Classroom experiments with artificial sweeteners: growing single crystals and simple calorimetry.

An easily accessible experimental set-up to grow large single crystals of two sweeteners readily available in supermarkets, erythritol and xylitol, is described. The crystallization of these compounds illustrates the principles of crystallization by evaporation. As these artificial sweeteners are also known to induce a cool sensation in the mouth, the crystal-growing experiment is complemented with a simple calorimetric set-up to demonstrate the endothermic nature of the dissolution of the selected sweeteners in a more qu-anti-tative approach by measuring the heat of dissolution (ΔH diss). First two unrelated cases of isolated sedoheptulokinase deficiency: A benign disorder?

We present the first two reported unrelated patients with an isolated sedoheptulokinase (SHPK) deficiency. The first patient presented with neonatal cholestasis, hypoglycemia, and anemia, while the second patient presented with congenital arthrogryposis multiplex, multiple contractures, and dysmorphisms. Both patients had elevated excretion of erythritol and sedoheptulose, and each had a homozygous nonsense mutation in SHPK. SHPK is an enzyme that phosphorylates sedoheptulose to sedoheptulose-7-phosphate, which is an important intermediate of the pentose phosphate pathway. It is questionable w

Molecular recognition of carbohydrates by a resorcinarene. Selective transport of alditols through a supported liquid membrane.

A supported liquid membrane (SLM) containing a resorcinarene carrier has been used for the selective transport of erythritol, threitol, ribitol and xylitol from concentrated (1.0-0.01 M) aqueous solutions. The membrane is made of a microporous polytetrafluoroethylene film impregnated with a 0.01 M solution of the carrier in CCl4. The permeabilities of the SLM for all alditols were calculated. On the basis of the flux dependence on the initial concentrations of carrier and alditol, the rate-determining step in the transport mechanism is shown to be the migration of the 1:1 carrier-carbohydrate

Evaluation of culture- and PCR-based methods for detecting Burkholderia pseudomallei in soil samples in Thailand.

BACKGROUND: Burkholderia pseudomallei is an environmental bacterium that causes melioidosis, a life-threatening disease prevalent in tropical regions. Accurate detection of B. pseudomallei in the environment is essential for identifying areas that pose an infection risk. This study aimed to evaluate culture- and real-time polymerase chain reaction (PCR)-based methods for detecting B. pseudomallei directly in soil samples, and to assess its environmental prevalence in relation to antibody levels in healthy individuals from Amnat Charoen, Northeast Thailand. METHODOLOGY/PRINCIPAL FINDINGS: Init

Effects of Adjunctive EDTA and Air-Polishing Powders on Periodontitis-Affected Root Surfaces: A SEM Study.

Background: Mechanical subgingival instrumentation remains the gold standard in periodontitis treatment; however, it may leave residual debris and induce surface alterations. Adjunctive strategies such as air polishing and ethylenediaminetetraacetic acid (EDTA) might optimize root surface conditions. Objective: To evaluate, by scanning electron microscopy (SEM), the effects of scaling and root planing (SRP) combined with EDTA, with or without adjunctive erythritol- or glycine-based air polishing, on root surface alterations and smear layer formation. Materials and Methods: Ten extracted human

Functional analysis of the methylerythritol phosphate pathway terminal enzymes IspG and IspH from Zymomonas mobilis.

UNLABELLED: Isoprenoids are a diverse family of compounds that are synthesized from two isomeric compounds, isopentenyl diphosphate and dimethylallyl diphosphate. In most bacteria, isoprenoids are produced from the essential methylerythritol phosphate (MEP) pathway. The terminal enzymes of the MEP pathway IspG and IspH are [4Fe-4S] cluster proteins, and in Zymomonas mobilis, the substrates of IspG and IspH accumulate in cells in response to O2, suggesting possible lability of their [4Fe-4S] clusters. Here, we show using complementation assays in Escherichia coli that even under anaerobic condi

The mevalonate and methylerythritol phosphate pathways as therapeutic targets in protozoan parasites: from moonlighting enzymes to metabolic plasticity.

The interplay between metabolic pathways and immune responses is critical for understanding host-parasite interactions in protozoan infections. This review examines the mevalonate (MVA) and methylerythritol phosphate (MEP) pathways-key routes for isoprenoid synthesis essential for sterol production and bioactive compounds. Beyond sustaining parasite survival, these pathways directly influence host immune signaling cascades, including NF-κB, MAPK, PI3K/Akt, and mTOR. A critical conceptual distinction is introduced: genuine "moonlighting" functions-where a single enzyme performs two distinct mol

Advancing Drug Discovery via High-Throughput Screening of Methylerythritol Phosphate Pathway Enzymes: Strategies and Prospects for Combating Resistance.

Antibiotic resistance and herbicide resistance have emerged as global threats, urgently necessitating the development of agents with novel modes of action and new targets. The methylerythritol phosphate (MEP) pathway is widely present in bacteria, plants, and apicomplexan parasites, yet absent in mammals. The metabolic products of this pathway are essential for cellular growth and metabolism. Consequently, the key enzymes therein serve as potential targets for developing new antimicrobials and herbicides. Establishing reliable high-throughput screening (HTS) strategies for enzyme inhibitors is

Aqueous and hydro-alcoholic media effects on polyols.

The ingestion of drug products with alcohol can have an adverse effect on drug levels in a patient's blood. The Food and Drug Agency (FDA) issued an alert in 2005 after hydromorphone was withdrawn from the market after clinical trials showed ingestion with alcohol to potentially result in lethal drug peak plasma concentrations. The potential impact of alcohol on extended release (ER) tablet matrices and the need to develop ER matrices robust to alcohol effects has then been of interest. This study investigated the compaction properties of polyols and their effect on drug release. Polyols (eryt

[Molecular characterization of Brucella abortus strain 19 and its application for controlling biologics].

Brucella abortus is the etiological agent of bovine brucellosis. The strain 19 used in vaccine elaboration can be identified through a deletion in the eri region associated with its susceptibility to erythritol. We optimized a PCR assay for specific characterization of this strain. The method described here is a rapid procedure that enables identification of B. abortus, and simultaneous differentiation of the strain 19 from other B. abortus biovar 1 strains. We applied the assay to detect the strain 19 in vaccines against B. abortus produced in Argentina. Thq results show this method could be

[Interaction of bacteria with mercuric compounds].

We investigated the interaction of mercuric compounds with the bacteria Corynebacterium ammoniagenes, Micrococcus luteus, and Mycobacterium smegmatus capable of producing hydroxylamines (R-NOH) and 2-C-methyl-D-erythritol-2,4-cyclopyrophosphate (MECP), which are prone to form free radicals. The interaction of these substances with Hg2+ ions and their dynamics during the mercuric poisoning of bacteria was studied by EPR and NMR. Under stress conditions induced by lowering pH or generation of active oxygen species, the bacteria and, especially, their mutants with enhanced sensitivity to oxidativ

Biochemical characteristics of clinical and environmental isolates of Burkholderia pseudomallei.

The biochemical characteristics of 213 isolates of Burkholderia pseudomallei from patients with melioidosis and 140 isolates from the soil in central and northeastern Thailand were compared. Whereas the biochemical profiles of all the clinical isolates were similar, all soil isolates from the central area and 25% of isolates from northeastern Thailand comprised a different phenotype. This was characterised by the ability to assimilate L-arabinose (100%), adonitol (100%), 5-keto-gluconate (90%) and D-xylose (84%), but failure to assimilate dulcitol (0%), erythritol (0%) and trehalose (10%). Clinical, microbiological, and experimental animal studies of Candida lipolytica.

Candida lipolytica was recovered from six patients in three different clinical centers. The index isolate caused a persistent fungemia with catheter-associated Candida thrombophlebitis, the second isolate was from a polymicrobial sinusitis, and the remaining four isolates were involved in tissue colonization. These and 20 other isolates were consistent in their morphological and physiological characteristics. All formed true hyphae and blastoconidia on cornmeal-Tween 80 agar and all assimilated glucose, glycerol, and erythritol. In a murine model of disseminated candidiasis, the index isolate

Evaluation of different debridement strategies for implant-abutment connections: An in vitro study.

STATEMENT OF PROBLEM: Technical complications, including abutment fracture, are a clinical reality. After retrieving failed components, re-establishing a reliable implant-abutment connection is essential, but recommendations on how to clean the interface are lacking. PURPOSE: The purpose of this in vitro study was to evaluate different clinical debridement strategies for removing titanium powder from an implant-abutment interface with regard to their efficacy and associated risk of damaging the interface. MATERIAL AND METHODS: Specimens (n=5) were assembled from previously cut implant halves

Nonviral vectors with a biosurfactant MEL-A promote gene transfection into solid tumors in the mouse abdominal cavity.

Recently, we showed that a biosurfactatnt 4-O-[(4',6'-di-O-acethyl-2',3'-di-O-alkanoyl)-beta-D-mannopyranosyl] meso-erythritol A (MEL-A) greatly increased the efficiency of gene transfection mediated by cationic liposomes in vitro. We then studied whether the high transfection efficiency of these liposomes is maintained in vivo for tumor cells in the mouse abdominal cavity. When a complex of the liposomes and plasmid DNA was injected intraperitoneally into C57BL/6J mice bearing B16/BL6 tumors, we found that the biosurfactant significantly increased liposome-mediated gene transfection to the mo

Purification and properties of beef liver aldehyde reductase catalyzing the reduction of D-erythrose 4-phosphate.

An aldehyde reductase catalyzing the NADPH-dependent reduction of D-erythrose 4-phosphate to D-erythritol 4-phosphate was purified from beef liver. It was proved to be homogeneous by polyacrylamide gel electrophoresis, sodium dodecyl sulfate polyacrylamide gel electrophoresis and ultracentrifugation analysis. The enzyme was proved to be a monomeric enzyme and its molecular weight was about 40,000. The enzyme was able to reduce not only tetroses but also trioses, aromatic aldehydes, D-glucuronate and succinic semialdehyde. Apparent Km-values for aromatic aldehydes were lower than those for tetr

Ethanol-induced mucosal injury in rabbit oesophagus.

The pathogenesis of oesophagitis associated with ethanol ingestion was studied experimentally by perfusing isolated rabbit oesophagus in situ with 20% and 40% (v/v) ethanol. Since ingestion of ethanol increases duodenogastric and gastro-oesophageal reflux, the effects of HCl and bile salts (in combination with ethanol) were also investigated. The severity of oesophageal mucosal damage was assessed by using transmucosal potential difference, net ion fluxes of H+ and Na+, and mucosal permeability to two neutral molecules of different sizes, 3H-H2O and 14C-erythritol, as indicators of mucosal int

A new family of enzymes catalyzing the first committed step of the methylerythritol 4-phosphate (MEP) pathway for isoprenoid biosynthesis in bacteria.

Isoprenoids are a large family of compounds with essential functions in all domains of life. Most eubacteria synthesize their isoprenoids using the methylerythritol 4-phosphate (MEP) pathway, whereas a minority uses the unrelated mevalonate pathway and only a few have both. Interestingly, Brucella abortus and some other bacteria that only use the MEP pathway lack deoxyxylulose 5-phosphate (DXP) reductoisomerase (DXR), the enzyme catalyzing the NADPH-dependent production of MEP from DXP in the first committed step of the pathway. Fosmidomycin, a specific competitive inhibitor of DXR, inhibited
