Extracellular vesicles and its advances in female reproduction.

Intercellular communication is an essential mechanism for development and maintenance of multicellular organisms. Extracellular vesicles (EVs) were recently described as new players in the intercellular communication. EVs are double-membrane vesicles secreted by cells and are classified according to their biosynthesis, protein markers and morphology. These extracellular vesicles contain bioactive materials such as miRNA, mRNA, protein and lipids. These characteristics permit their involvement in different biological processes. Reproductive physiology is complex and involves constant communicat

Extracellular Vesicles: A Therapeutic Option for Liver Fibrosis.

Extracellular vesicles (EVs) are a heterogeneous population of small membrane vesicles released by all types of cells in both physiological and pathological conditions. EVs shuttle different types of molecules and are able to modify the behavior of target cells by various mechanisms of action. In this review, we have summarized the papers present in the literature, to our acknowledge, that reported the EV effects on liver diseases. EVs purified from serum, stem cells, and hepatocytes were investigated in different experimental in vivo models of liver injury and in particular of liver fibrosis.

Extracellular Vesicles of Stem Cells to Prevent BRONJ.

Extracellular vesicles (EVs), several tens to hundreds of nanometers in size, are vesicles secreted by cells for intercellular communication. EVs released from mesenchymal stem cells (MSC-EVs) have the potential to treat multiple diseases. This study aimed to determine the effects of MSC-EVs on bisphosphonate-related osteonecrosis of the jaw (BRONJ), whose pathogenesis and treatment are not yet established. To this end, zoledronic acid (ZOL) was administered to bone marrow cells and fibroblasts in vitro. In vivo, a BRONJ model was produced by administering ZOL to rats and extracting teeth. Eac

[Research progress of extracellular vesicles in hepatic fibrosis].

Liver fibrosis is a pathophysiological process characterized by abnormal accumulation of connective tissues in the liver caused by chronic liver injuries, in which the activation and migration of hepatic stellate cells (HSCs) play a central role. Extracellular vesicles (EVs) are a class of nanoscale, bilayer lipid enveloped vesicles secreted by almost all cells. EVs are of great interest in liver pathology because they have been found to mediate the communication between cells and regulate cellular microenvironment via horizontal transfer of their cargoes. EVs carry bioactive cargoes including

Emerging Role of Extracellular Vesicles in Bone Remodeling.

Extracellular vesicles (EVs), as nanometer-scale particles, include exosomes, microvesicles, and apoptotic bodies. EVs are released by most cell types, such as bone marrow stem cells, osteoblasts, osteoclasts, and immune cells. In bone-remodeling microenvironments, EVs deliver specific proteins (e.g., tenascin C and Sema4D), microRNAs (e.g., miR-214-3p, miR-183-5p, and miR-196a), and other growth factors (e.g., bone morphogenetic protein 1 to 7 and transforming growth factor β1) to osteoblasts and regulate bone formation. In addition, EVs can deliver cytokines, such as RANK (receptor activator

Role of Extracellular Vesicles in Amyotrophic Lateral Sclerosis.

Amyotrophic Lateral Sclerosis (ALS) is the most common motor neuron disease in adults and primarily targets upper and lower motor neurons. The progression of the disease is mostly mediated by altered intercellular communication in the spinal cord between neurons and glial cells. One of the possible ways by which intercellular communication occurs is through extracellular vesicles (EVs) that are responsible for the horizontal transfer of proteins and RNAs to recipient cells. EVs are nanoparticles released by the plasma membrane and this review will describe all evidence connecting ALS, intercel

Extracellular vesicles induce protective immunity against Trichuris muris.

Gastrointestinal nematodes, such as Trichuris trichiura (human whipworm), are a major source of morbidity in humans and their livestock. There is a paucity of commercially available vaccines against these parasites, and vaccine development for T. trichiura has been impeded by a lack of known host protective antigens. Experimental vaccinations with T. muris (murine whipworm) soluble Excretory/Secretory (ES) material have demonstrated that it is possible to induce protective immunity in mice; however, the potential for extracellular vesicles (EVs) as a source of antigenic material has remained r

Emerging role of extracellular vesicles in musculoskeletal diseases.

Research into the biology of extracellular vesicles (EVs), including exosomes and microvesicles, has expanded significantly with advances in EV isolation techniques, a better understanding of the surface markers that characterize exosomes and microvesicles, and greater information derived from -omics approaches on the proteins, lipids, mRNAs, and microRNAs (miRNAs) transported by EVs. We have recently discovered a role for exosome-derived miRNAs in age-related bone loss and osteoarthritis, two conditions that impose a significant public health burden on the aging global population. Cardiac Extracellular Vesicles in Normal and Infarcted Heart.

Heart is a complex assembly of many cell types constituting myocardium, endocardium and epicardium that intensively communicate to each other in order to maintain the proper cardiac function. There are many types of intercellular intracardiac signals, with a prominent role of extracellular vesicles (EVs), such as exosomes and microvesicles, for long-distant delivering of complex messages. Cardiomyocytes release EVs, whose content could significantly vary depending on the stimulus. In stress, such as hypoxia, inflammation or injury, cardiomyocytes increase secretion of EVs. In hypoxic condition

Extracellular vesicles and reproduction-promotion of successful pregnancy.

Extracellular vesicles (EVs) are membrane-bound complexes secreted from cells under both physiological and pathological conditions. They contain proteins, nucleic acids and lipids and act as messengers for cell-cell communication and signalling, particularly between immune cells. EV research is a rapidly evolving and expanding field, and it appears that all biological fluids contain very large numbers of EVs; they are produced from all cells that have been studied to date, and are known to have roles in several reproductive processes. This review analyses the evidence for the role of EVs throu

Classification, functions, and clinical relevance of extracellular vesicles.

Both eukaryotic and prokaryotic cells release small, phospholipid-enclosed vesicles into their environment. Why do cells release vesicles? Initial studies showed that eukaryotic vesicles are used to remove obsolete cellular molecules. Although this release of vesicles is beneficial to the cell, the vesicles can also be a danger to their environment, for instance in blood, where vesicles can provide a surface supporting coagulation. Evidence is accumulating that vesicles are cargo containers used by eukaryotic cells to exchange biomolecules as transmembrane receptors and genetic information. Be

Gut microbiota-derived extracellular vesicles in Parkinson's disease.

The gut microbiota contributes to the pathophysiology of Parkinson's disease (PD), but the mechanism is not known. Extracellular vesicles (EVs), membrane-enclosed nanosized particles, contain various biomolecules, cross biological barriers, and reach sterile body compartments. We hypothesized that EVs released by the gut microbiota might influence PD pathophysiology. We isolated gut microbiota-derived EVs from the fecal samples of 20 patients with PD and 19 healthy controls. Patients with PD had higher concentrations of fecal EVs, and the mean particle size of EVs was larger. The 16S ribosomal

Food bioactives and extracellular vesicles: a narrative review.

Extracellular vesicles (EVs) are now recognized as key mediators of intercellular communication, transferring cargos such as proteins, lipids, and nucleic acids between cells and thereby influencing physiological and pathological processes. While the role of EVs has been extensively investigated in physiological and pathophysiological contexts, nutrition has only recently been recognized as an important modulator of endogenous EV biology. Emerging evidence indicates that nutrition and food bioactive compounds can influence EV biology through three distinct mechanisms: (i) modulation of the bio

Leveraging Tissue-Derived Extracellular Vesicles for Regenerative Medicine.

Tissue-derived extracellular vesicles (Ti-EVs) encapsulate the complex molecular signature of their native microenvironment, serving as critical mediators for tissue homeostasis and regeneration. This review provides a state-of-the-art overview of Ti-EV biology, critically evaluating current evidence from the liver, heart, adipose tissue, and skeletal muscle while identifying key challenges and future perspectives for clinical translation. We first categorize current isolation technologies by their validation status in the Ti-EV context, and then survey organ-specific functional profiles with

Transcriptomic and functional analysis of fibroid extracellular vesicles.

Exosomes were isolated from cultures of fibroid explants and matched myometrial explants, and their RNA cargo was analyzed by next-generation sequencing to profile both long and short RNA species. Fibroid-derived exosomes (Fib-EXO) expressed the canonical extracellular vesicle markers CD81 and CD63, and their size distribution (30-200 nm) was consistent with exosomal vesicles. The RNA cargo of Fib-EXO generally reflected that of its tissue of origin, although selective enrichment of specific transcripts, such as piR-1398740 and piR-333378, suggested active loading mechanisms. Long RNA sequenci
