Immunological tolerance during fetal development: from mouse to man.

The development of the adaptive immune system has been studied in the mouse primarily because it is easier to access fetal tissues and because there exists a rich array of probes for analysis of various components of the immune system. While much has been learned from this exercise, it is also clear that different species show substantial temporal variation in the development of the immune system during early life. In mice, for instance, mature α/β T cells first appear in the periphery during the final stages of fetal gestation and only increase in number after birth (Friedberg and Weissman, 1

[Smoking, fetal pulmonary development and lung disease in children].

Prenatal exposure to tobacco smoke is associated with a significant increase in respiratory symptoms and a decrease in lung function in children. Epidemiological studies have demonstrated that tobacco smoke exposure represents one of the most important respiratory risk factors in childhood. Nicotine crosses the placenta and activates the nicotinic receptors which are present in a wide range of lung cells during the prenatal period. In utero tobacco exposure is responsible for an increase in alveolar volumes and a decrease in airway walls caliber, which explains the decrease in airflow that is

Expression of CD21 and CD23 during human fetal development.

Neonates produce lower levels of IgE compared with adults. Diminished IL-4 production and impaired up-regulation of CD40L by neonatal T cells could explain this, however other regulators of IgE production, such as CD21 and CD23, could contribute to reduced circulating IgE levels during fetal development. Heparinized blood samples were collected from adults and from the umbilical cord at premature and term births. Whole blood flow cytometry was used to assess the percentage of T (CD3(+)) and B (CD19(+)) lymphocytes expressing CD21 and/or CD23 at 26-29 (n = 3), 30-33 (n = 7), 34-37 (n = 5), and

Fetal development in rhesus monkeys exposed prenatally to cocaine.

Using a timed-breeding protocol, one group of female rhesus monkeys was implanted subcutaneously with osmotic minipumps containing 0.3 mg/kg/h cocaine (N=18) or saline (N=18) from day 24 postconception through gestation. Another group received cocaine (N=12) or saline (N=8) from conception through day 42 of gestation. Mean levels of cocaine in maternal serum were approximately 150 ng/ml during pregnancy. A total of 56 pregnancies were documented in 42 adult monkeys, and 39 pregnancies completed full-term. Maternal food consumption and body weight increased during pregnancy, and there were no s

Lung development following diaphragmatic hernia in the fetal rabbit.

Diaphragmatic hernia was created in 39 rabbit fetuses on day 23 of gestation. Fifteen fetuses underwent a sham thoracotomy (SHAM). Thirty-nine non-operated littermates served as internal controls (CTR). Fetuses were harvested by Caesarean section on days 25, 27, 29 and 30 of gestation. Pulmonary response was evaluated by lung to body weight ratio (LBWR), morphometry, and density of type II pneumocytes. No difference was found between CTR and SHAM fetuses at term. CDH fetuses had smaller lungs (LBWR 0.014 +/- 0.004 versus 0.030 +/- 0.04 in CTR, P < 0.0001), a less complex acinus [mean terminal

Current topic: confined placental mosaicism and intrauterine fetal development.

Chromosomal mosaicism is the presence of two or more cell lines with different chromosomal complements in an individual derived from a single zygote (which is distinguishable from chimaerism in which an individual is derived from more than one zygote). Mosaicism may involve a number of different karyotypic abnormalities including those for whole sets of chromosomes (polyploidy, including triploidy and tetraploidy), trisomies, sex chromosomal anomalies and chromosomal rearrangements. Mosaicism itself may be present in all the tissues of the body and placenta, or it may only be seen in certain t

Fetal adrenal development during the second trimester of gestation.

Four hundred and fifty pairs of adrenals were studied from human fetuses ranging in age from 15 to 27 weeks of gestation (menstrual age). They were collected from spontaneous and medical abortions. The adrenal weight increased from 0.2 to 1.5 g during the second trimester. The left adrenal was significantly heavier than the right for the same gestational age. The length (L1), the height (L2) and the thickness (L3) of the adrenals were measured. The index of length (L1 x L2 x L3) correlated well with the fetal age, except for the Potter Syndrome in which it was decreased. This index could be a

Placental TRPV2 supports trophoblast function and normal fetal development.

While placental functioning arbitrates fetal outcome, the molecular players that underpin normal placental development remain poorly understood. Here, we provide evidence that the Transient Receptor Potential Vanilloid 2 (TRPV2) channel plays an important role in normal placental development, determining fetal size and survival. Its absence severely affects placental morphology, causing fetal growth restriction and embryonic lethality, which was found to be allocated to a non-redundant role in the trophoblast, but not in the embryo. CRISPR/Cas9-mediated Trpv2 deletion in mouse trophoblast stem

Maternal NKG2D-mediated immune activation shapes fetal neural development.

Maternal immune activation (MIA) shapes fetal growth and neurodevelopment, in part by disrupting immune homeostasis at the maternal-fetal interface, thereby contributing to developmental abnormalities such as autism spectrum disorder (ASD). In women with a history of influenza A virus (IAV) infection, we found an enrichment off CD56dimCD16+CD49a- natural killer (NK) cells displaying enhanced NKG2D expression in the decidua basalis, as assessed at delivery. Using IAV- and poly I:C-induced MIA mouse models, we demonstrated that MIA significantly increases the proportion of NK cells at the matern

Resveratrol mitigates diazinon-induced neurotoxicity during fetal brain development.

OBJECTIVES: Disruption of transcriptional programs governing fetal neurogenesis represents a critical but underexplored mechanism of diazinon (DZN)-induced developmental neurotoxicity. This research examines whether resveratrol (RV) influences the expression of genes associated with neurogenesis and survival after prenatal exposure to DZN. MATERIALS AND METHODS: Twenty-four pregnant Wistar rats were divided into DZN (40 mg/kg) group, RV (10 mg/kg), DZN+RV (40 mg/kg + 10 mg/kg), and Control. On day 21 of pregnancy, rats were cesarean sectioned, and the neonatal brains were examined using HPLC,

Prenatal stress, hormones, and fetal brain development: gender differences.

Prenatal stress is increasingly recognized as a critical factor influencing fetal brain development, with potential long-term risks for neurodevelopmental and psychiatric disorders. This review explores the effects of prenatal stress on the fetal brain, with particular emphasis on sex differences and the interplay between genetic, epigenetic, and environmental factors. The activation of the hypothalamic-pituitary-adrenal (HPA) axis in response to maternal stress leads to increased levels of glucocorticoids, which can cross the placenta and alter fetal brain structure and function. Key regions

Effect of Maternal Hyperglycemia on Fetal Pancreatic Islet Development.

Hyperglycemia during fetal development disturbs extracellular matrix (ECM) synthesis and deposition patterns, which disrupts organogenesis and adult organ function. Although the ECM cooperates in pancreas development, little is known about the effects of hyperglycemia on the pancreatic ECM during development. This study investigates the effect of severe maternal hyperglycemia on ECM composition and endocrine pancreas development in E19.0 mouse fetuses. Deposition patterns of pan-laminin, laminin (alpha 1 and gamma 1 chains) and integrin alpha 3 were evaluated by immunostaining. The proliferati

COVID-19 in pregnancy: implications for fetal brain development.

The impact of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection during pregnancy on the developing fetal brain is poorly understood. Other antenatal infections such as influenza have been associated with adverse neurodevelopmental outcomes in offspring. Although vertical transmission has been rarely observed in SARS-CoV-2 to date, given the potential for profound maternal immune activation (MIA), impact on the developing fetal brain is likely. Here we review evidence that SARS-CoV-2 and other viral infections during pregnancy can result in maternal, placental, and fetal im

Hot spots in fetal human penile and clitoral development.

To better understand how the human fetal penis and clitoris grows and remodels, we undertook an investigation to define active areas of cellular proliferation and programmed cell death spatially and temporally during development of human fetal external genitalia from the indifferent stage (8 weeks) to 18 weeks of gestation. Fifty normal human fetal penile and clitoral specimens were examined using macroscopic imaging, scanning electron microscopy and immunohistochemical localization for the cellular proliferation and apoptotic markers, Ki67 and Caspase-3, respectively. A number of hot spots of

Prenatal stress: Effects on fetal and child brain development.

The impact of stress on brain health begins in the womb. Both animal and human studies have found that prenatal maternal stress affects the brain and behavior of the offspring. Stressful life events, exposure to a natural disaster, and symptoms of maternal anxiety and depression increase the risk for the child having a range of emotional, behavioral and/or cognitive problems in later life. These include depression, anxiety, Attention Deficit Hyperactivity Disorder (ADHD), and/or conduct disorders. There is an increased risk for other outcomes also, including preterm delivery and reduced telome

Satellite rats are redundant in embryo-fetal development studies.

Routinely in many laboratories, satellite rats are added to embryo-fetal development (EFD) studies for pharmaceuticals to assess toxicokinetic (TK) properties, because it is assumed that collection of multiple blood samples with relatively large volumes might affect the study outcome. With recent refinement of blood sampling techniques, this belief requires reevaluation. The current work showed successful implementation of jugular vein blood sampling in an EFD rat study without satellite animals, thereby reducing the number of rats in standard EFD studies for pharmaceuticals by 20%. Although n

Malformations of Cortical Development: From Postnatal to Fetal Imaging.

Abnormal fetal corticogenesis results in malformations of cortical development (MCD). Abnormal cell proliferation leads to microcephaly or megalencephaly, incomplete neuronal migration results in heterotopia and lissencephaly, neuronal overmigration manifests as cobblestone malformations, and anomalous postmigrational cortical organization is responsible for polymicrogyria and focal cortical dysplasias. MCD comprises various congenital brain disorders, caused by different genetic, infectious, or vascular etiologies and is associated with significant neurological morbidity. Although MCD are rar

Risk and protective factors in maternal-fetal attachment development.

Prenatal attachment can be described as the parents' emotions, perceptions and behaviors that are related to the fetus. This relationship has been described as the most basic form of the human intimacy and represents the earlier internalized representation of the fetus that both parents typically acquire and elaborate during pregnancy. The quality of the relationship between an infant and his or her parent is an important factor influencing the child's later development, both cognitive and emotional. There is evidence - even though yet unclear - that demographic, perinatal and psychological va

Epilepsy drugs and effects on fetal development: Potential mechanisms.

Approximately 1% of all pregnancies are in woman with epilepsy. Although, the majority of children born to women with epilepsy are normal, they are at increased risk for malformations. Notably, the teratogenicity of antiepileptic drugs is a well-defined subject. The incidence of major malformations in offspring of mothers with epilepsy who were treated with AEDs is higher than women with untreated epilepsy and in the general population. These malformations include spina bifida, cleft palate, limb reduction defects, cardiac abnormalities, hypospadias, and gastrointestinal atresia. The exact mec
