Riluzole (RLZ) is a neuroprotective drug indicated for amyotrophic lateral sclerosis. To examine the feasibility of RLZ for repositioning as an anti-inflammatory bowel disease (IBD) drug, RLZ (2, 5, and 10 mg/kg) was administered orally to rats with colitis induced by 2,4-dinitrobenzenesulfonic acid. Oral RLZ was effective against rat colitis in a dose-dependent manner, which was statistically significant at doses over 5 mg/kg. To address safety issues upon repositioning and further improve anti-colitic effectiveness, RLZ was coupled with salicylic acid (SA) via an azo-bond to yield RLZ-azo-SA

BACKGROUND: Drug repositioning, finding new indications for existing drugs, has gained much recent attention as a potentially efficient and economical strategy for accelerating new therapies into the clinic. Although improvement in the sensitivity of computational drug repositioning methods has identified numerous credible repositioning opportunities, few have been progressed. Arguably the "black box" nature of drug action in a new indication is one of the main blocks to progression, highlighting the need for methods that inform on the broader target mechanism in the disease context.

BACKGROUND: Gastric cancer (GC) requires novel therapies. This study employed artificial neural networks (ANNs) to repurpose existing drugs against GC.
METHODS: ANN models predicted candidates from DrugBank using pharmacogenomic descriptors. Top compounds, selected by favorable IC50 and Z-scores, were screened in vitro (cytotoxicity in AGS/MKN-45 cells; toxicity in HDFs). Leads were validated in vivo in MKN-45 xenograft mice, assessing tumor markers (Ki67, CD44) and target genes.
RESULTS: Two leads, amitriptylinoxide (20313) and phytonadione (5284607), exhibited high in vitro potency and sel

The upregulation of interleukin-1 receptor-associated kinase 4 (IRAK4) drives pro-tumorigenic signaling across various malignancies. Currently available IRAK4 inhibitors are all limited by suboptimal selectivity and off-target toxicity. To develop non-toxic and mechanistically focused IRAK4 inhibitors, an in silico machine learning (ML) pipeline combined with structure-driven drug repositioning was employed. An IRAK4-targeting dataset of bioactive compounds was systematically filtered and curated to train a random forest (RF) regression model. Comparative cross-validation against 41 independen

MOTIVATION: Drug repositioning accelerates clinical translation by identifying new therapeutic indications for approved drugs. However, therapeutic associations in biomolecular networks often exist indirectly, through transitive chains and long-range mechanisms, rather than as directly observed links. Shallow methods are confined to direct similarity and miss such indirect associations, whereas deep graph neural networks suffer from over-smoothing and lose discriminative power in highly connected networks.
RESULTS: We propose a spatial-spectral collaborative framework. In the spatial domain,

Rosacea is a chronic dermatological condition that currently lacks a clear treatment approach due to an uncomprehensive knowledge of its pathogenesis. The main obstacle lies in understanding its etiology and the mode of action of the different drugs used. This study aims to clarify these aspects by employing drug repositioning. Using an in silico approach, we performed a transcriptomic analysis comparing samples from individuals with diverse types of rosacea to those from healthy controls to identify genes deregulated in this disease. Subsequently, we realized molecular docking and molecular d

BACKGROUND: Peripheral nerve injuries negatively impact the quality of life of patients, with no effective treatment available that accelerates sensorimotor recovery and promotes functional improvement and pain relief. The aim of this study was to evaluate the effects of diacerein (DIA) in an experimental mice model of sciatic nerve crush.
METHOD: In this study, male Swiss mice were used, randomly separated into six groups as follows: FO (false-operated + vehicle); FO + DIA (false-operated + diacerein 30 mg/kg); SNI (sciatic nerve injury + vehicle); SNI + DIA in doses of 3, 10 and 30 mg/kg (s

Pathogenic mosquito-borne viruses are a serious public health issue in tropical and subtropical regions and are increasingly becoming a problem in other climate zones. Drug repositioning is a rapid, pharmaco-economic approach that can be used to identify compounds that target these neglected tropical diseases. We have applied a computational drug repositioning method to five mosquito-borne viral infections: dengue virus (DENV), zika virus (ZIKV), West Nile virus (WNV), Japanese encephalitis virus (JEV) and Chikungunya virus (CHIV). We identified signature molecules and pathways for each virus

INTRODUCTION: Triple-Negative Breast Cancer (TNBC) lacks targeted therapies and is mainly treated with chemotherapeutics limited by toxicity and resistance. This study aims to identify and characterize cardiac steroids as potential anti-TNBC leads using phenotypic screening, drug-likeness evaluation, and in silico targetprofiling.
METHODS: A panel of natural products was screened against TNBC MDA-MB-231 cells using MTT assay and compared with conventional chemotherapeutics; selectivity indices were calculated using WI-38 fibroblasts. SwissADME was used to predict physiochemical and pharmacoki

Drug repositioning can accelerate therapeutic discovery, but computational drug-disease association prediction remains constrained by noisy multisource similarities, heterogeneous biomedical semantics, and sparse supervision. We present MVGCL, a type-aware dual-pathway framework that combines noise-robust multisource similarity fusion with heterogeneous biological network representation learning and aligns the two pathways through a distribution-aware hierarchical contrastive strategy to improve representation consistency under sparse and noisy supervision. Under 10-fold cross-validation proto

Esophageal squamous cell carcinoma (ESCC) is highly associated with cancer cachexia, a wasting syndrome lacking effective treatments. Existing animal models fail to capture key clinical and biological features of this condition. Here, we established a panel of patient-derived organoid xenograft (PDOX) models that authentically replicate the heterogeneity of ESCC-associated cachexia in immunodeficient mice. PDOX lines exhibited slow tumor growth compared with traditional ESCC xenografts. Heterogeneous cachexia phenotypes in PDOX-bearing mice, as compared with non-tumor-bearing mice, including b

Gaucher disease is a rare lysosomal storage disorder characterized by lipid accumulation. Emerging evidence suggests that patients with gaucher disease are at an increased risk of developing melanoma, yet the shared molecular mechanisms remain unclear. This study aimed to investigate and validate potential biomarkers commonly expressed in gaucher disease and melanoma through an integrated approach combining bioinformatics, in silico, and in vitro analyses. Gene expression profiles of gaucher disease and melanoma were downloaded from the Gene Expression Omnibus (GEO). Webgestalt was used to ide

Chagas disease is caused by the protozoan parasite Trypanosoma cruzi and affects 7 million people worldwide. Considering the side effects and drug resistance shown by current treatments, the development of new anti-Chagas therapies is an urgent need. T. cruzi hypoxanthine phosphoribosyltransferase (TcHPRT), the key enzyme of the purine salvage pathway, is essential for the survival of trypanosomatids. Previously, we assessed the inhibitory effect of different bisphosphonates (BPs), HPRT substrate analogues, on the activity of the isolated enzyme. BPs are used as a treatment for bone diseases a

Target-based design and repositioning are mainstream strategies of drug discovery. Numerous drug design and repositioning projects have been launched to fight the ongoing COVID-19 pandemic. The resulting drug candidates have often failed due to the misprediction of their target-bound structures. The determination of water positions of such structures is particularly challenging due to the large number of possible drugs and the diversity of their hydration patterns. To answer this challenge and help correct predictions, we introduce a new protocol HydroDock, which can build hydrated drug-target

Parkinson's disease (PD) is among the most prevalent neurodegenerative diseases. Available evidences support the view of PD as a complex disease, being the outcome of interactions between genetic and environmental factors. In face of diagnosis and therapy challenges, and the elusive PD etiology, the use of alternative methodological approaches for the elucidation of the disease pathophysiological mechanisms and proposal of novel potential therapeutic interventions has become increasingly necessary. In the present study, we first reconstructed the transcriptional regulatory networks (TN), cente

Objectives Malignant pleural mesothelioma (MPM) is an occupational disease mainly due to asbestos exposure. Effective therapies for MPM are lacking, making this tumour type a fatal disease. Materials and Methods In order to meet this need and in view of a future "drug repositioning" approach, here we screened five MPM (Mero-14, Mero-25, IST-Mes2, NCI-H28 and MSTO-211H) and one SV40-immortalized mesothelial cell line (MeT-5A) as a non-malignant model, with a library of 1170 FDA-approved drugs. Results Among several potential compounds, we found that fludarabine (F-araA) and, to a lesser extent,
