[Biochemical and histological changes in the hypothalamus in hormone-stimulated sheep].

The effects were investigated of a hormonal preparation of serum gonadotrophin (SC) on neurosecretion, PAS-positive mucopolysaccharides and on histological changes in the third cerebral ventricle; at the same time variations were determined of catecholamine concentrations of ependyma surface in the hypothalamic regions that control directly reproductive functions in ewes of the Merino and Wallachian breeds in the oestrus period. Ewe oestrus was synchronized by insertion of Ageline (Spofa) sponges into the vagina. A dose of 2000 IU of serum gonadotrophin (Bioveta, Ivanovice in Haná) was adminis

[Loss of catecholaminergic neuron in the hypothalamus of multiple system atrophy].

Autonomic failure is one of the primary clinical features of multiple system atrophy (MSA); while the hypothalamus is thought to be a higher center regulating the autonomic nervous system. In the hypothalamus, catecholamine depletion had been confirmed by neurochemical studies but no marked changes had yet been demonstrated by pathological studies. Using three MSA and three control cases, we evaluated the quantitative changes of catecholamine (CA)-containing neurons in the hypothalamus. As markers of CA neurons, neuromelanin (NM)- and tyrosin hydroxylase (TH)-containing neurons were stained by

Role of the hypothalamus in the control of atrial natriuretic peptide release.

Stimulation of the region antero-ventral to the third cerebral ventricle (AV3V) by a cholinergic drug, carbachol, and lesions of the AV3V have been demonstrated in previous studies to either augment or decrease sodium excretion, respectively. Atrial natriuretic peptide (ANP) dramatically increases renal sodium excretion and has been localized to brain areas previously shown to be involved in control of sodium excretion. Consequently, to evaluate a possible role of brain ANP in evoking the changes in renal sodium excretion that follow stimulations or lesions of the AV3V, we determined the effec

[Effects of CCK-8 of hypothalamus on antral motility in rats].

Cholecystokinin-octapeptide (CCK-8) of 10 ng were injected into lateral hypothalamus (LH) and ventromedial hypothalamus (VMH) in conscious rats, both of which brought about inhibition of antral motility recorded by a strain gauge, the actions were partly antagonised by bilateral vagotomy, but blocked by intravenous infusion of atropine or regitine. It is suggested that both vagus and sympathetic nerves be involved in mediation of function of central CCK-8. In addition, the unit spontaneous discharges of neurons in vagal nuclei were attenuated while CCK-8 injected into LH. On the contrary, the

In vivo release of angiotensin II from the rat hypothalamus.

Recent studies suggest that angiotensin II is released from neuronal tissue in vitro, but the occurrence of this phenomenon in the intact brain has not yet been demonstrated. To characterize the in vivo release of immunoreactive angiotensin II, push-pull cannulas were positioned in the anterior hypothalamus in 47 Sprague-Dawley rats (200-250 g) anesthetized with Inactin (100 mg/kg i.p.). Artificial cerebrospinal fluid was perfused at 20 microliters/min, and effluent samples were collected for 15-minute periods over 2 hours. Angiotensin II was detectable (greater than 2.5 pg/ml) in the push-pul

Cyto-, dendro- and fibro-architectonic studies on the chicken hypothalamus.

Based on the chicken hypothalamic analysis using the Nissl, Golgi-Cox and silver staining methods, a first attempt to make the cyto-, dendro- and fibro-architectonic atlases of the frontal planes at the same level was performed. In the preparations stained with Nissl method, the hypothalamus was subdivisible basically into the preoptic, rostral and caudal hypothalamic regions, each of which was composed of medial and lateral parts respectively. A well-defined boundaries between the preoptic and rostral hypothalamic regions was decided at the level of the rostral margin of the commissura rostra

Comparison of electrolytic and ibotenic acid lesions in the lateral hypothalamus.

Electrolytic lesions in the lateral hypothalamus (LH) seriously affect ingestive behavior and sensorimotor functions in the rat. We here report that bilateral infusions of the neurotoxin, ibotenic acid (IBO) in the LH yield a decrease in body weight, but not to the same extent as electrolytic lesions. The sensorimotor impairments were most severe after electrolytic lesions. When tested in a residential maze on days 5-7 and 18-20 after surgery, both lesioned groups showed no lack of motivation to seek food and water. Histological examination of the LH following IBO exposure revealed extensive d

Sexual maturation of the hypothalamus: pathophysiological aspects and clinical implications.

Sexual maturation in humans begins early in fetal life and culminates in adulthood when the gonads have acquired a full capacity for reproduction. It is remarkable that during this long process, the pituitary gonadal function, hence its hypothalamic control presents an alternative of activation and inhibition periods, during which the interrelations of the 3 components of the hypothalamic-pituitary-gonadal axis change gradually and inversely. The ontogeny of the hypothalamic-pituitary system, the varying activity of the reproductive endocrine system throughout sexual maturation and the develop

Distribution of tyrosine-hydroxylase-immunoreactive neurons in the hypothalamus of rats.

The distribution and morphology of cells containing tyrosine hydroxylase (TH) immunoreactivity in the hypothalamus of rats were studied by using a modified immunoperoxidase technique. The TH cell system is more complexly organized than was previously thought. On the basis of their clustering patterns, hypothalamic TH neurons could be subdivided into two groups: dorsal and ventral. The ventral group consists of a prominent aggregate of cells located in the caudal part of the arcuate nucleus. From here, cells extend around the caudal part of the ventromedial and dorsomedial nuclei and the base o

Exogenous vasopressin reverses hyperactivity in the hypothalamus of Brattleboro rats.

We have previously described an increase of cytochrome oxidase (COX) activity in discrete hypothalamic nuclei of the Brattleboro rat (BR). The present experiments were done to investigate whether this increase in COX activity could be eliminated by administration of exogenous arginine vasopressin (AVP). Continuous infusion of AVP to BR for 7 days corrected the polyuria and the urine hypoosmolality, as expected, and it also eliminated the COX hyperactivity in the paraventricular nucleus and nucleus circularis of the BR hypothalamus. These results suggest that the hyperactivity of these nuclei i

Purification of a glucagon releasing factor from the rat hypothalamus.

The purification of a peptide from the rat hypothalamus which is capable of stimulating the release of glucagon from the endocrine pancreas and therefore named glucagon releasing factor (GlRF), is described. Compositional analysis of the product obtained using well established techniques for peptide isolation revealed that GlRF appeared to consist of 30-31 amino acids. The significance of GlRF in the physiological regulation of glucagon secretion is yet to be determined but the existence of this very potent factor supports the concept of a hypothalamic-pancreatic neurohormonal link.

Presence of immunoreactive calcitonin in the hypothalamus and pituitary lobes of rats.

We have found calcitonin-like immunoreactive material in extracts of hypothalami from six-month old, male rats. The level of this immunoreactivity, 0.21 ng/hypothalamus, is substantial considering out lower limit of detection of 0.006 ng of rat calcitonin. However, the hypothalamus contains less calcitonin-like immunoreactive material than either the anterior lobe (1.16 ng) or neurointermediate lobe (0.81 ng) of the pituitary. Taken together these three sources of calcitonin-like immunoreactive material contain less than one thousandth the immunoreactivity found in the thyroid. The exact natur

Sexual differentiation of the hypothalamus in gonadal agenesis and testicular feminization.

Cyclical hypothalamic function was investigated in three patients with an XY karyotype and female external genitalia; in one of them we diagnosed gonadal agenesis, and in the other two testicular feminization. We studied the effect of estradiol and progesterone on gonadotropin release. The patient with gonadal agenesis had cyclical hypothalamic function, but this cyclical function was suppressed in the patients with testicular feminization in whom no LH secretion could be provoked by steroid stimulation. These observations support the concept that hypothalamic sexual differentiation is due to

Glioprotective Effects of Sulforaphane in Hypothalamus: Focus on Aging Brain.

Sulforaphane is a natural compound with neuroprotective activity, but its effects on hypothalamus remain unknown. In line with this, astrocytes are critical cells to maintain brain homeostasis, and hypothalamic astrocytes are fundamental for sensing and responding to environmental changes involved in a variety of homeostatic functions. Changes in brain functionality, particularly associated with hypothalamic astrocytes, can contribute to age-related neurochemical alterations and, consequently, neurodegenerative diseases. Thus, here, we investigated the glioprotective effects of sulforaphane on
