Characterization of a heme-protein responsive to hypoxia in Paracoccidioides brasiliensis.

Oxygen is fundamental to the life of aerobic organisms and is not always available to Paracoccidioides cells. During the life cycle stages, reduced oxygen levels directly affect general metabolic processes and oxygen adaptation mechanisms may play a fundamental role on fungal ability to survive under such condition. Heme proteins can bind to oxygen and participate in important biological processes. Several fungi, including Paracoccidioides, express a heme-binding globin (fungoglobin - FglA) presumable to regulate fungal adaptation to hypoxia. However, the characterization of fungoglobin in Par

Transcription factor CCG-8 as a new regulator in the adaptation to antifungal azole stress.

Antifungal azoles are widely used for controlling fungal infections. Fungi are able to change the expression of many genes when they adapt to azole stress, and increased expression of some of these genes can elevate resistance to azoles. However, the regulatory mechanisms behind transcriptional adaption to azoles in filamentous fungi are poorly understood. In this study, we found that deletion of the transcription factor gene ccg-8, which is known to be a clock-controlled gene, made Neurospora crassa hypersensitive to azoles. A comparative genome-wide analysis of the responses to ketoconazole

Comparative Proteomic Analysis of Pleurotus ostreatus Reveals Great Metabolic Differences in the Cap and Stipe Development and the Potential Role of Ca2+ in the Primordium Differentiation.

Pleurotus ostreatus is a widely cultivated edible fungus around the world. At present, studies on the developmental process of the fruiting body are limited. In our study, we compared the differentially expressed proteins (DEPs) in the stipe and cap of the fruiting body by high-throughput proteomics. GO and pathway analysis revealed the great differences in the metabolic levels, including sucrose and starch metabolism, and sphingolipid signaling and metabolism, and the differences of 16 important DEPs were validated further by qPCR analysis in expression level. In order to control the cap and

Harnessing gene expression to identify the genetic basis of drug resistance.

The advent of cost-effective genotyping and sequencing methods have recently made it possible to ask questions that address the genetic basis of phenotypic diversity and how natural variants interact with the environment. We developed Camelot (CAusal Modelling with Expression Linkage for cOmplex Traits), a statistical method that integrates genotype, gene expression and phenotype data to automatically build models that both predict complex quantitative phenotypes and identify genes that actively influence these traits. Camelot integrates genotype and gene expression data, both generated under

Isolation and characterization of extragenic mutations affecting the expression of the uroporphyrinogen decarboxylase gene (HEM12) in Sacharomyces cerevisiae.

Uroporphyrinogen decarboxylase (Uro-d; EC 4.1.1.37), the fifth enzyme in the heme biosynthetic pathway, which catalyzes the sequential decarboxylation of uroporphyrinogen to coproporphyrinogen, is encoded by the HEM12 gene in Saccharomyces cerevisiae. The HEM12 gene is transcribed into a major short mRNA and a minor longer one, approximately 1.35 and 1.55 kb, respectively, in size, and that differ in the 5' untranslated region. "Uroporphyric" mutants, which have no mutations in the HEM12 gene but accumulate uroporphyrinogen, a phenotype characteristic of partial Uro-d deficiency, were investig

Molecular and genetic analysis of the Cryptococcus neoformans MET3 gene and a met3 mutant.

The Cryptococcus neoformans MET3 cDNA (encoding ATP sulfurylase) was cloned by complementation of the corresponding met3 mutation in Saccharomyces cerevisiae. Sequence analysis showed high similarity between the deduced amino acid sequence of the C. neoformans Met3p and other fungal ATP sulfurylases. A C. neoformans met3 mutant was made by targeted insertional mutagenesis, which had the expected auxotrophic phenotype, and reconstituted the met3 mutant to Met(+). In vitro, the C. neoformans met3 mutant had a substantial defect in melanin formation, significantly reduced growth rate, and greatly

Transcriptional Downregulation of Methanol Metabolism Key Genes During Yeast Death in Engineered Pichia pastoris.

Pichia pastoris possesses the unique ability to utilize methanol as its sole carbon source, which makes it a proper host for producing various high-value-added products via metabolic engineering. Nevertheless, cell death has been observed during the fermentation of modified P. pastoris, with limited literature elucidating the underlying causes and mechanisms. Understanding the death mechanisms during methanol-based fermentation is crucial for optimizing fermentation strategies, enhancing the accumulation of target products, and reducing production costs. Here, we first sought to eliminate the

The transcription factor Ndt80 does not contribute to Mrr1-, Tac1-, and Upc2-mediated fluconazole resistance in Candida albicans.

The pathogenic yeast Candida albicans can develop resistance to the widely used antifungal agent fluconazole, which inhibits ergosterol biosynthesis, by the overexpression of genes encoding multidrug efflux pumps or ergosterol biosynthesis enzymes. Zinc cluster transcription factors play a central role in the transcriptional regulation of drug resistance. Mrr1 regulates the expression of the major facilitator MDR1, Tac1 controls the expression of the ABC transporters CDR1 and CDR2, and Upc2 regulates ergosterol biosynthesis (ERG) genes. Gain-of-function mutations in these transcription factors

Contribution of MylA in Conidiogenesis, Conidia Germination, and beta-Glucan Biosynthesis in Aspergillus fumigatus.

The conidia of Aspergillus fumigatus are the primary infectious propagules responsible for initiating invasive aspergillosis, and their development and physiology are closely regulated by numerous transcription factors. Our previous study demonstrated that the MYB-like transcription factor MylA plays a critical role in asexual sporulation and conidial viability in Aspergillus nidulans. However, the function of MylA in A. fumigatus has not yet been investigated. This study aimed to investigate the role of MylA, a member of the MYB-like transcription factor family, in conidiogenesis, conidial vi

Multifaceted roles of CcGpi7, a phosphoethanolamine transferase, in physiology and virulence of Corynespora cassiicola.

Gpi7 gene, encoding the catalytic subunit of GPI ethanolamine-phosphate (Etn-P) transferase II, is primarily involved in the synthesis, maturation, and sorting of GPI-anchored proteins (GPI-APs), thereby playing a crucial part in cell wall functions and host-pathogen interactions. This study aimed to investigate the role of Gpi7 (here designated CcGpi7) in Corynespora cassiicola, a devastating fungal pathogen causing leaf spot in cucumber and many other cash crops. We systematically identified and inventoried 138 GPI-APs in C. cassiicola, followed by a detailed structural characterization of t

The Role of Zinc in Copper Homeostasis of Aspergillus fumigatus.

Copper is an essential metal ion that performs many physiological functions in living organisms. Deletion of Afmac1, which is a copper-responsive transcriptional activator in A. fumigatus, results in a growth defect on aspergillus minimal medium (AMM). Interestingly, we found that zinc starvation suppressed the growth defect of the Δafmac1 strain on AMM. In addition, the growth defect of the Δafmac1 strain was recovered by copper supplementation or introduction of the CtrC gene into the Δafmac1 strain. However, chelation of copper by addition of BCS to AMM failed to recover the growth defect o

tRNA hypomodification facilitates 5-fluorocytosine resistance via cross-pathway control system activation in Aspergillus fumigatus.

Increasing antifungal drug resistance is a major concern associated with human fungal pathogens like Aspergillus fumigatus. Genetic mutation and epimutation mechanisms clearly drive resistance, yet the epitranscriptome remains relatively untested. Here, deletion of the A. fumigatus transfer RNA (tRNA)-modifying isopentenyl transferase ortholog, Mod5, led to altered stress response and unexpected resistance against the antifungal drug 5-fluorocytosine (5-FC). After confirming the canonical isopentenylation activity of Mod5 by liquid chromatography-tandem mass spectrometry and Nano-tRNAseq, we p

Exploring the interaction between endornavirus and Sclerotinia sclerotiorum: mechanisms of phytopathogenic fungal virulence and antivirus.

Hypovirulence-associated mycoviruses have the potential as biocontrol agents for plant fungal disease management, and exploration of the interactions between these mycoviruses and phytopathogenic fungi can provide opportunities to elucidate the underlying mechanisms of hypovirulence and antiviruses. We previously found that Sclerotinia sclerotiorum endornavirus 3 (SsEV3), belonging to the genus Betaendornavirus within the family Endornaviridae, confers hypovirulence on the phytopathogenic fungus Sclerotinia sclerotiorum, but the underlying mechanisms remains unclear. In this study, we found th

Overlapping coactivator function is required for transcriptional activation by the Candida glabrata Pdr1 transcription factor.

Azole resistance in the pathogenic yeast Candida glabrata is a serious clinical complication and increasing in frequency. The majority of resistant organisms have been found to contain a substitution mutation in the Zn2Cys6 zinc cluster-containing transcription factor Pdr1. These mutations typically lead to this factor driving high, constitutive expression of target genes like the ATP-binding cassette transporter-encoding gene CDR1. Overexpression of Cdr1 is required for the observed elevated fluconazole resistance exhibited by strains containing one of these hyperactive PDR1 alleles. While th

The conserved protein DopA is required for growth, drug tolerance and virulence in Aspergillus fumigatus.

The majority of Aspergillus fumigatus reproduction occurs asexually, with large numbers of conidiophores producing small hydrophobic conidia dispersed aerially. When healthy hosts inhale conidia, the mucosal cilia and phagocytosis by the innate immune system can remove them. However, in immunocompromised hosts, the conidia are not removed, which allows them to germinate, forming mycelium that invades host tissues and causes disease. Previously we isolated a white A. fumigatus A1j strain incapable of producing conidia and screened several genes (including dopA) with significant expression diffe

Only one of three hydrophobins (Hyd1-3) contributes to conidial hydrophobicity and insect pathogenicity of Metarhizium robertsii.

Class I/II hydrophobins constitute a family of small amphiphilic proteins that mediate cell hydrophobicity and adhesion to host or substrata and have pleiotropic effects in filamentous fungi. Here we report that only class I Hyd1 is essential for conidial hydrophobicity and insect pathogenicity among three hydrophobins (Hyd1-3) characterized in Metarhizium robertsii, an insect-pathogenic fungus. Aerial conidiation levels of three Δhyd1 mutants were much more reduced in 5-day-old cultures than in 7-day-old cultures, which were wettable (hydrophilic), but restored to a wild-type level in 15-day-

Candida auris: The Canary in the Mine of Antifungal Drug Resistance.

Candia auris has rapidly emerged as a fungal pathogen of worldwide importance. Its impact on global health is due in large part to the high frequency of multidrug resistance among C. auris clinical isolates. Although C. auris resistance to amphotericin B is an unusual feature of this organism, its notoriety should also serve as notice that other more commonly encountered fungal pathogens also show multidrug resistance. Here, we review the epidemiology and mechanisms of C. auris resistance and discuss why the emergence of C. auris provides justification for increased research into mechanisms of

A conserved Sas10/C1D domain protein, CaLrp1, is a critical regulator of hyphal development and pathogenicity in Candida albicans.

BACKGROUND: Candida albicans represents the predominant opportunistic fungal pathogen in clinical contexts, with its virulence contingent upon multilayered regulatory networks. CaLrp1 (orf19.5067) encodes a conserved Sas10/C1D domain-containing protein, though its precise biological role has remained uncharacterized. RESULTS: CaLrp1 deleted and complemented strains were constructed to systematically investigate its function in C. albicans. Abrogation of CaLrp1 resulted in delayed hyphal induction, a loss of rugose colony morphology, and a pronounced reduction in invasive growth on solid media

Dual regulatory roles of the zinc cluster transcription factor MrFTRP1 in development, stress tolerance, and virulence of Metarhizium robertsii.

The entomopathogenic fungus Metarhizium robertsii shows significant potential as a multifunctional biocontrol agent. The Zn(II)₂Cys₆ (C6) transcription factors (TFs) family is the largest class of transcription factors in M. robertsii, yet their functions are generally underexplored. In this study, we functionally characterized a putative C6 TF, designated MrFTRP1. Deletion of Mrftrp1 reduced conidial yield, which correlated with the downregulation of key central developmental regulators, including brlA, abaA, and wetA. Moreover, ΔMrftrp1 exhibited attenuated virulence against Galleria mellone

The zinc cluster transcription factor Znc1 regulates Rta3-dependent miltefosine resistance in Candida albicans.

UNLABELLED: Zinc cluster transcription factors (ZCFs) are a family of transcription regulators that are almost exclusively found in the fungal kingdom. Activating mutations in the ZCFs Mrr1, Tac1, and Upc2 frequently cause acquired resistance to the widely used antifungal drug fluconazole in the pathogenic yeast Candida albicans. Similar to a hyperactive Tac1, a constitutively active form of the ZCF Znc1 causes increased fluconazole resistance by upregulating the multidrug efflux pump-encoding gene CDR1. Hyperactive forms of both Tac1 and Znc1 also cause overexpression of RTA3, which encodes a
