Rabies, canine distemper, and canine parvovirus exposure in large carnivore communities from two Zambian ecosystems.

Disease transmission within and among wild and domestic carnivores can have significant impacts on populations, particularly for threatened and endangered species. We used serology to evaluate potential exposure to rabies virus, canine distemper virus (CDV), and canine parvovirus (CPV) for populations of African lions (Panthera leo), African wild dogs (Lycaon pictus), and spotted hyenas (Crocuta crocuta) in Zambia's South Luangwa National Park (SLNP) and Liuwa Plain National Park (LPNP) as well as community lands bordering these areas. In addition, domestic dogs in the study region were evalua

Molecular characterisation and histopathological analysis of canine distemper virus in a Malayan tiger (Panthera tigris jacksoni).

IMPORTANCE: Canine distemper virus (CDV) has a broad host range, spanning mammals of the order Carnivora and other orders, often proving fatal. The detection of CDV in a Malayan tiger (Panthera tigris jacksoni) in Malaysia in 2019 marks the first such case diagnosed in Malayan tigers, suggesting possible exposure from domestic animals or wildlife reservoirs. OBJECTIVE: This study aims to describe histological manifestations and characterise the CDV strain in the tiger to determine its molecular epidemiology and postulate viral pathogenicity. METHODS: Histopathology characterisation of the in

Influence of age and vaccination interval on canine parvovirus, distemper virus, and adenovirus serum antibody titers.

Canine core vaccine titer screenings are becoming increasingly popular in veterinary practice as a tool to guide vaccination decisions, despite a lack of supportive, peer-reviewed evidence-based literature. Additionally, it has been suggested that the canine core vaccine duration of host protective immunity can persist past the currently recommended vaccination interval. Thus, this study evaluated serum antibody titers against three core antigens in dogs with known vaccination histories and lifestyles, analyzing the effect of life stage, exposure risk, and time since last vaccination (TSLV). Canine distemper virus induces downregulation of GABAA,GABAB, and GAT1 expression in brain tissue of dogs.

The aim of the study was to determine the expression profiles of GABAA, GABAB, and GAT1 using RT-PCR and the immunoreactivity of GAT1 via immunohistochemical and immunofluorescence assays in CDV-infected brain tissue of dogs. For this purpose, dogs with CDV and dogs without CDV were selected. The mRNA transcript levels of GABAA, GABAB, and GAT1 were significantly downregulated in brain tissue in the CDV-infected group as compared with that in non-CDV-infected brain tissue in the control group (p < 0.01, p < 0.001). In addition, the immunoreactivity of GAT1 in CDV-infected brain tissue was sign

Reactive Oxygen Species Are Key Mediators of Demyelination in Canine Distemper Leukoencephalitis but not in Theiler's Murine Encephalomyelitis.

(1) Background: Canine distemper virus (CDV)-induced demyelinating leukoencephalitis (CDV-DL) in dogs and Theiler's murine encephalomyelitis (TME) virus (TMEV)-induced demyelinating leukomyelitis (TMEV-DL) are virus-induced demyelinating conditions mimicking Multiple Sclerosis (MS). Reactive oxygen species (ROS) can induce the degradation of lipids and nucleic acids to characteristic metabolites such as oxidized lipids, malondialdehyde, and 8-hydroxyguanosine. The hypothesis of this study is that ROS are key effector molecules in the pathogenesis of myelin membrane breakdown in CDV-DL and TMEV

Frequency of exposure of endangered Caspian seals to Canine distemper virus, Leptospira interrogans, and Toxoplasma gondii.

Canine distemper virus (CDV), Leptospira interrogans, and Toxoplasma gondii are potentially lethal pathogens associated with decline in marine mammal populations. The Caspian Sea is home for the endangered Caspian seal (Pusa caspica). In the late 1990s and early 2000s, CDV caused a series of mortality events involving at least several thousand Caspian seals. To assess current infection status in Caspian seals, we surveyed for antibodies to three pathogens with potential to cause mortality in marine mammals. During 2015-2017, we tested serum samples from 36, apparently healthy, Caspian seals, a

Occurrence of filaria in domestic dogs of Samburu pastoralists in Northern Kenya and its associations with canine distemper.

Samples of blood (serum, smears and blood preserved with ethanol) were collected from dogs during a vaccination campaign in northern Kenya in the years 2006 and 2007. Blood was screened for filarial parasites using molecular and microscopy methods and sera were tested for antibodies against canine distemper virus (CDV). Parasitological examination revealed the presence of two species of canine filariae: Acanthocheilonema dracunculoides and A. reconditum. The DNA from the former species was detected in 58% dogs sampled in 2006 and 36% dogs sampled in 2007, whereas the latter was found only in 4

The 2000 canine distemper epidemic in Caspian seals (Phoca caspica): pathology and analysis of contributory factors.

More than 10,000 Caspian seals (Phoca caspica) were reported dead in the Caspian Sea during spring and summer 2000. We performed necropsies and extensive laboratory analyses on 18 seals, as well as examination of the pattern of strandings and variation in weather in recent years, to identify the cause of mortality and potential contributory factors. The monthly stranding rate in 2000 was up to 2.8 times the historic mean. It was preceded by an unusually mild winter, as observed before in mass mortality events of pinnipeds. The primary diagnosis in 11 of 13 seals was canine distemper, character

Multiple sclerosis in the Faroe Islands. IV. The lack of a relationship between canine distemper and the epidemics of MS.

Clinical onset of multiple sclerosis (MS) occurred in 32 native resident Faroese between 1943 and 1973, comprising 3 consecutive epidemics of decreasing frequency. Relationship of MS with the appearance of canine distemper (CD) was explored by serologic studies, questionnaires, and veterinarian reports. Tested were sera from 12 MS patients and 112 controls among the 22 patients and 192 controls with questionnaires in 1978-1979. The daily treatment ledgers of the Veterinarian of the Faroes 1940-1961 were also reviewed and additional Faroese interviewed 1987-1988 as to CD. History of CD was dete

Intracerebral synthesis of antibodies to measles and distemper viruses in patients with subacute sclerosing panencephalitis and multiple sclerosis.

Serum and cerebrospinal fluid (CSF) measles and distemper antibody levels were quantified in a series of twenty patients (four subacute sclerosing panencephalitis (SSPE); ten multiple sclerosis (MS); six non-MS neurological cases) using independent competitive inhibition radioimmunoassays. These results were used in a Tourtellotte calculation to measure the intracerebral IgG synthesis to each virus. The results confirmed that in SSPE there is a greatly enhanced intracerebral measles antibody synthesis (6.0 mg/day). However, it was found that in SSPE this represents only part of a general syste

Region-Specific Apoptosis-Related Gene Expression Is Uncoupled from Viral RNA Load in Canine Distemper Neuropathogenesis.

Canine distemper virus (CDV) is a highly contagious morbillivirus associated with severe neurological disease in dogs. Although apoptosis is recognized as an important mechanism in CDV-associated neurodegeneration, the relationship between regional viral RNA load and apoptosis-related transcriptional responses remains poorly defined. This study aimed to quantify CDV RNA load and the expression of apoptosis-related genes, including the pro-apoptotic markers Bax, caspase-3 and caspase-8 and the anti-apoptotic marker Bcl-2, in the frontal cortex, hippocampus, and cerebellum of 21 dogs naturally i

Fatal Canine Distemper Virus Infections in Free-Ranging Black-Tufted Marmosets (Callithrix penicillata) in Central Brazil.

Canine distemper virus (CDV), a morbillivirus with a broad host range, has increasingly been reported in wildlife, including nonhuman primates. However, data on CDV infection in free-ranging Neotropical primates remain limited. Herein, we describe the pathologic, immunohistochemical, and molecular findings associated with CDV infection in three free-ranging black-tufted marmosets (Callithrix penicillata) from periurban areas of Palmas, Tocantins state, Brazil Between February and March 2023, residents reported sick or dead marmosets with facial cutaneous lesions. Post-vaccinal distemper-like disease in two dog litters with confirmed infection of vaccine virus strain.

Modified live canine distemper virus (CDV) vaccines are widely used and considered both safe and effective. Although there are occasional literature reports of suspected vaccine-induced disease, there are none where the vaccine strain has been identified in affected tissues. Here we describe two such cases in different litters. In litter A, five of ten puppies presented with fever, anorexia, vomiting, and diarrhea a few days post-vaccination. Four puppies died or were euthanized, and autopsy revealed atypical necrosis of the lymphoid tissue. In litter B, two of five puppies developed typical n

Agreement between In-Clinics and Virus Neutralization Tests in Detecting Antibodies against Canine Distemper Virus (CDV).

Core vaccinations and specific antibody titer evaluations are strongly recommended worldwide by all the vaccination guidelines. Virus neutralization (VN) is considered the gold standard for measuring antibody titer against canine distemper virus, but it is complex and time consuming, and the use of in-clinics tests would allow to obtain quicker results. The aim of this study was to evaluate the agreement of the commercial in-clinics VacciCheck test compared to VN. A total of 106 canine sera were analyzed using both methods. The best agreement was obtained using a protective threshold of ≥1:32.

A recombinant canine distemper virus expressing a modified rabies virus glycoprotein induces immune responses in mice.

Canine distemper virus (CDV) and rabies virus (RV) are two important pathogens of the dog. CDV, a member of the morbillivirus genus, has shown promise as an expression vector. The glycoprotein from RV is a main contributor to protective immunity and capable of eliciting the production of virus-neutralizing antibodies. In this study, we recovered an attenuated strain of canine distemper virus and constructed a recombinant virus, rCDV-RV-G, expressing a modified (R333Q) rabies virus glycoprotein (RV-G) of RV Flury strain LEP. RV-G expression by the recombinant viruses was confirmed. Furthermore,

Canine distemper virus and measles virus fusion glycoprotein trimers: partial membrane-proximal ectodomain cleavage enhances function.

The trimeric fusion (F) glycoproteins of morbilliviruses are activated by furin cleavage of the precursor F(0) into the F(1) and F(2) subunits. Here we show that an additional membrane-proximal cleavage occurs and modulates F protein function. We initially observed that the ectodomain of approximately one in three measles virus (MV) F proteins is cleaved proximal to the membrane. Processing occurs after cleavage activation of the precursor F(0) into the F(1) and F(2) subunits, producing F(1a) and F(1b) fragments that are incorporated in viral particles. We also detected the F(1b) fragment, inc

Seroprevalence of canine distemper in free-roaming dogs in buffer zone of Bardiya National Park, Nepal.

Canine distemper (CD), caused by Morbilivirus canis of Paramyxoviridae family, is a contagious and potentially fatal viral disease affecting a wide range of domestic and wild canid species. This study aimed to determine the seroprevalence of Canine Distemper Virus (CDV) and associated risk factors among free-roaming dogs in the buffer zone of Bardiya National Park (BNP), Nepal. A total of 91 serum samples were collected from free-roaming dogs and tested for CDV-specific IgG antibodies using an iELISA kit. The apparent seroprevalence was found to be 52.75% (95% CI: 42.5-63.0%), while the Rogan-

Circulation of arctic canine distemper virus in diarrheic dogs in Iran: molecular detection and phylogenetic characterization.

Canine distemper virus (CDV) is a pathogen that causes systemic infection in dogs and wildlife. Among the highly variable regions in the genome of the virus, the H gene is commonly sequenced for phylogenetic studies. The aim of the present study was to screen for the presence of CDV infection in dogs in Iran and to identify the genotype of the virus. Seventy-five dogs with gastrointestinal symptoms were screened via an immunochromatographic test. Fifty of them were positive. CDV was isolated from rectal swabs, and PCR was performed. The top 10 positive cases were subsequently selected for sequ
