Exploiting metabolic cell death for cancer therapy.

Resistance to cell death is a hallmark of cancer, driving tumour progression and limiting therapeutic efficacy. Metabolic cell death pathways have been identified as unique vulnerabilities in cancer, with ferroptosis being the most extensively studied, alongside the more recently discovered pathways of cuproptosis and disulfidptosis - each triggered by distinct metabolic perturbations. In this Review, we examine the molecular mechanisms and regulatory networks that govern these forms of metabolic cell death in cancer cells. We further examine the potential crosstalk between these pathways and

Disulfidoptosis as a Novel Mechanism of Neuronal Death: Insights from Creutzfeldt-Jakob Disease.

BACKGROUND: Sporadic Creutzfeldt-Jakob Disease (SCJD) is a severe neurodegenerative disorder characterized by rapid progression and extensive neuronal loss. Disulfidptosis is an innovative type of programmed cell demise characterized by an accumulation of disulfide bonds within the cellular cytoplasm, subsequently triggering functional disruption and cell demise. METHODS: Through literature review and analysis, we identified 18 candidate disulfidptosis-related genes (DRGs) involved in cellular processes. The dataset used for analysis, GSE124571, was obtained from the Gene Expression Omnibus d

[Role of programmed cell death in platinum resistance in ovarian cancer].

Ovarian cancer is the most lethal malignancy of the female reproductive system worldwide. Chemoresistance, particularly platinum resistance, is a major factor limiting improvement in prognosis, and its underlying mechanisms involve complex regulation of and escape from multiple programmed cell death pathways in cancer cells. Disulfidptosis is triggered by high expression of solute carrier family 7 member 11 (SLC7A11) under glucose starvation and shares upstream regulatory nodes with ferroptosis. Necrosis by sodium overload is driven by sodium ion overload mediated by transient receptor potenti

A TIGIT nanotrapping-guided STING-activatable immunometabolic strategy overcomes innate immune silence and T cell exhaustion in breast cancer.

Breast cancer exhibits a profoundly immunosuppressive tumor microenvironment (TME), where innate immune silence prevents antigen sensing and persistent T cell exhaustion limits effector responses, rendering most immunotherapies ineffective. Clinical profiling of 1093 The Cancer Genome Atlas (TCGA) cases identified a glucose-fueled glutathione (GSH)-glutathione peroxidase 4 (GPX4)-dihydrolipoamide S-acetyltransferase (DLAT) axis as a dominant metabolic shield that suppresses oxidative stress, and thereby enforces both stimulator of interferon genes (STING) silence and CD8+ T cell exclusion. CO-mediated restoration of mitochondrial redox homeostasis contributes to alleviating multiple programmed cell deaths in hepatic I/R injury.

Hepatic ischemia-reperfusion injury (HIRI) remains a life-threatening complication in liver surgery and transplantation with no clinically approved therapy. Here we develop a hyaluronic acid-modified, ROS-responsive nanoformulation (CPs@HA) that enables controllable delivery of carbon monoxide (CO) specifically to injured liver tissue. We show that mitochondrial dysfunction is closely associated with three distinct cell death pathways (ferroptosis, cuproptosis, and disulfidptosis) that occur in parallel during HIRI. CPs@HA restores mitochondrial homeostasis through activating the AMPK-SIRT1-PG

YTHDC1 Orchestrates Glucose and Glutamate Rewiring to Overcome Lethal Metabolic Stress in Triple-Negative Breast Cancer.

Developing effective therapies for triple-negative breast cancer (TNBC) requires the identification of key molecular regulators. Here, among the major m6A modulators, we find that only the reader YTHDC1 is specifically overexpressed in TNBC and correlates to unfavorable prognosis. Mechanistically, YTHDC1 orchestrates glucose and glutamine metabolism in an m6A-dependent manner to confer robust adaptability to lethal metabolic stress in TNBC cells. It not only stabilizes GLUT3 mRNA to sustain glucose uptake and NADPH production, but also reduces the mRNA stability of ATF4, thereby suppressing th

Triple-Synergistic Therapy with Cobalt-Pheophytin Coordination Micelles for Overcoming Drug Resistance in Pancreatic Cancers.

Pancreatic ductal adenocarcinoma (PDAC) represents one of the deadliest malignancies as KRAS (Kirsten rat sarcoma viral oncogene homolog) mutations continuously activate multiple downstream signaling pathways, leading to drug resistance. To overcome this issue, herein, we developed a cobalt-pheophytin (CoPheo) coordination micelle chelating two chemotherapeutic agents including ONC201 and Palbociclib (Pal), yielding CoPheo-ONC201-Pal-F127. Compared with other metals, cobalt exhibits stronger coordination ability and higher drug-loading efficiency. The resulting ligand bonds are both acid- and

Recent Advances in Natural Product Interventions on Cardiovascular Disease: The Role of Programmed Cell Death.

Cardiovascular diseases (CVDs) remain one of the leading causes of morbidity and mortality worldwide, with cardiomyocyte death playing an important role in myocardial injury, pathological cardiac remodeling, and the progression of heart failure. Programmed cell death (PCD), including apoptosis, pyroptosis, ferroptosis, cuproptosis, and disulfidptosis, plays an essential role in maintaining cellular homeostasis; however, its aberrant activation or dysregulation can contribute to cardiovascular injury and disease progression. Increasing evidence indicates that these PCD pathways are not independ

Exploring regulated cell death in renal cell carcinoma: mechanistic insights and therapeutic perspectives.

Renal cell carcinoma (RCC) is the third most prevalent urological malignancy, characterized by histological heterogeneity and complex molecular landscapes. Although established regimens, including surgical intervention, targeted therapies and immunotherapy, effectively manage early-stage disease, patients with advanced RCC still have a poor prognosis. The expanding spectrum of regulated cell death (RCD), ranging from established apoptosis and ferroptosis to emerging modalities like cuproptosis, disulfidptosis and ammonia-induced cell death, offers novel mechanistic insights into tumor vulnerab

Spatiotemporal microenvironment landscape and malignant epithelial pattern transition in breast ductal carcinoma progression.

BACKGROUND: Owing to the complexity of TME components and the heterogeneity of cancer cells, the relationship between the niches of TME and prognosis in breast ductal carcinoma remains unknown. The staged characteristics of corresponding cancer cell behaviors are unclear. Our study aims to reveal spatial structures and specific cellular information of TME and cancer cells subgroups during the progression from DCIS to IDC and lymph node metastasis. METHODS: Single-cell sequencing, spatial transcriptomics, bulk RNA sequencing datasets were used to explore the changes in microenvironmental compo

Crosstalk of metabolic cell death pathways in colorectal cancer: implications for precision therapy.

Colorectal cancer (CRC) remains one of the leading causes of cancer-related deaths worldwide, with therapeutic resistance being a critical bottleneck limiting patient outcomes. This necessitates novel intervention strategies beyond classical apoptosis paradigms. Metabolically-regulated cell death (particularly ferroptosis and cuproptosis), by targeting the metabolic vulnerabilities of tumor cells, offers new perspectives for overcoming cancer drug resistance. We provide a comprehensive overview of ferroptosis and cuproptosis in CRC, with a focused discussion on their crosstalk, therapeutic str

Programmed cell death: a promising management for Alzheimer's disease.

Epidemiological investigations have indicated that Alzheimer's disease (AD) has emerged as a predominant health challenge among older adults, with its mortality rate rising sharply each year. Nevertheless, measures capable of delaying or halting its clinical progression have remained largely unattained until recent years. Programmed cell death (PCD), defined as a genetically regulated and orderly mode of cellular demise, is pervasive throughout organismal development and essential for maintaining life homeostasis. As research on PCD in AD has expanded, accumulating evidence has shown that auto

Targeting ferroptosis in cancer: from mechanistic insights to therapeutic approaches.

Ferroptosis is a promising programmed cell death modality for cancer therapy, driven by iron overload and the accumulation of phospholipid peroxides that culminate in lethal membrane damage. Over the past decade, emerging evidence supports the concept that ferroptosis can be harnessed as an effective strategy to suppress tumor growth, particularly in therapy-resistant cancer cells undergoing epithelial-mesenchymal transition and in cancer stem cells. Given that ferroptosis is mechanistically and morphologically different from other known programmed cell death forms, increasing critical finding

The atypical E3 ligase HOIL-1 safeguards the ribosome during cellular stress.

The ribosome has emerged as a signalling hub that can sense metabolic perturbations and coordinate responses that either restore homeostasis or initiate cell death. The range of insults that signal via the ribosome and the mechanisms governing such cell fate decisions remain uncharacterized. Here we identify the atypical E3 ligase HOIL-1 as an unexpected node in the ribosome signalling network that resolves cellular stress. We find that truncating HOIL-1 mutations associated with dilated cardiomyopathy exacerbate cardiac dysfunction in mice and broadly sensitize cells to nutrient and translati

Poly(sodium lipoate) Particles with Nitroimidazole Modification for Disulfide Stress-Mediated Antitumor Metastasis.

Disulfidptosis, a form of programmed cell death triggered by disulfide stress, holds promise for antimetastatic strategies by promoting cytoskeletal collapse, while no relevant applied study was reported. Herein, we develop a disulfide stress inducer for antitumor metastasis by nitroimidazole-grafted poly(sodium lipoate) nanoparticles (NI@PSL). Upon thiol-mediated cellular uptake, NI@PSL degraded into dihydrolipoic acid (DHLA) and exposed grafted nitroimidazole (NI) in response to glutathione (GSH). DHLA formed aberrant disulfide bonds with cytoskeletal cysteine thiols. NI moieties were reduce
