Evaluating Neuropsychiatric Disease in Mouse Models of Systemic Lupus Erythematosus.

Primary neuropsychiatric manifestations in systemic lupus erythematosus (NPSLE) affect 20% to 40% of patients, significantly impacting quality of life and patient prognosis. However, the complex pathophysiology underlying these manifestations remains incompletely understood, partly due to challenges in distinguishing direct disease effects from secondary complications. Animal models are essential for investigating these mechanisms, but their effective utilization requires standardized behavioral assessment protocols, an area where the field currently lacks sufficient consensus. Investigating the behavioral impact of tropical almond (Terminalia catappa L.) consumption: Anxiolytic effects and memory preservation in aged rats.

Terminalia catappa L. (TC) has been traditionally used for its medicinal properties, including antioxidant and neuroprotective effects. Considering the increased vulnerability to cognitive decline and anxiety in aging, we investigated the potential impact of TC on anxiety-like behavior, memory, and oxidative stress in aged Wistar rats. Male Wistar rats were used, including young adults (70 days old) and aged animals (17 months old). The aged rats were allocated into three groups: control (n = 11), TC500 (n = 12), and TC1000 (n = 12). TC was administered by oral gavage at doses of 500 mg/kg or

Adolescent mice show anxiety- and aggressive-like behavior and the reduction of long-term potentiation in mossy fiber-CA3 synapses after neonatal maternal separation.

Exposure to maternal separation (MS) during early life is an identified risk factor for emotional disorders such as anxiety and depression later in life. This study investigated the effects of neonatal MS on the behavior and long-term potentiation (LTP) as well as basic synaptic transmission at hippocampal CA3-CA1 and mossy fiber (MF)-CA3 synapses in adolescent mice for 19days. When mice were adolescents, we measured depression, learning, memory, anxious and aggressive behavior using the forced swimming test (FST), Y-maze, Morris water maze (MWM), elevated plus maze (EPM), three consecutive da

Rapamycin Improves Recognition Memory and Normalizes Amino-Acids and Amines Levels in the Hippocampal Dentate Gyrus in Adult Rats Exposed to Ethanol during the Neonatal Period.

The mammalian target of rapamycin (mTOR), a serine/ threonine kinase, is implicated in synaptic plasticity by controlling protein synthesis. Research suggests that ethanol exposure during pregnancy alters the mTOR signaling pathway in the fetal hippocampus. Thus, we investigated the influence of pre-treatment with rapamycin, an mTORC1 inhibitor, on the development of recognition memory deficits in adult rats that were neonatally exposed to ethanol. In the study, male and female rat pups received ethanol (5 g/kg/day) by intragastric intubation at postanatal day (PND 4-9), an equivalent to the t

Indulging Curiosity: Preliminary Evidence of an Anxiolytic-like Effect of Castor Oil and Ricinoleic Acid.

In the process of validating the elevated zero maze, a common test of anxiety-like behavior, in our laboratory, we demonstrated an anxiolytic-like effect of castor oil and its primary component, ricinoleic acid. We tested the effects of vehicle and chlordiazepoxide in male mice in the elevated zero maze following a 30-min pretreatment time. Chlordiazepoxide is a United States Food and Drug Administration-approved drug that was previously shown to exert anxiolytic-like effects in both the elevated zero maze and elevated plus maze. Chlordiazepoxide was administered at doses of 5 or 10 mg/kg. We

Intermittent fasting promotes anxiolytic-like effects unrelated to synaptic mitochondrial function and BDNF support.

Anxiety disorders are linked to mitochondrial dysfunction and decreased neurotrophic support. Since anxiolytic drugs target mitochondria, non-pharmacological approaches to improve mitochondrial metabolism such as intermittent fasting (IF) may cause parallel behavioral benefits against anxiety disorders. Here, we investigated whether a chronic IF regimen could induce anxiolytic-like effects concomitantly to modulation in mitochondrial bioenergetics and trophic signaling in mice brain. A total of 44 Male C57BL/6 J mice (180 days old) were assigned to two dietary regimens: a normal, ad libitum di

Chlordiazepoxide reduces the generalised anxiety, but not the direct responses, of rats exposed to cat odor.

Rats were treated for 5 days with vehicle or chlordiazepoxide (CDP, 5, 10, or 20 mg/kg/day) and then exposed to a cloth impregnated with a neutral or cat odor. Those exposed to cat odor made significantly fewer contacts with the cloth and spent less time in contact with it and more time sheltering than those exposed to the neutral odor. CDP (5 mg/kg) significantly increased the time in contact with both odor cloths, but there were no other significant effects in the cat odor group. Rats previously exposed to cat odor showed significant anxiogenic responses in the social interaction and elevate

Effect of N-acetyl cysteine against aluminium-induced cognitive dysfunction and oxidative damage in rats.

Aluminium is a potent neurotoxin involved in the initiation and progression of various cognitive disorders like Alzheimer's disease. Chronic aluminium exposure induces oxidative stress and increases amyloid beta levels in vivo. The role of oxidative stress has been well-suggested in these cognitive problems. Therefore, the present study was designed to explore the possible role of N-acetyl cysteine against aluminium mediating cognitive dysfunction and oxidative stress in rats. Aluminium chloride (100 mg/kg, p.o.) was given to rats daily for 6 weeks. N-acetyl cysteine (per se; 50 and 100 mg/kg,

Kefir peptides exhibit antidepressant-like activity in mice through the BDNF/TrkB pathway.

Depression is a prevalent, stress-related mental disorder that can lead to serious psychiatric diseases with morbidity and high mortality. Although some functional fermented dairy drinks have promising anxiolytic and antidepressant effects, the mechanism is still not clear. To determine the antidepressant-like effect and the potential molecule mechanism of kefir peptides (KP), various behavioral tests, including the elevated plus maze test, open field test, forced swimming test, and tail suspension test, were used. Administration of 150 mg/kg KP in mice reduced the duration of immobility in th

The role of D2 dopamine receptors in oxytocin induced place preference and anxiolytic effect.

Neuropeptide oxytocin (OT) is involved in the regulation of social and non-social behaviour. The central nucleus of amygdala (CeA), part of the limbic system, plays an important role in learning, memory, anxiety and reinforcing mechanisms. CeA has been shown to be rich in OT receptors in rodents. Our previous findings indicated that OT in the rat CeA has a dose dependent rewarding and anxiolytic effect. The aim of our present study was to examine in the CeA the possible interaction of OT and D2 dopamine (DA) receptor antagonist Sulpiride on reinforcement in place preference test and on anxiety

The behavioral change of locomotor activity in a kaolin-induced hydrocephalus rat model: evaluation of the effect on the dopaminergic system with progressive ventricle dilatation.

Hydrocephalus is a pathological enlargement of the cerebral ventricle that results from an obstruction of the space containing cerebrospinal fluid (CSF) in the brain. Motor abnormalities, such as abnormal gait and posture, are frequently seen in patients with hydrocephalus. The present study was designed to investigate locomotor activity in the elevated plus maze behaviorally. Hydrocephalus was induced in Sprague-Dawley rats by injection of 0.1 ml of 20% kaolin solution into the cisterna magna (n=14). Control rats received the same volume of saline (n=12). The rats were sacrificed at 3 days an
