Glycinocins E-H, Antimicrobial Lipopeptides Produced by a Streptomyces Strain.

In an antifungal screening of extracts from microbial strains isolated from gypsum outcrops in Granada, Spain, four new cyclic lipopeptides, glycinocins E-H (1-4), were identified from Streptomyces sp. CA-297274 and exhibited potent activity against Zymoseptoria tritici, the causal agent of septoria tritici blotch of wheat. Isolation and structure elucidation performed using HR-MS/MS, 1D and 2D NMR, and Marfey's analyses, revealed that structures contained an Asp-Gly-Asp-Gly motif in the peptide scaffold, typical of calcium-dependent antibiotics. Genome sequencing and bioinformatic analysis of

Intrinsically disordered regions may lower the hydration free energy in proteins: a case study of nudix hydrolase in the bacterium Deinococcus radiodurans.

The proteome of the radiation- and desiccation-resistant bacterium D. radiodurans features a group of proteins that contain significant intrinsically disordered regions that are not present in non-extremophile homologues. Interestingly, this group includes a number of housekeeping and repair proteins such as DNA polymerase III, nudix hydrolase and rotamase. Here, we focus on a member of the nudix hydrolase family from D. radiodurans possessing low-complexity N- and C-terminal tails, which exhibit sequence signatures of intrinsic disorder and have unknown function. The enzyme catalyzes the hydr

The Influence of Ectoine on the Skin Parameters Damaged by a CO2 Laser.

Ectoine is a substance produced by extremophiles and is naturally used by them as protection against adverse environmental conditions in which they live. Scientific contributions discuss its excellent effect through cosmotropic properties, prevention of secondary messenger release in cells, and transcription factors. The influence on the lipid layer of the cell membrane and its preventive effect as a UV filter were also demonstrated. What is more, its anti-oxidative effect was established. Ectoine works as an immunostimulant and also has anti-inflammatory and anti-cancer properties. These attr

Driving a Stimuli-Responsive Wedge in the Packing of Phospholipid Membranes Using Bolaamphiphile Intercalants.

Bolaamphiphiles─amphiphilic molecules with polar groups at each of the two ends of a hydrophobic tail with pH-sensitive spontaneous molecular curvatures, endow membranes of extremophiles with an exquisite balance between stability (or robustness) and adaptability (or plasticity). But how the presence (or real-time insertion) of bolaamphiphiles influences lamellar lipid membranes is poorly understood. Using a combination of time-resolved confocal fluorescence microscopy, in situ small-angle X-ray and neutron scattering (SAXS and SANS), and neutron spin echo (NSE) measurements, we monitor here t

Aneurinibacillus thermoaerophilus CCM 8960 is a thermophilic bacterium isolated from compost in Brno. The bacterium accumulates polyhydroxyalkanoates (PHAs), a biodegradable and renewable alternative to petrochemical polymers. The bacterium reveals several features that make it a very interesting candidate for the industrial production of PHA. At first, due to its thermophilic character, the bacterium can be utilized in agreement with the concept of next-generation industrial biotechnology (NGIB), which relies on extremophiles. Second, the bacterium is capable of producing PHA copolymers conta

De novo transcriptome resources of the lichens, Dirinaria sp. UKM-J1 and UKM-K1 collected from Jerantut and Klang, Malaysia.

Lichen is a symbiotic organism that exists as a single composite body consisting of a mycobiont (fungus) and a photobiont (algae or a cyanobacterium). Many lichen species are considered as extremophiles due to their tolerance to radiation, desiccation, temperature and pollution. However, not all lichen species are tolerant to harsh environmental conditions as several species are sensitive for example to nitrogen, sulphur, acidity, heavy metals, halogens (e.g. fluoride) and ozone. Thus, to better understand why some lichens can withstand exposure to pollutants as opposed to those that are susce

Sustainable co-production of 1,3-PDO, ethanol and H2 from glycerol via dark fermentation by Citrobacter telavivensis T1.2D-1 isolated from the deep subsurface.

The increasing production of biodiesel has led to a surplus of glycerol, a polluting by-product in need of valorization. In this study, we demonstrate that Citrobacter telavivensis T1.2D-1, an extremophile bacterium isolated from the Iberian Pyrite Belt, effectively converts glycerol into valuable compounds via dark anaerobic fermentation. Genomic and bioinformatic analyses confirmed the presence of the dha and pdu operons, responsible for 1,3-propanediol (1,3-PDO) synthesis, and the hyc operon and fdhF gene involved in hydrogen (H2) production. Batch fermentations revealed that optimal yields

Characterization of a Novel GH3 β-Glucosidase with Alkali-Tolerant and Halophilic Properties from Pontixanthobacter luteolus SW-109.

β-Glucosidases are in high demand for industrial processes such as lignocellulosic saccharification and high-salt food fermentation. However, conventional enzymes often lose activity under high-salt and alkaline conditions, which constrains their broader applicability. In this study, a glycoside hydrolase family 3 (GH3) β-glucosidase (designated PlGH3) was cloned from Pontixanthobacter luteolus SW-109, and was heterologously expressed in Escherichia coli, followed by systematic characterization of its enzymatic properties and structural simulation. PlGH3 showed an optimal pH of 9.0 and optimal

Host-Microbiome Associations in Livebearing Fishes Adapted to Toxic Environments Rich in Hydrogen Sulfide.

Organisms inhabiting extreme environments must tolerate a variety of physiochemical stressors. In some cases, host-associated microbial communities facilitate the survival of their hosts in extreme environments, but extremophile symbioses have not been identified in vertebrates. We used 16S rRNA amplicon sequencing to investigate commonalities and differences in the gut bacterial communities of livebearing fishes (Poecilia mexicana species complex, Poeciliidae) that have repeatedly colonised toxic sulfide streams in southern Mexico. We found shared gut microbial taxa across habitat types and d

The Idiosyncratic Physiological Traits of the Naked Mole-Rat; a Resilient Animal Model of Aging, Longevity, and Healthspan.

The subterranean-dwelling naked mole-rat (Heterocephalus glaber) is an extremophilic rodent, able to thrive in the harsh underground conditions of sub-Saharan Northeast Africa. This pelage-free mammal exhibits numerous unusual ecophysiological features including pronounced tolerance of thermolability, hypoxia, hypercapnia and noxious substances. As a mammal, the naked mole-rat provides a proof-of-concept that age-related changes in physiology are avoidable. At ages far beyond their expected lifespans given both their body size and/or the timing of early developmental milestones, naked mole-rat

Convergent evolution of conserved mitochondrial pathways underlies repeated adaptation to extreme environments.

Extreme environments test the limits of life; yet, some organisms thrive in harsh conditions. Extremophile lineages inspire questions about how organisms can tolerate physiochemical stressors and whether the repeated colonization of extreme environments is facilitated by predictable and repeatable evolutionary innovations. We identified the mechanistic basis underlying convergent evolution of tolerance to hydrogen sulfide (H2S)-a toxicant that impairs mitochondrial function-across evolutionarily independent lineages of a fish (Poecilia mexicana, Poeciliidae) from H2S-rich springs. Using compar

From Glacier to Sauna: RNA-Seq of the Human Pathogen Black Fungus Exophiala dermatitidis under Varying Temperature Conditions Exhibits Common and Novel Fungal Response.

Exophiala dermatitidis (Wangiella dermatitidis) belongs to the group of the so-called black yeasts. Thanks in part to its thick and strongly melanized cell walls, E. dermatitidis is extremely tolerant to various kinds of stress, including extreme pH, temperature and desiccation. E. dermatitidis is also the agent responsible for various severe illnesses in humans, such as pneumonia and keratitis, and might lead to fatal brain infections. Due to its association with the human environment, its poly-extremophilic lifestyle and its pathogenicity in humans, E. dermatitidis has become an important mo

Extracellular polymeric substances mediate bioleaching/biocorrosion via interfacial processes involving iron(III) ions and acidophilic bacteria.

Extracellular polymeric substances seem to play a pivotal role in biocorrosion of metals and bioleaching, biocorrosion of metal sulfides for the winning of precious metals as well as acid rock drainage. For better control of both processes, the structure and function of extracellular polymeric substances of corrosion-causing or leaching bacteria are of crucial importance. Our research focused on the extremophilic bacteria Acidithiobacillus ferrooxidans and Leptospirillum ferrooxidans, because of the "simplicity" and knowledge about the interactions of these bacteria with their substrate/substr
