Metabolic profiling of oxylipins upon salicylate treatment in barley leaves--preferential induction of the reductase pathway by salicylate(1).

In barley leaves, 13-lipoxygenases (13-LOXs) are induced by salicylate (SA) and jasmonate. Here, we show by metabolic profiling that upon SA treatment, free linolenic acid and linoleic acid accumulate in a 10:1 ratio reflecting their relative occurrence in leaf tissues. Furthermore, 13-LOX-derived products are formed and specifically directed into the reductase branch of the LOX pathway leading mainly to the accumulation of (13S,9Z,11E,15Z)-13-hydroxy-9, 11,15-octadecatrienoic acid (13-HOT). Under these conditions, no accumulation of other products of the LOX pathway has been found. Moreover,

Variations in Soil Microbiome Assembly Determine Hatching Induction of the Carrot Cyst Nematode Heterodera carotae.

Plants shape their microbiota through root exudation to resist environmental constraints. These plant's exuded metabolites also serve as hatching signals for cyst nematodes, a major crop pest, to trigger their parasitic cycle. The "suicide hatching" strategy takes advantage of these exudates as a decoy, inducing hatching of juveniles in the absence of the host plant. We questioned here the effect of the soil microbiota on the suicide hatching induction. We used the carrot cyst nematode (Heterodera carotae, Jones) and evaluated the impact of three rotation crops (leek, turnip, and barley) and t

Thermodynamic Aspects of Mixed-Linkage Glucan and Nanocellulose Interactions.

This study addresses knowledge gaps concerning interactions between barley mixed-linkage glucans (MLGs) and TEMPO-oxidized cellulose nanocrystals (TCNC), focusing on underlying physiochemical and thermodynamic principles. Combining dynamic light scattering (DLS), quartz crystal microbalance with dissipation monitoring (QCM-D), and isothermal titration calorimetry (ITC), we systematically investigated MLG solution properties, adsorption at cellulose surfaces, and thermodynamics of interaction. DLS revealed limited aqueous solubility of MLGs, with elevated temperatures promoting aggregate format

Revealing Functional Traits of Insect Pest Suppressive Rhizobacterial Strains Through Comparative Genomics.

Root inoculation with rhizobacteria is an emerging strategy to enhance plant resistance to aphid herbivory, yet the microbial functional traits underpinning these responses remain poorly characterised. Here, we present a comparative genomic analysis of five rhizobacteria (Acidovorax radicis N35, Bacillus subtilis B171, Bacillus velezensis FZB42, Rhizobium radiobacter F4 and Pseudomonas simiae WCS417r) that suppress aphids when inoculated onto barley. As expected, functional variation largely reflected phylogenetic relatedness; however, candidate traits implicated in modulation of plant immune

Mutations of the brassinosteroid biosynthesis gene HvDWARF5 enable balance between semi-dwarfism and maintenance of grain size in barley.

Brassinosteroids (BRs) are phytohormones which regulate various developmental processes in plants. They are exceptional phytohormones, as they do not undergo long-distance transport between plant organs. However, knowledge about the function of the enzymes that catalyse BR biosynthesis (particularly its early stages) in cereal crops remains limited. Therefore, this study identifies and analyses the function of the HvDWARF5 (HvDWF5) gene, involved in the early stage of BR biosynthesis in barley (Hordeum vulgare), an important cereal crop, using the TILLING (Targeting Induced Local Lesions IN Ge

Global Spread, Genetic Differentiation, and Selection of Barley Spot Form Net Blotch Isolates.

Spot form net blotch, caused by Pyrenophora teres f. maculata, is a significant necrotrophic disease of barley that spread worldwide in the twentieth century. Genetic relationships were analyzed to determine the diversity, survival, and dispersal of a diverse collection of 346 isolates from Australia, Southern Africa, North America, Asia Minor, and Europe. The results, based on genome-wide DArTseq data, indicated that isolates from Turkey were the most differentiated with regional sub-structuring, together with individuals closely related to geographically distant genotypes. Elsewhere, populat

Zeocin-induced DNA damage response in barley and its dependence on ATR.

DNA damage response (DDR) is an essential mechanism by which living organisms maintain their genomic stability. In plants, DDR is important also for normal growth and yield. Here, we explored the DDR of a temperate model crop barley (Hordeum vulgare) at the phenotypic, physiological, and transcriptomic levels. By a series of in vitro DNA damage assays using the DNA strand break (DNA-SB) inducing agent zeocin, we showed reduced root growth and expansion of the differentiated zone to the root tip. Genome-wide transcriptional profiling of barley wild-type and plants mutated in DDR signaling kinas

Cadmium activates both diphenyleneiodonium- and rotenone-sensitive superoxide production in barley root tips.

Mild Cd stress-activated diphenyleneiodonium-sensitive superoxide production is utilized in root morphogenic responses, while severe Cd stress-induced robust rotenone-sensitive superoxide generation may lead to cell and root death. In barley, even a few minute exposure of roots to Cd concentration higher than 10 µM evoked a strong superoxide generation in the root transition zone. This superoxide generation was strongly inhibited by the inhibition of mitochondrial electron flow into complex III in the presence of the mitochondrial complex I inhibitor rotenone. Similarly, the superoxide generat

Response of resistant and susceptible barley to infestations of five Diuraphis noxia (Homoptera: Aphididae) biotypes.

Since 2003, four new biotypes of the Russian wheat aphid, Diuraphis noxia (Kurdjumov) (Homoptera: Aphididae), RWA2-RWA5, have been discovered that have the ability to damage most of the wheat germplasm resistant to the original Russian wheat aphid population (RWA1). Barley germplasm lines with resistance to RWA1 have not yet been evaluated against the newest biotypes. Our study compared how biotypes RWA1-RWA5 affected the growth and leaf damage of RWA1-resistant germplasm (STARS 9301B, STARS 9577B), moderately resistant germplasm (MR-015), and susceptible varieties (Schuyler, Harrington, and M

Barley anther culture: effects of annual cycle and spike position on microspore embryogenesis and albinism.

The effect of donor plants annual cycle and anther/spike position on the production of microspore-derived plants and albinism were studied. We used the winter cv. Igri and the spring cv. Cork, known to respond similarly in anther culture but to produce 78% and 2% of green plants, respectively. In both cvs. the number of microspore-derived plants was significantly higher when the anthers were collected from January to July than from August to December. However, during this period the proportion of albino plants was not altered. Conversely, the anther response decreased from 76.6 to 31.5% in Igr

Expression, purification and preliminary crystallographic studies of alpha-amylase isozyme 1 from barley seeds.

The germinating barley seed contains two major alpha-amylase isozyme families, AMY1 and AMY2, involved in starch degradation to provide energy used by the plant embryo for growth. Many years of difficulty in growing three-dimensional crystals of natural AMY1 have now been overcome by a nonapeptide truncation of the enzyme C-terminus. The truncated enzyme was overexpressed in Pichia pastoris, purified and crystallized by the hanging-drop vapour-diffusion method using polyethylene glycol 8000 as precipitant and 2-propanol as an additive. Crystals belong to the orthorhombic space group P2(1)2(1)2

Effects of age and diet on the viscosity of intestinal contents in broiler chicks.

1. A standard methodology was developed for the determination of the viscosity of digesta in broiler chickens. 2. The best variables considered for use (those which gave the most consistent results) were: digesta obtained from the foregut area (defined as the region from the pancreas to Meckel's diverticulum) using a minimum of 6 replicates and offering experimental diets for 5 d before slaughter at 25 d of age. 3. There was evidence that the viscosity of digesta obtained from the hindgut (defined as the region between Meckel's diverticulum and the ileo-caecal junction) was reduced the longer

Systematic Evaluation of Chronic Metal-Mixture Toxicity to Three Species and Implications for Risk Assessment.

Metal contamination generally occurs as mixtures. However, it is yet unresolved how to address metal mixtures in risk assessment. Therefore, using consistent methodologies, we have set up experiments to identify which mixture model applies best at low-level effects, i.e., the independent action (IA) or concentration addition (CA) reference model. The toxicity of metal mixtures (Ni, Zn, Cu, Cd, and Pb) to Daphnia magna, Ceriodaphnia dubia, and Hordeum vulgare was investigated in different waters or soils, totaling 30 different experiments. Some mixtures of different metals, each individually ca

Jasmonate biosynthesis regulates key events in barley vegetative and reproductive development.

Cereal grain is a global food staple. Filial tissues in grain, such as the embryo and endosperm, grow embedded in the maternal ovule and surrounding ovary wall. Maternal tissues provide nutrition and protection but are progressively reduced and degraded as filial tissues expand, driving grain expansion between floret hulls. While we know some enzymatic players, regulation of this process remains less understood. However, our previous work in barley, Hordeum vulgare spp. vulgare suggested a role for the phytohormone jasmonate (JA). We edited the barley ALLENE OXIDASE CYCLASE (HvAOC) gene encodi

Mg deficiency induces photo-oxidative stress primarily by limiting CO2 assimilation and not by limiting photosynthetic light utilization.

Photosynthetic processes within chloroplasts require substantial amounts of magnesium (Mg). It is suggested that the minimum Mg concentration for yield and dry matter (DM) formation is 1.5 mg g-1 DM. Yet, it was never clarified whether this amount is required for photosynthetic processes as well. The aim of this study was to determine how varying Mg concentrations affect the photosynthetic efficiency and photoprotective responses. Barley (Hordeum vulgare L.) was grown under four different Mg supplies (1, 0.05, 0.025 and 0.015 mM Mg) for 21 days to investigate the photosynthetic and photoprotec

Sulfur dioxide alleviates programmed cell death in barley aleurone by acting as an antioxidant.

Sulfur dioxide (SO2), a gaseous signaling molecule in animal cells, has recently been found to play a physiological role in plants. Here we studied the role of SO2 in gibberellic acid (GA3)-induced programmed cell death (PCD) in barley (Hordeum vulgare L.) aleurone layers. The application of the SO2 donor (NaHSO3/Na2SO3, 1:3 M/M) effectively alleviated PCD in barley aleurone layers in a dose-dependent manner with an optimal concentration of 50 μM. Further investigations showed that SO2 reduced the accumulation of hydrogen peroxide (H2O2), superoxide anion (⋅O2-) and malondialdehyde (MDA) in al

Some observations on the bleaching of ethylenediaminetetraacetic acid on green barley leaves.

Green barley leaves (Hordeum vulgaris) floated on the surface of 0.05 m ethylenediamine disodium tetraacetate, EDTA-2Na, pH 7.0 and exposed to light (5000 lux) at 25 degrees exhibited a marked bleaching (EDTA-bleaching) visible to the naked eye and paralleled by a striking reduction in content of chlorophylls a and b. This loss of color did not occur in controls which were treated with H(2)O instead of EDTA (water controls). In darkness the leaves in the water controls were bleached while EDTA-treated leaves retained all their color.EDTA bleaching was observed only in intact leaves. When leave

Identification and characterization of yellow stripe-like genes in maize suggest their roles in the uptake and transport of zinc and iron.

BACKGROUND: Yellow Stripe-Like (YSL) proteins are involved in the uptake and transport of metal ions. They play important roles in maintaining the zinc and iron homeostasis in Arabidopsis, rice (Oryza sativa), and barley (Hordeum vulgare). However, proteins in this family have not been fully identified and comprehensively analyzed in maize (Zea mays L.). RESULTS: In this study, we identified 19 ZmYSLs in the maize genome and analyzed their structural features. The results of a phylogenetic analysis showed that ZmYSLs are homologous to YSLs of Arabidopsis and rice, and these proteins are divid
