History of near-isogenic lines carrying genes for resistance to bacterial blight and blast in rice (Oryza sativa L.).

Bacterial blight and blast, caused by Xanthomonas oryzae pv. oryzae and Magnaporthe oryzae, respectively, are the two most devastating diseases of rice (Oryza sativa L.). Although the diseases were first documented in the 17th-19th centuries, systematic research on their pathology and host resistance began in the early 20th century and was significantly accelerated following the development of near-isogenic lines (NILs) carrying single resistance genes in universally susceptible backgrounds in the 1980s. Sets of NILs for bacterial blight (IRBB) and International Rice Research Institute (IRRI)-

Revealing the critical role of rare bacterial communities in shaping antibiotic resistance genes in saline soils through metagenomic analysis.

Salinity is considered one of the basic abiotic factors influencing the diversity and distribution of antibiotic resistance genes (ARGs) in soils, yet little is known about the distribution and driving factors of ARGs in naturally saline soils. In this study, metagenomic analysis was conducted to explore the intricate dynamics among soil salinity, microbial community structure and ARGs propagation, with a particular focus on the key contribution of rare potential-hosts of ARGs in light and heavy saline soils. The findings revealed that salinity was significantly negatively correlated with the

Melon CmRLCK VII-8 kinase genes CmRLCK27, CmRLCK30 and CmRLCK34 modulate resistance against bacterial and fungal diseases in Arabidopsis.

Receptor-like cytoplasmic kinases (RLCKs) represent a distinct class of receptor-like kinases crucial for various aspects of plant biology, including growth, development, and stress responses. This study delves into the characterization of RLCK VII-8 members within cucurbits, particularly in melon, examining both structural features and the phylogenetic relationships of these genes/proteins. The investigation extends to their potential involvement in disease resistance by employing ectopic overexpression in Arabidopsis. The promoters of CmRLCK VII-8 genes harbor multiple phytohormone- and stre

The REP2 repeats of the genome of Neisseria meningitidis are associated with genes coordinately regulated during bacterial cell interaction.

Interaction with host cells is essential in meningococcal pathogenesis especially at the blood-brain barrier. This step is likely to involve a common regulatory pathway allowing coordinate regulation of genes necessary for the interaction with endothelial cells. The analysis of the genomic sequence of Neisseria meningitidis Z2491 revealed the presence of many repeats. One of these, designated REP2, contains a -24/-12 type promoter and a ribosome binding site 5 to 13 bp before an ATG. In addition most of these REP2 sequences are located immediately upstream of an ORF. Among these REP2-associate

Global Analysis of the Co-Occurrence of Antimicrobial and Metal Resistance Genes in Bacterial Genomes From Dairy-Isolated Pathogens.

Metal contamination in dairy production can drive antimicrobial resistance co-selection independently of antibiotic use, yet the genomic architecture linking metal resistance and antimicrobial resistance across dairy-isolated pathogens remains largely uncharacterized globally. We analysed 3303 genomes from dairy environments across 52 countries, creating the first genome-resolved map of co-occurring resistance determinants in this critical agricultural interface. Metal/stress resistance genes were present at levels comparable to AMR genes (28.85% of isolates showed metal dominance), with arsen

Genome-wide characterization of the sugar transporter protein family identifies candidate genes for bacterial wilt resistance breeding in tobacco.

Sugar transporter proteins (STPs) play pivotal roles in hexose allocation and plant stress responses. However, systematic characterization of the STP family in tobacco (Nicotiana tabacum) and its involvement in Ralstonia solanacearum resistance remains unclear. In this study, 37 NtSTP genes were identified and classified into six groups, with Group VI being the most conserved and Group V exhibiting dicot-specific expansion. Gene structure and conserved motif analyses revealed that most NtSTP members possess the typical MFS_STP domain, although variations in exon-intron organization and motif c

Studies on the airborne bacterial communities and antimicrobial resistance genes in duck houses based on metagenome and PCR analysis.

The threat of antimicrobial resistance (AMR) is on the rise globally, especially with the development of animal husbandry and the increased demand for antibiotics. Livestock and poultry farms, as key sites for prevalence of antibiotic-resistant bacteria (ARB), can spread antimicrobial resistance genes (ARGs) through microbial aerosols and affect public health. In this study, total suspended particulate matter (TSP) and airborne culturable microorganisms were collected from duck houses in Tai'an, Shandong Province, and the bacterial communities and airborne ARGs were analyzed using metagenomics

Metagenomic characterization of bacterial community and antibiotic resistance genes found in the mass transit system in Seoul, South Korea.

Mass transit systems, including subways and buses, are useful environments for studying the urban microbiome, as the vast majority of populations in urban areas use public transportation. Microbial communities in urban environments include both human- and environment-associated bacteria that play roles in health and pathogen transmission. In this study, we used shotgun metagenomic sequencing to profile microbial communities sampled from various surfaces found in subway stations and bus stops within the Seoul mass transit system. The metagenomic approach and network analysis were used to invest

Integrated metagenomic and metatranscriptomic analyses of ultraviolet disinfection effects on antibiotic resistance genes and bacterial communities during wastewater treatment.

Ultraviolet (UV) disinfection is now widely implemented in wastewater treatment plants (WWTPs) worldwide, but its effect on antibiotic resistome of the surviving bacteria remains unclear. In this study, we employed high-throughput sequencing-based metagenomic and metatranscriptomic approaches to comprehensively elucidate the effects of UV disinfection on the shifts of bacterial community and antibiotic resistance genes (ARGs) on both DNA and mRNA levels in one WWTP. Metagenomic analyses revealed an insignificant change in the bacterial community after UV disinfection, while metatranscriptomic

Temporal changes of NF-κB signaling pathway genes in bacterial stimulated whole blood- a host mechanism associated with sepsis.

Bacterial sepsis affects both neonates and adults worldwide. There is no specific anti-sepsis treatment. Disease management mainly depends on early diagnosis. The gold standard blood culturing method is routinely practiced; it requires 24-48 h for confirmation. Understanding the disease mechanism may help in the early detection of sepsis. We studied the temporal change in NF-kB pathway genes in adult whole blood upon bacterial stimulations across time intervals (2-6 h). Four experimental conditions were investigated (1: Gram-positive, 2: Gram-negative, 3: Gram-positive + Gram-negative stimulat

Targeting novel genes for simultaneous detection of five fungal and bacterial agents from BAL samples using multiplex PCR assay.

The main purpose of our study was to evaluate multiplex PCR assay targeting novel genes for detection of five fungal and bacterial agents in BAL samples; because many fungi and bacteria that cause respiratory infections have similar clinical symptoms, diagnosing and differentiating them are therefore essential to controlling and treating them. A total of 100 BAL specimens from a mycobacterium and mycology laboratory were collected from patients suspected of having TB or other respiratory diseases. Novel DNA targets for Aspergillus, Nocardia, Cryptococcus, and Streptomyces were found using modi

Glycosylation genes expressed in seam cells determine complex surface properties and bacterial adhesion to the cuticle of Caenorhabditis elegans.

The surface of the nematode Caenorhabditis elegans is poorly understood but critical for its interactions with the environment and with pathogens. We show here that six genes (bus-2, bus-4, and bus-12, together with the previously cloned srf-3, bus-8, and bus-17) encode proteins predicted to act in surface glycosylation, thereby affecting disease susceptibility, locomotory competence, and sexual recognition. Mutations in all six genes cause resistance to the bacterial pathogen Microbacterium nematophilum, and most of these mutations also affect bacterial adhesion and biofilm formation by Yersi

The Potential Role of the Bacterial Persister Formation Gene ptsH in Hypervirulence Development in Klebsiella pneumoniae.

Persistence - i.e., the ability to exist in a metabolically inactive form - allows bacteria to accumulate genetic advantages. The evolution of the pathogenic potential of Klebsiella pneumoniae has led to the emergence of strains simultaneously characterized by increased aggressiveness (virulence) and prolonged survival in the host organism. This combination of properties contributes to the emergence of "superbugs," necessitating the search for specific markers that would make it possible to prevent the spread of highly adaptive clones. Hypervirulent K. pneumoniae (hvKp) strains represent a gro

Bacterial metabolic signatures in MASLD predicted through gene-centric studies in stool metagenomes.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a multifactorial condition in which the gut microbiome (GM) plays a central role. However, taxonomic associations derived from 16S ribosomal RNA (rRNA) gene studies have yielded inconsistent results, likely due to limited resolution and functional redundancy across taxa. We aimed to identify robust, functionally relevant microbial markers of MASLD using metagenomics and gene-centric profiling. METHODS: We analyzed 554 fecal metagenomes from three independent cohorts. Sequencing reads were quality-controlled and tax

nspA gene as a specific genetic marker for detection of Neisseria meningitidis causing bacterial meningitis.

Bacterial meningitis caused by Neisseria meningitidis which causes human brain meninges damage, is generally diagnosed from patient cerebrospinal fluid through microscopy, immunological assays, biochemical test, PCR, microarray and biosensors. However, these methods are expensive, time-consuming or non-confirmatory due to certain limitations. A quick PCR based method was developed for detection of bacterial meningitis caused by N. meningitidis using specific primers based on amplification of virulence nspA (Neisseria surface protein A) gene partial sequence (202 bp). The nspA gene amplicon cou
