We studied the uptake mechanism of mercuric chloride (Hg) and methylmercuric chloride (MeHg) in Aedes albopictus C6/36 cells. The uptake kinetics, together with the effect of temperature and a metabolic inhibitor (2, 4-dinitrophenol) on the mercury accumulation, were examined. Both amounts of internalized Hg and MeHg increased linearly with the extracellular concentration. Initially, the influx rate was high for both metal species but MeHg was found to accumulate seven times faster than Hg. Para-aminohippuric acid (PAH) accumulates against a concentration gradient in the ciliary body and independently in the iris of the rhesus monkey eye. This accumulation is inhibited by incubation of 0 degrees C and shows saturation kinetics in both tissues. Cyanide, ouabain, dinitrophenol, iodopyracet, and probenecid effectively depress PAH uptake in both tissues, but anaerobic incubation conditions have little effect on uptake in either tissue. Glycosides (alpha- and beta-D-glucosides and -D-galactosides) derived from three pesticides, 2-tert-butyl-2,4-dinitrophenol,2,6-dibromo-4-cyanophenol, and 5-(2,4-dichlorophenoxy)-2-nitrobenzoic acid, were synthesized from 2,3,4,6-tetra-O-chloroacetyl-D-gluco- and -D-galactopyranose by use of the Mitsunobu reaction. The kinetics of the Ca2+-ATPase of chicken skeletal myosin has been found to be of a biphasic type. DNP and SCN- increased Ca2+-ATPase activity, whereas CN- and HCO3- acted as inhibitors in the concentration range used. Upon raising substrate concentration, activation and inhibition constants for those modifiers responded also with an elevation of their values.

A model of cellular response to irradiation involving adenosine diphosphate ribosylation (ADP-ribosylation) is proposed. Its main assumptions are (a) control of accessibility of sites for ADP-ribosylation in chromatin by free Ca2+/Mg2+ ratio; and (b) regulation of the Ca2+/Mg2+ ratio by factors affecting intracellular free Ca2+ concentration; the regulation would be mediated by mitochondria. The model seeks to explain the mechanism of action of radiomodifiers such as caffeine, local anaesthetics, polyamines and 2,4-dinitrophenol.

Chlamydospores of Fusarium sulphureum were formed within 72 h by incubating macroconidia at 37 degrees C in stationary culture in a medium containing mannitol and inorganic salts. The conversion process was dependent on inoculum level, temperature, aeration, and the presence of an external source of nitrogen and carbon. Chlamydospore formation was not inhibited by an uncoupler of oxidative phosphorylation, 2,4-dinitrophenol, or by inhibitors of bacterial cell wall synthesis; however, it was affected by inhibitors of respiration and of protein and chitin synthesis.

Iron chelated to the exochelins from Mycobacterium neoaurum was taken up by a suspension of M. leprae, prepared from the liver of an infected armadillo, over 15 hr. No uptake occurred when the iron was chelated with exochelins from M. bovis BCG or M. smegmatis or to a single exochelin from M. vaccae. Uptake appeared to be by facilitated diffusion since it was not inhibited by either HgCl2, NaN3, or 2,4-dinitrophenol. This was similar to the mode of uptake of ferriexochelin into M. neoaurum itself.

The influx of 2,4-dichlorophenoxyacetic acid (2,4-D) into Chinese hamster ovary (CHO) cells was studied. The cells mainly took up but did not metabolize the undissociated form of the herbicide. The uptake of 2,4-D was carried out against a concentration gradient and was inhibited by sodium azide and dinitrophenol. The results presented here show that the herbicide influx was an active, energy dependent process. (Na+ + K+)ATPase does not seem to be involved because ouabain, an inhibitor of the enzyme, did not affect the 2,4-D uptake.

Like the nitrate transport system, the nitrite uptake system in Neurospora crassa is induced by either nitrate or nitrite. This induction is prevented by cycloheximide, puromycin, or 6-methyl purine. The K(m) for nitrite of the induced nitrite uptake system is 86 muM, and the V(max) is 100 mumol of nitrite per g (wet weight) per h. Nitrite uptake is inhibited by metabolic poisons such as arsenate, dinitrophenol, cyanide, and antimycin A.
