A Historical Review of Brain Drug Delivery.

The history of brain drug delivery is reviewed beginning with the first demonstration, in 1914, that a drug for syphilis, salvarsan, did not enter the brain, due to the presence of a blood-brain barrier (BBB). Owing to restricted transport across the BBB, FDA-approved drugs for the CNS have been generally limited to lipid-soluble small molecules. Drugs that do not cross the BBB can be re-engineered for transport on endogenous BBB carrier-mediated transport and receptor-mediated transport systems, which were identified during the 1970s-1980s. By the 1990s, a multitude of brain drug delivery tec

Structural patterns and transcriptional effects of integrated Epstein-Barr Virus revealed by long-read sequencing and RNA-sequencing in nasopharyngeal carcinoma.

Integration of Epstein-Barr virus (EBV) DNA into the human genome is a critical event in nasopharyngeal carcinogenesis. Here, we comprehensively characterize large-scale virus-human integration events in three EBV-positive nasopharyngeal carcinoma (NPC) cell lines and one patient NPC tumor using Nanopore long-read sequencing technology. We identified four distinct integration types, with Type A being particularly prevalent, characterized by the integration of a single fragment of the EBV genome followed by human DNA. Our findings reveal the involvement of multiple integration events in inducin

Updated Gene Therapy for Renal Inborn Errors of Metabolism.

Inborn errors of metabolism (IEMs) are a group of disorders resulting from defects in enzymes in metabolic pathways. These disorders impact the processing of metabolites, leading to a wide array of effects on each organ system. Advances in genetic screening have allowed for the early identification and intervention of IEMs, traditionally in the form of enzyme replacement or vitamin supplementation. However, many IEMs disrupt essential metabolic pathways where simple supplementation proves ineffective, resulting in substantial disease burden. In the case of renal IEMs, metabolic pathway disrupt

Legalomics: why microbiology matters to the promotion of just societies.

Despite profound socioeconomic and ecological consequences, criminal justice remains an overlooked dimension of sustainability. Sustainable Development Goal (SDG) 16 calls for promoting inclusive societies and ensuring justice for all. Yet the SDG 16 framework largely ignores the biological and structural determinants of behavior related to justice involvement. This perspective article argues that microbial ecology plays a foundational role in cognition and behavior-factors central to justice outcomes. Advances in neuromicrobiology and omics technologies show how microbial disruptions linked t

New insights into the potential mechanisms of spermatogenic failure in patients with idiopathic azoospermia.

Idiopathic azoospermia (IA) refers to azoospermia without a clear aetiology. Due to the unclear aetiology and pathological mechanism of IA, there is no effective treatment for IA. The development of assisted reproductive and microsperm extraction technologies has brought hope to patients with IA with fertility problems. However, there are still many patients with IA whose testes lack healthy sperm, causing infertility. Therefore, it is key to identify how testicular spermatogenic failure can be reversed to promote spermatogenesis in patients with IA to resolve fertility problems; these goals a

Inactivation mechanisms of cold plasma on Bacillus cereus biofilms and its application in simulated meat processing environments.

Bacterial biofilms are a common source of contamination in the food industry, often forming on surfaces such as equipment and pipelines, and contributing to the spread of foodborne diseases. This study aims to evaluate the effectiveness of an atmospheric pressure plasma jet (APPJ) in inactivating Bacillus cereus biofilms on glass surfaces and to explore the underlying sterilization mechanisms. Meanwhile, the formation conditions of biofilms in the beef slaughtering and processing environments are simulated to assess the application potential of plasma technology in the food industry. Results s

Navigating Challenges and Adapting Innovations in Acute Kidney Injury and Kidney Failure.

Acute kidney injury and kidney failure represent critical health challenges, requiring continuous and specialized care. The impact of natural disasters, such as earthquakes, floods, and hurricanes, alongside man-made crises like armed conflicts and pandemics, exacerbates the complexities of managing these conditions. Disasters disrupt health care infrastructure, cause resource shortages, and pose ethical dilemmas in care allocation, often leading to surges in acute kidney injury cases and interruptions in kidney failure treatment. This article examines these challenges and underscores the impo

A Novel Approach to Calcium Destruction in Coronary and Peripheral Blood Vessels: Intravascular Lithotripsy.

Lithotripsy has been used for decades in the treatment of kidney stones and gallstones, in which ultrasound shock waves generated outside of the body are used to physically break up hardened masses. In the past decade, intravascular lithotripsy (IVL), a technology developed by Shockwave Medical Inc. (Santa Clara, CA), has emerged as a novel therapy for the treatment of vascular calcification. In the coronary blood vessels, IVL modifies arterial calcium and enables percutaneous coronary interventions to be performed in a safe and consistent manner, and in the peripheral blood vessels, IVL can b

Time to Abandon Antimicrobial Approaches in Wound Healing: A Paradigm Shift.

Antimicrobial approaches (eg, antibiotics and antiseptics) have been used for decades in the treatment of infected wounds, ulcers, and burns. However, an increasing number of meta-analyses have raised questions regarding the therapeutic value of these approaches. Newer findings show that the body actively hosts an ecosystem of bacteria, fungi, viruses, and mites on its outer surfaces, known as the microbiome, as part of its defense against pathogens. Antimicrobials would disrupt this system and thereby work against the strategy the body has chosen. Recently, a new technology, micropore particl

Cas9/sgRNA selective targeting of the P23H Rhodopsin mutant allele for treating retinitis pigmentosa by intravitreal AAV9.PHP.B-based delivery.

P23H is the most common mutation in the RHODOPSIN (RHO) gene leading to a dominant form of retinitis pigmentosa (RP), a rod photoreceptor degeneration that invariably causes vision loss. Specific disruption of the disease P23H RHO mutant while preserving the wild-type (WT) functional allele would be an invaluable therapy for this disease. However, various technologies tested in the past failed to achieve effective changes and consequently therapeutic benefits. We validated a CRISPR/Cas9 strategy to specifically inactivate the P23H RHO mutant, while preserving the WT allele in vitro. We, then,

Symbiotic AI and equitable digital health: toward trustworthy and inclusive health ecosystems.

Digital health has accelerated rapidly through advances in telemedicine, wearable technologies, artificial intelligence (AI), remote monitoring, and interoperable healthcare infrastructures. Despite these developments, substantial inequities persist among underserved populations, including rural and remote communities, older adults, low-socioeconomic groups, and geographically isolated populations exposed to infrastructure disruptions caused by avalanches, landslides, storms, flooding, or prolonged power outages. Current digital health approaches frequently emphasize technological innovation a

SILENCE: a new forward genetic technology.

Silencing induced by long terminal repeat (LTR)-encoded cis-acting response element, termed SILENCE, is a forward genetic system that allows for conditional, epigenetic control of host-gene transcription. This new research tool is independent of gene mutation or disruption, does not require complementation, and conditional gene repression appears complete at the level of protein function. SILENCE functions in hypodiploid cells and is a platform technology with broad applications in gene discovery.

The Gut Microbiome as a Reservoir for Antimicrobial Resistance.

This review will consider the gut as a reservoir for antimicrobial resistance, colonization resistance, and how disruption of the microbiome can lead to colonization by pathogenic organisms. There is a focus on the gut as a reservoir for β-lactam and plasmid-mediated quinolone resistance. Finally, the role of functional metagenomics and long-read sequencing technologies to detect and understand antimicrobial resistance genes within the gut microbiome is discussed, along with the potential for future microbiome-directed methods to detect and prevent infection.

Imaging blood-brain barrier disruption in neuroinflammation and Alzheimer's disease.

The blood-brain barrier (BBB) is the neurovascular structure that regulates the passage of cells and molecules to and from the central nervous system (CNS). Alzheimer's disease (AD) is a neurodegenerative disorder that is associated with gradual breakdown of the BBB, permitting entry of plasma-derived neurotoxins, inflammatory cells, and microbial pathogens into the CNS. BBB permeability can be visualized directly in AD patients using imaging technologies including dynamic contrast-enhanced and arterial spin labeling magnetic resonance imaging, and recent studies employing these techniques hav

Assemblages of excess and pleasures: The sociosexual uses of online and chemical technologies among men who have sex with men.

Chemicals have penetrated everyday lives of men who have sex with men as never before, along with new online and mobile technologies used to seek pleasures and connections. Poststructuralist (including queer) explorations of these new intensities show how bodies exist in the form of (political) surfaces able to connect with other bodies and with other objects where they may find/create a function (e.g., reproduce or disrupt hegemonies). This federally funded netnographic study explored how a variety of chemicals such as recreational drugs, pharmaceuticals and steroids are contributing to the c

Heat-induced antigen retrieval for immunohistochemical reactions in routinely processed paraffin sections.

The development of heat-induced antigen (epitope) retrieval (HIER) technologies has led to dramatic improvements in our ability to detect antigens in formalin fixed, archival tissues. Paradoxically, wet heat treatment at temperatures greater than 95 degrees C in appropriate buffer solutions can reconstitute the antigenicity of many proteins that have been rendered nonreactive during the fixation and paraffin embedding process, which heretofore could only be identified in fresh or frozen tissues. The reason for this effect is unclear, but it has been suggested that the vigorous heat treatment p

Knockoff: Druggable Cleavage of Membrane Proteins.

Comparative cell biology relies on methods that disrupt protein function. Traditional approaches target the gene that encodes the protein of interest via conventional knockout (KO) methodology, conditional Cre-lox system, or recently, flexible protocols based on CRISPR/Cas9. However, these technologies lack precise temporal control (hours), whereby the slow half-lives of proteins may confound measurements, possibly resulting in misleading phenotypes. Targeting the protein itself bypasses issues pertaining to protein half-life, resulting in more acute disruption. An ideal system would enable co

RNA therapeutics 2.0: Expanding the landscape from mRNA vaccines to splicing modulators and beyond.

RNA therapeutics have progressed into a disruptive drug class quickly, replacing a variety of primary experimental agents which included vaccines, antisense oligonucleotides (ASOs), small interfering RNAs (siRNAs), aptamers and RNA editing systems. First-generation modalities, demonstrated by fomivirsen and pegaptanib were limited by vulnerability to nuclease attack, inefficient delivery and immune stimulation were treated with clinical feasibility. Recent clinical achievements, including mRNA vaccinations against COVID-19, have been based on developments in backbone chemistry, nucleoside modi

Multi-sensor fusion and segmentation for autonomous vehicle multi-object tracking using deep Q networks.

Autonomous vehicles, often known as self-driving cars, have emerged as a disruptive technology with the promise of safer, more efficient, and convenient transportation. The existing works provide achievable results but lack effective solutions, as accumulation on roads can obscure lane markings and traffic signs, making it difficult for the self-driving car to navigate safely. Heavy rain, snow, fog, or dust storms can severely limit the car's sensors' ability to detect obstacles, pedestrians, and other vehicles, which pose potential safety risks. So, we have presented a multi-sensor fusion and
