Gut Immunity and Microbiota Dysbiosis Are Associated with Altered Bile Acid Metabolism in LPS-Challenged Piglets.

Bacterial infections are among the major factors that cause stress and intestinal diseases in piglets. Lipopolysaccharide (LPS), a major component of the Gram-negative bacteria outer membrane, is commonly employed for inducing an immune response in normal organisms for convenience. The association between LPS stimulation and gut immunity has been reported. However, the effects of gut immunity on microbial homeostasis and metabolism of host, especially bile acid and lipid metabolism in piglets, remain unclear. Hence, in the current study, we elucidated the effect of gut immunity on microbial ba

Effect of bile acids on 1,2-dimethylhydrazine-induced colon cancer in rats.

The effects of intercepting a part of the enterohepatic circulation of bile acids by ileal resection or bypass on the quantitative and qualitative changes of fecal bile acids were investigated in rats, and the relationship of these changes to the development of DMH-induced colon cancer was studied. The daily fecal total bile acid level was increased in all intercepted groups, especially those with long resection and exclusion. Total bile acid levels played a greater role in the development of DMH-induced colon cancer than did primary or secondary bile acid levels individually. Our findings sug

Physical Activity Modulating Lipid Metabolism in Gallbladder Diseases.

Physical activity encompasses a series of overall benefits on cardiovascular health and metabolic disorders. Research has recently focused on the hepatobiliary tract, as an additional target of the health-related outcomes of different types of physical exercise. Here, we focus on the global features of physical activity with respect to exercise modality and intensity, and on studies linking physical activity to lipid metabolism, gallbladder diseases (gallstones, symptoms, complications and health-related quality of life), gallbladder motor-function, enterohepatic circulation of bile acids, and

Intestinal Failure-Associated Liver Disease.

Intestinal failure-associated liver disease (IFALD) is characterized by either liver steatosis or cholestasis and may develop in patients on long-term home parenteral nutrition for chronic intestinal failure. The pathogenesis of IFALD is multifactorial and includes gastrointestinal disease-related, parenteral nutrition-related, and systemic-related factors. Alteration of bile acid enterohepatic circulation, gut microbiome, and intestinal permeability, seem to be the main mechanisms. Patients forced to a total oral fasting regimen are at greater risk. Parenteral nutrition overfeeding and/or of

[An experimental model of continuous biliary and vascular access in the free rat: a study of the changes in biliary function under the influence of the parenteral administration of amino acids].

Total parenteral nutrition is associated with different hepatic alterations such as cholestasis, steatosis or cholelithiasis. To study such alterations anaesthetised rats, in which the enterohepatic circulation of bile acids is interrupted, have been widely used. A model of free moving rat with venous and biliary catheterization has been developed in this work. This overcomes the interferences induced by anaesthesia as well the alterations in hydrostatic pressure or restraint-induced stress present in other models. The use of our animal model has confirmed previous data from anesthetised rats

Ileitis abolishes tolerogenic functions of the enterohepatic bile acid pool.

Bile acids (BAs) regulate lipid uptake, epithelial integrity, and immune responses in the gut. Hepatocytes synthesize primary BAs, which microbiota metabolize into secondary metabolites. Together, these species form a composite pool that circulates enterohepatically between the liver and intestines. Here, we show that immunoregulatory outputs of the enterohepatic BA pools involve competition between multiple BA species for individual nuclear receptors and are dependent on intestinal health. In healthy mice, the primary BA, tauro β muricholic acid (tβMCA) antagonized RORγt-mediated Th17 functio

Treatment of experimental imipramine and desipramine poisoning in the rat.

The influence of orally administered activated charcoal on organ concentrations of parenteral imipramine and desipramine was investigated. Ancillary distribution experiments indicated that the gastroenteral cycle of these substances might be more important than the enterohepatic cycle. Nevertheless the effectiveness of repeated activated charcoal dosage in lowering antidepressant concentrations of visceral organs is unpredictable. This is interpreted as a consequence of predominant binding of these drugs in the tissues, in contrast to drugs like acetosal and the barbiturates, which are distrib

Kinetics of bile acid metabolism in experimental blind loop syndrome.

In an experimental model of the contaminated small bowel syndrome, Eriksson's washout technique was used to study bile salt metabolism. Bile salt secretion rate was increased from 204-7 +/- SD 36-5 mumol/100 g body weight/24 h to 329-9 +/- 146-6, pool size from 33-4 +/- 5-4 mumol/100 g BW to 42-5 +/- 7-2, and biliary flow from 6-6 +/- 1-72 ml/100 g BW/24 h to 8-4 +/- 2.1. Detailed analysis of the washout curves gave indirect evidence of a short-circuit jejunohepatic shunt in the model contaminated small bowel syndrome. Parameters of this shunt were calculated with 9-1 mumol/100 g BW for 'short

Gut Microbiota-Related Cellular and Molecular Mechanisms in the Progression of Nonalcoholic Fatty Liver Disease.

Nonalcoholic fatty liver disease (NAFLD) is one of the most common and increasing liver diseases worldwide. NAFLD is a term that involves a variety of conditions such as fatty liver, steatohepatitis, or fibrosis. Gut microbiota and its products have been extensively studied because of a close relation between NAFLD and microbiota in pathogenesis. In the progression of NAFLD, various microbiota-related molecular and cellular mechanisms, including dysbiosis, leaky bowel, endotoxin, bile acids enterohepatic circulation, metabolites, or alcohol-producing microbiota, are involved. Currently, diagno

Intestinal absorption of 25-hydroxyvitamin D and osteomalacia in primary biliary cirrhosis.

Bone histology and intestinal absorption of 25-hydroxyvitamin D3 (25-OHD) were investigated in 11 patients with primary biliary cirrhosis (P.B.C.). 4 patients had osteomalacia, and all these had received long-term cholestyramine. Plasma-25-hydroxyvitamin-D (25-OHD) concentrations after an oral dose of 25-OHD3 were significantly lower in the patients with P.B.C. (especially those with osteomalacia) than in normal controls. Serum calcium and urinary calcium excretion were lower, and serum-alkaline-phosphatase higher, in patients with osteomalacia. It is suggested that absorption of 25-OHD underg

[Bone disease in primary biliary cirrhosis].

Primary biliary cirrhosis(PBC)is a chronic autoimmune cholestatic liver disease. Metabolic bone disease is recognized in a complication of chronic liver disease, particularly in PBC. Bone disease in PBC includes osteoporosis and, osteomalacia which is more frequent in advanced liver disease. It is important that PBC occurs mainly in middle-aged women who are highest risk group in primary osteoporosis. In patients with PBC, the dysfunction in enterohepatic circulation of bile acids is associated with the impaired absorption of fats and fat soluble vitamins. Vitamin D and K deficiency leads to o

Ursodeoxycholic acid improves cholestasis in infants with cystic fibrosis.

OBJECTIVE: To describe two infants with cholestatic jaundice treated with ursodeoxycholic acid (UDCA). CASE SUMMARY: Two infants with cystic fibrosis (CF)-associated hepatobiliary disease, manifesting as cholestatic jaundice and elevated liver enzymes within the first 6 weeks of life, had improved biochemical indices of liver function following treatment with UDCA 20-40 mg/kg/d. DISCUSSION: To our knowledge, this is the first report of UDCA treatment in infants with CF-associated cholestatic jaundice. Infants and children require treatment with increased doses of UDCA to compensate for reduc

Daily fluctuation of lithogenicity in bile in the postoperative period in cholecystectomized patients.

The lithogenicity of bile samples taken over 14 hrs from 7 a.m. to 9 p.m. on two consecutive days at 4-5 hrs intervals has been measured in 7 freshly colecystectomized patients on the 7th to 9th postoperative days. The values have also been estimated additionally in 4 patients 1 and 2 hrs after meal intake. It was found that the lithogenicindex is higher at 16 and 21 hrs than at 7 and 11 hrs. The lithogenicity began to increase 2 hrs after lunch. The observed difference between our results and those of other authors could be due either to the changed enterohepatic circulation before the begin

Quantifying Forms and Functions of Enterohepatic Bile Acid Pools in Mice.

BACKGROUNDS & AIMS: Bile acids (BAs) are core gastrointestinal metabolites with dual functions in lipid absorption and cell signaling. BAs circulate between the liver and distal small intestine (i.e., ileum), yet the dynamics through which complex BA pools are absorbed in the ileum and interact with host intestinal cells in vivo remain poorly understood. Because ileal absorption is rate-limiting in determining which BAs in the intestinal lumen gain access to host intestinal cells and receptors, and at what concentrations, we hypothesized that defining the rates and routes of ileal BA absorptio

Anti-diabetic effect of dicaffeoylquinic acids is associated with the modulation of gut microbiota and bile acid metabolism.

INTRODUCTION: The human gut microbiome plays a pivotal role in health and disease, notably through its interaction with bile acids (BAs). BAs, synthesized in the liver, undergo transformation by the gut microbiota upon excretion into the intestine, thus influencing host metabolism. However, the potential mechanisms of dicaffeoylquinic acids (DiCQAs) from Ilex kudingcha how to modulate lipid metabolism and inflammation via gut microbiota remain unclear. OBJECTIVES AND METHODS: The objectives of the present study were to investigate the regulating effects of DiCQAs on diabetes and the potential

Total alditols from Cistanche deserticola attenuate functional constipation by regulating bile acid metabolism.

HEADINGS ETHNOPHARMACOLOGICAL RELEVANCE: Functional constipation (FC), characterized by chronic constipation, significantly impacts physiological function and induces psychological stress in patients. However, current clinical treatment options for FC are currently limited. Cistanche deserticola, a traditional Chinese medicine that promotes intestinal moisture and bowel relaxation, contains cistanche total alditol extract (CTAE) as its primary active extract. However, the production of CTAE, its overall efficacy, and potential mechanisms for treating FC have yet to been investigated. AIM OF T

Cross-tissue multi-omics analyses reveal the gut microbiota's absence impacts organ morphology, immune homeostasis, bile acid and lipid metabolism.

The gut microbiota influences host immunity and metabolism, and changes in its composition and function have been implicated in several non-communicable diseases. Here, comparing germ-free (GF) and specific pathogen-free (SPF) mice using spatial transcriptomics, single-cell RNA sequencing, and targeted bile acid metabolomics across multiple organs, we systematically assessed how the gut microbiota's absence affected organ morphology, immune homeostasis, bile acid, and lipid metabolism. Through integrated analysis, we detect marked aberration in B, myeloid, and T/natural killer cells, altered m

Enterohepatic recirculation of trichloroethanol glucuronide as a significant source of trichloroacetic acid. Metabolites of trichloroethylene.

Trichloroacetic acid (TCA) is a metabolite of trichloroethylene (TRI) thought to contribute to its hepatocarcinogenic effects in mice. Recent studies have shown that peak blood concentrations of TCA do not occur until approximately 12 hr after an oral dose of TRI; however, blood concentrations of TRI reach a maximum within 1 hr and is nondetectable after 2 hr. The objective of this study was to examine quantitatively enterohepatic recirculation of trichloroethanol (TCEOH) and TCA as a possible mechanism responsible for the delayed production of TCA. Jugular vein, duodenum, and bile duct-cannul
