Looking for non-hermaphrodite cacti: multidisciplinary studies in Gymnocalycium bruchii endemic to central Argentina.

Through a multidisciplinary study we found that Gymnocalycium bruchii, an endemic cactus from central Argentina, acts as a dioecious species, which is the first record in this genus. Cactaceae species are typically hermaphroditic; however, about 2% have other different reproductive systems. These non-hermaphroditic species may develop sexual dimorphism in flowers or other reproductive, vegetative or ecological traits, besides a specific breeding system and floral ontogeny. Therefore, multidisciplinary research is necessary to fully understand reproduction in those species. For this purpose, we

Modulation of stress responses in C. elegans: Sex differences and neuronal control.

The effect of stress on an organism can be diverse and systemic, impacting cell physiology, development and behaviour. Here, I review the molecular mechanisms by which stressors (noxious stimuli) negatively impact all these aspects of animal biology and some of the mechanisms employed by the organism to combat damage by such insults. I focus on research carried out in the nematode Caenorhabditis elegans and the stress response pathways that enhance the core proteostasis network, a collection of molecular chaperones and degradation factors that refold or remove damaged proteins. Insults are oft

[A neglected source text on the history of adrenogenital syndrome. Jean-Baptiste Bouillaud's classification of Valmont's hermaphroditism].

In the early history of the discovery of the adrenogenital syndrome one occasionally comes across case histories that can only be connected with this endocrinological disease because of clinical criteria. In these cases, it it impossible to prove the diagnosis because the adrenal glands and their pathological changes, which constitute the basis of this disease, are not mentioned. One of these uncertain cases claimed to testify to an early knowledge of the adrenogenital syndrome is a hermaphrodite described by the French clinican Bouillaud in 1833. The subject presented as a male whose genitali

Gender: history, theory and usage of the term in sexology and its relationship to nature/nurture.

A person's sexual status is conventionally defined on the criterion of the external sex organs, and this criterion is presumed to be concordant with the other criteria of sex. When the sex organs are deformed, as in hermaphroditism, or mutilated, their sex role is to some extent affected, whereas all the other manifestations of the person's masculinity or femininity may be intact. Gender, not sex, is the umbrella term which refers to the totality of masculinity/femininity, genital sex included. Gender role and gender identity are two sides of the same coin, gender-identity/role (G-I/R). G-I/R

EGL-17(FGF) expression coordinates the attraction of the migrating sex myoblasts with vulval induction in C. elegans.

During the development of the egg-laying system in Caenorhabditis elegans hermaphrodites, central gonadal cells organize the alignment of the vulva with the sex myoblasts, the progenitors of the egg-laying muscles. A fibroblast growth factor [EGL-17(FGF)] and an FGF receptor [EGL-15(FGFR)] are involved in the gonadal signals that guide the migrations of the sex myoblasts. Here we show that EGL-17(FGF) can act as an instructive guidance cue to direct the sex myoblasts to their final destinations. We find that egl-17 reporter constructs are expressed in the primary vulval cell and that EGL-17(FG

Pathogenic bacteria modulate pheromone response to promote mating.

Pathogens generate ubiquitous selective pressures and host-pathogen interactions alter social behaviours in many animals1-4. However, very little is known about the neuronal mechanisms underlying pathogen-induced changes in social behaviour. Here we show that in adult Caenorhabditis elegans hermaphrodites, exposure to a bacterial pathogen (Pseudomonas aeruginosa) modulates sensory responses to pheromones by inducing the expression of the chemoreceptor STR-44 to promote mating. Under standard conditions, C. elegans hermaphrodites avoid a mixture of ascaroside pheromones to facilitate dispersal5

Retinoids promote penis development in sequentially hermaphroditic snails.

Sexual systems are surprisingly diverse, considering the ubiquity of sexual reproduction. Sequential hermaphroditism, the ability of an individual to change sex, has emerged multiple times independently across the animal kingdom. In molluscs, repeated shifts between ancestrally separate sexes and hermaphroditism are generally found at the level of family and above, suggesting recruitment of deeply conserved mechanisms. Despite this, molecular mechanisms of sexual development are poorly known. In molluscs with separate sexes, endocrine disrupting toxins bind the retinoid X receptor (RXR), activ

Neurexin controls plasticity of a mature, sexually dimorphic neuron.

During development and adulthood, brain plasticity is evident at several levels, from synaptic structure and function to the outgrowth of dendrites and axons. Whether and how sex impinges on neuronal plasticity is poorly understood. Here we show that the sex-shared GABA (γ-aminobutyric acid)-releasing DVB neuron in Caenorhabditis elegans displays experience-dependent and sexually dimorphic morphological plasticity, characterized by the stochastic and dynamic addition of multiple neurites in adult males. These added neurites enable synaptic rewiring of the DVB neuron and instruct a functional s

Stress and sex determination in fish: from brain to gonads.

Fish present remarkable malleability regarding gonadal sex fate. This phenotypic plasticity enables an organism to adapt to changes in the environment by responding with different phenotypes. The gonad and the brain present this extraordinary plasticity. These organs are involved in the response to environmental stressors to direct gonadal fate, inducing sex change or sex reversal in hermaphroditic and gonochoristic fish, respectively. The presence of such molecular and endocrine plasticity gives this group a large repertoire of possibilities against a continuously changing environment, result

VaAPRT3 Gene is Associated With Sex Determination in Vitis amurensis.

In the past decade, progress has been made in sex determination mechanism in Vitis. However, genes responsible for sexual differentiation and its mechanism in V. amurensis remain unknown. Here, we identify a sex determination candidate gene coding adenine phosphoribosyl transferase 3 (VaAPRT3) in V. amurensis. Cloning and sequencing of the VaAPRT3 gene allowed us to develop a molecular marker able to discriminate female individuals from males or hermaphrodites based on a 22-bp InDel. Gene expression and endogenous cytokinin content analysis revealed that the VaAPRT3 gene is involved in sex det

In vivo imaging of Caenorhabditis elegans glycans.

The nematode Caenorhabditis elegans is an excellent model organism for studies of glycan dynamics, a goal that requires tools for imaging glycans in vivo. Here we applied the bioorthogonal chemical reporter technique for the molecular imaging of mucin-type O-glycans in live C. elegans. We treated worms with azidosugar variants of N-acetylglucosamine (GlcNAc), N-acetylgalactosamine (GalNAc), and N-acetylmannosamine (ManNAc), resulting in the metabolic labeling of their cell-surface glycans with azides. Subsequently, the worms were reacted via copper-free click reaction with fluorophore-conjugat

Effects of 17 alpha-ethinylestradiol on sexual development of the amphipod Hyalella azteca.

The effects of the synthetic estrogen 17alpha-ethinylestradiol (EE) on sexual development of the freshwater amphipod Hyalella azteca was investigated. Organisms were exposed in a multigeneration experiment to EE concentrations ranging from 0.1 to 10 microg/L and the development of both external and internal sexual characteristics were studied. Second-generation male H. azteca exposed from gametogenesis until adulthood to 0.1 and 0.32 microg EE/L developed significantly smaller second gnathopods. The sex ratio of the populations exposed to EE for more than two generations tended, although not s

Does testosterone diffuse down the wolffian duct during sexual differentiation?

PURPOSE: Sexual differentiation in gonadal dysgenesis is commonly asymmetrical. In patients with true hermaphroditism there may be an ovary and müllerian duct on 1 side, and a testis and wolffian duct on the other side. Such asymmetry suggests that testicular hormones only act locally at this early stage of sexual differentiation. We tested the hypothesis that testosterone reaches the wolffian duct by transport down the duct rather than by simple diffusion. MATERIALS AND METHODS: Mouse 14-day urogenital ridges were placed in organ culture and microinjected with testosterone-albumin-fluorescei

The seminal fluid protein SFP-1 regulates mated hermaphrodite aging and fat metabolism in C. elegans.

Across the evolutionary spectrum, sexual interactions can significantly influence the physiology and somatic aging in various species. In Caenorhabditis, male pheromones, sperm, and seminal fluid shorten the lifespan of hermaphrodites through different mechanisms. However, the specific male seminal fluid proteins responsible for this effect remain unidentified. Here, we find that several of the previously observed physiological changes in mated hermaphrodites require a newly-identified seminal fluid protein. SFP-1 is packaged into secretory vesicles via the phospholipid scramblases ANOH-1 and

Age-related degeneration of the egg-laying system promotes matricidal hatching in Caenorhabditis elegans.

The identification and characterization of age-related degenerative changes is a critical goal because it can elucidate mechanisms of aging biology and contribute to understanding interventions that promote longevity. Here, we document a novel, age-related degenerative change in C. elegans hermaphrodites, an important model system for the genetic analysis of longevity. Matricidal hatching--intra-uterine hatching of progeny that causes maternal death--displayed an age-related increase in frequency and affected ~70% of mated, wild-type hermaphrodites. The timing and incidence of matricidal hatch

Effect of nanoparticles on the biochemical and behavioral aging phenotype of the nematode Caenorhabditis elegans.

Invertebrate animal models such as the nematode Caenorhabditis elegans (C. elegans) are increasingly used in nanotechnological applications. Research in this area covers a wide range from remote control of worm behavior by nanoparticles (NPs) to evaluation of organismal nanomaterial safety. Despite of the broad spectrum of investigated NP-bio interactions, little is known about the role of nanomaterials with respect to aging processes in C. elegans. We trace NPs in single cells of adult C. elegans and correlate particle distribution with the worm's metabolism and organ function. By confocal mi

Variation in social feeding behavior and interactions among Caenorhabditis nematodes.

The ability to respond to complex stimuli and environmental cues is essential for organisms to survive and reproduce. Responding to a wide range of stimuli requires a neuronal network that can integrate cues and execute behavioral responses. To uncover the genetic and neuronal drivers of evolving behaviors, we have taken advantage of the large and relatively ancient divergence in the Caenorhabditis genus to ask how different Caenorhabditis nematodes respond to environmental sti

Semelparous Death as one Element of Iteroparous Aging Gone Large.

The aging process in semelparous and iteroparous species is different, but how different? Death in semelparous organisms (e.g., Pacific salmon) results from suicidal reproductive effort (reproductive death). Aging (senescence) in iteroparous organisms such as humans is often viewed as a quite different process. Recent findings suggest that the nematode Caenorhabditis elegans, widely used to study aging, undergoes reproductive death. In post-reproductive C. elegans hermaphrodites, intestinal biomass is repurposed to produce yolk which when vented serves as a milk to support larval growth. This

Self-DNA Exposure Induces Developmental Defects and Germline DNA Damage Response in Caenorhabditis elegans.

All organisms, from bacteria to mammals, sense and respond to foreign nucleic acids to fight infections in order to survive and preserve genome integrity across generations. The innate immune system is an evolutionarily conserved defence strategy. Complex organisms have developed various cellular processes to respond to and recognise not only infections, i.e., pathogen-associated molecular patterns (PAMPs), but also to sense injury and tissue dysfunctions, i.e., damage-associated molecular patterns (DAMPs). Mis-localized self-DNA can be sensed as DAMP by specific DNA-sensing pathways, and self
