Cystathionine γ-Lyase Attenuates Vascular Smooth Muscle Cell Senescence via Foxm1-Gas1 Pathway to Mediate Arterial Stiffness.

Aims: Arterial stiffness, a hallmark of vascular aging, significantly contributes to hypertension and impaired organ perfusion. Vascular smooth muscle cell (VSMC) dysfunction, particularly VSMC senescence and its interaction with stiffness, is crucial in the pathogenesis of arterial stiffness. Although hydrogen sulfide (H2S) and its key enzyme cystathionine γ-lyase (CSE) are known to play roles in cardiovascular diseases, their effects on arterial stiffness are not well understood. Methods & Results: First, we observed a downregulation of CSE/H2S in the aortic media during biological aging and

Wnt Site Signaling Inhibitor Secreted Frizzled-Related Protein 3 Protects Mitral Valve Endothelium From Myocardial Infarction-Induced Endothelial-to-Mesenchymal Transition.

Background The onset and mechanisms of endothelial-to-mesenchymal transition (EndMT) in mitral valve (MV) leaflets following myocardial infarction (MI) are unknown, yet these events are closely linked to stiffening of leaflets and development of ischemic mitral regurgitation. We investigated whether circulating molecules present in plasma within days after MI incite EndMT in MV leaflets. Methods and Results We examined the onset of EndMT in MV leaflets from 9 sheep with inferior MI, 8 with sham surgery, and 6 naïve controls. Ovine MVs 8 to 10 days after inferior MI displayed EndMT, shown by in

Fork head box M1 regulates vascular endothelial growth factor-A expression to promote the angiogenesis and tumor cell growth of gallbladder cancer.

BACKGROUND: Gallbladder cancer (GBC) is an aggressive type of biliary tract cancer that lacks effective therapeutic targets. Fork head box M1 (FoxM1) is an emerging molecular target associated with tumor progression in GBC, and accumulating evidence suggests that vascular endothelial growth factor (VEGF) promotes various tumors by inducing neoangiogenesis. AIM: To investigate the role of FoxM1 and the angiogenesis effects of VEGF-A in primary GBC. METHODS: Using immunohistochemistry, we investigated FoxM1 and VEGF-A expression in GBC tissues, paracarcinoma tissues and cholecystitis tissues.

FOXM1 expression is induced by the brain microenvironment and supports CRC brain metastatic adaptation.

Colorectal cancer (CRC) is a leading cause of cancer-related mortality in the Western world. While brain metastasis (BMs) are relatively uncommon in CRC, they represent the fourth most frequent cause of BMs overall, and their occurrence is increasingly recognized as a complication in advanced CRC, associated with poor prognosis and limited treatment options. The brain microenvironment presents unique metabolic challenges, including low oxygen levels and restricted lipid availability. In order to survive in this hostile environment, CRC cells must acquire metabolic adaptations allowing them to

Plakophilin 3 drives acinar cell transformation and promotes cancer initiation and progression in pancreas.

We discovered that Plakophilin 3 (PKP3) was expressed only at the apical membrane of duct cells in the pancreas, and its expression was induced in acinar-to-ductal metaplasia (ADM), pancreatic intraepithelial neoplasia (PanIN), and pancreatic ductal adenocarcinoma (PDAC) cells. PKP3 was predominantly localized on the cell membrane in ADM and both in the membrane and cytoplasm in PanIN, while PDAC cells expressed PKP3 in the nucleus rather than in the cytoplasm or membrane. PKP3 expression transformed acinar cells into duct-like cells and promoted the malignant behavior of PDAC cells. Mechanist

Vagus-macrophage-hepatocyte link promotes post-injury liver regeneration and whole-body survival through hepatic FoxM1 activation.

The liver possesses a high regenerative capacity. Liver regeneration is a compensatory response overcoming disturbances of whole-body homeostasis provoked by organ defects. Here we show that a vagus-macrophage-hepatocyte link regulates acute liver regeneration after liver injury and that this system is critical for promoting survival. Hepatic Foxm1 is rapidly upregulated after partial hepatectomy (PHx). Hepatic branch vagotomy (HV) suppresses this upregulation and hepatocyte proliferation, thereby increasing mortality. In addition, hepatic FoxM1 supplementation in vagotomized mice reverses the

Deregulation of FoxM1b leads to tumour metastasis.

The forkhead box M1b (FoxM1b) transcription factor is over-expressed in human cancers, and its expression often correlates with poor prognosis. Previously, using conditional knockout strains, we showed that FoxM1b is essential for hepatocellular carcinoma (HCC) development. However, over-expression of FoxM1b had only marginal effects on HCC progression. Here we investigated the effect of FoxM1b expression in the absence of its inhibitor Arf. We show that transgenic expression of FoxM1b in an Arf-null background drives hepatic fibrosis and metastasis of HCC. We identify novel mechanisms of FoxM

Dexamethasone-Induced Liver Enlargement Is Related to PXR/YAP Activation and Lipid Accumulation but Not Hepatocyte Proliferation.

Dexamethasone (Dex), a widely prescribed anti-inflammatory drug, was reported to induce liver enlargement (hepatomegaly) in clinical practice and in animal models. However, the underlying mechanisms are not elucidated. Dex is a known activator of pregnane X receptor (PXR). Yes-associated protein (YAP) has been implicated in chemically induced liver enlargement. Here, the roles of PXR and YAP pathways were investigated in Dex-induced hepatomegaly. Upregulation of PXR downstream proteins, including cytochrome P450 (CYP) 3A11, 2B10, and organic anion transporter polypeptide 2 (OATP2), indicated P

Immune microenvironment and treatment response of dMMR gastric cancer: Heterogeneity and implications for immunotherapy.

Deficient mismatch repair (dMMR) in gastric cancer is associated with a high tumor mutation burden (TMB) and increased neoantigen load, which may enhance sensitivity to immunotherapy. Nevertheless, heterogeneous responses among patients with dMMR suggest complex tumor immune microenvironment (TIME) regulation. The present study aimed to integrate a case of dMMR gastric cancer with multi-cohort bioinformatic analyses to assess TIME determinants of immunotherapy outcomes, focusing on hematological immune cell subsets. Pre- and post-immunotherapy TIME changes were evaluated in a patient with dMMR

Targeting FoxM1 effectively retards p53-null lymphoma and sarcoma.

The forkhead box transcription factor FOXM1 is considered to be a promising target for cancer therapy. However, the significance of FOXM1 in tumors harboring mutation in p53, which is very common, is unclear. In this study, we investigated the efficacy of FoxM1 targeting in spontaneous p53-null tumors using genetic ablation as well as using a peptide inhibitor of FOXM1. We show that conditional deletion of FoxM1 inhibits growth of the p53-null thymic lymphoma and sarcoma cells. In addition, deletion of FoxM1 induces apoptotic cell death of the p53-null tumors, accompanied by reduced expression

The Preclinical Effects and Mechanisms of Biofield Therapy on Pancreatic Cancer Cell Growth and Metastasis.

BACKGROUND: The high mortality of pancreatic ductal adenocarcinoma (PDAC) underscores the need for novel treatments. We investigated the potential role of human biofield therapy (BT) in modifying tumorigenic processes in murine and human PDAC cells through a series of in vitro and in vivo studies. METHODS: Cell viability and ultrastructure changes were examined by PrestoBlue assay and Transmission Electron Microscope, respectively. Cell voltage potentials were assessed using DiBAC4 staining. Both FOXM1 CRISPR KO and Tet-on FOXM1 overexpressing PDAC cells were used for the mechanistic studies.

The role of forkhead box M1-methionine adenosyltransferase 2 A/2B axis in liver inflammation and fibrosis.

Methionine adenosyltransferase 2 A (MAT2A) and MAT2B are essential for hepatic stellate cells (HSCs) activation. Forkhead box M1 (FOXM1) transgenic mice develop liver inflammation and fibrosis. Here we examine if they crosstalk in male mice. We found FOXM1/MAT2A/2B are upregulated after bile duct ligation (BDL) and carbon tetrachloride (CCl4) treatment in hepatocytes, HSCs and Kupffer cells (KCs). FDI-6, a FOXM1 inhibitor, attenuates the development and reverses the progression of CCl4-induced fibrosis while lowering the expression of FOXM1/MAT2A/2B, which exert reciprocal positive regulation

Reprogramming tumour-associated macrophages in breast cancer via si-FOXM1-loaded lipid nanoparticles enhances immune checkpoint inhibitor efficacy.

BACKGROUND AND PURPOSE: The polarization state of tumour-associated macrophages (TAMs) plays a crucial role in breast cancer treatment outcomes, particularly in enhancing the efficacy of immune checkpoint inhibitors by transitioning TAMs from the M2 to M1 phenotype. This study aims to investigate whether delivering si-Forkhead Box M1 (FOXM1) efficiently into tumour cells via lipid nanoparticles (LNPs) can silence FOXM1 expression, up-regulate Kruppel-like factor 15 (KLF15) and thereby modulate TAM polarization to enhance the antitumor immune response against breast cancer. EXPERIMENTAL APPROA

Efficient Inhibition of FOXM1 Expression and Viability of High-grade Meningioma Cells by Domatinostat-mediated Dual Targeting of HDAC1 and HDAC2.

BACKGROUND/AIM: High-grade meningiomas are tumors with high recurrence rates and poor prognoses, and the development of effective pharmacological therapies remains a major challenge. Forkhead box protein M1 (FOXM1), a transcription factor known to play a key role in the malignancy of high-grade meningiomas, has gained attention as a potential therapeutic target. Class I histone deacetylase (HDAC) inhibitors have been shown to suppress FOXM1 expression and cell proliferation in various cancers; however, their efficacy in high-grade meningiomas remains unclear. The present study investigated the

Vorinostat Inhibition of FOXM1 Oncogenic Signaling Is Associated With the Downregulation of MYCN Transcription in Metastatic Retinoblastoma.

Vorinostat (suberoylanilide hydroxamic acid, SAHA) is a histone deacetylase (HDAC) class I/II inhibitor that has been evaluated in clinical trials for efficacy in pediatric cancers. Previous studies demonstrated cellular responses to SAHA in retinoblastoma (RB), a pediatric intraocular cancer. However, its mechanism of action remains incompletely understood. In this report, we demonstrate that SAHA inhibition of cell proliferation and induction of cell death was associated with the downregulation of the FOXM1 oncogene in the MYCN-driven RB1-/RB1-cell lines WERI-RB1 and Y79, of which Y79 exhibi

Targeting p53-Driven FOXM1 Suppresses Tumor Growth and Synergistically Sensitizes to Chemotherapy in Triple-Negative Breast Cancer Models.

Triple-negative breast cancer (TNBC) is characterized by a lack of estrogen, progesterone, and HER2 receptors; an aggressive phenotype; high rates of early relapse and metastasis; and the worst mortality rates among all breast cancer subtypes. Currently, there is no effective curative targeted therapy for TNBC and chemotherapy remains the primary treatment for TNBC. Therefore, there is a critical need to develop highly effective, novel therapies to improve patient survival. We previously validated FOXM1, a proto-oncogenic transcription factor, for the first time as a potential molecular target

Lian-Mei-Yin formula alleviates diet-induced hepatic steatosis by suppressing Yap1/FOXM1 pathway-dependent lipid synthesis.

Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disease, with a global prevalence of 25%. Patients with NAFLD are more likely to suffer from advanced liver disease, cardiovascular disease, or type II diabetes. However, unfortunately, there is still a shortage of FDA-approved therapeutic agents for NAFLD. Lian-Mei-Yin (LMY) is a traditional Chinese medicine formula used for decades to treat liver disorders. It has recently been applied to type II diabetes which is closely related to insulin resistance. Given that NAFLD is another disease involved in insulin resistance
