Validation of the Q87.11 ICD Code for Prader-Willi Syndrome.

BACKGROUND: Prader-Willi syndrome (PWS) is a rare genetic disorder that is identified by the ICD-10 code Q87.11. Although this code has been used in several large studies, its diagnostic validity has not been evaluated. This study assessed the accuracy of the Q87.11 code for identifying individuals with PWS. METHODS: The electronic health record (EHR) of a large health system was reviewed for patients with the Q87.11 code. Additionally, a 10% random sample of patients with free-text evidence of possible PWS but no code was reviewed. All charts were assessed for PWS using expert review. Diagno

The role of chronogenetics and of gemellology in the prevention of genetic morbidity.

The prevention of hereditary diseases after birth is essentially based on the detection of these diseases at their preclinical level. This may be made possible through a careful pedigree examination and possible identification of a "genealogical alarm", as well as a detailed clinical examination of the subject and possible identification of preclinical signs of the pathogenesis, i.e., of a "clinical alarm". The respective roles of chronogenetics and of gemellology in this approach are reviewed.

Protecting cells at the genetic level and simulating unauthorized access via a biohackathon.

The protection of high-value cell lines (assets) relies on physical security by limiting access to samples. We present a cybersecurity-inspired platform that protects biological assets at the genetic level. This technology uses a permutation lock design where an asset can only be decrypted using an authentication code r from a search space composed of n objects on a defined keypad. Here, the genetic asset is designed as a scrambled DNA sequence, and the code is a temporal pattern of small molecules that regulate sets of recombinases that can unscramble a DNA sequence into the desired final seq

Epigenetic alterations in rheumatoid arthritis fibroblast-like synoviocytes.

Rheumatoid arthritis is an immune-mediated disease that primarily affects diarthrodial joints. Susceptibility and severity of this disease are influenced by nongenetic factors, such as environmental stress, suggesting an important role of epigenetic changes. In this review, we summarize the epigenetic changes (DNA methylation, histone modification and miRNA expression) in fibroblast-like synoviocytes, which are the joint-lining mesenchymal cells that play an important role in joint inflammation and damage. We also review the effects of these epigenetic changes on rheumatoid arthritis pathogene

Epigenetic changes and their implications in autoimmune hepatitis.

BACKGROUND: The genetic risk of autoimmune hepatitis is insufficient to explain the observed risk, and epigenetic changes may explain disparities in disease occurrence in different populations within and between countries. The goal of this review was to examine how epigenetic changes induced by the environment or inherited as a phenotypic trait may affect autoimmune hepatitis and be amenable to therapeutic intervention. MATERIALS AND METHODS: Pertinent abstracts were identified in PubMed by multiple search terms. The number of abstracts reviewed was 1689, and the number of full-length article

The functional landscape of coding variation in the familial hypercholesterolemia gene LDLR.

Variants in the familial hypercholesterolemia gene LDLR-the most important genetic driver of cardiovascular disease-can raise circulating low-density lipoprotein (LDL) cholesterol concentrations and increase the risk of premature atherosclerosis. Definitive classifications are lacking for nearly half of clinically encountered LDLR missense variants, limiting interventions that reduce disease burden. We tested the impact of ~17,000 (nearly all possible) LDLR coding variants on both LDLR cell-surface abundance and LDL uptake, yielding sequence-function maps that recapitulate known biochemistry,

Epigenetics of atopic dermatitis: pathogenesis and therapeutic prospects.

Atopic dermatitis (AD) is a chronic inflammatory skin disease that poses a serious threat to human physical and mental health. Its main clinical features include itching, lichenification, and dry skin. The etiology of AD is not yet fully understood, and its pathogenesis is only partially elucidated. An increasing body of evidence suggests that this refractory disease is associated with epigenetic alterations, including specific modifications in immune cells and keratinocytes that play a key role in the pathogenesis of AD. The latest advances in epigenetic research have provided new insights in

Management strategies for fluoride toxicity and the critical role of epigenetic therapy.

Fluoride is an essential trace element primarily known for its role in preventing dental caries; however, excessive exposure can lead to adverse health effects. Emerging research suggests possible links between fluorosis and neurodevelopmental disorders, including reduced cognitive function and increased prevalence of ADHD. Its impact on liver health such as hepatotoxicity and hormone-related infertility also raises concern. Moreover, high fluoride exposure has been associated with various toxicities, including skeletal fluorosis, reproductive toxicity, and neurotoxicity, potentially driven by

Historical Roles of Selenium and Selenoproteins in Health and Development: The Good, the Bad and the Ugly.

Selenium is a fascinating element that has a long history, most of which documents it as a deleterious element to health. In more recent years, selenium has been found to be an essential element in the diet of humans, all other mammals, and many other life forms. It has many health benefits that include, for example, roles in preventing heart disease and certain forms of cancer, slowing AIDS progression in HIV patients, supporting male reproduction, inhibiting viral expression, and boosting the immune system, and it also plays essential roles in mammalian development. The role of epigenetics and non-coding RNAs in autophagy: A new perspective for thorough understanding.

Autophagy is a major self-degradative intracellular process required for the maintenance of homeostasis and promotion of survival in response to starvation. It plays critical roles in a large variety of physiological and pathological processes. On the other hand, aberrant regulation of autophagy can lead to various cancers and neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and Crohn's disease. Emerging evidence strongly supports that epigenetic signatures, related non-coding RNA profiles, and their cross-talking are significantly associated with the control of aut

Epigenetic Regulation of Spermatogonial Stem Cell Homeostasis: From DNA Methylation to Histone Modification.

Spermatogonial stem cells(SSCs)are the ultimate germline stem cells with the potential of self-renewal and differentiation, and a dynamic balance of SSCs play an essential role in spermatogenesis. During the gene expression process, genomic DNA and nuclear protein, working together, contribute to SSC homeostasis. Recently, emerging studies have shown that epigenome-related molecules such as chromatin modifiers play an important role in SSC homeostasis through regulating target gene expression. Here, we focus on two types of epigenetic events, including DNA methylation and histone modification,

Epigenetic dysfunctional diseases and therapy for infection and inflammation.

Even though the discovery of the term 'epigenetics' was in the 1940s, it has recently become one of the most promising and expanding fields to unravel the gene expression pattern in several diseases. The most well studied example is cancer, but other diseases like metabolic disorders, autism, or inflammation-associated diseases such as lung injury, autoimmune disease, asthma, and type-2 diabetes display aberrant gene expression and epigenetic regulation during their occurrence. The change in the epigenetic pattern of a gene may also alter gene function because of a change in the DNA status. Co

Hyperplastic callus formation in osteogenesis imperfecta.

Osteogenesis imperfecta, an inherited disorder of connective tissues, affects roughly (OI) 4000 people in Germany (11). The main clinical symptoms are fragile bones, progressing skeletal deformities, generalized osteoporosis and short stature. Incidentally, the clinical manifestations can range from perinatal lethal forms to phenotypical normal adults. In many instances the underlying causes of the disease are mutations in gene coding for collagen I, the predominant protein in most connective tissues. Fracture healing is usually not impaired, although in a unique group of OI-patients, a tumor-

Measuring histone modifications across different cell types in the central nervous system.

This chapter explores recent advancements in single-cell epigenomics, with a focus on techniques for studying histone modifications and transcription factors at unprecedented resolution. Traditional bulk methods, like chromatin immunoprecipitation sequencing, have provided valuable insights but lack the ability to resolve subtle variations and rare cell types within complex tissues. Emerging single-cell techniques, such as single-cell cleavage under targets and tagmentation (scCUT&Tag), overcome these limitations by enabling precise mapping of chromatin modifications at the individual cell lev

Codepoietic biological evolution: From the origin of life to eukaryogenesis.

Evolution is an intrinsic property of autopoietic systems performing natural computation, which is based on an internal formal self-description representing the arrangement of digital and analog coding structures. Autopoietic systems are capable of assigning new values to previously unproven (ambiguous) statements, which occurs, in particular, in conditions beyond the limits of adaptability. This process, defined by Marcello Barbieri as codepoiesis, occurs through the introduction of new codes or the rearrangement of existing ones, increasing the computing power of biological systems. Formally

Histone Modifications and DNA Methylation in Psoriasis: A Cellular Perspective.

In recent years, epigenetic modifications have attracted significant attention due to their unique regulatory mechanisms and profound biological implications. Acting as a bridge between environmental stimuli and changes in gene activity, they reshape gene expression patterns, providing organisms with regulatory mechanisms to respond to environmental changes. A growing body of evidence indicates that epigenetic regulation plays a crucial role in the pathogenesis and progression of psoriasis. A deeper understanding of these epigenetic mechanisms not only helps unveil the molecular mechanisms und

Vitiligo and Epigenetics: From Pathogenesis to Clinical Applications.

Vitiligo is a multifactorial chronic depigmentary disorder characterised by the loss of functional melanocytes. Its pathogenesis involves complex interactions among genetic predisposition, environmental factors, and autoimmune dysregulation. In recent years, extensive research has highlighted the critical role of epigenetic regulation in vitiligo development, including aberrant DNA methylation, dysregulated histone modifications, and non-coding RNA (ncRNA) expression disturbances. These abnormal epigenetic modifications contribute to disease progression by disrupting melanogenesis, promoting o

Uncoupling histone modification crosstalk by engineering lysine demethylase LSD1.

Biochemical crosstalk between two or more histone modifications is often observed in epigenetic enzyme regulation, but its functional significance in cells has been difficult to discern. Previous enzymatic studies revealed that Lys14 acetylation of histone H3 can inhibit Lys4 demethylation by lysine-specific demethylase 1 (LSD1). In the present study, we engineered a mutant form of LSD1, Y391K, which renders the nucleosome demethylase activity of LSD1 insensitive to Lys14 acetylation. K562 cells with the Y391K LSD1 CRISPR knockin show decreased expression of a set of genes associated with cell
