Chromosome breakage by pairs of closely linked transposable elements of the Ac-Ds family in maize.

Chromosome breaks and hence chromosomal rearrangements often occur in maize stocks harboring transposable elements (TEs), yet it is not clear what types of TE structures promote breakage. We have shown previously that chromosomes containing a complex transposon structure consisting of an Ac (Activator) element closely linked in direct orientation to a terminally deleted or fractured Ac (fAc) element have a strong tendency to break during endosperm development. Here we show that pairs of closely linked transposons with intact ends, either two Ac elements--a common product of Ac transposition--o

Toward unveiling transcriptome dynamics and regulatory modules at the maternal/filial interface of developing maize kernel.

The basal region of maize (Zea mays) kernels, which includes the pedicel, placenta-chalazal, and basal endosperm transfer layers, serves as the maternal/filial interface for nutrient transfer from the mother plant to the developing seed. However, transcriptome dynamics of this maternal/filial interface remain largely unexplored. To address this gap, we conducted high-temporal-resolution RNA sequencing of the basal and upper kernel regions between 4 and 32 days after pollination and deeply analyzed transcriptome dynamics of the maternal/filial interface. Solid-state NMR spectroscopy reveals polishing-induced supramolecular disorder in sake rice starch.

While the brewing suitability of sake rice is influenced by its structural features, the molecular basis of its starchy core (shinpaku) remains unresolved. Here, Yamadanishiki, a widely used sake rice cultivar, polished to 40% was characterized using solid-state nuclear magnetic resonance (NMR) spectroscopy, scanning electron microscopy (SEM), gas-chromatography-mass spectrometry (GC-MS), and amylopectin chain-length analysis. Solid-state 13C NMR revealed progressive disruption of A-type crystalline order with polishing, particularly in shinpaku, whereas the amylopectin backbone remains unchan

Using Synchrotron-Based Approaches To Examine the Foliar Application of ZnSO4 and ZnO Nanoparticles for Field-Grown Winter Wheat.

The effects of foliar-applied ZnO nanoparticles (ZnO NPs) and ZnSO4 on the winter wheat ( Triticum aestivum L.) grain yield and grain quality were studied under field conditions, with the distribution and speciation of Zn within the grain examined using synchrotron-based X-ray fluorescence microscopy and X-ray absorption spectroscopy. Although neither of the two Zn compounds improved the grain yield or quality, both increased the grain Zn concentration (average increments were 5 and 10 mg/kg for ZnSO4 and ZnO NP treatments, respectively). Across all treatments, this Zn was mainly located withi

Autoantibodies from a patient with scleroderma CREST recognized kinetochores of the higher plant Haemanthus.

Human autoantibodies from a patient with scleroderma CREST (calcinosis, Raynaud phenomenon, esophageal dismotility, sclerodactyly, telangiectasia) were used to immunostain kinetochores on chromosomes in endosperm of the seed of the monocot Haemanthus katherinae Bak. Kinetochores of mitotic chromosomes and prekinetochores of interphase cells were specifically stained using conventional indirect immunofluorescence procedures as well as a nonfading immunogold-silver-enhanced technique and analyzed by fluorescence and video microscopy. In interphase, prekinetochores were either single or double st

Elevated vitamin E content improves all-trans β-carotene accumulation and stability in biofortified sorghum.

Micronutrient deficiencies are common in locales where people must rely upon sorghum as their staple diet. Sorghum grain is seriously deficient in provitamin A (β-carotene) and in the bioavailability of iron and zinc. Biofortification is a process to improve crops for one or more micronutrient deficiencies. We have developed sorghum with increased β-carotene accumulation that will alleviate vitamin A deficiency among people who rely on sorghum as their dietary staple. However, subsequent β-carotene instability during storage negatively affects the full utilization of this essential micronutrie

The hub transcription factor ZmbZIP29 controls the speed of seed germination by promoting starch degradation within the maize embryo.

Starch serves as a crucial energy source for seed germination. The classic starch degradation model focuses primarily on the endosperm during late germination, but little is known about starch mobilization within the embryo during early germination stages. Using transcriptomics and weighted gene co-expression network analysis, we identified ZmbZIP29 as a hub transcription factor that regulates early germination. ZmbZIP29 transcripts accumulate primarily in the aleurone layer, scutellum, radicle, and coleoptile during early germination. Our findings indicated that the maize embryo uses its loca

Stratification overcomes ABA-mediated seed dormancy by uncoupling RGL2/ABI5 inhibition from α-amylase expression.

Cold-pretreatment (stratification) is widely employed to overcome seed dormancy and enhance germination, however, its underlying molecular mechanisms remain elusive. We tested the hypothesis that stratification alleviates the repression of α-amylase expression, a critical step for endosperm starch mobilization during germination. Stratification at 4 °C effectively overcame dormancy in wild-type Arabidopsis thaliana, the ABA-catabolism mutant cyp707a2, the GA-insensitive mutant sleepy1, and the cyp707a2 sleepy1 double mutant, but failed to rescue the dormancy phenotype of the GA-biosynthesis mu

Physicochemical, microbiological, and microstructural changes in germinated wheat grain.

This study investigates the physicochemical, microbiological, and microstructural changes in soft wheat grain during germination under varying moisture conditions: moderately dry, moist, and wet. Pre-harvest sprouting can severely compromise grain quality and usability; however, understanding germination-induced changes offers insights into potential utilization strategies. Physical parameters-including thousand-kernel weight, test weight, and falling number-showed strong correlation with germination time, decreasing by 8.2%, 22%, and 74%, respectively. The Effects of Photoperiodic Transcription Factor OsPRR37 on Grain Filling and Starch Synthesis During Rice Caryopsis Development.

Grain filling governs grain weight formation in rice, while starch biosynthesis during this process critically determines both grain quality and yield. In this study, we characterized the heading date regulator OsPRR37 on regulating grain development, starch metabolism, and starch physicochemical properties. The osprr37 mutants exhibited undesirable agronomic traits, including reduced plant height, decreased grain thickness, lower 1000-grain weight, and diminished yield. Moreover, mutant endosperm displayed irregular starch packing, aberrant granules morphology, and decreased granule diameter.

Senescence-induced expression of cytokinin reverses pistil abortion during maize flower development.

Maize is a monoecious species that produces imperfect (unisexual), highly derived flowers called florets. Within the spikelet, the basic repeating unit of the maize inflorescence, the spikelet meristem gives rise to an upper and a lower floret. Although initially bisexual, floret unisexuality is established through selective organ elimination. In addition, the lower floret of each ear spikelet is aborted early in its development, leaving the upper floret to mature as the only pistillate floret. Expression from the cytokinin-synthesizing isopentenyl transferase (IPT) enzyme under the control of

ZINC FINGER PROTEIN 1 and 8 interact with polycomb repressive complex 2 to repress class B and C floral organ identity genes.

Precise control of gene expression is essential for establishing and maintaining cell identities in response to endogenous signals and environmental cues in multicellular organisms. An example is the regulation of the class B and C floral organ identity genes-APETALA3 (AP3), PISTILLATA (PI), and AGAMOUS (AG)-which are repressed in leaves and activated in floral meristems. Their repression in leaves is associated with CURLY LEAF (CLF), a core component of the polycomb repressive complex 2 (PRC2). However, since CLF is expressed in both leaf and floral primordia, its target specificity is likely

The splicing factor SPR2 interacts with the RNA-binding protein PPR8 to facilitate mitochondrial intron splicing and is essential for maize seed development.

Intron splicing plays a crucial role in organellar gene expression in plants, yet its mechanism remains poorly understood. We previously reported a small PPR protein, SPR2, which is a general splicing factor in the splicing of over half of the mitochondrial introns via interaction with PPR-SMR1, a small MutS-related domain protein. To gain insight into the mechanism, we identified SPR2-interacting proteins using immunoprecipitation and mass spectrometry (IP-MS). One of the proteins is a P-type PPR protein with unknown function, named PPR8. PPR8 is targeted to mitochondria, and protein interact

Mass spectrometry-based quantification of immunostimulatory gliadin proteins and peptides in coloured wheat varieties: Implications for Celiac Disease.

Pigmented wheat varieties (Triticum aestivum spp.) are getting increasingly popular in modern nutrition and thoroughly researched for their functional and nutraceutical value. The colour of these wheat grains is caused by the expression of natural pigments, including carotenoids and anthocyanins, that can be restricted to either the endosperm, pericarp and/or aleurone layers. While contrasts in phytochemical synthesis give rise to variations among purple, blue, dark and yellow grain's antioxidant and radical scavenging capacities, little is known about their influence on gluten proteins expres

Identification of proteinaceous inhibitors of a cysteine proteinase (an Arg-specific gingipain) from Porphyromonas gingivalis in rice grain, using targeted-proteomics approaches.

Porphyromonas gingivalis is known to be a major etiologic agent in the onset and progression of chronic periodontitis. Among various virulence factors that this bacterium produces, Arg- and Lys-specific cysteine proteinases (gingipains) are believed to be major determinants of the pathogenicity of P. gingivalis. Here, we report on our finding that there are inhibitors of these cysteine proteinases in a rice protein fraction. Comprehensive affinity chromatography and MS analyses resulted in the identification of 17 Arg-gingipain (Rgp)-interacting proteins in the rice endosperm. Of these, four p

[Comparative toxicity of different cereals for subjects intolerant of gluten].

Coeliac disease is caused by prolamines, the storage proteins of some cereals, located in the endosperm. Cereals do not all have the same toxicity. The four wheat prolamine groups (alpha, beta, gamma and omega gliadins), visible in electrophoresis at acid pH, have been isolated and their toxicities compared by observing the morphological changes in intestinal biopsies cultured in vitro when peptic-tryptic digests of the studied proteins were added to the culture medium. The toxicity was found to be mainly located in the alpha and beta-gliadins and in peptides of 5 to 10 000 molecular weight. P

ARABIDOPSIS Bsister and SEEDSTICK MADS-box transcription factors modulate maternal nutrient flow for seed development in Arabidopsis.

Successful seed development in Angiosperms depends on the coordinated transport and allocation of sugars from maternal tissues to the developing embryo and endosperm. In Arabidopsis thaliana, ovules function as carbohydrate sink organs, accumulating starch in both gametophytic and sporophytic domains prior to fertilization. During early embryogenesis, the stored starch is then mobilized. Despite extensive knowledge of starch metabolism in photosynthetic tissues, the regulatory mechanisms governing sugar transport in reproductive organs remain poorly understood. Recent studies have identified f

Parent-of-origin specific allelic expression in outbreeding Arabidopsis arenosa identifies antagonistic parental enrichment in protein degradation pathways.

In plants, the epigenetic phenomenon of parent-of-origin allele-specific expression occurs mainly in the triploid endosperm. Although well studied in inbreeding Arabidopsis thaliana, genomic imprinting has been less investigated in outcrossers. In order to investigate a wider role of parental-specific allelic expression, we have analyzed imprinting in whole seeds of the obligate outbreeder Arabidopsis arenosa. High-throughput analysis of imprinting in outbreeding species is hampered by the lack of reference genomes and available sequenced accessions. High degree of allelic variation in outbree

Embryonic Development and Potential Causes of Reproductive Failure in Tilia taishanensis S. B. Liang.

In this study, the rare and endangered tree species Tilia taishanensis S. B. Liang was selected as the research material, and its embryonic developmental characteristics and abortion mechanism were systematically observed using scanning electron microscopy and resin semi-thin sectioning techniques. The results showed that its pollen was bicellular, the anther wall belonged to the dicotyledonous type with a secretory tapetum, and asynchronous meiosis occurred during microsporogenesis. The pistil possessed a pentalocular ovary and anatropous ovules, and the embryo sac exhibited a typical seven-c

A universal design of restructured dimer antigens: Development of a superior vaccine against the paramyxovirus in transgenic rice.

The development of vaccines, which induce effective immune responses while ensuring safety and affordability, remains a substantial challenge. In this study, we proposed a vaccine model of a restructured "head-to-tail" dimer to efficiently stimulate B cell response. We also demonstrate the feasibility of using this model to develop a paramyxovirus vaccine through a low-cost rice endosperm expression system. Crystal structure and small-angle X-ray scattering data showed that the restructured hemagglutinin-neuraminidase (HN) formed tetramers with fully exposed quadruple receptor binding domains
