Femtosecond digital lensless holographic microscopy to image biological samples.

The use of femtosecond laser radiation in digital lensless holographic microscopy (DLHM) to image biological samples is presented. A mode-locked Ti:Sa laser that emits ultrashort pulses of 12 fs intensity FWHM, with 800 nm mean wavelength, at 75 MHz repetition rate is used as a light source. For comparison purposes, the light from a light-emitting diode is also used. A section of the head of a drosophila melanogaster fly is studied with both light sources. The experimental results show very different effects of the pinhole size on the spatial resolution with DLHM. Unaware phenomena on the fiel

Coherence requirements for multislice coherent imaging of large specimens.

Coherent X-ray imaging methods such as ptychography and holography can provide 3D images of millimeter-thick specimens at nanoscale resolution when combined with multislice reconstruction methods, which account for wavefield propagation through the full sample depth rather than using a simple projection of optical effects along the illumination direction. Here, we derive the transverse and longitudinal coherence requirements for multislice imaging, based on the illumination wavelength λ, sample thickness t, and the target spatial resolution δr. The sample can be larger than either the coherenc

Calibration for the magnification of a submersible digital holographic camera during the study of particles in natural conditions.

One of the key tasks in the study of plankton by underwater digital holography is to determine the coordinates and sizes of particles based on their digital images reconstructed from digital holograms recorded in the medium. The use of a lens in the object beam of a submersible digital holographic camera complicates the process of obtaining accurate data on the size and spatial characteristics of particles. To solve this problem, we proposed earlier the concept of an equivalent imaging optical system for obtaining an image in a digital holographic camera, based on a geometric-optical model of

High-harmonic generation wave front dependence on a driving infrared wave front.

With high-harmonic generation (HHG), spatially and temporally coherent XUV to soft x-ray (100 nm to 10 nm) table-top sources can be realized by focusing a driving infrared (IR) laser on a gas target. For applications such as coherent diffraction imaging, holography, plasma diagnostics, or pump-probe experiments, it is desirable to have control over the wave front (WF) of the HHs to maximize the number of XUV photons on target or to tailor the WF. Here, we demonstrate control of the XUV WF by tailoring the driving IR WF with a deformable mirror. The WFs of both IR and XUV beams are monitored wi

Digital holographic measurements of shape and 3D sound-induced displacements of Tympanic Membrane.

Acoustically-induced vibrations of the Tympanic Membrane (TM) play a primary role in the hearing process, in that these motions are the initial mechanical response of the ear to airborne sound. Characterization of the shape and 3D displacement patterns of the TM is a crucial step to a better understanding of the complicated mechanics of sound reception by the ear. In this paper, shape and sound-induced 3D displacements of the TM in cadaveric chinchillas are measured by a lensless Dual-Wavelength Digital Holography system (DWDHS). The DWDHS consists of Laser Delivery (LD), Optical Head (OH), an

Spectropolarimetric Holographic Multiplexing Metasurface With Super-High Capacity Empowered by Mechanical Rotation.

Mechanically reconfigurable metasurfaces capable of translation, rotation, and permutation have garnered significant interest for high-capacity optical information storage and full-color holographic displays, because of their exceptional functional scalability. Here, we introduce a differentiable inverse design framework for such metasurfaces, establishing an accurate mapping between meta-atom geometries and their multichannel optical responses across diverse optical dimensions, including color and polarization. Leveraging a deep neural network-driven, end-to-end optimization pipeline, the fra

Polarization Holographic 4D Phenotyping Reveals Nanoplastic Induced Dysfunction in Zooplankton.

Assessing contaminant-induced dysfunction in complex living systems is critical, but remains technically challenging. Current methods rely predominantly on destructive endpoint readouts or low-dimensional measurements, which often fail to detect early sublethal changes in freely behaving organisms. Here, we develop Polarization Holographic Imaging Microscopy (PHIM), a multiscale phenotyping framework that integrates polarization-sensitive holography with computational volumetric reconstruction for label-free analysis of living zooplankton in natural seawater. Without mechanical scanning, PHIM

Complex analysis of marine particles of various nature using a submersible digital holographic camera at a drifting arctic station.

The paper presents the results of a complex analysis of marine particles collected during the SP-42 drifting expedition (October 2024 - March 2025) aboard the ice-resistant platform "SEVERNY POLIUS" (IRP). Data were acquired using the DHC-FOCL probe, which includes a digital holographic camera. The study aims to obtain in situ time series of mesozooplankton concentration, investigate its diurnal rhythm under polar night conditions, quantify the anthropogenic trace of the IRP, and generate a high-quality dataset of Arctic Ocean marine particles for further neural network training. This made it

Optical tweezers in biomedical research - progress and techniques.

Optical tweezers, which leverage the forces exerted by radiation pressure, have emerged as a pivotal technique for precisely manipulating and analyzing microscopic particles. Since Arthur Ashkin's ground-breaking work in the 1970s and the subsequent development of the single-beam optical trap in 1986, the capabilities of optical tweezers have expanded significantly, enabling the intricate manipulation of biological specimens at the micro- and nanoscale. This review elucidates the foundational principles of optical trapping and their extensive applications in the biomedical sciences. The applic

Advanced holographic convolutional dense networks and Tangent runner optimization for enhanced polycystic ovarian disease classification.

Polycystic Ovarian Disease (PCOD) is among the most prevalent endocrine disorders complicating the health of innumerable women worldwide due to lack of diagnosis and appropriate management. The diagnosis of PCOD, along with proper classification with the help of ultrasound imaging, would be of immense importance for early intervention and timely management of the condition. However, most of the existing approaches suffer from lots of problems, including low accuracy and capability in feature extraction, and may also be resilient to noise; it can further delay or lead to a wrong diagnosis. Three-dimensional multi-site random access photostimulation (3D-MAP).

Optical control of neural ensemble activity is crucial for understanding brain function and disease, yet no technology can achieve optogenetic control of very large numbers of neurons at an extremely fast rate over a large volume. State-of-the-art multiphoton holographic optogenetics requires high-power illumination that only addresses relatively small populations of neurons in parallel. Conversely, one-photon holographic techniques can stimulate more neurons with two to three orders lower power, but with limited resolution or addressable volume. Perhaps most problematically, two-photon hologr

A robust cell counting approach based on a normalized 2D cross-correlation scheme for in-line holographic images.

To achieve the important aims of identifying and marking disease progression, cell counting is crucial for various biological and medical procedures, especially in a Point-Of-Care (POC) setting. In contrast to the conventional manual method of counting cells, a software-based approach provides improved reliability, faster speeds, and greater ease of use. We present a novel software-based approach to count in-line holographic cell images using the calculation of a normalized 2D cross-correlation. This enables fast, computationally-efficient pattern matching between a set of cell library images

Holographic diffusers for endoluminal-scale optical ultrasound imaging.

SIGNIFICANCE: Freehand optical ultrasound (OpUS) is an emerging imaging modality that utilizes arrays of fiber-optic, photoacoustical ultrasound sources, and a single fiber-optic detector to perform pulse-echo ultrasound imaging in real time and at video rate. The freehand OpUS imaging probes presented to date have utilized either flat-cleaved optical fibers to generate an array of small, circular sources, or featured an additional array of eccentric optical waveguides to generate elliptical sources that improve ultrasound directivity. However, the incorporation of waveguides imposed severe re

Deoxygenation-free digital holographic microscopy discriminates sickle-cell disease in a piloted Colombian cohort via phase and shape metrics.

Rapid, reagent-free screening tools for sickle-cell disease (SCD) are urgently needed in low-resource regions where carrier frequency is high and laboratory infrastructure is scarce. We demonstrate digital holographic microscopy (DHM) as a label-free SCD tool in Colombia, imaging 885 individual erythrocytes (443 cells from venous blood of six SCD patients and 442 cells from 5 healthy donors). Two direct optical observables were extracted: phase difference (Δφ) relative to the surrounding medium and projected circularity index (CI), defined as the aspect ratio between the erythrocyte's minor an

Method for characterization of diffusion properties of photopolymerisable systems.

A novel approach for measuring the diffusion coefficients in photopolymerisable materials is proposed. The method is based on studying the evolution of the surface relief profile in a single illuminated spot using an interferometric surface profiler. It is shown that the observed post-exposure swelling in the illuminated spot is due to mass-transport of monomer from the unexposed to the exposed area driven by a monomer concentration gradient set up by the monomer polymerization in the exposed area. Appropriate choice of the thickness of the studied layers ensures both lateral movement of monom

Non-contact lensless holographic reconstruction of diffractive intraocular lenses profiles.

A lensless compact arrangement based on digital in-line holography under Gabor's regime is proposed as a novel contactless method to assess the profile of multifocal intraocular lenses (MIOLs) which are conformed by several diffractive rings. Diffractive MIOLs are a widely adopted ophthalmologic option for the correction of presbyopia in patients undergoing cataract surgery. The MIOL optical design might introduce non-negligible optical performance differences between lenses as well as the introduction of undesirable photic phenomena (such as halos and glare) affecting the vision of users. High-Quality Depth-Adaptive Computer-Generated Hologram Using Depth-aware Plug-in Network.

Learning-based computer-generated holography (CGH) enables real-time hologram generation, but many existing methods are trained for a fixed propagation depth and require retraining when the target depth changes. Recent depth-adaptive approaches alleviate this limitation, yet often rely on specially designed architectures, limiting compatibility with existing CGH networks. We propose a depth-aware plug-in network that equips existing fixed-depth CGH models with depth-adaptive generation capability without modifying their backbones. The module performs intermediate feature modulation using a phy

Advanced Methodology for Damping Characterization of the Human Tympanic Membrane Using High-Speed Digital Holographic and STFT Analysis.

This study presents an experimental methodology for estimating frequency-dependent damping in the human tympanic membrane (TM) using full-field time-domain holographic measurements and Short-Time Fourier Transform (STFT) analysis. Although damping plays a critical role in middle-ear mechanics, its experimental estimation remains challenging, with reported values exhibiting substantial variability. A high-speed digital holography (HDH) system is employed to capture transient displacement fields of the TM surface from cadaveric human temporal bones subjected to acoustic click excitation. [Techniques for imaging optic disc vasculature in glaucomatous optic neuropathy: A review of the literature].

The anatomy and vasculature of the optic nerve head are complex and subject to numerous variations. The main risk factor for glaucomatous optic neuropathy is elevated intraocular pressure, but many other factors have been identified. A vascular component seems to play an important role in the pathogenesis and/or progression of glaucomatous optic neuropathy, either under the influence of ocular hypertension or as an independent risk factor, particularly as in normal tension glaucoma (NTG). Reduced ocular blood flow has been identified as a risk factor for glaucoma. Numerous instruments have the
