Development of an efficient in vitro micropropagation and biochemical profiling of Salvia halophila, an endemic Turkish sage.

BACKGROUND: Salvia halophila Hedge is an endemic sage species confined to the salt steppe ecosystems of Central Anatolia (Türkiye), particularly around the Lake Tuz Basin, and is classified as Endangered (EN) by the IUCN. Its narrow distribution, low seed germination rates, and habitat pressures threaten its survival and restrict the sustainable exploitation of its valuable phytochemicals, such as rosmarinic acid and other phenolics. In vitro micropropagation represents a strategic approach for producing genetically uniform and phytochemically rich plant material, thereby supporting both ex si

The molecular regulation of axillary bud fate determination and outgrowth into branch crown in strawberry involves BRC1.

In strawberry, the axillary bud (AXB) can produce either an elongated stem called stolon giving a daughter-plant (asexual reproduction) or an inflorescence-bearing branch crown (BC; sexual reproduction). The AXB fate depends on node position on the axis and on genetic and environmental factors. Here, in Fragaria vesca, we addressed the largely unanswered question of how molecular factors determine AXB fate. To get insights into the mechanisms already at play in a morphologically indistinguishable (undifferentiated) AXB, depending on its fate, we combined the phenotypic characterization of AXB

A single nucleotide mutation in GID1c disrupts its interaction with DELLA1 and causes a GA-insensitive dwarf phenotype in peach.

Plant stature is one important factor that affects the productivity of peach orchards. However, little is known about the molecular mechanism(s) underlying the dwarf phenotype of peach tree. Here, we report a dwarfing mechanism in the peach cv. FenHuaShouXingTao (FHSXT). The dwarf phenotype of 'FHSXT' was caused by shorter cell length compared to the standard cv. QiuMiHong (QMH). 'FHSXT' contained higher endogenous GA levels than did 'QMH' and did not response to exogenous GA treatment (internode elongation). These results indicated that 'FHSXT' is a GA-insensitive dwarf mutant. A dwarf phenot

The effect of seed priming on yield and germination properties of quinoa (Chenopodium quinoa Willd).

Seed priming is a widely used pre-germination technique that enhances sprouting efficiency, synchrony, and the initial development of plantlets, particularly under suboptimal environmental conditions. This study aimed to evaluate the impact of different seed priming treatments on the germination, growth, and yield performance of quinoa (Chenopodium quinoa Willd.) through a series of laboratory, greenhouse, and field experiments conducted during the 2024 cropping season. The laboratory experiment was arranged in a completely randomized design (CRD) with four replications. Treatments included po

The regulatory role of DELLA protein DkGAI1 in persimmon dwarfism.

Persimmon (Diospyros kaki Thunb.) is an important fruit crop in East Asia with significant commercial potential. However, dwarfing cultivation in persimmon trees is rarely applied due to the lack of excellent dwarfing rootstocks and cultivars. 'Nantongxiaofangshi' (Diospyros kaki Linn.cv. Nantongxiaofangshi) is an excellent persimmon cultivar suitable for dwarfing cultivation due to its distinct dwarfing characteristics, but the underlying mechanism remains unclear. In this study, we further identified the DkGAI1 gene, which encodes a DELLA protein, as a candidate gene regulating dwarfing in '

Gibberellic acid promotes single-celled fiber elongation through the activation of two signaling cascades in cotton.

The agricultural green revolution spectacularly enhanced crop yield through modification of gibberellin (GA) signaling. However, in cotton, the GA signaling cascades remain elusive, limiting our potential to cultivate new cotton varieties and improve yield and quality. Here, we identified that GA prominently stimulated fiber elongation through the degradation of DELLA protein GhSLR1, thereby disabling GhSLR1's physical interaction with two transcription factors, GhZFP8 and GhBLH1. Subsequently, the resultant free GhBLH1 binds to GhKCS12 promoter and activates its expression to enhance VLCFAs b

Revisit and explore the ethylene-independent mechanism of sex expression in cucumber (Cucumis sativus).

This review provides a thorough and comprehensive perspective on the topic of cucumber sexual expression. Specifically, insights into sex expression mediated by pathways other than ethylene are highlighted. Cucumber (Cucumis sativus L.) is a common and important commercial crop that is cultivated and consumed worldwide. Additionally, this species is commonly used as a model for investigating plant sex expression. Cucumbers exhibit a variety of floral arrangements, comprising male, female, and hermaphroditic (bisexual) flowers. Generally, cucumber plants that produce female flowers are typicall

The immunosuppressive, growth-hindering, hepatotoxic, and oxidative stress and immune related-gene expressions-altering effects of gibberellic acid in Oreochromis niloticus: A mitigation trial using alpha-lipoic acid.

This study aimed to examine the effects of gibberellic acid (GBA) on growth, hemato-biochemical parameters related to liver functions, digestive enzymes, and immunological response in Oreochromis niloticus. Besides, the probable underlying mechanisms were explored by assessing antioxidant, apoptotic, and immune-related gene expression. Furthermore, the likelihood of restoration following alpha-lipoic acid (LIP) dietary supplementation was explored. The fish (average initial weight 30.75 ± 0.46) were equally classified into four groups: the control group, the LIP group (fed on a basal diet plus

Manipulating GA-Related Genes for Cereal Crop Improvement.

The global population is projected to experience a rapid increase in the future, which poses a challenge to global food sustainability. The "Green Revolution" beginning in the 1960s allowed grain yield to reach two billion tons in 2000 due to the introduction of semi-dwarfing genes in cereal crops. Semi-dwarfing genes reduce the gibberellin (GA) signal, leading to short plant stature, which improves the lodging resistance and harvest index under modern fertilization practices. Here, we reviewed the literature on the function of GA in plant growth and development, and the role of GA-related gen

Characterization of GA20ox genes in tall and dwarf types coconut (Cocos nucifera L.).

Coconuts (Cocos nucifera L.) are divided by the height into tall and dwarf types. In many plants the short phenotype was emerged by mutation of the GA20ox gene encoding the enzyme involved in gibberellin (GA) biosynthesis. Two CnGA20ox genes, CnGA20ox1 and CnGA20ox2, were cloned from tall and dwarf types coconut. The sequences, gene structures and expressions were compared. The structure of each gene comprised three exons and two introns. The CnGA20ox1 and CnGA20ox2 genes consisted of the coding region of 1110 and 1131 bp, encoding proteins of 369 and 376 amino acids, respectively. Their amino

Phytohormone-microbiome interactions and epigenetic regulation of plant stress responses and priming.

Plants continuously face fluctuating environmental conditions, requiring tightly coordinated regulatory systems to balance growth and stress responses. This review synthesizes current knowledge on phytohormones as central integrators of plant-microbiome interactions, highlighting their dual role as internal regulators and ecological gatekeepers that shape microbiome assembly and function. Phytohormones, such as auxins, cytokinins, gibberellins, abscisic acid, ethylene, salicylic acid, and jasmonates, dynamically regulate plant development, immunity, and rhizosphere chemistry, thereby influenci

Minimising toxicity of cadmium in plants--role of plant growth regulators.

A range of man-made activities promote the enrichment of world-wide agricultural soils with a myriad of chemical pollutants including cadmium (Cd). Owing to its significant toxic consequences in plants, Cd has been one of extensively studied metals. However, sustainable strategies for minimising Cd impacts in plants have been little explored. Plant growth regulators (PGRs) are known for their role in the regulation of numerous developmental processes. Among major PGRs, plant hormones (such as auxins, gibberellins, cytokinins, abscisic acid, jasmonic acid, ethylene and salicylic acid), nitric o

Comparative Transcriptomic, Anatomical and Phytohormone Analyses Provide New Insights Into Hormone-Mediated Tetraploid Dwarfing in Hybrid Sweetgum (Liquidambar styraciflua × L. formosana).

Polyploid breeding is an effective approach to improve plant biomass and quality. Both fast growth and dwarf types of in vitro or ex vitro plants are produced after polyploidization. However, little is known regarding the dwarf type mechanism in polyploids grown in vitro. In this study, the morphological and cytological characteristics were measured in tetraploid and diploid hybrid sweetgum (Liquidambar styraciflua × L. formosana) with the same genetic background. RNA sequencing (RNA-Seq) was used to analyse shoot and root variations between tetraploid and diploid plants; important metabolites

Seed dormancy and germination of three Corydalis species, disjunct relict species in East Asia.

Disjunct congeneric species are common in the temperate flora of Europe, East Asia and eastern North America. However, it is unclear whether the kind of seed dormancy is maintained after divergence. This study aimed to determine the type of seed dormancy of three Corydalis species native to Korea and examine whether the dormancy trait has been conserved after long-term separation. We examined embryo growth and germination characteristics of C. remota, C. incisa and C. speciosa through phenology experiments, controlled temperature experiments, stratification treatments and GA3 application. Plant growth regulators and fertilizer increase phytoextraction efficiency of Tagetes minuta under lead-contaminated soil.

Soil contamination with heavy metals severely threatens food security and human health worldwide. Heavy metal contamination must be controlled through sustainable and economical technologies. To enhance the phytoextraction of lead (Pb), the plants with resistance against Pb stress and maximum production of biomass could serve as a potential candidate for the phytoremediation of Pb-contaminated soils. The current study was conducted to illuminate the effect of gibberellic acid (GA3), indole acetic acid (IAA), and fertilizers (NPK) on plant growth and Pb phytoextraction capabilities of Tagetes m

Chemical armed allies promote plant resistance to pathogen: The contribution of arbuscular mycorrhizal fungi to an invasive weed.

Invasive weeds often possess strong resistance to biotic stresses, which causes huge ecological problems. Both plant growth regulators (PGRs) and arbuscular mycorrhizal (AM) fungi contribute to plant growth and resistance. However, their combined interactions in invasive plants' defense remain poorly understood. To address this knowledge gap, the invasive weed Alternanthera philoxeroides was treated with gibberellins (GA) and paclobutrazol (PAC), inoculated with Clariodeoglous etunicatum to test its response to pathogenic fungi. We found that these two PGRs suppressed AM fungi colonization. Bo

Dwarfism mechanism in Malus clonal rootstocks.

The dwarfing mechanism in apple clonal rootstocks is driven by complex interactions between anatomical, hormonal, genetic, and phenolic factors, offering potential for advanced genetic manipulation to optimize tree size and enhance orchard productivity. The widespread adoption of dwarfing rootstocks is pivotal to modern commercial apple (Malus × domestica Borkh) orchards due to their ability to control tree size, shorten the juvenile period, and enhance reproductive growth and overall productivity. The underlying mechanisms of rootstock-induced dwarfism are multifaceted and involve interaction

A stress-associated protein OsSAP8 modulates gibberellic acid biosynthesis by reducing the promotive effect of transcription factor OsbZIP58 on OsKO2.

Gibberellic acid (GA) is a vital phytohormone for plant growth and development. GA biosynthesis is a complex pathway regulated by various transcription factors. Here we report a stress-associated protein 8 (OsSAP8), negatively involved in GA biosynthesis. Overexpression of OsSAP8 in rice resulted in a semi-dwarfism phenotype and reduced endogenous GA3 content. In contrast, an OsSAP8 knockout mutant exhibited higher endogenous GA3 content and slightly increased plant height. Sub-cellular localization analysis of OsSAP8 showed that it could enter the nucleus. Based on electrophoretic mobility sh

Dwarf and Increased Branching 1 controls plant height and axillary bud outgrowth in Medicago truncatula.

Optimizing plant architecture is an efficient approach for breeders to increase crop yields, and phytohormones such as gibberellins (GAs) play an important role in controlling growth. Medicago truncatula is a model legume species, but the molecular mechanisms underlying its architecture are largely unknown. In this study, we examined a tobacco retrotransposon Tnt1-tagged mutant collection of M. truncatula and identified dwarf and increased branching 1 (dib1), which exhibited extreme dwarfism and increased numbers of lateral branches. By analysis of the flanking sequences of Tnt1 insertions in
