Goat farming plays an important role in the Sri Lankan rural economy. Sri Lankan indigenous (SLI) goats and their crossbreds are reared mainly under extensive management and indiscriminately exposed to pathogens and parasites. This study was designed to evaluate resistance to haemonchosis in SLI goats and their Jamnapari crossbreds (JCB) in the dry zone of Sri Lanka. Twenty SLI and 20 JCB 4-month-old male goats were artificially challenged with 5000 H. contortus L3 larvae. Faecal egg counts (FEC), body weights, FAffa MAlan CHArt (FAMACHA®) scores, packed cell volumes (PCV), red blood cell coun

Most small ruminant farms in tropical climates are plagued by Haemonchus contortus, a hematophagous, abomasal parasite. Heavy burdens of this parasite can cause anemia, hypoproteinemia, weight loss, and mortality in susceptible animals. Haemonchus contortus is becoming a major health concern in New World camelids as well, namely llamas (Llama glama) and alpacas (Vicugna pacos), yet little research has been conducted regarding its prevalence or pathology in these species. Herein, we present a retrospective review of llamas and alpacas that were admitted to The University of Georgia Veterinary T

Previously, chemical analysis of gastric fundic mucin showed that infection of sheep with Haemonchus contortus or Teladorsagia circumcincta changed the proportions of monosaccharides and decreased terminal mucin fucosylation and sialylation. To identify the effects of these parasites on the two mucin-secreting cell lineages, fundic and antral tissues were collected for histochemistry from 69 lambs aged from 3-4 to 9-10 months-of-age which had received a single infection of either H. contortus or T. circumcincta and euthanased at Day 21 or 28 post- infection respectively. All fundic tissues wer

OBJECTIVES: We have found that in a laboratory strain of the nematode parasite of ruminants, Haemonchus contortus, repeated ivermectin treatment, in vivo in sheep, selected against TTC, coding for Phe, and for TAC, coding for Tyr, at codon 200 in beta-tubulin isotype 1. This 200Tyr single nucleotide polymorphism (SNP) has been associated with benzimidazole anthelmintic resistance. In this study, we investigated the alpha-tubulin and beta-tubulin sequences of 17 different field and laboratory strains/isolates of H. contortus with known treatment history and status for susceptibility or resistan

Haemonchus contortus commences feeding on host blood by day 11 of infection, which leads to the presence of blood in the host's faeces. This study examined the capacity for a faecal occult blood (FOB) test to determine the severity of H. contortus infection in sheep at pasture, and to predict a rise in worm egg count (WEC) as infection matures. Diluted faeces were assayed with Bayer Hemastix and the change in colour of the reagent patch was scored on a 9-point scale from 1 (negative) to 5 in half unit increments. Performance of the test was compared with four benchmarks for severe infection: (

Fractionated Haemonchus longistipes crude antigen revealed 7 protein fractions at molecular weights of 93, 74, 67, 56, 32, 26 and 16 kDa. The bands at 56, 32 and 26 kDa reacted specifically with H. longistipes hyperimmune sera prepared in rabbit (1:100 dilution) using EITB technique. The eluted concentrated fractions as well as crude worm antigens were used in diagnosis of natural infection of Gastro-Intestinal Nematodes (GIN) only or associated with other parasites in camels using ELISA technique. H. longistipes crude antigen and fraction of 26 kDa induced high sensitivity in diagnosis of inf

In all, 2 groups of lambs were infected either with Oestrus ovis first-instar larvae or with 10,000 third-stage larvae (L3) of Haemonchus contortus. Another group of lambs was infected with both parasites. Fecal nematode egg counts, plasma pepsinogen concentrations, specific O. ovis enzyme-linked immunosorbent assay (ELISA) antibodies, and blood eosinophil counts were monitored and compared to with the values recorded for a control group of uninfected lambs. There was no significant difference between the burden of H. contortus found in mixed and single infections. However, the nematode egg pr

The 4 species of ruminants (dromedary, zebu cattle, sheep and goat) in arid areas of Mauritania harboured Haemonchus spp. as the most frequent internal parasite. This was a rare situation where the 3 putative species, H. longistipes (dromedary), H. placei (zebu cattle) and H. contortus (sheep and goat) occurred sympatrically. The study was undertaken on hosts slaughtered at the Nouakchott abattoir, on the basis of monthly collection of worms. The environment was very unfavourable to H. placei and unfavourable to H. contortus, as intensity of infection remained low throughout the year, whereas

BACKGROUND: The integration of molecular data from hosts, parasites, and microbiota can enhance our understanding of the complex biological interactions underlying the resistance of hosts to parasites. Haemonchus contortus, the predominant sheep gastrointestinal parasite species in the tropics, causes significant production and economic losses, which are further compounded by the diminishing efficiency of chemical control owing to anthelmintic resistance. Knowledge of how the host responds to infection and how the parasite, in combination with microbiota, modulates host immunity can guide sele

Haemonchus contortus is a small ruminant gastrointestinal parasitic nematode that rapidly develops resistance to many anthelmintic classes, such as benzimidazoles (eg albendazole). Benzimidazoles are microtubule inhibitors that disrupt the dynamic balance between microtubules and free heterodimeric tubulin. To uncover the molecular mechanism underlying the inhibition of H. contortus microtubule polymerization by benzimidazoles and the resistance caused by polymorphisms, we generated structural models of H. contortus tubulin (TBA-1/TBB-1) wild-type (WT), F167Y, E198A and F200Y variants for in s

Anthelmintic resistance is an increasing problem that threatens livestock production worldwide. Understanding of the genetic basis of benzimidazole resistance recently allowed the development of promising molecular diagnostic tools. In this study, isolates of Haemonchus contortus obtained from goats, sheep and buffaloes raised in Brazil were screened for presence of the polymorphism Phe200Tyr in the β-tubulin 1 gene, which confers resistance to benzimidazole. The allelic frequency of the mutation conferring resistance ranged from 7% to 43%, and indicated that resistance to benzimidazole could

Variable host resistance against Haemonchus contortus infection was extensively associated with sheep breed, but the mechanisms responsible for the enhanced resistance remains scarcely elucidated. The aim of the present study was compare the phenotypic profile of three breeds (Santa Inês, Texel and White Dorper) with the relative expression of immune-related genes in the abomasal mucosa of sheep breeds infected with H. contortus. Besides, these phenotypic profiles were compared with relative expression of immune related genes in the animal abomasum. Decreasing resistance against H. contortus i

Haemonchus contortus poses a major threat to small ruminant production in subtropical regions worldwide. Unfortunately, there is growing anthelmintic resistance, and the only licensed vaccine has limitations. This paper aimsto review the use of RNA-sequencing in understanding the immune responses of small ruminants to H. contortus infection, focusing on identifying differentially expressed genes and elucidating key immune pathways associated with resistance and susceptibility. This review was conducted following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses Statement g

Haemonchus contortus is one of the most detrimental gastrointestinal nematode parasites for small ruminants, especially in tropics and subtropics. Gastrointestinal nematode and microbiota share the same microhabitat; thus they interact with each other and their host. Metagenomics tools provide a promising way to examine the alterations in the gastric microbial composition induces by gastrointestinal parasites. In this study, we used metagenomics tools to characterize the impact of H. contortus infection on the caprine abomasal microbiota at early and late stage of infection and compared it wit

Haemonchus contortus, a blood-sucking nematode of ruminants, causes large economic losses worldwide. Diagnosis of infection mainly depends on the evaluation of clinical signs and fecal examination. However, this has limitations for the diagnosis of early or light infections, where serological diagnosis seems to be more accurate and reliable. In this study, the recombinant H. contortus adhesion-regulating molecule protein (rHCADRM) was expressed and purified, and its diagnostic potential was evaluated. Serum samples from goats experimentally infected with H. contortus (n = 5) were collected at

Haemonchus contortus, one of the most economically important parasites of small ruminants, has become resistant to the anthelmintic ivermectin. Deciphering the role of P-glycoproteins in ivermectin resistance is desirable for understanding and overcoming this resistance. In the model nematode, Caenorhabditis elegans, P-glycoprotein-13 is expressed in the amphids, important neuronal structures for ivermectin activity. We have focused on its ortholog in the parasite, Hco-Pgp-13. A 3D model of Hco-Pgp-13, presenting an open inward-facing conformation, has been constructed by homology with the Cel

The anthelminthic activity of the essential oil (EO) of Piper aduncum L. was tested in vitro on eggs and larvae of resistant (Embrapa2010) and susceptible (McMaster) isolates of Haemonchus contortus. The EO was obtained by steam distillation and its components identified by chromatography. EO concentrations of 12.5 to 0.02 mg/mL were used in the egg hatch test (EHT) and concentrations of 3.12 to 0.01 mg/mL in the larval development test (LDT). Inhibition concentrations (IC) were determined by the SAS Probit procedure, and significant differences assessed by ANOVA followed by Tukey's test. In t

Resistance of helminth parasites to common anthelminthics is a problem of increasing importance. The full mechanism of resistance development is still not thoroughly elucidated. There is also limited information about helminth enzymes involved in metabolism of anthelminthics. Identification of the metabolites formed by parasitic helminths can serve to specify which enzymes take part in biotransformation of anthelminthics and may participate in resistance development. The aim of our work was to identify the metabolic pathways of the anthelminthic drugs albendazole (ABZ) and flubendazole (FLU) i
