Six-coordinate and five-coordinate Fe(II)(CN)(2)(CO)(x) thiolate complexes (x = 1, 2): synthetic advances for iron sites of [NiFe] hydrogenases.

The dicyanodicarbonyliron(II) thiolate complexes trans,cis-[(CN)(2)(CO)(2)Fe(S,S-C-R)](-) (R = OEt (2), N(Et)(2) (3)) were prepared by the reaction of [Na][S-C(S)-R] and [Fe(CN)(2)(CO)(3)(Br)](-) (1). Complex 1 was obtained from oxidative addition of cyanogen bromide to [Fe(CN)(CO)(4)](-). In a similar fashion, reaction of complex 1 with [Na][S,O-C(5)H(4)N], and [Na][S,N-C(5)H(4)] produced the six-coordinate trans,cis-[(CN)(2)(CO)(2)Fe(S,O-C(5)H(4)N)](-) (6) and trans,cis-[(CN)(2)(CO)(2)Fe(S,N-C(5)H(4))](-) (7) individually. Photolysis of tetrahydrofuran (THF) solution of complexes 2, 3, and 7

Characterization of the CO oxidation/H2 evolution system of Rhodospirillum rubrum. Role of a 22-kDa iron-sulfur protein in mediating electron transfer between carbon monoxide dehydrogenase and hydrogenase.

The response of the membrane-associated carbon monoxide dehydrogenase (CODH) from Rhodospirillum rubrum to solubilization by detergents and organic solvents, the properties of solubilized CODH, and the mechanism for coupling CO oxidation to hydrogen evolution via a CO-induced hydrogenase activity have been investigated. The release of CODH by a variety of ionic and nonionic detergents occurs in a redox-dependent fashion: CODH is solubilized in the presence of low-potential reductants (dithionite, CO, and H2) but is resistant to solubilization from membranes prepared in the absence of reductant

Characterization of the active site of a hydrogen sensor from Alcaligenes eutrophus.

A third hydrogenase was recently identified in the proteobacterium Alcaligenes eutrophus as a constituent of a novel H2-sensing multicomponent regulatory system. This regulatory hydrogenase (RH) has been overexpressed in cells deficient in both the NAD+-reducing [NiFe]-hydrogenase and the membrane-bound [NiFe]-hydrogenase. EPR, FTIR and activity studies of membrane-free extracts revealed that the RH has an active site much like that of standard [NiFe]-hydrogenases, i.e. a Ni-Fe site with two CN- groups and one CO molecule. Its catalytic power is low, but the RH is always active, insensitive to

[Fe2(SR)2(mu-CO)(CNMe)6]2+ and analogues: a new class of diiron dithiolates as structural models for the H(ox)Air state of the fe-only hydrogenase.

Low-temperature oxidation of Fe(2)(S(2)C(n)H(2n)(CNMe)(6-x)(CO)x (n = 2, 3; x = 2, 3) affords a family of mixed carbonyl-isocyanides of the type [Fe(2)(S(2)C(n)H(2n)(CO)x(CNMe)(7-x)](2+). The degree of substitution is controlled by the RNC/Fe ratio, as well as the degree of initial substitution at iron, with tricarbonyl derivatives favoring more highly carbonylated products. The structures of the monocarbonyl derivatives [Fe(2)(S(2)C(n)H(2n))(mu-CO)(CNMe)(6)](PF(6))(2) (n = 2,3) established crystallographically and spectroscopically, are quite similar, with Fe---Fe distances of ca. 2.5 A, alth

Use of the 2,3-bis(diphenylphosphino)-N-phenylmaleimide ligand as a redox [Fe4S4] cubane surrogate in di-iron complexes related to [FeFe]-hydrogenases: influence of the dithiolate bridge on the redox behaviour.

The redox behaviours of two complexes with the 2,3-bis(diphenylphosphino)-N-phenylmaleimide (bmi) ligand related to the active site of [FeFe]-hydrogenases, [Fe2(CO)4(κ2-bmi)(μ-pdt)] (1) and [Fe2(CO)4(κ2-bmi)(μ-adtBn)] (2) [pdt = propane-1,3-dithiolate (μ-S(CH2)3S) and adtBn = N-benzyl-azapropane-1,3-dithiolate (μ-SCH2N{CH2(C6H5)}CH2S)], were studied. The complexes were synthesized by photolysis and they were characterized by IR, NMR (1H, 13C, 31P) spectroscopy and elemental analyses. The structures of 1 and 2 in the solid state were solved by X-ray diffraction analysis. Their electrochemical b

Spin distribution of the H-cluster in the H(ox)-CO state of the [FeFe] hydrogenase from Desulfovibrio desulfuricans: HYSCORE and ENDOR study of (14)N and (13)C nuclear interactions.

Hydrogenases are enzymes which catalyze the reversible cleavage of molecular hydrogen into protons and electrons. In [FeFe] hydrogenases the active center is a 6Fe6S cluster, referred to as the "H-cluster." It consists of the redox-active binuclear subcluster ([2Fe](H)) coordinated by CN(-) and CO ligands and the cubane-like [4Fe-4S](H) subcluster which is connected to the protein via Cys ligands. One of these Cys ligands bridges to the [2Fe](H) subcluster. The CO-inhibited form of [FeFe] hydrogenase isolated from Desulfovibrio desulfuricans was studied using advanced EPR methods. In the H(ox)

Hydrogen as an energy source for the human pathogen Bilophila wadsworthia.

The gram-negative anaerobic gut bacterium Bilophila wadsworthia is the third most common isolate in perforated and gangrenous appendicitis, being also found in a variety of other infections. This organism performs a unique kind of anaerobic respiration in which taurine, a major organic solute in mammals, is used as a source of sulphite that serves as terminal acceptor for the electron transport chain. We show here that molecular hydrogen, one of the major products of fermentative bacteria in the colon, is an excellent growth substrate for B. wadsworthia. We have quantified the enzymatic activi

Methanogenesis involves direct hydride transfer from H2 to an organic substrate.

Certain anaerobic microorganisms evolved a mechanism to use H2 as a reductant in their energy metabolisms. For these purposes, the microorganisms developed H2-activating enzymes, which are aspirational catalysts in a sustainable hydrogen economy. In the case of the hydrogenotrophic pathway performed by methanogenic archaea, 8e- are extracted from 4H2 and used as reducing equivalents to convert CO2 into CH4. Under standard cultivation conditions, these archaea express [NiFe]-hydrogenases, which are Ni-dependent and Fe-dependent enzymes and heterolytically cleave H2 into 2H+ and 2e-, the latter

Di-iron aza diphosphido complexes: mimics for the active site of Fe-only hydrogenase, and effects of changing the coordinating atoms of the bridging ligand in [Fe2[mu-(ECH2)2NR](CO)6].

The bis(phosphido)-bridged complex [Fe(2)(mu-PPhH)(2)(CO)(6)] (1) undergoes double deprotonation to give the phosphorus-centered dianionic derivative [Fe(2)(mu-PPh)(2)(CO)(6)](2)(-) (2) which in turn reacts with the tertiary base RN(CH(2)Cl)(2) to give [Fe(2)[(PPhCH(2))(2)NR](CO)(6)] (3) in moderate yield. Treatment of 3 with HBF(4).Et(2)O affords the N-protonated species [Fe(2)[(PPhCH(2))(2)NHR](CO)(6)] BF(4) (4). The structural changes to the heavy atom skeleton of 3 arising from protonation are slight, the most obvious being a ca. 0.03 Alengthening of the N-C bonds.

Hydrogen Metabolism in Helicobacter pylori Plays a Role in Gastric Carcinogenesis through Facilitating CagA Translocation.

UNLABELLED: A known virulence factor of Helicobacter pylori that augments gastric cancer risk is the CagA cytotoxin. A carcinogenic derivative strain, 7.13, that has a greater ability to translocate CagA exhibits much higher hydrogenase activity than its parent noncarcinogenic strain, B128. A Δhyd mutant strain with deletion of hydrogenase genes was ineffective in CagA translocation into human gastric epithelial AGS cells, while no significant attenuation of cell adhesion was observed. The quinone reductase inhibitor 2-n-heptyl-4-hydroxyquinoline-N-oxide (HQNO) was used to specifically inhibit

Respiratory hydrogen use by Salmonella enterica serovar Typhimurium is essential for virulence.

Based on available annotated gene sequence information, the enteric pathogen salmonella, like other enteric bacteria, contains three putative membrane-associated H2-using hydrogenase enzymes. These enzymes split molecular H2, releasing low-potential electrons that are used to reduce quinone or heme-containing components of the respiratory chain. Here we show that each of the three distinct membrane-associated hydrogenases of Salmonella enterica serovar Typhimurium is coupled to a respiratory pathway that uses oxygen as the terminal electron acceptor. Cells grown in a blood-based medium express

Selective inhibition of NikA mediated Ni(II) import in E. coli by the Indium(III)-EDTA complex.

Nickel is a required nutrient for bacteria to produce [NiFe]-hydrogenase and urease enzymes. [NiFe]-hydrogenase catalyzes the reversible conversion of hydrogen into protons and electrons and urease catalyzes the hydrolysis of urea into carbon dioxide and ammonia-both key in bacterial pathogenesis. As such, nickel trafficking and homeostasis are interesting targets for potential antibacterial strategies. In E. coli, NikA binds a Ni(II)-(L-His)2 chelate in the periplasm and delivers this complex to the NikBCDE transporter. Blocking Ni(II) uptake by NikA would prevent the biosynthesis of active [

Regulation of a catabolic enzyme whose substrate is always available.

Certain hydrogenases are capable of converting the trace amounts of molecular hydrogen constantly present in Earth's atmosphere to supply cells with a little energy during periods of starvation. Recently, Kropp and colleagues investigated the regulation of one of these "high-affinity" hydrogenases in response to different growth conditions for the ubiquitous soil bacterium Mycobacterium smegmatis (A. Kropp, J. D. Archer, M. Jespersen, T. D. Watts, et al., mSystems 11:e01678-25, 2026, https://doi.org/10.1128/msystems.01678-25). A mutation in the gylR gene, whose product acts as a positive trans

Disrupting the mtr-operon in Methanosarcina acetivorans enables methyl-reducing methanogenesis with hydrogen and serine as the alternative electron donors.

Methanosarcina acetivorans is a model methanogen because of its metabolic versatility and genetic tractability. This microbe is not known to natively utilize hydrogen as a catabolic electron donor, despite its genome encoding hydrogenases, and the fact that this microbe can be used to heterologously express functional hydrogenases. Its native hydrogenases are expressed at a very low level and are suggested to have a role in recycling hydrogen that is produced as a byproduct of nitrogen fixation. To explore whether hydrogen can act as a catabolic electron donor, we utilized a previously constru

Use of molecular hydrogen as an energy substrate by human pathogenic bacteria.

Molecular hydrogen is produced as a fermentation by-product in the large intestine of animals and its production can be correlated with the digestibility of the carbohydrates consumed. Pathogenic Helicobacter species (Helicobacter pylori and H. hepaticus) have the ability to use H(2) through a respiratory hydrogenase, and it was demonstrated that the gas is present in the tissues colonized by these pathogens (the stomach and the liver respectively of live animals). Mutant strains of H. pylori unable to use H(2) are deficient in colonizing mice compared with the parent strain. On the basis of a

Effect of culture conditions on producing and uptake hydrogen flux of biohydrogen fermentation by metabolic flux analysis method.

In this work, metabolic flux analysis (MFA) method was used to estimate the effects of the culture conditions on both the producing and uptake hydrogen flux inside the cell of Klebsiella pneumoniae ECU-15. The results indicated that higher temperature could reduce the amount of the uptake hydrogen and enhance the hydrogen production from the NADH pathway. Moreover, both the producing hydrogen flux from formate and the uptake hydrogen flux were attained to the maximum at pH 7.0-7.5. The producing hydrogen flux was higher at 5g/L initial glucose than that of the other concentrations, and the upt

Mononuclear Ni(III) complexes [NiIII(L)(P(C6H3-3-SiMe3-2-S)3)]0/1- (L = thiolate, selenolate, CH2CN, Cl, PPh3): relevance to the nickel site of [NiFe] hydrogenases.

The stable mononuclear Ni(III)-thiolate complexes [NiIII(L)(P(C6H3-3-SiMe3-2-S)3)]- (L = SePh (2), Cl (3), SEt (4), 2-S-C4H3S (5), CH2CN (7)) were isolated and characterized by UV-vis, EPR, IR, SQUID, CV, 1H NMR, and single-crystal X-ray diffraction. The increased basicity (electronic density) of the nickel center of complexes [NiIII(L)(P(C6H3-3-SiMe3-2-S)3)]- modulated by the monodentate ligand L and the substituted groups of the phenylthiolate rings promotes the stability and reactivity. Electron flow in hydrogenotrophic methanogens under nickel limitation.

Methanogenic archaea are the main producers of the potent greenhouse gas methane1,2. In the methanogenic pathway from CO2 and H2 studied under laboratory conditions, low-potential electrons for CO2 reduction are generated by a flavin-based electron-bifurcation reaction catalysed by heterodisulfide reductase (Hdr) complexed with the associated [NiFe]-hydrogenase (Mvh)3-5. F420-reducing [NiFe]-hydrogenase (Frh) provides electrons to the methanogenic pathway through the electron carrier F420 (ref. 6). Here we report that under strictly nickel-limited conditions, in which the nickel concentration
