Effects of atrazine and endosulfan sulphate on the ecdysteroid system of Daphnia magna.

The ecdysteroid system is used by crustaceans and other arthropods as the major endocrine signalling molecules, regulating processes such as molting and embryonic development. The aim of this study was to evaluate the ecdysteroid activity of two pesticides (atrazine and endosulfan sulphate), with distinct modes of action and which act in the juvenoid system of the crustacean Daphnia magna as weak juvenoid compounds. To assess the ecdysteroid activity, we first exposed maternal daphnids and embryos to nominal concentrations of the pesticides and determined the effect promoted by these pesticide

Ontogeny of the partial secondary wall of the otoccipital region of the endocranium in prehatching Alligator mississippiensis (Archosauria, Crocodylia).

The ontogeny of the posterior otic and anterior occipital portions of the neural endocranium of prehatching Alligator mississippiensis was investigated by reconstruction from sectioned material. In Stage 6 of this species, in which the endochondral ossification of the otoccipital region of the neural endocranium is only in its very early stage, two bony outgrowths-laminae-are present at the external wall of the posterior portion of the neural endocranium. The anterior lamina arises from the external surface of the basal plate at the level of the posterior margin of the subcapsular process; the

Postparietal and prehatching ontogeny of the supraoccipital in Alligator mississippiensis (Archosauria, Crocodylia).

The first record of the postparietal bone of Alligator mississippiensis, documented by transverse histological sections, is presented. It is the first evidence of the presence of this bone within Recent reptiles. The postparietal is present in a specimen with a head length of 32.3 mm. The bone is a small dermal plate lying ventrally and posteriorly to the posterior margin of the parietal and dorsally to the trabecular bone, forming a dorsal surface of the supraoccipital portion of the neural endocranium. The trabecular bone develops perichondrally from the dorsal surface of the tectal cartilag

Size-dependent pigmentation-pattern formation in embryos of Alligator mississippiensis: time of initiation of pattern generation mechanism.

The pigmentation pattern of Alligator mississippiensis was examined. The number of white stripes on the dorsal side of embryos (stages 21-28) and hatchlings from eggs incubated at 30 degrees C (100% females) and 33 degrees C (100% males) was recorded. Total length, nape-rump length and tail length were recorded for each embryo and hatchling. The number of white stripes was affected by incubation temperature but not sex; hatchlings incubated at 33 degrees C had two more white stripes than those at 30 degrees C, despite being the same length. Five female hatchlings produced at 33 degrees C by ma

Triclabendazole sulfoxide causes stage-dependent embryolethality in zebrafish and mouse in vitro.

BACKGROUND: Fascioliasis and paragonimiasis are widespread foodborne trematode diseases, affecting millions of people in more than 75 countries. The treatment of choice for these parasitic diseases is based on triclabendazole, a benzimidazole derivative which has been suggested as a promising drug to treat pregnant women and children. However, at the moment, this drug is not approved for human use in most countries. Its potential adverse effects on embryonic development have been scarcely studied, and it has not been assigned a pregnancy category by the FDA. Thus, to help in the process of ris

Thermal responses and climate change implications of spring and autumn spawning Patagonian squid (Doryteuthis gahi) embryos.

Ocean warming affects ectotherm physiological and phenological processes, potentially creating mismatches between early life stages and their prey. Seasonal spawning cohorts are thought to provide flexibility in responding to environmental variability, but if there is seasonal adaptation between these cohorts, then they may respond to ocean warming differently, affecting species resilience and potentially impacting the wider food web. We tested the response to warming of egg masses and paralarvae from two spawning cohorts (autumn and spring) of the Patagonian squid (Doryteuthis gahi). Treated

Combined effects of incubation temperature and lipopolysaccharide exposure on immune response modulation in the quail gut.

Embryogenesis is a critical programming window in birds, during which environmental factors can reshape gene expression and influence adaptive capacity after hatching. However, little is known about how embryonic thermal manipulation modulates intestinal immune and metabolic responses to inflammatory challenge later in life. This study evaluated the effects of embryonic thermal manipulation on the intestinal transcriptomic response of Japanese quail (Coturnix coturnix japonica) challenged with lipopolysaccharide (LPS). A 2 × 2 factorial design was applied, including two incubation temperatures

Ferroptosis-driven neurobehavioral impairment in zebrafish embryos: Mechanistic links to developmental dibutyl phthalate exposure.

Dibutyl phthalate (DBP), a ubiquitous chemical contaminant detected in various environmental matrices, poses growing concerns regarding its ecotoxicity and public health risks. In this study, zebrafish (Danio rerio) model was used to investigate the ferroptosis-mediated neurotoxicity elicited by iron dyshomeostasis and lipid peroxidation, upon developmental DBP exposure. DBP-induced ferroptosis compromised zebrafish embryogenesis, leading to reduced tail bent rate and heart rate, as well as elevated mortality and deformity. Larval neurobehavioral deficits, assessed through spontaneous locomoti

Evaluating the toxicity of PEGylated multiwalled carbon nanotubes in zebrafish embryos: insights for biomedical and environmental safety.

Polyethylene glycol-Multiwalled carbon nanotubes (PEG-MWCNTs) hold significant potential for biomedical applications, including diagnostics and controlled drug delivery. However, their toxicity allied to the synthesis process remains a critical concern. Residual metal impurities from the synthesis process are suggested as potential contributors to the observed toxicity. This study evaluates the cellular, genomic, and morphological toxicity of PEG-MWCNTs in zebrafish embryos. MWCNTs were synthesized via chemical vapor deposition, purified with nitric acid, and functionalized with PEG-6000. They

A fast in situ hybridization chain reaction method in Drosophila embryos and ovaries.

The in situ hybridization chain reaction (isHCR) is a powerful method for visualizing mRNA in many species. We present a rapid isHCR method for Drosophila embryos and ovaries. Ethylene carbonate was added to the hybridization buffer to facilitate the hybridization reaction, and a modified short hairpin DNA was used in the amplification reaction; these modifications decreased the RNA staining time from 3 days to 1 day. This method is compatible with immunohistochemistry and can detect multiple mRNAs. The proposed method could significantly reduce staining time for Drosophila researchers using i

Descriptive embryological insights of the colorectum of quail embryos with concern to its functional morphology.

BACKGROUND: Quail is an interesting emerging bird species gaining attention in developmental embryology research due to its small size, quick lifespan, and fast growth rate. These characteristics make quail an ideal model for examining the development of the gastrointestinal tract. Consequently, the embryonic development of the colorectum was conducted to provide a comprehensive understanding of its functions in digestion, absorption, and immunity. METHODOLOGY: The morphological anatomy and microscopical structure of the colorectal wall of 74 embryos were studied using light and scanning elec

Early ontogenesis from embryo to juvenile in Senegalese sole Solea senegalensis under laboratory conditions.

Senegalese sole, Solea senegalensis, is a flatfish of high commercial value in the world. It has been identified as an interesting and promising species for marine commercial aquaculture diversification in Europe for at least four decades and was introduced to China in 2003. Early ontogenesis from embryo to juvenile stages in S. senegalensis was analysed under controlled laboratory conditions to provide morphological information for aquaculture. From 0 to 59 days post hatching (dph), 10-20 larvae were sampled and measured each day (0-17 dph) or every 2-6 days (17-59 dph). Morphological charact

Time-course transcriptome analysis discloses PFDMO2OA (C8 HFPO-TA)-induced developmental malformations and cardiovascular toxicities in zebrafish.

PFDMO2OA (C8 HFPO-TA), a novel substitute for perfluorooctanoic acid (PFOA), has been frequently detected in surface waters. However, information on its toxicity remains scarce. In the present study, zebrafish embryos were exposed to varying concentrations of PFDMO2OA, ranging from 80 to 800 mg/L, until 120 h post-fertilization (hpf) to explore its potential developmental toxicities. The LC50 value for mortality was 505.9 mg/L, comparable to that of PFOA (over 500 mg/L), suggesting a lack of safety of PFDMO2OA compared to PFOA. At 120 hpf, PFDMO2OA exposure led to various malformations in embr

Embryonic Exposure to Organophosphate Flame Retardants (OPFRs) Differentially Induces Cardiotoxicity in Rare Minnow (Gobiocypris rarus).

Organophosphorus flame retardants (OPFRs) such as triphenyl phosphate (TPHP) and tris(1,3-dichloro-2-propyl) phosphate (TDCIPP) were reported to impair cardiac function in fish. However, limited information is available regarding their cardiotoxic mechanisms. Using rare minnow (Gobiocypris rarus) as a model, we found that both TPHP and TDCIPP exposures decreased heart rate at 96 h postfertilization (hpf) in embryos. Atropine (an mAChR antagonist) can significantly attenuate the bradycardia caused by TPHP, but only marginally attenuated in TDCIPP treatment, suggesting that TDCIPP-induced bradyc

Improved methodology for fixation and preparation of Aedes aegypti embryos.

The yellow fever mosquito Aedes aegypti is a major disease vector and an increasingly popular emerging model research organism. We present here an improved protocol for the collection, fixation, and preparation of A. aegypti embryos for immunohistochemical and in situ hybridization studies. The processing of A. aegypti embryos for such studies is complicated by the inability to easily remove the vitelline membrane, which prevents the reagents needed for staining from reaching their targets, and which furthermore obscures visualization of the embryo since the membrane is highly sclerotized. Metabolomics analysis of the potential toxicological mechanisms of diquat dibromide herbicide in adult zebrafish (Danio rerio) liver.

Although diquat is a widely used water-soluble herbicide in the world, its sublethal adverse effects to fish have not been well characterised. In this study, histopathological examination and biochemical assays were applied to assess hepatotoxicity and combined with gas chromatography-mass spectrometry (GC-MS)-based metabolomics analysis to reveal overall metabolic mechanisms in the liver of zebrafish (Danio rerio) after diquat exposure at concentrations of 0.34 and 1.69 mg·L-1 for 21 days. Results indicated that 1.69 mg·L-1 diquat exposure caused cellular vacuolisation and degeneration with n

Avermectin induces cardiac toxicity in early embryonic stage of zebrafish.

Avermectin is a widely used insecticide, and it is mainly effective against animal parasites and insects. Given its extensive use in agriculture, a large amount of avermectin is accumulated in natural waters. Avermectin is a neurotoxin that affects the autonomous behavior of zebrafish and inhibits neurological responses in invertebrates via GABA-chloride channels. In this study, we used zebrafish as a model organism to explore the lethal teratogenic effects of different avermectin concentrations. We found that 50-μg/L avermectin could cause significant malformation abnormalities during the dev

Pyridaben induced cardiotoxicity during the looping stages of zebrafish (Danio rerio) embryos.

Pyridaben is a widely used acaricide in agriculture and reaches a high concentration (97 μg/L) in paddy water for a short time when pyridaben was applied to rice. However, its toxicity to aquatic organisms is still poorly understood. Therefore, we assessed the pyridaben cardiotoxicity to aquatic organisms using the zebrafish (Danio rerio) model. We found that pyridaben is highly toxic to aquatic organisms, and LC50 of pyridaben for zebrafish at 72 hpf was 100.6 μg/L. Pyridaben caused severe cardiac malformations and functional abnormalities. Morphologic abnormity included severe pericardial ed

Growing up gator: a proteomic perspective on cardiac maturation in an oviparous reptile, the American alligator (Alligator mississippiensis).

We recently described lasting changes in the cardiac proteome of American alligators (Alligator mississippiensis) reared under hypoxic conditions, that resemble what embryos encounter in natural nests. While these changes were consistent with functional differences in cardiac performance induced by developmental hypoxia, the magnitude of this response was dwarfed by a much greater effect of development alone (76% of the total differentially abundant proteins). This means that substantial differences in relative steady-state protein expression occur in the hearts of alligators as they mature fr
