[Mechanisms of apoptosis induced by trichloroethylene in normal human epidermis keratinocytes].

OBJECTIVE: To explore the potential mechanism of trichloroethylene (TCE)-induced apoptosis in normal human epidermis keratinocyte (NHEK) by assaying the Caspase activities, mitochondrial membrane potential (DeltaPsim) and apoptosis in vitro. METHODS: NHEK was exposed to TCE and Caspases-3, 8 and 9 activities were determined using a commercial assay kit. Apoptosis and DeltaPsim were detected by flow cytometry (FCM) after double-stained with annexin-V and PI, Rh123 and PI respectively. NHEK was pretreated with inhibitor of Caspase-3 or 9 to verify the activation of Caspases by TCE treatment. R

The melanocytorrhagic hypothesis of vitiligo tested on pigmented, stressed, reconstructed epidermis.

Common generalized vitiligo is an acquired depigmenting disorder characterized by a chronic and progressive loss of melanocytes from the epidermis and hair follicles. We previously proposed a new theory that vitiligo involves the chronic detachment and transepidermal loss of melanocytes caused by autoimmune, neural and impaired redox mechanisms associated with mechanical trauma. In this study, we reconstructed epidermis on dead de-epidermized dermis with normal and/or non-segmental non-lesional vitiligo (NSV) cells and tested catecholamines or sera or hydrogen peroxide. Under unstressed condit

Adrenergic and cholinergic control in the biology of epidermis: physiological and clinical significance.

The presence of an autocrine adrenergic and cholinergic intra/intercellular signal transduction network in the human epidermis contributes significantly to homeostatic and compensatory responses regulating vital functions in keratinocytes and melanocytes. The ligands produced control autocrine and paracrine loops to initiate responses through cognate receptors expressed within the same or adjacent cells. The epidermal adrenergic signal controls calcium homeostasis, cell growth, differentiation, motility, and pigmentation via the beta2 and alpha1 adrenoceptors. The cholinergic system is highly

[Inhibition of promyelocytic leukemia gene by tazarotene in hyperproliferative epidermis of psoriasis].

OBJECTIVE: To investigate the mechanism of tazarotene against active psoriasis vulgaris. METHODS: A randomized, controlled trial was conducted in 43 patients with active psoriasis vulgaris, who were divided into tazarotene and control groups. Promyelocytic leukemia (PML) mRNA in active psoriatic lesions before and 14 days after tazarotene treatment was detected by in situ hybridization. RESULTS: PML mRNA expression was detected not only in the basal layer (86.96%), but also in the suprabasal layers of the epidermis in the manner of focal expression (78.26%). After tazarotene treatment, virtu

A case of erythrokeratoderma variabilis: loosened gap junctions in the acanthotic epidermis.

A 15-year-old Japanese female without contributory personal or family medical history had demonstrated irregular, keratotic plaques in the lower extremities since infancy that had been gradually enlarging. The keratotic plaques showed partial erythematous change, which altered shape over a relatively short period, leaving pigmentation. The biopsy specimen taken from the erythematous, keratotic plaque showed typical church-spire-like papillomatosis with acanthosis, and thickening of granular and horny layers. Gene analysis targeting connexin 30.3 and 31, based on the diagnosis of erythrokeratod

Permanent repigmentation of piebaldism by erbium:YAG laser and autologous cultured epidermis.

BACKGROUND: Several surgical techniques have been proposed for the treatment of piebaldism. These procedures, however, are poorly suited for the treatment of large leucodermal lesions, can cause scars and require multiple donor sites. Recently, it has been reported that autologous cultured epidermis induces scarless repigmentation of large vitiligo lesions, using a single small donor site. OBJECTIVES: To induce permanent repigmentation of large achromic lesions in patients suffering from piebaldism by means of autologous cultured epidermal grafts using a rapid, simple and non-invasive surgica

Differential TNFalpha mRNA regulation detected in the epidermis of leprosy patients.

The epidermis is an important site of the immunoinflammatory response in the skin. In the present study, the expression of cytokine and ICAM-1 (intercellular adhesion molecule-1) genes was evaluated by RT-PCR in the epidermis isolated from biopsies from 25 reactional leprosy patients. TNFalpha and IL-6 mRNAs were detected in all individuals during the reactional state (reversal reaction or erythema nodosum leprosum), IL-8 message was detected in 66.6% and 62.5% of the patients, IL-12 mRNA was present in 91.6% and 62.5% and ICAM-1 in 100% and 71.4%, respectively. In addition, when skin biopsies

Immunocytochemical analysis of beta keratins in the epidermis of chelonians, lepidosaurians, and archosaurians.

Beta (beta) keratins are present only in the avian and reptilian epidermises. Although much is known about the biochemistry and molecular biology of the beta keratins in birds, little is known for reptiles. In this study we have examined the distribution of beta keratins in the adult epidermis of turtle, lizard, snake, tuatara, and alligator using light and electron immunocytochemistry with a well-characterized antiserum (anti-beta(1) antiserum) made against a known avian scale type beta keratin. In lizard, snake, and tuatara epidermis this antiserum reacts strongly with the beta-layer, more w

The reconstructed epidermis with melanocytes: a new tool to study pigmentation and photoprotection.

The reconstruction of the epidermal melanin unit ex vivo has been achieved during the last decade, using the combination of previous cell culture techniques. The system reviewed is basically a modification of the Pruniéras model, using the air-liquid interface to grow differentiated keratinocytes, with the addition of 5% melanocytes in the seeding suspension, as well as the use of a more adapted culture medium. Repeated UVB irradiation induces a stimulation of melanogenesis macroscopically, and increases melanin concentration and melanosome transfer in reconstructs. These results have been rep

Ultraviolet radiation-induced p53 responses in the epidermis are differentiation-dependent.

BACKGROUND: The tumour suppressor, p53, is recognized as a crucial molecule in regulating cellular responses to various DNA-damaging agents. Very early on in the development of nonmelanoma cancers p53 is mutated or lost, suggesting that p53 is crucial in protecting normal keratinocytes from the harmful effects of ultraviolet (UV) radiation. OBJECTIVE: Using two mouse models, one with multiple copies of mutant p53 and the other a p53 "knockout," our laboratory has examined a role for p53 in UV-induced DNA damage and determined if these effects are differentiation dependent. CONCLUSION: We out

Ultrastructural identification of basic abnormalities as clues to genetic disorders of the epidermis.

The present article discusses specific, directly gene-dependent ultrastructural markers of dominantly inherited epidermal disorders that serve as clues to their underlying molecular genetic abnormalities. These are epidermolysis bullosa simplex Koebner and Weber-Cockayne with rupture or non-assembly of basal cell keratins and point mutations in keratins 5 and 14. Clumping of basal cell keratins is pathognomonic of EB Dowling-Meara and caused by mutations in hot spots of the rod domain of K5 and K 14. Clumps and aggregates of basal keratins occur side by side in the same cell and thus do not in

Enhanced association of plasminogen/plasmin with lesional epidermis of bullous pemphigoid.

The distribution of plasminogen/plasmin, the central proteolytic component of the plasminogen activator/plasmin system was analysed in lesional skin of bullous pemphigoid by using monoclonal antibodies (MAbs) specific for distinct epitopes of the plasminogen/plasmin molecule. Four groups of MAbs were used: (i) MAbs HD-PG 1 and HD-PG 2, specific for epitopes associated with the lysine-binding sites I (kringle domain 1 + 2 + 3) and II (kringle domain 4) of plasminogen/plasmin, (ii) MAbs HD-PG 6 and HD-PG 7, specific for the lysine binding site I only, (iii) MAbs HD-PG 12 (formerly designated P 2

Corroborative evidence that alopecia areata involves the epidermis and the sebaceous glands.

By using combined autoradiography and computerized image analysis, keratinocytes, sebaceous cells, and endothelial cells of the papillary dermis appear affected in alopecia areata. The atrophy and the reduction in the number of cells synthesizing DNA in these tissues resemble the changes observed in the hair follicles.

Flow cytometry for separation of keratinocyte subpopulations from the viable epidermis.

Human epidermal cell suspensions were analyzed and sorted with flow cytometry. The desmosome and differentiation-related KM48 monoclonal antibody was used for indirect immunofluorescence and permitted staining of keratinocytes at various stages of the cell maturation. Intensity of the staining correlated with the degree of differentiation. Three sorting gates were chosen to obtain subpopulations which varied distinctly in KM48 expression. The flow cytometry-sorted cells were characterized by their ultrastructural appearance and by the bullous pemphigoid antigen expression. According to the ult

Effects of sunlight exposure on Langerhans cells and melanocytes in human epidermis.

The effects of 12 daily, half-hour exposures to sunlight on epidermal Langerhans cells (LC), and melanocytes were studied in 31 healthy volunteers. Nineteen were of Celtic and 12 of mixed European descent. Sunlight appeared to have both a direct, transient, detrimental effect and an indirect stimulating effect on the LC, and a direct and an indirect stimulating effect on the melanocytes. The mixed Europeans had a somewhat greater increase in the melanocyte population density following exposure to sunlight than the Celts. However, the number of LC fell following sunlight exposure regardless of

The biochemical and immunocytochemical characterisation of an antigen on the membrane of basal cells of the epidermis.

A murine monoclonal antibody, LICR-LON-23.10, which had been raised against a well-differentiated squamous cell carcinoma cell line (LICR-LON-HN5), recognises an antigen which is present on the membrane of basal cells of the epidermis. The tissue distribution of the antigen, as defined using immunohistochemical techniques, suggests that it is expressed preferentially on cells which are adjacent to a basement membrane. In squamous cell carcinomas, the antigen is expressed uniformly on undifferentiated cells, but in areas of keratinisation, the antigen is absent. The antigen recognised by the an

Comparison of prekeratin and keratin polypeptides in normal and psoriatic human epidermis.

High-resolution electrophoresis has been used to extend previous observations on the polypeptide composition of keratins in psoriatic epidermis. We have compared psoriatic scale keratins with normal and with scale extracts from several different epidermal disorders. Uninvolved psoriatic epidermis contained prekeratin and keratin of normal profile (68, 60, 58, 52 kDa and 66, 58, 55 kDa, respectively). Prekeratin from involved psoriatic epidermis showed a variable quantitative reduction in the 68-kDa polypeptide and an altered expression of smaller polypeptides (Mr 40 000-55 000). Keratin from t
