OCT Angiography to Predict Geographic Atrophy Progression using Choriocapillaris Flow Void as a Biomarker.

PURPOSE: To investigate the relationship between choriocapillaris (CC) flow void (FV) percentage and geographic atrophy (GA) growth rate, and study how variations in FV percentage surrounding GA predict regional GA growth. METHODS: This prospective, longitudinal study enrolled subjects with GA secondary to nonexudative age-related macular degeneration. Optical coherence tomography angiography imaged the CC and FV percentage was evaluated using a validated algorithm. GA growth rate was measured as the difference in the square root of GA area divided by the months between baseline and follow-up

Role of Deep Learning-Quantified Hyperreflective Foci for the Prediction of Geographic Atrophy Progression.

PURPOSE: To quantitatively measure hyperreflective foci (HRF) during the progression of geographic atrophy (GA) secondary to age-related macular degeneration (AMD) using deep learning (DL) and investigate the association with local and global growth of GA. METHODS: Eyes with GA were prospectively included. Spectral-domain optical coherence tomography (SDOCT) and fundus autofluorescence images were acquired every 6 months. A 500-μm-wide junctional zone adjacent to the GA border was delineated and HRF were quantified using a validated DL algorithm. HRF concentrations in progressing and nonprogr

Quantifying Choriocapillaris Flow Voids in Patients With Geographic Atrophy Using Swept-Source OCT Angiography.

BACKGROUND AND OBJECTIVE: To compare choriocapillaris (CC) flow voids (FVs) throughout the macula in patients with age-related macular degeneration (AMD) and geographic atrophy (GA) to age-similar controls using swept-source optical coherence tomography angiography (SS-OCTA). PATIENTS AND METHODS: In this cross-sectional study, 12 subjects with GA secondary to nonexudative AMD and 12 age-similar controls participated. SS-OCTA was performed using a 6 mm × 6 mm scanning pattern. CC FVs were calculated using a one-standard deviation thresholding method developed from a normal database. RESULTS:

Vision-related quality of life is selectively affected by comorbidities in patients with geographic atrophy.

BACKGROUND: The atrophic late stage of age-related macular degeneration (AMD) is termed geographic atrophy (GA), and affects visual acuity (VA) as well as quality of life (QoL). Previous studies have found that best-corrected VA (BCVA), the standard vision assessment often underrepresents functional deficits. Therefore, the purpose of this study was to evaluate the correlation between atrophic lesion size, VA and QoL measured with the National Eye Institute Visual Function Questionnaire (VFQ-39) in a Danish population. Moreover, we wanted to evaluate the correlation between comorbidities, beha

METHODS: Patients with GA due to AMD were included. Fundus autofluorescence (FAF) intensity, spectral-domain optical coherence tomography (OCT), color fundus photography, and FLIO imaging were carried out at baseline and approximately 5 years of follow-up. The Role of Dopaminergic Agonists in Geographic Atrophy: Converging Evidence From Clinical and Experimental Data.

PURPOSE: Levodopa (L-DOPA) is an emerging drug repurposing candidate for age-related macular degeneration (AMD). Basic and clinical studies identify a potential role for L-DOPA to prevent and treat neovascular AMD. However, only one prior study has been performed in geographic atrophy (GA). METHODS: We performed a retrospective cohort analysis from the Sight Outcomes Research Collaborative database, including eyes with a diagnosis of early- or intermediate-stage AMD in 1 eye. We compared eyes exposed to any dopamine agonist (DA), dopamine receptor D2 agonist, and L-DOPA with eyes with no DA e

Equity reporting in systematic reviews and meta-analysis for geographic atrophy: a PROGRESS-Plus assessment.

CONTEXT: As systematic reviews and meta-analyses (SRMAs) are crucial for treatment development, they must provide guidelines that represent diverse patient demographics to promote equitable health care. As new research and treatment modalities are being developed for geographic atrophy (GA), establishing an equitable research foundation is becoming vitally important to physicians as they personalize their treatment plans. OBJECTIVES: This analysis aims to determine whether SRMAs pertaining to GA are reporting equity-related items utilizing the PROGRESS-Plus framework. METHODS: We conducted a

Functional endpoints in patients with geographic atrophy: What to consider when designing a clinical trial.

There is an unmet need in patients with geographic atrophy (GA) for treatments that preserve and improve functional vision to maintain their independence and quality of life. The limited number of available treatments for GA have demonstrated structural benefits, but none have consistently shown significant functional outcomes in clinical trials. Currently, best-corrected visual acuity (BCVA) is considered the gold standard functional endpoint in clinical trials; however, in the case of GA, it cannot fully evaluate visual impairment or treatment response, particularly in fovea-sparing GA lesio

IMPORTANCE: Initial intravenous immunoglobulin (IVIG)-refractory status and prolonged fever are established risk factors for the development of coronary artery abnormalities (CAAs) among patients with acute-phase Kawasaki disease (KD). However, whether different risk factors exist for initial unresponsiveness to IVIG and CAA development remains unclear. Imaging geographic atrophy: integrating structure and function to better understand the effects of new treatments.

Geographic atrophy (GA) is an advanced and irreversible form of age-related macular degeneration (AMD). Chronic low grade inflammation is thought to act as an initiator of this degenerative process, resulting in loss of photoreceptors (PRs), retinal pigment epithelium (RPE) and the underlying choriocapillaris. This review examined the challenges of clinical trials to date which have sought to treat GA, with particular reference to the successful outcome of C3 complement inhibition. Currently, optical coherence tomography (OCT) seems to be the most suitable method to detect GA and monitor the e
