Physiological response to drought and genome wide characterization of PYL-PP2C genes in Eucalyptus spp.

Climate change has intensified droughts, affecting Eucalyptus productivity. These species are water-demanding and understanding their ABA-mediated stomatal closure mechanisms is key to improve drought response through PYL and PP2C gene regulation. This study analyzed the physiological responses to drought in two Eucalyptus genotypes (G1 and G2) and characterized PYL and PP2C genes, including expression analysis and Genome-wide structure. G2 exhibited greater physiological stability and more efficient stomatal regulation under drought conditions, whereas G1 showed increased vulnerability. A tot

Physiological responses of tomato callus and regenerated plants under PEG-induced stress during in vitro selection for drought tolerance.

BACKGROUND: Drought stress is a major constraint limiting tomato (Solanum lycopersicum L.) productivity worldwide. In vitro selection using polyethylene glycol (PEG)-induced osmotic stress provides a controlled approach for identifying stress-responsive genotypes. METHODS: This study aimed to select local tomato varieties using an in vitro selection system based on PEG 6000-induced osmotic stress (0-8%). Callus cultures were induced from root explants and grown on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L NAA. Physiological responses were assessed using relative growth rate (RGR), c

From stress signaling to yield stability: physiological and molecular mechanisms of wheat resilience to heat and drought stress.

Wheat resilience depends on coordinated signaling, reproductive protection, and source-sink regulation, providing a framework to breed robust trait combinations that stabilize yield under combined heat and drought. Climate change is increasing the frequency and severity of heat and drought events, posing a major threat to wheat productivity, yield stability, and food security. Because these stresses often coincide in the field, their combined effects can impair growth, reproductive development, grain filling, and final yield more severely than either stress alone. Wheat resilience under such c

Comparative Transcriptomic Analysis of Water-Deficit Responses in Japonica Hybrid Rice 'Dianheyou 615'.

Water deficit severely limits rice productivity. The elite Dian (D1)-type hybrid japonica rice 'Dianheyou 615 (ZH1)' exhibits exceptional drought adaptation in high-altitude rainfed uplands of the Yungui Plateau, yet the underlying molecular mechanisms remain unknown. We compared phenotypic and transcriptomic responses of ZH1 and six other japonica cultivars under well-watered and water-deficit conditions. Water-deficit stress significantly impaired agronomic traits across all cultivars; however, ZH1 uniquely maintained relatively stable flag leaf morphology and seed-setting rate, and displaye

Genome-wide characterization of BiP gene family and functional analysis of VvBiP3 in Vitis vinifera.

Drought stress exacerbated by climate change severely impairs the growth and quality of horticultural crops. Grapevine (Vitis vinifera L.) is an economically vital fruit species, and exploring stress-tolerant genes lays a foundation for its genetic improvement and sustainable cultivation. BiP proteins serve as pivotal endoplasmic reticulum (ER) molecular chaperones and participate widely in plant abiotic stress acclimation, yet the genomic features and drought-associated functions of grapevine BiP family genes remain largely unclear. In this study, three VvBiP members were identified genome-wi

The transcription factor CsPAT1 from tea plant (Camellia sinensis) is involved in drought tolerance by modulating phenylpropanoid biosynthesis.

Tea plants, in particular, leafy cash crops, prefer warm and humid climates. Our previous work identified CsPAT1 as a facilitator of lignin biosynthesis in tea plants. The specific role of CsPAT1 in tea plants' abiotic stress response remains unclear. In this study, we found that the expression of CsPAT1 in tea plants was induced under drought, cold, heat, and ABA treatments. CsPAT1 transgenic Arabidopsis lines displayed enhanced drought tolerance compared with wild-type (WT) controls. The SOD and POD activities, proline content, and expression levels of drought-responsive genes were significa

Screening and Bioinformatic Analysis of Drought-Adaptive-Response Genes in Wild Cerasus humilis.

This study aimed to screen genes associated with drought adaptation in Cerasus humilis and analyze their sequence characteristics and evolutionary relationships. The results could provide a theoretical basis for exploring the molecular basis underlying drought adaptation of Cerasus humilis. Two-year-old wild Cerasus humilis plants were cultivated in a greenhouse, and irrigation was completely withheld for 20 consecutive days. Leaves were sampled and a cDNA library of wild Cerasus humilis was constructed. By utilizing high-throughput screening in yeast combined with mannitol-induced simulated d

Climate-resilient crops: Lessons from xerophytes.

Developing climate-resilient crops is critical for future food security and sustainable agriculture under current climate scenarios. Of specific importance are drought and soil salinity. Tolerance traits to these stresses are highly complex, and the progress in improving crop tolerance is too slow to cope with the growing demand in food production unless a major paradigm shift in crop breeding occurs. In this work, we combined bioinformatics and physiological approaches to compare some of the key traits that may differentiate between xerophytes (naturally drought-tolerant plants) and mesophyte

Physiological-Biochemical Characteristics and Omics Landscape of Drought Tolerance in Chlorella sp. from the Taklamakan Desert of China.

Microalgae endemic to desert ecosystems possess exceptional stress-resistance traits; however, the physiological and molecular drought-adaptation mechanisms of desert-dwelling Chlorella remain largely uncharacterized. Herein, we combined physiological-biochemical assays, ultrastructural observation, transcriptomics and metabolomics to dissect drought responses in Taklamakan-desert-origin Chlorella sp. under polyethylene glycol 6000 (PEG-6000)-simulated mild (10%), moderate (20%), and severe (30%) drought stress, with mechanistic focus on moderate-drought conditions. Moderate drought repressed

Peanut-Colonized Piriformospora indica Enhanced Drought Tolerance by Modulating the Enzymes and Expression of Drought-Related Genes.

Peanut (Arachis hypogaea L.) is an important cash and oil seed crop, mostly distributed in arid and semi-arid areas. In recent years, due to the influence of atmospheric circulation anomalies and other factors, drought has become frequent and increasingly serious in China. This has posed serious challenges to peanut production. The objective of this study was to investigate the potential of the endophytic fungus Piriformospora indica to form a symbiotic relationship with peanut plants and to evaluate the drought tolerance of P. indica-colonized peanut plants subjected to a simulated drought st

H2S is involved in drought-mediated stomatal closure through PLDα1 in Arabidopsis.

PLDα1 promoted H2S production by positively regulating the expression of LCD. Stomatal closure promoted by PLDα1 required the accumulation of H2S under drought stress. Phospholipase Dα1 (PLDα1) acting as one of the signal enzymes can respond to drought stress. It is well known that hydrogen sulfide (H2S) plays an important role in plant responding to biotic or abiotic stress. In this study, the functions and relationship between PLDα1 and H2S in drought stress resistance in Arabidopsis were explored. Our results indicated that drought stress promotes PLDα1 and H2S production by inducing the ex

Genome-wide characterization and drought-responsive expression profiling of plasma membrane intrinsic proteins (PIPs) in Caragana korshinskii.

Plasma membrane intrinsic proteins (PIPs) are an important subfamily of aquaporins that function as membrane channels that facilitate water transport across the plasma membrane in plants. Although the molecular mechanisms of PIPs have been extensively studied in model species, their characteristics and functions remain poorly characterized in Caragana korshinskii. Here, 13 CkPIP genes were identified through genome-wide analysis. CkPIPs were clustered into PIP1 (seven members) and PIP2 (six members) subfamilies. They exhibited conserved physicochemical properties, contained cis-acting elements

Evaluation of Silicon and Proline Application on the Oxidative Machinery in Drought-Stressed Sugar Beet.

Drought stress deleteriously affects growth, development and productivity in plants. So, we examined the silicon effect (2 mmol) and proline (10 mmol) individually or the combination (Si + proline) in alleviating the harmful effect of drought on total phenolic compounds, reactive oxygen species (ROS), chlorophyll concentration and antioxidant enzymes as well as yield parameters of drought-stressed sugar beet plants during 2018/2019 and 2019/2020 seasons. Our findings indicated that the root diameter and length (cm), root and shoot fresh weights (g plant-1) as well as root and sugar yield signi

Exploring the Genetic Basis of Drought Tolerance in Alhagi camelorum: A Comprehensive Transcriptome Study of Osmotic Stress Adaptations.

Alhagi camelorum, a desert shrub known for its impressive drought tolerance, exhibits notable resilience under arid conditions. However, the underlying mechanisms driving its drought resistance remain largely unexplored. This study aims to investigate these mechanisms by exposing A. camelorum to osmotic stress using varying polyethylene glycol (PEG) concentrations (1%, 5%, 10%) in a controlled laboratory setting. Growth analysis revealed significant inhibition and phenotypic changes with increasing PEG levels. Transcriptomic analysis, including differentially expressed gene identification, GO

Identification and functional annotation of drought-induced molecular pathways in sugarcane using suppression subtractive hybridization.

BACKGROUND: Wild ancestors of sugarcane, in particular Saccharum spontaneum, are inherently tolerant to abiotic stress. Interspecific hybrids derived from S. spontaneum possess agronomically desirable traits in addition to their potential for sustaining themselves in drought-stressed conditions. METHODS AND RESULTS: In our study, suppression subtractive hybridisation was used to identify drought-responsive candidate transcripts (DRCTs) under water deficit conditions in AS 04-1687 and TS-GS 2024, which were validated by physiological and molecular expression analysis. The amplicon size of the

Physiological responses and morphological adjustments in provenances of two woodland species in a chronic drought experiment.

Drought-induced tree dieback is impacting forest and woodland ecosystems. One approach to mitigate this is to plant better adapted genotypes of species from warmer and drier climates. This requires understanding drought adaptation both interspecifically and intraspecifically, which we investigated in this common garden experiment. Six genotypes from two related Eucalyptus species, Eucalyptus microcarpa Maiden and Eucalyptus melliodora A.Cunn. ex Schauer, were selected from geographically disjunct locations, five of which encompass the drier ranges of each species. One-year-old saplings were pl

Integrated transcriptomic and biochemical analyses reveal mechanisms of drought adaptation in strawberry (Fragaria × ananassa L.).

Drought is a major abiotic stress that limits strawberry productivity worldwide, yet the underlying physiological and molecular mechanisms of cultivar-specific tolerance remains poorly understood. This study integrated physiological, biochemical, and transcriptomic analyses to dissect drought responses of two contrasting cultivars of Fragaria × ananassa: drought-tolerant 'Kara' and drought-sensitive 'E22'. Plants were exposed to both progressive water-deficit (Def) and PEG-induced osmotic stress under controlled greenhouse conditions. Physiologically, the two cultivars diverged substantially:
