Multifunction of the ER P-Type Calcium Pump Spf1 During Hyphal Development in Candida albicans.

Candida albicans is one of the most important fungal pathogens. Hyphal development is required for the virulence of this pathogen. Our previous study has revealed that Spf1, an ER P-type calcium pump, plays an important role in hyphal development. However, the detailed mechanisms by which this protein functions in this process remain to be investigated. In this study, we found that loss of Spf1 led to decreased growth biomass under the hypha-inducing condition, suggesting a role of this protein in maintaining hyphal growth rate. Actin staining further revealed that the spf1Δ/Δ mutant showed at

Neutrophil extracellular traps capture the human pathogen, Candida albicans, in blood and delay hyphal transformation.

In tissues, neutrophils neutralize Candida albicans through phagocytosis and delay C. albicans hyphae growth by deploying neutrophil extracellular traps (NETs). However, in the bloodstream, the dynamic interactions between NETs and C. albicans are far less understood. Here, we employ a microfluidic assay and measure a significant increase in intact NETs in blood within 3 h after adding C. albicans yeast or hyphae. We show that C. albicans yeast can be captured efficiently on NETs, thereby delaying the transition to hyphal growth. We measure higher amounts of intact NETs in blood samples from i

Fungal pathogenesis: Sugar affects shape.

Morphological transitions underlie fungal pathogenicity. Transitions to pathogenic growth forms are well studied, but morphological reversions are not, despite their importance for fungal commensalism. A new study addresses this, revealing mechanisms by which a bacterial polyketide triggers hypha-to-yeast morphogenesis in Candida albicans.

A practical method to detect mycoses of the nails.

The authors present a method that allows for reliable identification of fungal structures in nails with suspected colonization by fungi. The method is based on the well-known technique of heating nail fragments in 20% potassium hydroxide, but its details allow for reliable and quick processing of samples, when convenient. The method requires simple equipment and is designed for individual practices but might be employed as is or with minor technical improvements by a dermatology department. Minor changes will make it feasible to simultaneously process several samples. The results that were obt

Fungi growing on jute fabrics deteriorating under weather exposure and in storage.

Special media suitable for the isolation of fungi growing on jute were developed. Species that would better resist sunlight, such as organisms with dark hyphae and spores or closed fruit bodies, predominated on weather-exposed fabrics. Several strongly cellulolytic organisms not previously implicated in fiber decomposition were isolated from this source. On the other hand, the fungi obtained from jute materials damaged in storage were mostly of the familiar types. Most of the newly isolated fungi are generally missed on ordinary media, but they probably play an important role in the natural de

Lack of vessel wall elastolysis in human invasive pulmonary aspergillosis.

In experimental studies, the apparent ability of Aspergillus fumigatus isolates to produce elastase in agar plates correlates with their ability to cause invasive pulmonary aspergillosis in mice pretreated with cortisone. Thus, elastase production may govern the pathogenicity of particular isolates. If this is so, then disruption of the elastic layers within blood vessel walls in invasive aspergillosis would be expected. To test this hypothesis, tissue blocks were prepared from nine patients with invasive pulmonary aspergillosis. Separate but immediately adjacent histological sections were sta

A histological and ultrastructural comparison of the sulfur granule of the actinomycosis and actinobacillosis.

The sulfur granules of actinomycosis and actinobacillosis were studied histologically and ultrastructurally. The sulfur granule of actinomycosis was large, and oval or horsehoe-shaped. In it, there were a number of gram-positive, filamentous or short rod-like hyphae beneath clubs. Ultrastructurally, the center of the sulfur granule was composed of a cluster of hyphae, and the periphery was surrounded by clubs arranged radially. The clubs were made of electron-dense fine granules and had a degenerated hypha at the center. These granules varied in amount from small to large. As for the host reac

Morphotype transition and sexual reproduction are genetically associated in a ubiquitous environmental pathogen.

Sexual reproduction in an environmental pathogen helps maximize its lineage fitness to changing environment and the host. For the fungal pathogen Cryptococcus neoformans, sexual reproduction is proposed to have yielded hyper virulent and drug resistant variants. The life cycle of this pathogen commences with mating, followed by the yeast-hypha transition and hyphal growth, and it concludes with fruiting body differentiation and sporulation. How these sequential differentiation events are orchestrated to ensure developmental continuality is enigmatic. Here we revealed the genetic network of the

The role of the C. albicans transcriptional repressor NRG1 during filamentation and disseminated candidiasis is strain dependent.

Candida albicans is one of the most common causes of superficial and invasive fungal diseases in humans. Its ability to cause disease is closely linked to its ability to undergo a morphological transition from budding yeast to filamentous forms (hyphae and pseudohyphae). The extent to which C. albicans strains isolated from patients undergo filamentation varies significantly. In addition, the filamentation phenotypes of mutants involving transcription factors that positively regulate hyphal morphogenesis can also vary from strain to strain. Here, we characterized the virulence, in vitro and in

Multiple discharging sinuses: an unusual presentation caused by Absidia corymbifera.

A case of zygomycosis presenting with non-healing multiple discharging sinuses in a diabetic patient is reported here. The debrided tissue on histopathological examination revealed dense infiltration with aseptate fungal hyphae. Potassium hydroxide mount showed hyaline aseptate hyphae suggestive of zygomycosis. On culture, Absidia corymbifera was isolated. The patient responded to surgical debridement and therapy with amphotericin B followed by itraconazole.

Cutaneous phaeohyphomycosis caused by Exophiala spinifera in a patient with systemic lupus erythematosus.

Exophiala spinifera can induce both phaeohyphomycosis and chromomycosis. To date there have been 18 human infections caused by E. spinifera in the English literature. A case of E. spinifera-induced phaeohyphomycosis in a patient with systemic lupus erythematosus (SLE) is described. Direct microscopic examination of the pus showed branched, septate and chained hyphae and spores. A dark green velvety colony grew on Sabouraud dextrose agar. Slide culture showed branched, septate hyphae and spine-like annellated conidiophores. Histopathological biopsy revealed yellowish brown hyphae and spores. Th

Gastrointestinal basidiobolomycosis. Morphologic findings in a cluster of six cases.

We describe the histopathologic features of 6 cases of gastrointestinal basidiobolomycosis examined at 4 Phoenix, AZ, area hospitals during the last 4 years. Resected stomach and intestinal specimens were characterized by marked mural thickening with fibrosis, prominent tissue eosinophil infiltration and palisading granulomatous inflammation around pale fungal hyphae. In 2 cases, there was colonic perforation. Basidiobolus ranarum hyphae (associated with spore-like spherules in 4 cases) were identified within tissue sections; the irregularly branched, thin-walled, occasionally septated hyphae

Three-Dimensional Ultrastructure of Arabidopsis Cotyledons Infected with Colletotrichum higginsianum.

We used serial block-face scanning electron microscopy (SBF-SEM) to study the host-pathogen interface between Arabidopsis cotyledons and the hemibiotrophic fungus Colletotrichum higginsianum. By combining high-pressure freezing and freeze-substitution with SBF-SEM, followed by segmentation and reconstruction of the imaging volume using the freely accessible software IMOD, we created 3D models of the series of cytological events that occur during the Colletotrichum-Arabidopsis susceptible interaction. We found that the host cell membranes underwent massive expansion to accommodate the rapidly g

Filamentous Histoplasma capsulatum endocarditis involving mitral and aortic valve porcine bioprostheses.

We report a case of histoplasmosis capsulati endocarditis simultaneously infecting a mitral and aortic porcine bioprostheses. Histologically, the fungus demonstrated a diversity of morphologies, ranging from typical, intracellular, 2- to 5-micron yeast cells, to septate and branching hyphae, to bizarre hyphae with vesicular swellings. Cultures obtained from both valves were positive in four days. The combination of unexpected histologic appearance and rapid growth in culture may pose problems in diagnosis.

Taste Dysfunction in Oral Candidiasis: Impact of Candida Carriage and Hyphal Presence.

BACKGROUND: Taste dysfunction is a common symptom of oral candidiasis; however, its underlying mechanisms remain unclear. OBJECTIVES: This study aimed to determine whether Candida albicans carriage, particularly its hyphal phenotype, is associated with impaired taste sensitivity. PATIENTS/METHODS: Fifty-seven participants were divided into three groups: noncarriers (n = 20), carriers without hyphae (n = 20), and carriers with hyphae (n = 17). Tongue biofilm samples were collected for Candida culture and smear tests. Salivary flow rates were measured, and taste sensitivity was assessed using

Aspergillus fumigatus-Stimulated Human Corneal Epithelial Cells Induce Pyroptosis of THP-1 Macrophages by Secreting TSLP.

Fungal keratitis (FK) is a keratopathy caused by pathogenic fungal infection. The aim of this work is to explore the role of thymic stromal lymphopoietin (TSLP) in FK. Human corneal epithelial cells (HCECs) were treated with Aspergillus fumigatus hyphae, and we found that TSLP was highly expressed and secreted in the hyphae-treated HCECs. Hyphae-treated HCECs or TSLP treatment enhanced the expression of caspase-1 P20, GSDMD-N (p30), IL-1β, and IL-18 in the human THP-1 macrophages. The influence conferred by hyphae-treated HCECs or TSLP treatment was rescued by TSLP neutralizing antibody or VX-
