Isolation and molecular characterization of a novel picornavirus from baitfish in the USA.

During both regulatory and routine surveillance sampling of baitfish from the states of Illinois, Minnesota, Montana, and Wisconsin, USA, isolates (n = 20) of a previously unknown picornavirus were obtained from kidney/spleen or entire viscera of fathead minnows (Pimephales promelas) and brassy minnows (Hybognathus hankinsoni). Following the appearance of a diffuse cytopathic effect, examination of cell culture supernatant by negative contrast electron microscopy revealed the presence of small, round virus particles (∼ 30-32 nm), with picornavirus-like morphology. Amplification and sequence an

An Unlikely Case of Blastomycosis in an Elderly Female: A Diagnostic Challenge in a Non-endemic Region.

Blastomyces dermatitidis is a fungus typically found in the soil of endemic regions such as the Midwest, concentrating in areas like Ohio, Mississippi, and the Great Lakes area. The systemic infection caused by inhaling Blastomyces dermatitidis is known as blastomycosis. The frequency of blastomycosis in non-endemic regions is increasing for a variety of speculated reasons, such as higher rates of immunosuppressed individuals and possible climate. Due to clinician unfamiliarity, misdiagnosis of blastomycosis is common, which potentiates worsening systemic infections. This study shows the clini

Diagnosing Blastomycosis: A Review of Laboratory Methods and Clinical Utility.

Blastomycosis, caused by dimorphic fungi of the Blastomyces genus, is endemic to regions in North America, including the Great Lakes and other parts of Canada and the United States of America. The infection primarily occurs through the inhalation of airborne conidia from contaminated soil and decaying organic matter. Pulmonary involvement is most common, but dissemination to other organs such as the skin and bones can occur, especially in immunocompromised individuals. Diagnosis is challenging due to its clinical overlap with other diseases. Culture remains the gold-standard diagnostic method,

Epidemiological study of Rift Valley fever virus in Kigoma, Tanzania.

Rift Valley fever virus (RVFV) is an acute, zoonotic viral disease caused by a Phlebovirus, which belongs to the Bunyaviridae family. Among livestock, outbreaks of the disease are economically devastating. They are often characterised by large, sweeping abortion storms and have significant mortality in adult livestock. The aim of the current study was to investigate RVFV infection in the Kigoma region, which is nestled under the hills of the western arm of the Great Rift Valley on the edge of Lake Tanganyika, Tanzania. A region-wide serosurvey was conducted on non-vaccinated small ruminants (s

Effects of organochlorine chemicals on the reproductive outcome of humans who consumed contaminated Great Lakes fish: an epidemiologic consideration.

Three sets of studies of the impacts of human exposure to PCB contaminated fish from the Great Lakes basin--the Michigan Sports Fisherman Cohort, the Michigan Maternal/Infant Cohort, and the Wisconsin Maternal/Infant Cohort-were evaluated using the epidemiologic criteria of Susser (1986). The studies were compared against each other, and against comparable data from other geographic locales. A total of seven major categories of exposure sequelae were evaluated. These ranged from the effects of primary exposure to contaminants upon maternal health status, to effects from secondary intrauterine

Mycobacterium tuberculosis complex Lineage 8 causes drug resistant pulmonary tuberculosis also in South Kivu, Eastern Democratic Republic of Congo.

BACKGROUND: Lineage 8 (L8) occupies a unique phylogenetic position as a sister clade to all other known Mycobacterium tuberculosis complex (MTBC) lineages. Although the initial identification of L8 strains was limited to Rwanda and Uganda, the complete geographical distribution of this lineage remains undetermined due to limited genomic surveillance across eastern and central Africa. METHODS: We performed whole genome sequencing (WGS) on MTBC isolates from rifampicin-resistant tuberculosis (TB) patients' sputum in Bukavu, South Kivu province, eastern Democratic Republic of Congo (DRC). Post-glacial colonization and isolation shape fine-scale population structure in Venustaconcha ellipsiformis: implications for population-level conservation.

The spatial distribution of genetic diversity in freshwater organisms reflects a complex interplay of historical and contemporary evolutionary processes. In North America, repeated Pleistocene glaciations reshaped aquatic landscapes, influencing dispersal, isolation, and secondary contact. Freshwater mussels (order Unionida), which rely on host-mediated larval dispersal and often occupy fragmented habitats, are particularly sensitive to these processes. Venustaconcha ellipsiformis, a widespread but imperiled mussel species, provides a model for evaluating how glacial history and river evolutio

Role of candidate gene variants in modulating the risk and severity of alcoholic hepatitis.

BACKGROUND: Alcoholic hepatitis (AH) is a severe and life-threatening alcohol-associated liver disease. Only a minority of heavy drinkers acquires AH and severity varies among affected individuals, suggesting a genetic basis for the susceptibility to and severity of AH. METHODS: A cohort consisting of 211 patients with AH and 176 heavy drinking controls was genotyped for five variants in five candidate genes that have been associated with chronic liver diseases: rs738409 in patatin-like phospholipase domain-containing protein 3 (PNPLA3), rs72613567 in hydroxysteroid 17-beta dehydrogenase 13 (

Modeling sorption dynamics and ecotoxicological risk of microplastics and polybrominated diphenyl ethers in Arctic and subarctic freshwater.

Microplastics can act as sinks and carriers for hydrophobic organic contaminants, including polybrominated diphenyl ethers (PBDEs), increasing their bioavailability and potential harm to Arctic and subarctic freshwater biota. However, the sorption dynamics, drivers of co-contamination, and combined toxicity of microplastics and PBDEs remain poorly understood. This study develops a Bayesian Network (BN) to assess PBDE sorption onto microplastics and evaluate the ecotoxicological risk of PBDE-enriched microplastics with an application to northern pike in Great Slave Lake, Canada. The model integ

[A small-mammal study in the northeastern Kara Kum for leptospirosis infectivity].

Search for natural foci of leptospirosis was carried out in 1987-1989 in humid biotopes of Tashauz Province, Turkmenistan. Such potential carriers of leptospirosis as house mice (Mus musculus) and tamarisk gerbils (Meriones tamariscinus) are widely spread in this area, and the size of their population can amount to great numbers. The sharpest fluctuations in the population size are characteristic of house mice inhabiting the shores of water collecting ponds and lakes in the regions of minimally used pastures. A moderate number of house mice was registered in spring and autumn at the area of ir

Species distribution models for the eastern blacklegged tick, Ixodes scapularis, and the Lyme disease pathogen, Borrelia burgdorferi, in Ontario, Canada.

The blacklegged tick, Ixodes scapularis, is established in several regions of Ontario, Canada, and continues to spread into new geographic areas across the province at a rapid rate. This poses a significant public health risk since I. scapularis transmits the Lyme disease-causing bacterium, Borrelia burgdorferi, and other pathogens of potential public health concern. The objective of this study was to develop species distribution models for I. scapularis and B. burgdorferi to predict and compare the potential distributions of the tick vector and the Lyme disease pathogen as well as the ecologi

Assessing consequences of anthropogenic hybridization: Great Lakes mallard movement and resource selection.

BACKGROUND: The release of domesticated animals is resulting in anthropogenic hybridization with their wild congeners and there is concern that these interactions are a risk to conservation. In North America, large-scale releases of domesticated mallards have continued for over a century, yet the potential ecological consequences are not well understood. Towards this, we assessed whether hybridization between game-farm and wild mallards was associated with variation in movement and resource selection behavior, which in turn can affect survival and reproduction. METHODS: We coupled genomic, te

Epidemiology of Eastern Massasauga (Sistrurus catenatus) Snakebites in the Great Lakes States, 1800-2015.

INTRODUCTION: The eastern massasauga rattlesnake (Sistrurus catenatus) has the northernmost distribution of any venomous snake in the United States and presents a potential, but little-studied, risk to humans. METHODS: We quantified the reported incidence of massasauga bites to humans in the Great Lakes states by searching historic and contemporary media reports between 1800 and 2015. RESULTS: We identified 848 reported massasauga bites across the region, most of which were concentrated along the border of Indiana and Michigan. The number of bites per decade increased into the late 1800s as

Thiamine Levels in Muscle and Eggs of Adult Pacific Salmon from the Fraser River, British Columbia.

Multiple species and stocks of Pacific salmon Oncorhynchus spp. have experienced large declines in the number of returning adults over a wide region of the Pacific Northwest due to poor marine survival (low smolt-to-adult survival rates). One possible explanation for reduced survival is thiamine deficiency. Thiamine (vitamin B1 ) is an essential vitamin with an integral role in many metabolic processes, and thiamine deficiency is an important cause of salmonid mortality in the Baltic Sea and in the Laurentian Great Lakes. To assess this possibility, we (1) compared muscle thiamine content over

Diagnosis and treatment of Lyme disease.

Lyme disease is the most common tick-borne disease in the United States. This review details the risk factors, clinical presentation, treatment, and prophylaxis for the disease. Information was obtained from a search of the PubMed and MEDLINE databases (keyword: Lyme disease) for articles published from August 31, 1997, through September 1, 2007. Approximately 20,000 cases of Lyme disease are reported annually. Residents of the coastal Northeast, northwest California, and the Great Lakes region are at highest risk. Children and those spending extended time outdoors in wooded areas are also at

Investigation of ground level and remote-sensed data for habitat classification and prediction of survival of Ixodes scapularis in habitats of southeastern Canada.

In southeastern Canada, most populations of Ixodes scapularis Say, the Lyme disease vector, occur in Carolinian forests. Climate change projections suggest a northward range expansion of I. scapularis this century, but it is unclear whether more northerly habitats are suitable for I. scapularis survival. In this study, we assessed the suitability of woodlands of the Lower Great Lakes/St. Lawrence Plain region for I. scapularis by comparing tick egg survival in four different woodlands. Woodlands where I. scapularis are established, and sand dune where I. scapularis do not survive, served as po

Efficient CRISPR/Cas9-mediated genome editing of phytoene desaturase in Musa-AAA: a critical step for genetic improvement of east African highland bananas.

East African highland bananas (EAHBs), locally referred to as "matooke", are an important staple crop in Uganda. The EAHBs have a triploid genome (AAA) with a large phenotypic diversity in the Great Lakes region of Africa and are challenged by both abiotic and biotic factors. The EAHBs have been improved through conventional breeding and genetic engineering though facing challenges such as genetic drag of unfavorable traits and complex regulatory processes, respectively. Therefore, a more precise approach for crop improvement such as genome editing is highly recommended. In the current study,

Patterns of adverse childhood experiences and mental health outcomes among American Indians with type 2 diabetes.

BACKGROUND: Extensive research demonstrates that Adverse Childhood Experiences (ACEs) are highly interconnected and have numerous health consequences well into adulthood. Yet, there is a dearth of focused research that examines ACEs and health inequities for American Indians (AIs). OBJECTIVE: To assesses the prevalence of ten types of childhood adversities, explore constellations of exposures, and examine whether there are differential risks of mental health outcomes according to sub-group classification. Modeling the potential impact of storm surge and sea level rise on coastal archaeological heritage: A case study from Georgia.

Climate change poses great risks to archaeological heritage, especially in coastal regions. Preparing to mitigate these challenges requires detailed and accurate assessments of how coastal heritage sites will be impacted by sea level rise (SLR) and storm surge, driven by increasingly severe storms in a warmer climate. However, inconsistency between modeled impacts of coastal erosion on archaeological sites and observed effects has thus far hindered our ability to accurately assess the vulnerability of sites. Modeling of coastal impacts has largely focused on medium-to-long term SLR, while obse

Air toxics exposure from vehicle emissions at a U.S. border crossing: Buffalo Peace Bridge Study.

The Peace Bridge in Buffalo, New York, which spans the Niagara River at the east end of Lake Erie, is one of the busiest U.S. border crossings. The Peace Bridge plaza on the U.S. side is a complex of roads, customs inspection areas, passport control areas, and duty-free shops. On average 5000 heavy-duty diesel trucks and 20,000 passenger cars traverse the border daily, making the plaza area a potential "hot spot" for emissions from mobile sources. In a series of winter and summer field campaigns, we measured air pollutants, including many compounds considered by the U.S. Environmental Protecti
