Osteogenic activities of genistein derivatives were influenced by the presence of prenyl group at ring A.

Our recent report indicated that the crude extract from stem bark of Erythrina variegata L. (Leguminosae) (EV) exerted beneficial effects against osteoporosis induced by estrogen deficiency in vivo. Follow-up phytochemical study has isolated genistein-derivatives mainly in the form of prenylgenistein from this extract, including 6-prenylgenistein, 8-prenylgenistein, and 6, 8-diprenylgenistein. The present study was performed to investigate the structure-function relationship of these compounds on osteoblastic proliferation, differentiation and mineralization in UMR 106 cells. Our results showe

Effects of dietary genistein on nutrient use and mineral status in heat-stressed quails.

Genistein is a powerful antioxidant and plays a role in calcium and bone metabolism. We evaluated the efficacy of dietary supplementation with genistein on the nutrient use and mineral concentrations in tibia and serum of quails reared at high environmental temperature (34 degrees C). Two hundred and forty Japanese quails (10 days old) were randomly assigned to 8 treatment groups consisting of 10 replicates of 3 birds. The birds were kept in a temperature-controlled room at 22 degrees C (Thermoneutral, TN groups) or 34 degrees C (for 8 h/d; 09.00 am-05.00 pm; Heat stress, HS groups). Birds wer

Effects of exposure to genistein during pubertal development on the reproductive system of male mice.

Genistein, a soybean-originated isoflavone, is widely consumed by humans for putative beneficial health effects but its estrogenic activity may affect adversely the development of the male reproductive system. Twenty-one days old ICR mice weaned from dams fed with a casein-based AIN-76A diet during gestation and lactation were exposed to genistein (2.5 and 5.0 mg/kg/day, p.o.) for 5 weeks. 17beta-Estradiol (7.5 microg/kg/day) and corn oil were used for the positive and negative vehicle controls, respectively. The animals were fed the casein-based AIN-76A diet throughout the experiment. There w

Genistein affects ER beta- but not ER alpha-dependent gene expression in the hypothalamus.

Isoflavone phytoestrogens are growing increasingly popular because of their reported cardiovascular and anticarcinogenic properties, but the effects of these compounds in the brain are largely unknown. In a previous study, we found that an isoflavone supplement, containing a mixture of soy phytoestrogens, inhibited estrogen-dependent female sexual behavior and was antiestrogenic for both ER alpha- and ER beta-dependent gene expression in the hypothalamus. Here we examined the impact of the soy isoflavone genistein, a major component of the supplement, on estrogen-dependent female sexual behavi

Molecular and Cellular Mechanisms Underlying the Therapeutic Effects of Genistein in Sepsis: A Systematic Review.

Sepsis is known to be a potentially fatal condition associated with immune dysregulation and systemic inflammatory response. Genistein is one of the naturally occurring isoflavones that have been studied extensively as immune modulators. This systematic review identifies the role of genistein in sepsis. A literature search of PubMed, Embase, Scopus, Web of Science, and Google Scholar (to December 2025) revealed articles examining the role of genistein in sepsis-related disorders. Following deduplication (n = 219 of 298), 79 articles were screened, of which 49 articles were removed based upon t

Preventive and therapeutic effects of genistein and daidzein on anxiety-like behaviors in ovariectomized rats.

Estrogen has demonstrated beneficial effects; however, it can also have unfavorable effects. Phytoestrogens are present in many consumable products and commonly used as supplements. These are of interest as they may have beneficial effects on mood with fewer undesirable effects on reproductive tissues. This study investigated the anxiolytic-like effects of the phytoestrogens genistein and daidzein on ovariectomized (Ovx) rats and their effects on the expression of uterine estrogen receptors (ER) and brain monoamines. In experiment 1, Ovx rats received either vehicle, 17β-estradiol, or 0.25 - 1

Beneficial effects of the phytoestrogen genistein on hippocampal impairments of spontaneously hypertensive rats (SHR).

Hippocampal neuropathology is a recognized feature of the spontaneously hypertensive rat (SHR). The hippocampal alterations associate with cognitive impairment. We have shown that hippocampal abnormalities are reversed by 17β-estradiol, a steroid binding to intracellular receptors (estrogen receptor α and β subtypes) or the membrane-located G-protein coupled estradiol receptor. Genistein (GEN) is a neuroprotective phytoestrogen which binds to estrogen receptor β and G-protein coupled estradiol receptor. Here, we investigated whether GEN neuroprotection extends to SHR. For this purpose, we trea

Cell cycle is disturbed in mucopolysaccharidosis type II fibroblasts, and can be improved by genistein.

Mucopolysaccharidoses (MPSs) are inherited metabolic diseases caused by mutations resulting in deficiency of one of enzymes involved in degradation of glycosaminoglycans (GAGs). These compounds accumulate in cells causing their dysfunctions. Genistein is a molecule previously found to both modify GAG metabolism and modulate cell cycle. Therefore, we investigated whether the cell cycle is affected in MPS cells and if genistein can influence this process. Fibroblasts derived from patients suffering from MPS types I, II, IIIA and IIIB, as well as normal human fibroblasts (the HDFa cell line) were

In vitro effect of genistein on DNA damage in leukocytes from mucopolysaccharidosis IVA patients.

Mucopolysaccharidosis IVA is a lysosomal storage disorder leading to an increase in glycosaminoglycans storage. Genistein is an isoflavone capable to inhibit glycosaminoglycans production. The objective of this study was to analyze the in vitro effect of different concentrations of genistein on DNA injury in mucopolysaccharidosis IVA patients. The lower concentration tested (10 μM) showed a significant increase on DNA injury in vitro, although higher concentrations (30 μM and 50 μM) showed higher DNA damage.

Genistein decreases liver fibrosis and cholestasis induced by prolonged biliary obstruction in the rat.

Fibrosis accompanies most chronic liver disorders and is a major factor contributing to hepatic failure. Therefore, the need for an effective treatment with the aim of modifying the clinical course of this disease is evident. The aim of this work is to determine whether genistein, which has been shown to modulate the physiology and pathophysiology of liver, is able to decrease experimental liver fibrosis and cholestasis. In male Wistar rats, the common bile duct was ligated. Administration of genistein (5 microg rat-1, day-1, p.o.) began four weeks after biliary obstruction and continued for a

Deciphering Angiogenic Drivers in Hepatocellular Carcinoma: From Prognostic Signature Construction to Genistein-Mediated Inhibition.

Hepatocellular carcinoma (HCC) is an aggressive malignancy with poor prognosis and a lack of reliable biomarkers. In this study, we integrated weighted gene co-expression network analysis, machine-learning-based prognosticmodelling, single-cell RNA sequencing, spatial transcriptomics, and molecular docking to identify angiogenesis-related drivers in HCC. SCAF1 was identified as a hub gene and was significantly upregulated in HCC tissues compared with normal liver tissues. High SCAF1 expression was closely associated with advanced clinicopathological features and unfavourable survival outcomes.

Targeted expression of Glycine max isoflavone synthase enhances daidzein and genistein content in soybean.

Isoflavonoids are key secondary metabolites in leguminous plants that play essential roles in plant physiology and provide significant health benefits to humans. In the isoflavone biosynthetic pathway, isoflavone synthase (IFS) catalyzes the conversion of naringenin and liquiritigenin into the bioactive isoflavones genistein and daidzein. This study aimed to enhance genistein and daidzein accumulation in soybean seeds through genetic engineering.
