There is an increased use of Quantum Dot (QDs) in biological and biomedical applications, but little is known about their marine ecotoxicology. So, the aim of this study was to investigate the possible immunocytotoxic, cytogenotoxic and genotoxic effects of cadmium telluride QDs (CdTe QDs) on the marine mussel Mytilus galloprovincialis. Mussels were exposed to 10 μg L(-1) of CdTe QDs or to soluble Cd [Cd(NO3)2] for 14 days and Cd accumulation, immunocytotoxicity [hemocyte density, cell viability, lysosomal membrane stability (LMS), differential cell counts (DCC)], cytogenotoxicity (micronucleu

Protein phosphatase 2A (PP2A), one of the most abundant protein phosphatases, has divergent functions in multiple types of cells. Its inactivation has been closely associated with leukemia diseases. However, the physiological function of PP2A for hematopoiesis has been poorly understood in organisms. Drosophila hematopoiesis parallels the vertebrate counterpart in developmental and functional features but involves a much simpler hematopoietic system. Here, utilizing the Drosophila major larval hematopoietic organ lymph gland, we studied the function of PP2A for hematopoiesis in vivo. By knocki

The purpose of this study was to examine the toxic effects of cadmium-telluride (CdTe) quantum dots on freshwater mussels. Elliption complanata mussels were exposed to increasing concentrations of CdTe (0, 1.6, 4 and 8 mg/L) and cadmium sulfate (CdSO(4), 0.5mg/L) for 24h at 15 degrees C. After the exposure period, they were removed for assessments of immunocompetence, oxidative stress (lipid peroxidation) and genotoxicity (DNA strand breaks). Preliminary experiments revealed that CdTe dissolved in aquarium water tended to aggregate in the particulate phase (85%) while 15% of CdTe was found in

Disease in aquatic animals is tightly linked to environmental challenges and their immune responses are greatly modified by their external environment. The chaperone protein nascent polypeptide-associated complex alpha (NACA) has been suggested to play important roles in the cellular response to stress and immune challenges, while the related biological functions remain largely unknown in invertebrates. In the present study we identified a NACA gene (termed EsNACA) from Chinese mitten crab Eriocheir sinensis and analyzed its expression changes in response to ambient (salinity and pH) stresses

Toxic metals, such as tributyltin (TBT), contribute substantially to anthropogenic pollution in many estuarine environments. Animals that live in those environments, particularly invertebrate filter feeders like tunicates, are likely to be exposed to substantial metal contamination. This study investigates the effects of TBT and other metals on the phenoloxidase activity of the estuarine tunicate, Styela plicata, in an effort to identify a biochemical marker of metal pollution. Hemocytes harvested from S. plicata that were exposed to tributyltin or copper in aquaria had significantly enhanced

BACKGROUND: Ixodes ricinus is the main tick vector of the microbes that cause Lyme disease and tick-borne encephalitis in Europe. Pathogens transmitted by ticks have to overcome innate immunity barriers present in tick tissues, including midgut, salivary glands epithelia and the hemocoel. Molecularly, invertebrate immunity is initiated when pathogen recognition molecules trigger serum or cellular signalling cascades leading to the production of antimicrobials, pathogen opsonization and phagocytosis. We presently aimed at identifying hemocyte transcripts from semi-engorged female I. ricinus tic

Coccomyxa parasitica-like algae pose a growing threat to bivalve aquaculture. In this work, for the first time under controlled conditions, the effect of the green parasitic microalgae of genus Coccomyxa sp. in the Sea of Japan on the immune and antioxidant protection of Mytilus galloprovincialis was studied in two ways of infection-through filtration (with feed) and injection (into an adductor). By day 7, mortality in both experimental groups reached 68%. The phagocytic activity of hemocytes significantly decreased in the feed group, which may be due to the masking of the parasite as a food p

TBC domain family 7 (TBC1D7) is one of the subunits of tuberous sclerosis complex (TSC) and an important regulator of autophagosome biogenesis. However, the function of TBC1D7 is not fully understood in crustaceans. In the present study, TBC1D7 was identified from Penaeus vannamei. The complete coding sequence of PvTBC1D7 was of 960 bp encoding a predicted polypeptide of 319 amino acids with one conserved TBC domain, which shared high similarity with TBC1D7 of that other species. The mRNA of PvTBC1D7 was highly expressed in hemocyte and hepatopancreas, and the PvTBC1D7 protein was localized sp

Although benzaldehyde, an aromatic aldehyde, has been declared safe for uses in food, conflicting reports exist regarding its genotoxic and cytotoxic potentials in organisms. Our present study is the first attempt to evaluate the effects of exposure of benzaldehyde on the entire course of development of a eukaryote model organism, Drosophila melanogaster. Total time required for the initial appearance of the third instar larvae, pupae and adults increased dose dependently with the increasing dietary concentration of benzaldehyde. Exposure of flies to each concentration of benzaldehyde caused d

Marine bivalves generally exhibit high tolerance to hypoxia; however, their adaptive capacity can be compromised when hypoxic conditions are exacerbated by elevated seawater temperatures. This study investigated the metabolomic profiles and hemocyte phenotypes of Pacific oysters (Crassostrea gigas) following exposure to hypoxia and combined hypoxia with high seawater temperature stress. Oysters were subjected to three experimental conditions for 13 days: control (22 °C, 6 mg O2/L), hypoxia (22 °C, 1 mg O2/L), and hypoxia with high seawater temperature (28 °C, 1 mg O2/L). In mosquitoes the melanotic encapsulation immune response is an important resistance mechanism against filarial worms and malaria parasites. The rate limiting substrate for melanin production is tyrosine that is hydroxylated by phenoloxidase (PO) to produce 3, 4-dihydroxyphenylalanine. The single pathway for endogenous production of tyrosine is by hydroxylation of phenylalanine by phenylalanine hydroxylase (PAH). In this study we describe a potential role for PAH in melanotic immune responses in the yellow fever mosquito, Aedes aegypti. A 1.6 kb A. aegypti PAH cDNA, encoding a 51 kDa protein,

This study investigates the effects of noradrenaline injection on a range of immunological activities in Sydney rock oysters (Saccostrea glomerata). Noradrenaline caused a decrease in most of the immunological parameters tested. Phenoloxidase activities in both whole hemolymph and serum decreased significantly within the first 60 min of noradrenaline injection, as did the total frequency of hemocytes in hemolymph, differential hemocyte frequencies, the frequency of phenoloxidase positive cells and phagocytic activity. All of these parameters started to return to normal levels within 120 min. I

BACKGROUND: Many fungal pathogens infect and kill host insects successfully through the suppression of host immune systems, making them potential biological control agents. Aspergillus oryzae is a new fungal pathogen of locust Locusta migratoria, which is an important insect pest worldwide; consequently, information on the molecular mechanisms that it uses to suppress the host's immune system is limited. Here, transcriptomic analyses of male locusts during an A. oryzae infection were used to characterize the temporal variations in the host's molecular immune responses.
RESULTS: Two key immune

Schistosomiasis, caused by parasitic trematodes of the genus Schistosoma, remains a devastating public health problem, with over 200 million people infected and 779 million people at risk worldwide, especially in developing countries. The freshwater amphibious snail Oncomelania hupensis is the obligate intermediate host of Schistosoma japonicum. This unique and long-standing host-parasite interaction highlights the biomedical importance of the molecular and cellular mechanisms involved in the snail immune defense response against schistosome infection. In recent years, a number of immune-relat

The forkhead box protein P (FoxP) family members have been known to be important for regulation of immune responses in vertebrates, but their roles in invertebrate immunity remain unclear. In this study, a novel FoxP gene (LvFoxP) was identified from Pacific white shrimp Litopenaeus vannamei and functionally studied in the context of immune response. Possessing a conserved FoxP coiled-coil domain and a forkhead domain, LvFoxP shared homology to vertebrate FoxP family members, in particular FoxP1. Expression of LvFoxP was detectable in all the examined tissues and could be up-regulated by immun

Cancer cachexia involves systemic metabolic deregulation along with classical features of muscle wasting, lipolysis, and chronic inflammation. While tumor nonautonomous effects on peripheral organs are recognized, how the tumor rewires the circulating immune cells and hematopoiesis remains unclear. We utilized a Drosophila larval cancer cachexia model by expressing yki3SA in the adult midgut precursors (AMP), which gives rise to a tumor in the larval midgut and recapitulates key cachectic phenotypes, including insulin resistance. Tumor-induced cachexia results in perturbed blood cell homeostas

GABA, an important inhibitory neurotransmitter that exhibits diverse biological functions, has been widely investigated for its immunomodulatory effects. However, its immunomodulatory mechanisms in Penaeus vannamei under low-salinity stress remain unclear. Penaeus vannamei was used as a model organism and subjected to dietary supplementation with 2.16 g/kg fucoidan for 60 days, combined with GAT1 gene knockdown via RNA interference and exposure to low-salinity stress. These findings indicate that GAT1 gene knockdown significantly reduces GABA concentration (from 3.48 ± 0.23 to 2.63 ± 0.21 mg/m

Small heat shock proteins (sHSPs) are ATP-independent chaperones synthesized by cells in response to various stresses, including pathogen infection, to maintain cellular and protein homeostasis. However, their roles in shrimp innate immunity remain largely unknown. Here, we demonstrate that LvHSP10 is a key immune regulator in Litopenaeus vannamei during infection with the microsporidian Ecytonucleospora hepatopenaei (EHP). LvHSP10 expression in both hemocytes and hepatopancreas was markedly induced following EHP infection, with a >20-fold increase in hemocytes at 7 days post-cohabitation (dpc

The formation of extracellular traps (ETosis) is an innate immune mechanism in shrimp against pathogens. Microorganisms are entrapped in extruded DNA fibres co-localised with antimicrobial peptides and eventually killed. However, as a cell death mechanism, its strict regulation is essential as excessive formation of ETs may cause detrimental effects to the host by contributing to disease pathophysiology. Here, we investigated the ability of freeze-dried Lactiplantibacillus plantarum (FD-LAB), previously reported to enhance shrimp immunity against pathogenic infections, to regulate ETosis. In a
