients with pemphigus vulgaris (PV). PV is an auto

Persistent desmoglein-1 downregulation and periostin accumulation in histologic remission of eosinophilic esophagitis.

BACKGROUND: Patients with eosinophilic esophagitis (EoE) require long-lasting resolution of inflammation to prevent fibrostenosis and dysphagia. However, the dissociation between symptoms and histologic improvement suggests persistent molecular drivers despite histologic remission. OBJECTIVE: We characterized persisting molecular alterations in pediatric patients with EoE using tissue transcriptomics and proteomics. METHODS: Esophageal biopsy samples (n = 247) collected prospectively during 189 endoscopies from pediatric patients with EoE (n = 36, up to 11 follow-up endoscopies) and pediatri
