The impact of exposure to environmental contaminant on hepatocellular lipid metabolism.

Increasing evidences show that ubiquitous perfluorooctanoic acid (PFOA), a representative environmental pollutant, is found to be linked to lipid dysmetabolism. However, the biological mechanism behind this outcome remains uninvestigated. In the present study, we established the PFOA-injured liver in mice to explore the underlying mechanism associated with PFOA-induced lipid disturbance in the liver via a group of biochemical and molecular assays. As results, PFOA-exposed mice showed increased transaminase (ALT), reduced triglyceride and free fatty acid contents in serum, as well as elevated l

Technical and clinical validation of an environmental exposure unit for ragweed.

BACKGROUND: Allergic rhinitis is a common condition, with ragweed pollen one of the more prevalent aeroallergens. Environmental exposure units such as the Allergen BioCube® are valuable models for clinical allergy studies. A study was conducted to validate the Allergen BioCube for uniform ragweed pollen concentrations and clinically relevant sign and symptom responses to ragweed exposure. METHODS: Ragweed pollen concentrations were measured on 3 consecutive days in the Allergen BioCube and verified by Rotorod collection and continuous laser particle count measurements. Subjects (N=10) were ex

Children's Cancer and Environmental Exposures: Professional Attitudes and Practices.

BACKGROUND: Epidemiologic studies worldwide have provided substantial evidence of the contributions of environmental exposures to the development of childhood cancer, yet this knowledge has not been integrated into the routine practice of clinicians who care for children with this disease. To identify the basis of this deficit, we sought to assess the environmental history-taking behavior and perceptions of environmental health among pediatric hematologists and oncologists. PROCEDURE: A web-based survey was sent from June to October 2012 to 427 pediatric oncologists, fellows, and nurse practi

[Biomonitoring of exposure to chemical products in environmental and workplace toxicology].

Biological monitoring, also called biomonitoring, is a process to prevent and assess health risk for individuals exposed to chemical products present in environment or through workplace exposure. Biomonitoring is most often a monitoring of exposure or of effect. The exposure monitoring is currently the most widespread in toxicology. It involves the determination in biological fluids of different biomarkers, most of which are biomarkers of internal dose. These biological indicators are typically measured in blood and urine, but other biological samples can be analyzed. They are used to assess t

Fate and transport of mercury in environmental media and human exposure.

Mercury is emitted to the atmosphere from various natural and anthropogenic sources, and degrades with difficulty in the environment. Mercury exists as various species, mainly elemental (Hg(0)) and divalent (Hg(2+)) mercury depending on its oxidation states in air and water. Mercury emitted to the atmosphere can be deposited into aqueous environments by wet and dry depositions, and some can be re-emitted into the atmosphere. The deposited mercury species, mainly Hg(2+), can react with various organic compounds in water and sediment by biotic reactions mediated by sulfur-reducing bacteria, and

Survey of environmental exposure to asbestos in the town of Buyuktatlar, Turkey.

Asbestos-related cases admitted to Chest Diseases Clinic of Kahramanmaras Sutcu Imam University Medical Faculty generally came from Buyuktatlar, a town adjacent to the Afsin district of Kahramanmaraş city. This led us to screen for lung pathologies due to exposure to environmental asbestos in the town of Buyuktatlar. Six hundred and eighty-five inhabitants older than 30 years living in the town were identified as the study group and 488 persons (71·2%) were accepted for evaluation. Then, chest X-rays of these cases were taken and bilateral pleural plaque, bilateral pleural thickening, reticula

Chronic kidney disease associated with environmental toxins and exposures.

People are exposed to various potentially toxic agents and conditions in their natural and occupational environments. These agents may be physical or chemical, may enter the human body through oral, inhalational, or transdermal routes, and may exert effects on all organ systems. Several well-known as well as lesser known associations exist between chronic kidney disease (CKD) and both environmental agents and conditions, such as heavy metals, industrial chemicals, elevated ambient temperatures, and infections. The effects of these agents may be modulated by genetic susceptibility and other com

The assessment of environmental and occupational exposure to hazardous substances by biomonitoring.

BACKGROUND: Modern biomonitoring has expanded beyond its origins in occupational medicine to cover a wide variety of diagnostic procedures and assessments of environmental pollution, ranging from exposure to heavy metals and chemicals to the effects of pesticides and environmental tobacco smoke. In biomonitoring, the overall pollutant load and hazardous exposure of an organism is quantitatively determined, by monitoring the pollutants themselves, their metabolic products and/or conjugates with protein or DNA, in either serum, urine or other body fluids, as well as tissue samples in exceptional

Observations on environmental asbestos exposure in a high risk area.

BACKGROUND AND OBJECTIVE: Environmental asbestos exposure is causally associated with various pulmonary pathologies. In Turkey, one of the most important sources of asbestos exposure is dust originating from the walls of homes whitewashed with white stucco. The main asbestos types implicated are tremolite and, to a lesser extent, chrysotile. This study investigated the presence and effect of environmental asbestos exposure in a small village in Isparta, Turkey. METHODS: Samples of asbestos mine ore, whitewashed plaster from the interior walls of the houses and whitesoil from the outside walls

Children's cognitive health: the influence of environmental chemical exposures.

The potential exists for developmental exposure of children to myriad chemicals, many of which are known to be neurotoxic. Some, such as the organophosphate pesticides, are specifically designed to attack the central nervous system. Despite the known and suspected risks associated with such exposures, critical aspects of the dose-response relationships are unknown or, at best, poorly characterized for the overwhelming majority of chemicals. Among the major knowledge gaps for most chemicals are the critical window(s) of vulnerability, the threshold or "no observed adverse effect level," and the

Environmental lead exposure and urate excretion in the general population.

PURPOSE: Chronic occupational exposure to lead is related to low urate excretion and a high incidence of gout in lead workers. However, whether chronic low-level environmental lead exposure influences urate excretion in the general population remains unknown. SUBJECTS AND METHODS: We studied 111 healthy subjects with normal renal function (serum creatinine level < or =1.4 mg/dL) and no previous lead exposure or systemic diseases. All subjects had their blood lead levels measured, received ethylenediaminetetraacetic acid mobilization tests to assess their body lead burdens, and were investigat

Use of environmental tobacco smoke constituents as markers for exposure.

The 16-City Study analyzed for gas-phase environmental tobacco smoke (ETS) constituents (nicotine, 3-ethenyl pyridine [3-EP], and myosmine) and for particulate-phase constituents (respirable particulate matter [RSP], ultraviolet-absorbing particulate matter [UVPM], fluorescing particulate matter [FPM], scopoletin, and solanesol). In this second of three articles, we discuss the merits of each constituent as a marker for ETS and report pair-wise comparisons of the markers. Neither nicotine nor UVPM were good predictors for RSP. However, nicotine and UVPM were good qualitative predictors of each

Assessment of environmental asbestos exposure in Turkey by bronchoalveolar lavage.

Environmental or domestic exposure to asbestos fibers originating from local soil is responsible for a high incidence of diseases in large rural areas of Turkey. Bronchoalveolar lavage fluids (BALF) were obtained for 65 Turkish subjects originating from these areas and for 42 Turkish controls. Asbestos bodies (ABs) and uncovered fibers (UFs) were quantified by phase contrast light microscopy. Total fiber burden was determined by transmission electron microscopy. The main asbestos types disclosed were tremolite and to a lesser extent chrysotile. AB and fiber concentrations were higher in enviro

[Problems of environmental exposure to toxic effects of glutaraldehyde in medicine].

Glutaraldehyde is an antibacterial and antiviral agent commonly used for cleaning and disinfection of endoscopic equipment. The accepted glutaraldehyde air-concentration is 0.2 ppm. The irritant action, allergic potential and possible neurotoxicity of glutaral have been reported. Red eyes, cough, headache and running nose are most frequent symptoms of glutaral activity. The incidence of bronchial asthma and contact dermatitis has also been reported. In order to avoid occupational hazards of glutaraldehyde closed washing-disinfection systems, proper ventilation (20 cycles per hour), vapour reco

Maternal medications and environmental exposures as risk factors for gastroschisis.

In a case-control study of gastroschisis, we evaluated the risks associated with mother's first-trimester use of medications and with hobby or occupational exposures for 110 cases and 220 controls without a birth defect. Mothers of cases and controls were age-matched. For hobby or occupational exposures, we found significantly elevated risks for high levels of solvents (odds ratio (OR) = 3.8; 95% confidence interval (CI) = 1.6-9.2) and for colorants (OR = 2.3; 95% CI = 1.3-4.0). For medications, we found significantly elevated risks for two strong cyclooxygenase inhibitors, aspirin (OR = 4.7;

Environmental exposure to chromium compounds in the valley of León, México.

The effects on the environment and health of the operation of a chromate compounds factory and tanneries in the León valley in central México are discussed. Sampling and analysis of chromium were performed in water, soil, and human urine. Groundwater has been polluted in an area of about 5 km2 by the leaching of a solid factory waste, which results in concentrations up to 50 mg/l of hexavalent chromium. The plume shape and extension appear to be controlled by the prevailing well extraction regime. Total chromium was detected in the soil around the factory as a result of both aerial transport a

Exposure of children with cystic fibrosis to environmental tobacco smoke.

BACKGROUND: In children, passive exposure to environmental tobacco smoke has been associated with growth suppression and an increased frequency of respiratory tract infections. On the assumption that this association would be more pronounced in children with chronic pulmonary disease, we examined the growth, nutritional status, lung function, and clinical condition of children with cystic fibrosis in relation to their exposure to environmental tobacco smoke. METHODS: We studied 43 children (age, 6 to 11 years) on entry to a summer camp and then again after two weeks in this smoke-free environ

Isopropanol exposure: environmental and biological monitoring in a printing works.

Occupational exposure to isopropanol was studied in 12 workers by testing environmental air, alveolar air, venous blood, and urine during their work shift. Isopropanol, which ranged in environmental air between 7 and 645 mg/m3, was detected in alveolar air, where it ranged between 4 and 437 mg/m3, but not in blood or in urine. Alveolar isopropanol concentration (Ca) was significantly correlated with environmental isopropanol concentration (Ci) at any time of exposure. The value of the arithmetical Ca/ci ratio was 0.418 (SD 0.101). Acetone, which is a metabolite of isopropanol, was found in alv

Synergistic Environmental Exposures and the Airways Capturing Complexity in Humans: An Underappreciated World of Complex Exposures.

Paradoxically, the vast majority of research models intended to understand the relationship between exogenous exposures and lung disease are reduced to a single inhalant. This approach is understandable given the practical challenges of investigation, but it is problematic in terms of translation to the real-world human condition. Furthermore, use of data from such models can lead to underestimation of effect, which may adversely influence regulatory imperatives to protect public health based on the most robust information. Efforts to incrementally introduce layers of complexity to observation
