Vascular smooth muscle cell metabolic reprogramming and phenotypic remodeling in atherosclerosis.

Metabolic reprogramming of vascular smooth muscle cells (VSMC) is emerging as a central driver of atherosclerotic plaque heterogeneity and instability. VSMCs undergo phenotypic remodeling into osteogenic, macrophage-like, foam cell-like, or pro-inflammatory states through metabolic reprogramming, which actively drives vascular calcification, lipid accumulation, and extracellular matrix degradation. In this review, we summarize the various phenotypes of VSMCs observed during AS development and describe potential molecular pathways linking metabolic reprogramming to phenotypic remodeling. We hig

Bruceine A induces cell growth inhibition and apoptosis through PFKFB4/GSK3β signaling in pancreatic cancer.

Pancreatic cancer is one of the most aggressive cancers with a poor prognosis and 5-year low survival rate. In the present study, we report that bruceine A, a quassinoid found in Brucea javanica (L.) Merr. has a strong antitumor activity against human pancreatic cancer cells both in vitro and in vivo. Human proteome microarray reveals that 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 4 (PFKFB4) is the candidate target of bruceine A and both fluorescence measurement and microscale thermophoresis suggest bruceine A binds to PFKFB4. Bruceine A suppresses glycolysis by inhibiting PFKFB4, l

Non-complexed four cascade enzyme mixture: simple purification and synergetic co-stabilization.

Cell-free biosystems comprised of synthetic enzymatic pathways would be a promising biomanufacturing platform due to several advantages, such as high product yield, fast reaction rate, easy control and access, and so on. However, it was essential to produce (purified) enzymes at low costs and stabilize them for a long time so to decrease biocatalyst costs. We studied the stability of the four recombinant enzyme mixtures, all of which originated from thermophilic microorganisms: triosephosphate isomerase (TIM) from Thermus thermophiles, fructose bisphosphate aldolase (ALD) from Thermotoga marit

SOME PROPERTIES OF FRUCTOSE 1,6-DIPHOSPHATASE OF RAT LIVER AND THEIR RELATION TO THE CONTROL OF GLUCONEOGENESIS.

1. Fructose 1,6-diphosphatase has been purified tenfold from rat liver. The final preparation was not contaminated by either glucose 6-phosphatase or phosphofructokinase. The properties of the enzyme have been investigated in an attempt to define factors that could be of revelance to metabolic control of fructose 1,6-diphosphatase activity. 2. The metal ions Fe(2+), Fe(3+) and Zn(2+) inhibited the activity of fructose 1,6-diphosphatase even in the presence of an excess of mercaptoethanol; other metal ions tested had no effect. The inhibition produced by Zn(2+) was reversed by EDTA, but that pr

Discovery of FBP1 as novel therapeutic target and asiatic acid-hydrogen sulfide donors accelerate diabetic wound healing.

INTRODUCTION: Wound healing impairment is highly prevalent in diabetes and frequently progresses to serious complications, including refractory ulcers and necessitated amputations. RNA sequencing in methylglyoxal (MGO)-injured HaCaT cells implicated fructose-1,6-bisphosphatase 1 (FBP1) in suppressing keratinocyte proliferation and migration, identifying it as a potential therapeutic target. OBJECTIVES: This study aimed to validate FBP1 as a therapeutic target for diabetic wounds and evaluate asiatic acid (AA) and its novel hydrogen sulfide (H2S)-donor derivatives, designed to enhance efficacy

Coagulanolide modulates hepatic glucose metabolism in C57BL/KsJ-db/db mice.

Increased hepatic glucose output is one of the major causes of fasting hyperglycemia in diabetic patients. In this study, we investigated the mechanism of action of coagulanolide on hepatic glucose, regulating enzymes in type 2 diabetic C57BL/KsJ-db/db (db/db) mice. Coagulanolide is an active component of Withania coagulans fruit. Oral administration of coagulanolide for 3 weeks decreases fasting blood glucose and plasma insulin significantly, and it improves glucose tolerance in the db/db mice group. The enzyme activity and protein expression of glucokinase and pyruvate kinase was significant

Reduced cardiac fructose 2,6 bisphosphate increases hypertrophy and decreases glycolysis following aortic constriction.

This study was designed to test whether reduced levels of cardiac fructose-2,6-bisphosphate (F-2,6-P(2)) exacerbates cardiac damage in response to pressure overload. F-2,6-P(2) is a positive regulator of the glycolytic enzyme phosphofructokinase. Normal and Mb transgenic mice were subject to transverse aortic constriction (TAC) or sham surgery. Mb transgenic mice have reduced F-2,6-P(2) levels, due to cardiac expression of a transgene for a mutant, kinase deficient form of the enzyme 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFK-2) which controls the level of F-2,6-P(2). FBP1-Mediated Compartmentalization of Glycolytic Enzyme Complexes Suppresses PKM2 Function to Ameliorate MASLD Progression.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is driven by aberrant hepatocyte glycolytic reprogramming. Fructose-1,6-bisphosphatase 1 (FBP1), a key gluconeogenic enzyme, has poorly defined roles in MASLD progression. Here, we show that FBP1 is consistently downregulated during MASLD progression, with a progressive decline from MASL to MASH in clinical specimens and key preclinical models. Mechanistically, FBP1 directly interacts with glycolytic enzymes Pyruvate kinase M2 (PKM2), Enolase 1 (ENO1), and Lactate Dehydrogenase A (LDHA) to form compartmentalized complexes, suppre

Nuclear Translocation of PFKFB3 Promotes Disuse-Induced Muscle Atrophy via Scaffolding Nedd4-Mediated JunB Ubiquitination.

Disuse-induced muscle atrophy is characterized by coordinated metabolic remodeling and enhanced protein degradation, yet the molecular link between these processes remains unclear. Here, we identify 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (PFKFB3) as a critical regulator of this condition. Using a unilateral hindlimb immobilization mouse model, metabolomics, and functional assays in vivo and in vitro, we demonstrate that PFKFB3 is markedly upregulated during muscle atrophy and promotes myofiber wasting. Mechanistically, PFKFB3 predominantly localizes to the nucleus and functions

Nesfatin-1 as a crucial mediator of glucose homeostasis in the reptile, Hemidactylus flaviviridis.

Nesfatin-1 is a crucial regulator of energy homeostasis in mammals and fishes, however, its metabolic role remains completely unexplored in amphibians, reptiles, and birds. Therefore, present study elucidates role of nesfatin-1 in glucose homeostasis in wall lizard wherein fasting stimulated hepatic nucb2/nesfatin-1, glycogen phosphorylase (glyp), phosphoenolpyruvate carboxykinase (pepck), and fructose 1,6-bisphosphatase (fbp), while feeding upregulated pancreatic nucb2/nesfatin-1 and insulin, suggesting towards tissue-specific dual role of nesfatin-1 in glucoregulation. Metabolite interactions in the bacterial Calvin cycle and implications for flux regulation.

Metabolite-level regulation of enzyme activity is important for microbes to cope with environmental shifts. Knowledge of such regulations can also guide strain engineering for biotechnology. Here we apply limited proteolysis-small molecule mapping (LiP-SMap) to identify and compare metabolite-protein interactions in the proteomes of two cyanobacteria and two lithoautotrophic bacteria that fix CO2 using the Calvin cycle. Clustering analysis of the hundreds of detected interactions shows that some metabolites interact in a species-specific manner. We estimate that approximately 35% of interactin

Ocimum tenuiflorum mitigates iron-induced testicular toxicity via modulation of redox imbalance, cholinergic and purinergic dysfunctions, and glucose metabolizing enzymes activities.

Oxidative stress is a primary culprit in the pathophysiology of infertility conditions in males. This study investigated the effects of Ocimum tenuiflorum on redox imbalance, cholinergic and purinergic dysfunctions and glucose dysmetabolism in oxidative-mediated testicular toxicity using in vitro, ex vivo and in silico models. Induction of oxidative testicular injury was carried out by incubating normal testicular tissue with 0.1 mM FeSO4 and treated by co-incubating with different concentrations of O. tenuiflorum infusion for 30 min at 37°C. O. tenuiflorum displayed significant ferric reducin

Alteration of hepatic enzyme activities in obese KK mice treated with monosodium aspartate.

Changes in body weight, concentrations of urine glucose, blood glucose, plasma insulin and FFA and hepatic enzyme activities were investigated in KK and C57BL mice treated with monosodium-L-aspartate (MSA). MSA was administered subcutaneously to neonates at a dose of 4 mg/g body weight. The MSA-treated KK and C57BL mice were remarkably obese at 10 weeks of age. The average plasma insulin concentration in the control KK mice was 73.6 microU/ml, over 4 times higher than in the control C57BL mice. In the control KK mice, hepatic glucokinase (GK) activity was quite low, and fructose-1,6-bisphospha

[Enzyme activity of gluconeogenesis in dietary thiamine deficiency].

Activity of the main enzymes of gluconeogenesis under food thiamine deficiency was studied in tissues of satiated and 48-hour starved rats. Starvation of control rats (with no vitamin B1-deficiency) led to increased activity of glucose 6-phosphatase (G-6-P) in the liver, kidney and small intestinal mucosa, and of phosphoenolpyruvate carboxykinase (PEPCK) in the liver and kidneys. Fructose 1,6-diphosphatase (F-D-P) activity in the control animals was not changed in the liver and kidneys but decreased in the small intestinal mucosa. Starvation of the test animals (with vitamin B1-deficiency) was

[Hydrogen sulfide acted as a downstream signal was involved in the regulation of salicylic acid on photosynthesis of cucumber seedlings under low temperature and low light intensity].

Both salicylic acid (SA) and hydrogen sulfide (H2S) play an important role in regulating plant growth and development and physiological metabolism under abiotic stresses. As signal molecules, the interaction between them in regulating cucumber photosynthesis under low temperature and low light is still unclear. Here, we examined the regulation and interaction of SA and H2S on photosynthesis in cucumber seedlings under low temperature (8 ℃/5 ℃, day/night) and low light (100 μmol·m-2·s-1). Seedlings were foliar-sprayed with SA, sodium sulfide (NaHS, H2S donor), and their scavenger or biosynthesi
