Thyroid Dysfunction and Alopecia Areata: A Genetic Prediction Causality Analysis Study.

Источник пометил публикацию как отозванную

BACKGROUND: Observational studies have suggested a correlation between alopecia areata (AA) and thyroid dysfunction (TD). However, the causal relationship between AA and TD remains uncertain. The purpose of this study is to investigate the causal relationship between these two conditions. Understanding the potential causal relationship between AA and TD is valuable for elucidating the pathogenesis of AA and for designing innovative methods to prevent and treat AA and its related complications. METHODS: All data for this two-sample Mendelian randomization (MR) study were sourced from public da

Prolactin - a pleiotropic factor in health and disease.

The principal role of prolactin in mammals is the regulation of lactation. Prolactin is a hormone that is mainly synthesized and secreted by lactotroph cells in the anterior pituitary gland. Prolactin signalling occurs via a unique transmembrane prolactin receptor (PRL-R). The structure of the PRL-R has now been elucidated and is similar to that of many biologically fundamental receptors of the class 1 haematopoietic cytokine receptor family such as the growth hormone receptor. The PRL-R is expressed in a wide array of tissues, and a growing number of biological processes continue to be attrib

MVMRmode: Introducing an R package for plurality valid estimators for multivariable Mendelian randomisation.

BACKGROUND: Mendelian randomisation (MR) is the use of genetic variants as instrumental variables. Mode-based estimators (MBE) are one of the most popular types of estimators used in univariable-MR studies and is often used as a sensitivity analysis for pleiotropy. However, because there are no plurality valid regression estimators, modal estimators for multivariable-MR have been under-explored. METHODS: We use the residual framework for multivariable-MR to introduce two multivariable modal estimators: multivariable-MBE, which uses IVW to create residuals fed into a traditional plurality vali

Multiple Sclerosis and Hematologic Malignancies: A Bidirectional Mendelian Randomization Study.

Observational studies have reported associations between multiple sclerosis (MS) and hematologic malignancies (HM), but findings remain inconsistent. We conducted a bidirectional two-sample Mendelian randomization (MR) analysis using publicly available genome-wide association summary statistics for MS (n = 115,803) and HM (n = 218,792). Primary causal estimates were obtained with the inverse-variance-weighted (IVW) estimator, complemented by prespecified sensitivity analyses for heterogeneity and pleiotropy. Genetically proxied liability to MS was associated with higher odds of leukemia (unspe

Causal relationship between Alzheimer's disease and unstable angina: a bidirectional Mendelian randomization analysis.

BACKGROUND: Research from observational studies has demonstrated a link between Alzheimer's disease (AD) and a higher risk of cardiovascular disease (CVD). Uncertainty surrounds the exact genetic cause of AD and coronary heart disease, particularly unstable angina (UA). Mendelian randomization (MR) analysis was used to examine the causal genetic link between AD and UA to evaluate the impact of AD on UA. METHODS: The purpose of the bidirectional MR analysis was to investigate the link between exposure and illness causation. Genetic instrumental variables for AD were obtained from European popu

Causal associations of white blood cell subtypes with age-related hearing loss: A Mendelian randomization study.

While elevated white blood cell (WBC) counts are associated with auditory decline, the causal nature of this relationship remains elusive due to potential residual confounding. We utilized 2-sample Mendelian randomization (MR) to assess potential causal relationships between 5 genetically predicted WBC subtypes and the risk of age-related (ARHL) and sensorineural hearing loss. Two-sample MR analyses were conducted using summary statistics from large-scale genome-wide association studies. Causal estimates were assessed via inverse variance weighted, weighted median, weighted mode, and MR-Egger

Genetically predicted circulating concentrations of micronutrients and risk of autoimmune thyroiditis: a Mendelian randomized study.

BACKGROUND: Micronutrients play pivotal roles in modulating various aspects of the immune response. However, the existing literature on the association between micronutrients and autoimmune thyroiditis (AIT) remains limited and contentious. To address this gap, we conducted Mendelian randomization (MR) to investigate potential links between genetically predicted concentrations of six micronutrients (Copper (Cu), Iron (Ir), Calcium (Ca), Vitamin D (VD), Vitamin C (VC), Zinc (Zn)) and the risk of AIT. METHOD: Utilizing summary statistics from genome-wide association studies (GWAS) in individual

Efficient Mendelian randomization analysis with self-adaptive determination of sample structure and multiple pleiotropic effects.

Mendelian randomization (MR) has emerged as a highly valuable tool for inferring the causal effects of exposures on outcomes in observational studies using genetic variants, typically single-nucleotide polymorphisms (SNPs), as instrumental variables (IVs). Standard MR typically involves three steps: inputs of genome-wide association studies (GWASs) for both exposure and outcome, determination of IVs, and inference of causal effects. However, existing methods fail to simultaneously account for characteristics of GWAS data, uncertainty surrounding the validity of SNPs as IVs, and efficiency of e

Association between rheumatoid arthritis-interstitial lung disease and lung cancer: a two -sample Mendelian randomization study with East Asian genetic instruments.

OBJECTIVES: Epidemiological studies consistently report an increased risk of lung cancer among rheumatoid arthritis patients. Proposed risk factors include male sex, smoking, and rheumatoid arthritis-associated interstitial lung disease. However, whether a direct genetic causal link exists between rheumatoid arthritis-associated interstitial lung disease and lung cancer remains unexplored. We used Mendelian randomization with East Asian genetic instruments to evaluate this relationship. METHODS: We conducted a two-sample Mendelian randomization in which genetic instruments for rheumatoid arth

Common genetic mechanisms between obesity and COVID-19 severity: unravelling pleiotropic loci and biological pathways.

COVID-19 and obesity are complex conditions marked by immune and metabolic dysfunction, with the former still ranking among the leading causes of death from infectious diseases worldwide and the latter reaching pandemic proportions. Clinical evidence consistently shows that obesity increases the risk of severe COVID-19, yet the biological mechanisms underlying this association remain unclear. Given their physiological and clinical overlap, they may share genetic pathways. We investigated genetic variants jointly associated with body mass index (BMI) and COVID-19 using publicly available genome

Identification of pleiotropy at the gene level between psychiatric disorders and related traits.

Major mental disorders are highly prevalent and make a substantial contribution to the global disease burden. It is known that mental disorders share clinical characteristics, and genome-wide association studies (GWASs) have recently provided evidence for shared genetic factors as well. Genetic overlaps are usually identified at the single-marker level. Here, we aimed to identify genetic overlaps at the gene level between 7 mental disorders (schizophrenia, autism spectrum disorder, major depressive disorder, anorexia nervosa, ADHD, bipolar disorder and anxiety), 8 brain morphometric traits, 2

Immune cell phenotypes in the context of diabetic nephropathy: A Mendelian randomization analysis.

To clarify whether specific immune cell phenotypes exert a causal influence on diabetic nephropathy (DN), we conducted a Mendelian randomization (MR) investigation using large-scale genetic datasets. Summary-level data for immune cell traits and DN were obtained from recent genome-wide association studies and the FinnGen consortium. Genetic instruments were selected under the core MR assumptions, ensuring strong relevance and minimal confounding. The inverse-variance weighted approach served as the primary analytical framework. Complementary analyses, including MR-Egger regression, Cochran Q t

Haplotype analysis reveals pleiotropic disease associations in the HLA region.

The human leukocyte antigen (HLA) region plays an important role in human health through its involvement in immune cell recognition and maturation. While genetic variation in the HLA region is associated with many diseases, the pleiotropic patterns of these associations have not been systematically investigated. Here, we developed a haplotype approach to investigate disease associations phenome wide for 412,181 Finnish individuals and 2,459 diseases. Across the 1,035 diseases with a genome-wide association study association, we found a 17-fold average per-SNP enrichment of hits in the HLA regi

Leveraging pleiotropy identifies common-variant associations with selective IgA deficiency.

Selective IgA deficiency (SIgAD) is the most common inborn error of immunity (IEI). Unlike many IEIs, evidence of a role for highly penetrant rare variants in SIgAD is lacking. Previous SIgAD studies have had limited power to identify common variants due to their small sample size. We overcame this problem first through meta-analysis of two existing GWAS. This identified four novel common-variant associations and enrichment of SIgAD-associated variants in genes linked to Mendelian IEIs. SIgAD showed evidence of shared genetic architecture with serum IgA and a number of immune-mediated diseases

Implicating genes, pleiotropy, and sexual dimorphism at blood lipid loci through multi-ancestry meta-analysis.

BACKGROUND: Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery. RESULTS: To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N = 1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six ge

The search for modifier genes in Huntington disease - Multifactorial aspects of a monogenic disorder.

It is becoming increasingly evident that the underlying mutation of a single locus is often insufficient for the prediction of the comprehensive phenotype in human Mendelian disorders, implicating that there is no clear distinction between monogenic and complex traits. By definition, monogenic traits show a classic pattern of inheritance and are strongly influenced by variation within a single gene. However, many Mendelian traits that result in genetic disorders can have phenotypes that differ in subtle or profound ways such as severity, onset age and other associated phenotypic characteristic
