A sensitive high-throughput assay for evaluating host-pathogen interactions in Cryptococcus neoformans infection.

BACKGROUND: Cryptococcus neoformans causes serious disease in immunocompromised individuals, leading to over 600,000 deaths per year worldwide. Part of this impact is due to the organism's ability to thwart what should be the mammalian hosts' first line of defense against cryptococcal infection: internalization by macrophages. Even when C. neoformans is engulfed by host phagocytes, it can survive and replicate within them rather than being destroyed; this ability is central in cryptococcal virulence. It is therefore critical to elucidate the interactions of this facultative intracellular patho

Exosomes as Central Mediators of Host-Pathogen Interactions in Periodontitis: A Systematic Literature Review.

Periodontitis is a chronic inflammatory disease that compromises the periodontium and is among the most prevalent oral conditions worldwide. Recent studies highlight exosomes - small extracellular vesicles carrying nucleic acids, lipids, metabolites, and proteins - as key mediators of intercellular communication contributing to disease progression. The objective of this review was to systematically analyze the existing scientific literature regarding the role of exosomes in the pathogenesis of periodontitis. This systematic review was conducted in accordance with the Preferred Reporting Items

Host-Pathogen Interactions and Correlated Factors That Are Affected in Replicative-Aged Cryptococcus neoformans.

Cryptococcus neoformans is a facultative intracellular fungal pathogen. Ten-generation-old (10GEN) C. neoformans cells are more resistant to phagocytosis and killing by macrophages than younger daughter cells. However, mechanisms that mediate this resistance and intracellular parasitism are poorly understood. Here, we identified important factors for the intracellular survival of 10GEN C. neoformans, such as urease activity, capsule synthesis, and DNA content using flow cytometry and fluorescent microscopy techniques. The real-time visualization of time-lapse imaging was applied to determine t

Exploration of drug repurposing for Mpox outbreaks targeting gene signatures and host-pathogen interactions.

Monkeypox (Mpox) is a growing public health concern, with complex interactions within host systems contributing to its impact. This study employs multi-omics approaches to uncover therapeutic targets and potential drug repurposing opportunities to better understand Mpox's molecular pathogenesis. We developed an in silico host-pathogen interaction (HPI) network and applied weighted gene co-expression network analysis (WGCNA) to explore interactions between Mpox and host proteins. Subtype-specific host-pathogen protein-protein interaction networks were constructed, and key modules from the HPI a

Characterization of Porphyromonas gingivalis sialidase and disruption of its role in host-pathogen interactions.

Key to onset and progression of periodontitis is a complex relationship between oral bacteria and the host. The organisms most associated with severe periodontitis are the periodontal pathogens of the red complex: Tannerella forsythia, Treponema denticola and Porphyromonas gingivalis. These organisms express sialidases, which cleave sialic acid from host glycoproteins, and contribute to disease through various mechanisms. Here, we expressed and purified recombinant P. gingivalis sialidase SiaPG (PG_0352) and characterized its activity on a number of substrates, including host sialoglycoprotein

Direct detection of bacteremia by exploiting host-pathogen interactions of lipoteichoic acid and lipopolysaccharide.

Bacteremia is a leading cause of death in sub-Saharan Africa where childhood mortality rates are the highest in the world. The early diagnosis of bacteremia and initiation of treatment saves lives, especially in high-disease burden areas. However, diagnosing bacteremia is challenging for clinicians, especially in children presenting with co-infections such as malaria and HIV. There is an urgent need for a rapid method for detecting bacteremia in pediatric patients with co-morbidities to inform treatment. In this manuscript, we have developed and clinically validated a novel method for the dire

Functional role of the type 1 pilus rod structure in mediating host-pathogen interactions.

Uropathogenic E. coli (UPEC), which cause urinary tract infections (UTI), utilize type 1 pili, a chaperone usher pathway (CUP) pilus, to cause UTI and colonize the gut. The pilus rod, comprised of repeating FimA subunits, provides a structural scaffold for displaying the tip adhesin, FimH. We solved the 4.2 Å resolution structure of the type 1 pilus rod using cryo-electron microscopy. Residues forming the interactive surfaces that determine the mechanical properties of the rod were maintained by selection based on a global alignment of fimA sequences. We identified mutations that did not alter

Cyclic di-GMP in E. coli: Roles in pathogenesis, host-pathogen interactions, and therapeutic potentials.

Cyclic di-GMP (c-di-GMP) is a universal bacterial second messenger that orchestrates the transition between motile and sessile lifestyles, thereby shaping microbial physiology, virulence, and persistence. In Escherichia coli (E. coli), c-di-GMP functions as a central integrator of environmental cues, dynamically regulating motility, biofilm formation, energy metabolism, and pathogenicity. Here, we provide a comprehensive synthesis of recent advances in c-di-GMP signaling in E. coli, spanning local signaling modules that control curli and cellulose biosynthesis to global regulatory circuits lin

Molecular interactions between ticks (Ixodidae) and cattle: Salivary proteins, host immune modulation, and pathogen transmission.

Ticks (Ixodidae) are major ectoparasites of cattle and highly effective vectors of pathogens responsible for babesiosis, theileriosis, and anaplasmosis. Their capacity to remain attached and blood feed for prolonged periods is underpinned by a finely regulated molecular dialogue at the tick cattle skin interface, driven primarily by the continuous injection of saliva throughout feeding. Tick saliva is a complex and dynamically regulated mixture of proteins, peptides, and small molecules that counter host hemostatic, inflammatory, and immune defenses, thereby ensuring uninterrupted blood uptake

Skin and Wound Infection Models to Test Host-Pathogen Interactions and Evaluate Topical Antimicrobial Treatments.

There is a wide variety of models to study different aspects of infections such as cell lines, organoid cultures, ex vivo models, or even experiments with animals. Each of which mirrors a rising complexity level, and reflect the in vivo condition in humans to a varying degree. When infection models are used to test the therapeutic efficacy and tolerability of antimicrobials or antiseptics, the right choice of model is crucial to truly extract meaningful information and apply these findings to the in vivo circumstances in humans. In this chapter, we report on protocols for using infection model

Multiscale modeling of host-pathogen interactions and mucociliary clearance during non-tuberculous mycobacterial pulmonary infection.

Non-tuberculous mycobacterial (NTM) infections are a clinical challenge in cystic fibrosis (CF), where impaired mucociliary clearance and altered mucus rheology promote bacterial colonization despite host immune responses. Understanding how bacterial growth, immune cell dynamics, and mucus transport regulate infection progression is difficult because these processes interact across spatial and temporal scales. We develop a computational framework bridging a mechanistic agent-based model (ABM) of NTM infection with a spatially resolved partial differential equation (PDE) model. The PDE model co

The battle within: how Pseudomonas aeruginosa uses host-pathogen interactions to infect the human lung.

Pseudomonas aeruginosa is a versatile Gram-negative pathogen known for its ability to invade the respiratory tract, particularly in cystic fibrosis patients. This review provides a comprehensive analysis of the multifaceted strategies for colonization, virulence, and immune evasion used by P. aeruginosa to infect the host. We explore the extensive protein arsenal of P. aeruginosa, including adhesins, exotoxins, secreted proteases, and type III and VI secretion effectors, detailing their roles in the infective process. We also address the unique challenge of treating diverse lung conditions tha

Host-pathogen interactions in the Plasmodium-infected mouse liver at spatial and single-cell resolution.

Upon infecting its vertebrate host, the malaria parasite initially invades the liver where it undergoes massive replication, whilst remaining clinically silent. The coordination of host responses across the complex liver tissue during malaria infection remains unexplored. Here, we perform spatial transcriptomics in combination with single-nuclei RNA sequencing over multiple time points to delineate host-pathogen interactions across Plasmodium berghei-infected liver tissues. Our data reveals significant changes in spatial gene expression in the malaria-infected tissues. These include changes re

Regulation of host/pathogen interactions in the gastrointestinal tract by type I and III interferons.

Interferons (IFNs) are an integral component of the host innate immune response during viral infection. Recent advances in the study of type I and III IFNs suggest that though both types counteract viral infection, type III IFNs act predominantly at epithelial barrier sites, while type I IFNs drive systemic responses. The dynamics and specific roles of type I versus III IFNs have been studied in the context of infection by a variety of enteric pathogens, including reovirus, rotavirus, norovirus, astrovirus, and intestinal severe acute respiratory syndrome coronavirus 2, revealing shared patter

When the Going Gets Rough: The Significance of Brucella Lipopolysaccharide Phenotype in Host-Pathogen Interactions.

Brucella is a facultatively intracellular bacterial pathogen and the cause of worldwide zoonotic infections, infamous for its ability to evade the immune system and persist chronically within host cells. Despite the frequent association with attenuation in other Gram-negative bacteria, a rough lipopolysaccharide phenotype is retained by Brucella canis and Brucella ovis, which remain fully virulent in their natural canine and ovine hosts, respectively. While these natural rough strains lack the O-polysaccharide they, like their smooth counterparts, are able to evade and manipulate the host immu

Phenotypic switching in Candida tropicalis alters host-pathogen interactions in a Galleria mellonella infection model.

Candida tropicalis is a human pathogen associated with high mortality rates. We have reported a switching system in C. tropicalis consisting of five morphotypes - the parental, switch variant (crepe and rough), and revertant (crepe and rough) strains, which exhibited altered virulence in a Galleria mellonella model. Here, we evaluate whether switching events may alter host-pathogen interactions by comparing the attributes of the innate responses to the various states. All switched strains induced higher melanization in G. mellonella larvae than that induced by the parental strain. The galiomic

Non-coding RNAs in Host-Pathogen Interactions: Subversion of Mammalian Cell Functions by Protozoan Parasites.

Pathogens have evolved mechanisms to modulate host cell functions and avoid recognition and destruction by the host damage response. For many years, researchers have focused on proteins as the main effectors used by pathogens to hijack host cell pathways, but only recently with the development of deep RNA sequencing these molecules were brought to light as key players in infectious diseases. Protozoan parasites such as those from the genera Plasmodium, Toxoplasma, Leishmania, and Trypanosoma cause life-threatening diseases and are responsible for 1000s of deaths worldwide every year. Some of t

Molecular mechanisms involved in vascular interactions of the Lyme disease pathogen in a living host.

Hematogenous dissemination is important for infection by many bacterial pathogens, but is poorly understood because of the inability to directly observe this process in living hosts at the single cell level. All disseminating pathogens must tether to the host endothelium despite significant shear forces caused by blood flow. However, the molecules that mediate tethering interactions have not been identified for any bacterial pathogen except E. coli, which tethers to host cells via a specialized pillus structure that is not found in many pathogens. Furthermore, the mechanisms underlying tetheri

Early host-pathogen interactions in the liver and spleen during systemic murine listeriosis: an overview.

Systemic listeriosis initiated by parenteral inoculation of mice with Listeria monocytogenes has been used extensively as a model infection for studying mammalian host defense against intracellular bacterial pathogens in general. Most effort has been expended on trying to understand the requirement for specific T cell-mediated immunity for combatting infection with this pathogen. By contrast, non-specific defenses have received much less attention. However, it is now obvious that these early innate defenses are critically important for the well-being of the host. If these early defenses fail t
