NKB/NK3 system negatively regulates the reproductive axis in sexually immature goldfish (Carassius auratus).

To ascertain the significance of the Neurokinin B/Tachykinin 3 receptor (NKB/NK3) system in goldfish reproduction, two cDNAs encoding tachykinin 3 receptors, namely tacr3a and tacr3b, were cloned. Subsequent studies revealed that the downstream signalling of both Tac3rs can be activated by different NKB peptides, suggesting that the cloned receptors are biologically functional in goldfish. RT-PCR analysis showed that tacr3s are widely expressed in brain regions. During the gonadal development, tacr3a and tacr3b exhibited different expression patterns in the hypothalamus and pituitary. The acti

Sema-3A indirectly disrupts the regeneration process of goldfish optic nerve after controlled injury.

BACKGROUND: Neurons of adult mammalian CNS are prevented from regenerating injured axons due to formation of a non-permissive environment. The retinal ganglion cells (RGC), which are part of the CNS, share this characteristic. In sharp contrast, the RGC of lower vertebrates, such as fish, are capable of re-growing injured optic nerve axons, and achieve, through a complex multi-factorial process, functional vision after injury. Semaphorin-3A (sema-3A), a member of the class 3 semaphorins known for its repellent and apoptotic activities, has previously been shown to play a key role in the format

Immune response and expression analysis of cathepsin K in goldfish during Aeromonas hydrophila infection.

The innate immunity and expression profiles of cathepsins D were determined in the goldfish (Carassius auratus) tissues after challenge with a fish pathogen Aeromonas hydrophila. The innate immunity of reactive oxygen species (ROS) and reactive nitrogen species (RNS) were determined by peripheral blood leucocytes. Blood and tissue samples of the muscle, gills, liver, kidney, heart, spleen, and intestine were sampled at 1, 3, 6 and 12 h post-infection for cathepsin D expression by semi-quantitative RT-PCR. The ROS and RNS production did not significantly increase at 1 h post-challenged goldfish

Profiling neuroendocrine gene expression changes following fadrozole-induced estrogen decline in the female goldfish.

Teleost fish represent unique models to study the role of neuroestrogens because of the extremely high activity of brain aromatase (AroB; the product of cyp19a1b). Aromatase respectively converts androstenedione and testosterone to estrone and 17beta-estradiol (E2). Specific inhibition of aromatase activity by fadrozole has been shown to impair estrogen production and influence neuroendocrine and reproductive functions in fish, amphibians, and rodents. However, very few studies have identified the global transcriptomic response to fadrozole-induced decline of estrogens in a physiological conte

Prevention of Ichthyophthirius multifiliis infestation in goldfish (Carassius auratus) by potassium ferrate(VI) treatment.

Ichthyophthirius multifiliis is an important freshwater teleost pathogen that often leads to significant economic losses to the aquaculture industry. The purpose of this study was to assess the acute toxicity of potassium ferrate(VI) to I. multifiliis theront and the concentration needed to prevent I. multifiliis infestation in goldfish, Carassius auratus. Five hundred theronts were exposed to concentrations of potassium ferrate(VI) in each well of a 96-well microtiter plate and observed for 4h to determine the acute toxicity. Results showed that the exposure of I. multifiliis theronts to pota

Appetite-suppressing effects of urotensin I and corticotropin-releasing hormone in goldfish (Carassius auratus).

Fish urotensin I (UI), a member of the corticotropin-releasing hormone (CRH) family of peptides, is a potent inhibitor of food intake in mammals, yet the role of UI in the control of food intake in fish is not known. Therefore, to determine the acute effects of UI on appetite relative to those of CRH, goldfish were given intracerebroventricular (i.c.v.) injections of carp/goldfish UI and rat/human CRH (0.2-200 ng/g) and food intake was assessed for a 2-hour period after the injection. UI and CRH both suppressed food intake in a dose-related manner and UI (ED50 = 3.8 ng/g) was significantly mor

Assessment of the Risk of Microplastics on Gill and Gut Health and Subsequent Pathogen Susceptibility in the Goldfish Model.

Microplastics are pervasive pollutants in aquatic ecosystems, yet their effects on fish tissues remain insufficiently characterized. Our study investigates the impact of polystyrene microplastics (0.5 and 2 μm) on the gill and intestinal tissues of goldfish (Carassius auratus), with a focus on inflammatory responses and pathogen susceptibility. Following two weeks of exposure, histological and molecular analyses revealed increased filament cartilage thickness in gills, enhanced villus thickness and goblet cell numbers in intestines, and upregulation of immune- and oxidative stress-related gene

Histological and Molecular Characterization of the Inferior Olivary Nucleus and Climbing Fibers in the Goldfish, Carassius auratus.

The cerebellum receives inputs via the climbing fibers originating from the inferior olivary nucleus in the ventral medulla. In mammals, the climbing fibers entwine and terminate onto both major and peripheral branches of dendrites of the Purkinje cells. In this study, the inferior olivary nucleus and climbing fiber in the goldfish were investigated with several histological techniques. By neural tracer application to the hemisphere of the cerebellum, labeled inferior olivary neurons were found in the ventral edge of the contralateral medulla. Kainate stimulated Co + + uptake and gephyrin immu

Molecular identification of the Dyn/Kor system and its potential role in the reproductive axis of goldfish.

To ascertain the significance of the dynorphin/kappa-opioid receptor (Dyn/Kor) system in fish reproduction, prodynorphin (pdyn) cDNA was cloned from goldfish. Two Dyn peptides (DynA and DynB) are present in the goldfish prodynorphin precursor. Both DynA and DynB are biologically active as they are able to functionally interact with the goldfish Kor expressed in cultured eukaryotic cells to suppress forskolin-induced CRE promoter activity. RT-PCR analysis showed that pdyn is widely expressed in brain regions, with the highest expression in hypothalamus. During ovarian development, hypothalamic

Effect of green light spectra on the reduction of retinal damage and stress in goldfish, Carassius auratus.

We investigated the effect of light spectra on retinal damage and stress in goldfish using green (530 nm) and red (620 nm) light emitting diodes (LEDs) at three intensities each (0.5, 1.0, and 1.5 W/m(2)). We measured the change in the levels of plasma cortisol and H2O2 and expression and levels of caspase-3. The apoptotic response of green and red LED spectra was assessed using the terminal transferase dUTP nick end labeling (TUNEL) assay. Stress indicator (cortisol and H2O2) and apoptosis-related genes (caspase-3) decreased in green light, but increased in red light with higher light intensi

Orexin A enhances locomotor activity and induces anxiogenic-like action in the goldfish, Carassius auratus.

Orexin acts as an orexigenic factor for the regulation of appetite and rhythmicity in rodents. In goldfish, intracerebroventricular (ICV) administration of orexin A has been shown to affect not only food intake, but also locomotor activity. However, as there is still no information regarding the effect of orexin A on emotional behavior in goldfish, we investigated the effect of orexin A on psychomotor activity in this species. Intracerebroventricular administration of synthetic orexin A at 2 and 4pmol/g body weight (BW) enhanced locomotor activity, and this enhancement by orexin A at 4pmol/g B

Function of otolith organ in goldfish revealed from analysis of eye movement induced by acceleration.

An otolith organ on ground behave as a detector of both gravity and linear acceleration, and play an important role in controlling posture and eye movement for tilt of the head or translational motion. On the other hand, a gravitational acceleration ingredient to an otolith organ disappears in microgravity environment. However, linear acceleration can be received by otolith organ and produce a sensation that is different from that on Earth. It is suggested that in microgravity signal from the otolith organ may cause abnormality of posture control and eye movement. Therefore, the central nervou

Neurolin, the goldfish homolog of DM-GRASP, is involved in retinal axon pathfinding to the optic disk.

Young axons of new retinal ganglion cells (RGCs) in the continuously growing goldfish retina fasciculate with one another and their immediate forerunners on their path toward the optic disk and along the optic nerve. They express the immunoglobulin superfamily cell adhesion molecules (CAMs) neurolin (DM-GRASP) and the L1-like E587 antigen. Repeated injections of Fab fragments from polyclonal antisera against neurolin (neurolin Fabs) into the eye of 3. 4-cm-long and rapidly growing goldfish caused highly aberrant pathways of young RGC axon subfascicles in the dorsal retina. Many axons grew in c

Growth hormone-releasing hormone immunoreactivity in the brain, pituitary, and pineal of the goldfish, Carassius auratus.

Using antisera directed against carp growth hormone-releasing hormone (cGHRH), the distribution of immunoreactive (ir) structures in the brain, pituitary, and pineal of the goldfish, Carassius auratus, was investigated. The antisera were produced in rabbits by administration of cGHRH(1-44)-NH(2) followed by cGHRH(1-45)-OH for different periods of time, both coupled to human alpha-globulins via bisdiazotized benzidine as immunogen. Immunoreactive perikarya were visualized in the nucleus preopticus periventricularis (NPP), nucleus lateralis tuberis (NLT), nucleus of the lateral lemniscus (NLL),

Growth hormone-dependent potentiation of gonadotropin-stimulated steroid production by ovarian follicles of the goldfish.

The possible involvement of growth hormone (GH) in the regulation of ovarian function in the goldfish was investigated by determining the effects of common carp GH on steroid production by vitellogenic and preovulatory ovarian follicles incubated in vitro. Carp GH acts in a dose-dependent manner to potentiate the actions of common carp gonadotropin (GtH) on the production of 17 beta-estradiol and testosterone by vitellogenic ovarian follicles and the actions of human chorionic gonadotropin (hCG) on testosterone production by preovulatory ovarian follicles. Carp GH alone had no effect on basal

Association of spermine and 4S RNA during axonal transport in regenerating optic nerves of goldfish.

Experiments were designed to determine whether polyamines are bound to 4S RNA and then transported axonally along regenerating optic axons of goldfish. In one set of experiments, inhibition of retinal RNA synthesis by intraocular injections of 10 microgram of cordycepin, blocked the axonal transport of both [3H]RNA and [14C]spermidine by about 65%, 6 and 14 days after injection. Intraocular injections of vinblastine, (0.1, 0.5 or 1.0 microgram) an agent which interrupts axonal transport of proteins, had no effect on retinal RNA synthesis nor on the amount of [14C]spermidine incorporated into t

The fine structure of nerve cell bodies and their myelin sheaths in the eighth nerve ganglion of the goldfish.

The eighth cranial nerve ganglion consists of bipolar nerve cell bodies each occupying part of an internodal segment. The perikaryal sheaths range from a single layer of Schwann cell cytoplasm on the smallest cells to typical thick compact myelin on the largest. On most perikarya, the sheath displays an intermediate form, consisting of multiple layers of Schwann cell cytoplasm (loose myelin), or of loose and compact myelin continuous with each other. Internodes beyond the one containing the cell body bear only compact myelin. In loose myelin the thickness of each layer of Schwann cell cytoplas

Integrative transcriptomic and metabolomic analysis of body color formation in transparent goldfish (Carassius auratus).

The mechanism underlying the transparent body coloration in goldfish (Carassius auratus) remains unclear. This study employed untargeted metabolomics and transcriptomics analyses of scales, skin, and muscle tissues from transparent goldfish (T) and common goldfish (L), revealing that the transparent phenotype stems from systemic disruption of the purine metabolism network. Metabolomic analysis revealed significant downregulation of guanine and its precursor guanosine across all tissues. Correspondingly, transcriptomic analysis further demonstrated tissue-specific dysregulation of key genes inv
