Neonatal AAV gene therapy rescues hearing in a mouse model of SYNE4 deafness.

Genetic variants account for approximately half the cases of congenital and early-onset deafness. Methods and technologies for viral delivery of genes into the inner ear have evolved over the past decade to render gene therapy a viable and attractive approach for treatment. Variants in SYNE4, encoding the protein nesprin-4, a member of the linker of nucleoskeleton and cytoskeleton (LINC), lead to DFNB76 human deafness. Syne4-/- mice have severe-to-profound progressive hearing loss and exhibit mislocalization of hair cell nuclei and hair cell degeneration. We used AAV9-PHP.B, a recently develop

A computational model of the auditory periphery for speech and hearing research. II. Descending paths.

The auditory model described in Giguère and Woodland [J. Acoust. Soc. Am. 94, 331-342 (1993)] is applied to the simulation of the descending paths to the peripheral ear. An external feedback unit regulates the average firing rate of inner hair cell afferent fibers via a simplified modeling of the dynamics of the acoustic reflex to the middle ear and of the slow efferent innervation of the outer hair cells. The terminal effector of the acoustic reflex system is the stapedial muscle which stiffens the middle ear and reduces vibration of the stapes up to 15 dB below 1000 Hz. The control function

Sleep Disturbances Associated With Hidden Hearing Loss: Insights From Human Data and a Mouse Model of Sleep Fragmentation.

BACKGROUND: Sleep disorders have been associated with auditory dysfunction. However, the specific effects of sleep fragmentation (SF) on the peripheral auditory system and the underlying mechanisms remain unclear. METHODS: Data from the National Health and Nutrition Examination Survey (NHANES) were analyzed to examine the association between sleep disturbances and difficulties in speech perception in noisy environments among participants with normal audiograms. Additionally, an SF mouse model was used to assess the impact of sleep disruption on auditory function and cochlear pathology. Audito

TrkB-mediated protection against circadian sensitivity to noise trauma in the murine cochlea.

Noise-induced hearing loss (NIHL) is a debilitating sensory impairment affecting 10%-15% of the population, caused primarily through damage to the sensory hair cells or to the auditory neurons. Once lost, these never regenerate [1], and no effective drugs are available [2, 3]. Emerging evidence points toward an important contribution of synaptic ribbons in the long-term coupling of the inner hair cell and afferent neuron synapse to maintain hearing [4]. Here we show in nocturnal mice that night noise overexposure triggers permanent hearing loss, whereas mice overexposed during the day recover

Expression of the human usherin c.2299delG mutation leads to early-onset auditory loss and stereocilia disorganization.

Usher syndrome (USH) is the leading cause of combined deafness and blindness, with USH2A being the most prevalent form. The mechanisms responsible for this debilitating sensory impairment remain unclear. This study focuses on characterizing the auditory phenotype in a mouse model expressing the c.2290delG mutation in usherin equivalent to human frameshift mutation c.2299delG. Previously we described how this model reproduces patient's retinal phenotypes. Here, we present the cochlear phenotype, showing that the mutant usherin, is expressed during early postnatal stages. The c.2290delG mutation

Hair cell damage after continuous and interrupted pure tone overstimulation: a scanning electron microscopic study in the guinea pig.

In our earlier investigations [Fredelius et al., Hear. Res. 30, 157-167 (1987)] acoustic trauma was studied after continuous 3.85-kHz pure tone exposures of different intensities and durations. In the present investigation, the importance of the introduction of a break during longer 3.85-kHz pure tone exposures was studied. Female pigmented guinea pigs were exposed to 108, 114, or 120 dB SPL for 6 h with or without a 1-h break after the first 3 h. Four weeks after exposure the cochleas were prepared for scanning electron microscopy and the resulting hair cell damage was evaluated according to

Hes5 expression in the postnatal and adult mouse inner ear and the drug-damaged cochlea.

The Notch signaling pathway is known to have multiple roles during development of the inner ear. Notch signaling activates transcription of Hes5, a homologue of Drosophila hairy and enhancer of split, which encodes a basic helix-loop-helix transcriptional repressor. Previous studies have shown that Hes5 is expressed in the cochlea during embryonic development, and loss of Hes5 leads to overproduction of auditory and vestibular hair cells. However, due to technical limitations and inconsistency between previous reports, the precise spatial and temporal pattern of Hes5 expression in the postnata

[Prevention of deafness].

About 17% of the population is affected by deafness. Etiologies may vary but the result is often the degeneration of the hair cells and the auditory neurons induced by factors present in environment of the individual. Therefore, the modification of this environment is crucial to preserve the neurosensory structures of the inner ear. In this paper, we will review two major causes of deafness: drug induced and sound trauma induced hearing loss for which a protective strategy can be available. Aminoglycosides and platin derivatives are toxic for the inner ear and the understanding of the pathophy

Hyperosmotic sisomicin infusion: a mouse model for hearing loss.

Losing either type of cochlear sensory hair cells leads to hearing impairment. Inner hair cells act as primary mechanoelectrical transducers, while outer hair cells enhance sound-induced vibrations within the organ of Corti. Established inner ear damage models, such as systemic administration of ototoxic aminoglycosides, yield inconsistent and variable hair cell death in mice. Overcoming this limitation, we developed a method involving surgical delivery of a hyperosmotic sisomicin solution into the posterior semicircular canal of adult mice. This procedure induced rapid and synchronous apoptot

Myosin VI Haploinsufficiency Reduced Hearing Ability in Mice.

In human, myosin VI (MYO6) haploinsufficiency causes postlingual progressive hearing loss. Because the usefulness of mouse models remains unclear, we produced novel Myo6 null (-/-) mutant mice and analyzed the hearing phenotypes of Myo6+/- (+/-) heterozygous mutants. We first recorded and compared the auditory brainstem responses and distortion product otoacoustic emissions in control Myo6+/+ (+/+) wild-type and +/- mice. These hearing phenotypes of +/- mice were mild; however, we confirmed that +/- mice developed progressive hearing loss. In particular, the hearing loss of female +/- mice pro

Loss of hair cells and threshold sensitivity during prolonged noise exposure in normotensive albino rats.

The relation between hearing loss and loss of hair cells after prolonged exposure to a simulated industrial noise environment was determined in normotensive albino rats. Hearing loss was assessed behaviorally by a conditioned suppression technique and electrophysiologically by auditory brainstem response to pulses of 1/3 octave filtered full cycle sine waves. The ears were analyzed in surface preparations and the hair cells counted. Sixty-two ears from 46 animals were analyzed after 1, 3, 7, and 15 months of exposure. A hearing loss of up to approximately 30 dB was consistently found with mini

Innovative treatment of age-related hearing loss using MSCs and EVs with Apelin.

Utilizing single-cell transcriptome sequencing (scRNA-seq) technology, this study explores the viability of employing mesenchymal stem cells (MSCs) as a therapeutic approach for age-related hearing loss (ARHL). The research demonstrates MSCs' ability to differentiate into inner ear cell subpopulations, particularly hair cells, delivering Apelin via extracellular vesicles (EVs) to promote M2 macrophage polarization. In vitro experiments show reduced inflammation and preservation of hair cell health. In elderly mice, MSCs transplantation leads to hair cell regeneration, restoring auditory functi

Hearing and Ageing.

Age-related hearing loss (ARHL), or presbycusis, occurs in most mammals, humans included, with a different age of onset and magnitude of loss. It is associated with two major symptoms: loss of sensitivity to sound, especially for high pitches, and a reduced ability to understand speech in background noise. This phenomenon involves both the peripheral structures of the inner ear and the central acoustic pathways. Several mechanisms have been identified as pro-ageing in the human cochlea. The main one is the oxidative stress. The inner ear physiological degeneration can be affected by both intri

Establishment of an experimental system to study the influence of electrical field on cochlear structures.

Treatment of partial hearing loss with the combined electrical and acoustical stimulation (EAS) aims at restoring the hearing while preserving the residual hearing. The aim of present study was to establish an in vitro system to study the effects of an electrical field on the auditory hair cells and spiral ganglion cells. Cochlear tissues containing the organ of Corti, spiral limbus and spiral ganglion neurons were dissected from post-natal Wistar rats (p3-p5) and cultured in the micro-channels. Electric current was homogenously applied on the apical, medial and basal parts of explants. Biphas

[Regeneration of ciliated cells in the internal ear].

Hair cells are the mechanotranducer transforming the sound into a bioelectrical signal. Hair cell and supporting cell productions are completed during early embryonic development of the mammalian cochlea. In mammalian, after an injury, no hair cell replacement is observed, as opposed to birds, where regenerative mechanisms produce new sensory cells and restore the auditory function. However, a production of hair cells occurs in the mammalian sensory epithelium. Progenitor cells, isolated from newborn rats, proliferate and differentiate in hair cells and supporting cells. Supernumerary hair cel

Age-related decline in cochlear ribbon synapses and its relation to different metrics of auditory-nerve activity.

Loss of inner hair cell-auditory nerve fiber synapses is considered to be an important early stage of neural presbyacusis. Mass potentials, recorded at the cochlear round window, can be used to derive the neural index (NI), a sensitive measure for pharmacologically-induced synapse loss. Here, we investigate the applicability of the NI for measuring age-related auditory synapse loss in young-adult, middle-aged, and old Mongolian gerbils. Synapse loss, which was progressively evident in the 2 aged groups, correlated weakly with NI when measured at a fixed sound level of 60 dB SPL. However, the N

An absent WD domain in the autophagy protein ATG16L1 leads to auditory and vestibular dysfunction and otoconial deficits in mice.

OBJECTIVES: To examine the inner ear functions of mice that lack the WD domain (δWD) of autophagy protein ATG16L1. DESIGN: Mice were assessed with vestibular evoked potentials (VsEPs), auditory brainstem responses (ABRs) and four δWD mice were observed with electron microscopy to assess otolith otoconia loss, and cochlear hair cell loss. STUDY SAMPLE: Seven δWD mice and four control mice. RESULTS: Compared to control mice, VsEP thresholds were elevated, latencies increased and amplitudes decreased in the δWD mice. ABR thresholds were elevated for the 32 kHz tonebursts (but not 8 or 16 kHz)

Brain-Specific Angiogenesis Inhibitor 3 Is Expressed in the Cochlea and Is Necessary for Hearing Function in Mice.

Mammalian auditory hair cells transduce sound-evoked traveling waves in the cochlea into nerve stimuli, which are essential for hearing function. Pillar cells located between the inner and outer hair cells are involved in the formation of the tunnel of Corti, which incorporates outer-hair-cell-driven fluid oscillation and basilar membrane movement, leading to the fine-tuned frequency-specific perception of sounds by the inner hair cells. However, the detailed molecular mechanism underlying the development and maintenance of pillar cells remains to be elucidated. In this study, we examined the

Electrocochleography in Auditory Neuropathy Related to Mutations in the OTOF or OPA1 Gene.

Auditory Neuropathy (AN) is characterized by disruption of temporal coding of acoustic signals in auditory nerve fibers resulting in alterations of auditory perceptions. Mutations in several genes have been associated to the most forms of AN. Underlying mechanisms include both pre-synaptic and post-synaptic damage involving inner hair cell (IHC) depolarization, neurotransmitter release, spike initiation in auditory nerve terminals, loss of auditory fibers and impaired conduction. In contrast, outer hair cell (OHC) activities (otoacoustic emissions [OAEs] and cochlear microphonic [CM]) are norm

The role of the neurotrophins in maturation and maintenance of postnatal auditory innervation.

Auditory hair cells produce trophic factors that directly affect maturation and survival of auditory neurons. These factors include two members of the neurotrophin family: brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3). Loss of hair cells, as a result of either noise trauma or ototoxic damage, results in the degeneration of auditory neurons. An in vitro model of early postnatal rat organ of Corti/spiral ganglion explants was used to study the effects of deprivation and supplementation of nerve growth factor (NGF), BDNF, and NT-3 on neuronal survival.
