Engineering bacteria for cancer immunotherapy by inhibiting IDO activity and reprogramming CD8+ T cell response.

Inhibiting indoleamine 2,3 dioxygenase (IDO) for anticancer therapy has garnered significant attention in recent years. However, current IDO inhibitors face significant challenges which limit their clinical application. Here, we genetically engineered a high tryptophan-expressing Clostridium butyricum (L-Trp CB) strain that can colonize tumors strictly following systemic administration. We revealed that butyrate produced by L-Trp CB can inhibit IDO activity, preventing tryptophan catabolism and kynurenine accumulation in tumors. In addition, the large released tryptophan by L-Trp CB can provid

Classifying cGAS-STING Activity Links Chromosomal Instability with Immunotherapy Response in Metastatic Bladder Cancer.

UNLABELLED: The cGAS-STING pathway serves a critical role in anticancer therapy. Particularly, response to immunotherapy is likely driven by both active cGAS-STING signaling that attracts immune cells, and by the presence of cancer neoantigens that presents as targets for cytotoxic T cells. Chromosomal instability (CIN) is a hallmark of cancer, but also leads to an accumulation of cytosolic DNA that in turn results in increased cGAS-STING signaling. To avoid triggering the cGAS-STING pathway, it is commonly disrupted by cancer cells, either through mutations in the pathway or through transcrip

The Italian Network for Tumor Bio-Immunotherapy (NIBIT) Foundation: ongoing and prospective activities in immuno-oncology.

The ongoing revolution in cancer immunotherapy stems from the knowledge that distinct immune-checkpoints regulate the physiological crosstalk between and among immune cells by delivering inhibitory or activating signals. These notions, and the availability of mAb directed to diverse immune-checkpoint molecules, have led to a significant clinical improvement in cancer treatment. In this scenario, further achievements are undoubtedly to be expected from the contribution of novel, proof-of-principle clinical trials designed to explore the therapeutic efficacy of new immunotherapy-based combinatio

Activation of complement by monoclonal antibodies that target cell-associated β₂-microglobulin: implications for cancer immunotherapy.

β₂-Microglobulin (β2M), the light chain of the class I major histocompatibilty complex (MHC-I), is a promising tumor target for monoclonal antibodies (mAbs) in cancer immunotherapy. Several reports indicate that chelation of cell-associated β2M by specific mouse mAbs promotes tumor cell destruction by inducing apoptosis or other cytotoxic signaling pathways. Human mAbs employed in cancer therapy are usually IgG1, which mediates cell-killing by effector mechanisms including complement dependent cytotoxicity (CDC). The analogous mouse IgG2a and IgG2b isotypes are similarly effective in activatin

Changes in basophil activation during grass-pollen sublingual immunotherapy do not correlate with clinical efficacy.

BACKGROUND: Biomarkers predicting the safety and efficacy of sublingual immunotherapy (SLIT) remain to be established. METHODS: Eighty-nine patients with allergic rhinoconjunctivitis to grass pollen received either a placebo or five-grass-pollen daily tablet sublingually for 4 months. Following exposure in an allergen challenge chamber, clinical responders and nonresponders were identified individually by evaluating their rhinoconjunctivitis total symptom score (RTSS). Activation of peripheral blood basophils was measured by cytofluorometry before and after 2 or 4 months of immunotherapy, bas

Simultaneous expression of T-cell activating antigens in renal cell carcinoma: implications for specific immunotherapy.

PURPOSE: The activation of antigen specific T cells by tumor associated antigens (TAA) might be a promising treatment strategy for patients with renal cell carcinoma (RCC). We analyzed TAA expression in patients with RCC as well as the prevalence of fitting HLA phenotypes and calculated the percent of patients eligible for peptide vaccination trials. MATERIALS AND METHODS: A total of 41 RCC samples from primary tumors were analyzed for TAA expression by reverse transcriptase-polymerase chain reaction. Genes of interest were MAGE-1, MAGE-3, G250 and PRAME since peptides derived from these gene

Phase I study of the adoptive immunotherapy of human cancer with lectin activated autologous mononuclear cells.

In previous in vitro studies, the authors showed that phytohemagglutinin (PHA) stimulated peripheral blood lymphocytes (PBL) from cancer patients to generate cells that were lytic for fresh autologous tumor but not for lymphocytes or lymphoblasts. Thus, after IRB approval, a phase I clinical protocol was instituted in cancer patients who had failed all other therapy to determine the toxicity and effects, in vivo, of the infusion of large numbers of such PHA activated autologous PBL. Ten patients were treated on the protocol, six with sarcoma, one with melanoma, and three with colorectal cancer

Engineering Subcutaneous Ectopic Tertiary Lymphoid Structures as Personalized Immune-Activation Niches for Cancer Immunotherapy.

Tumor-associated tertiary lymphoid structures (TLSs) can promote anti-tumor immune responses and survival. However, intratumor TLS formation and maturation are limited in tumors with a "cold" immune phenotype, including reduced infiltration of immune cells and a suppressive tumor microenvironment (TME). Here, we present a personalized immunotherapy strategy utilizing a subcutaneously injected, bioengineered hydrogel (scTLS) that acts as an ectopic immune-activation center, effectively decoupled from the suppressive TME. Native immune cells and antigen-presenting cells (APCs) with autologous an

Compartment-specific PPARδ activity controls the outcome of cancer-immune cell competition and immunotherapy efficacy.

Most patients with solid tumors fail to sustain antitumor immunity, in part because immune cells cannot outcompete cancer cells inside the immunosuppressive tumor microenvironment. Here, we show the lipid-sensing transcription factor PPARδ is a compartment-specific determinant of cancer-immune fitness balance and immunotherapy efficacy. Activating PPARδ in cancer cells promotes their growth and impairs antitumor immunity, whereas activating it in immune cells drives tumor suppression and overrides tumor-promoting effects of cancer-intrinsic activation. Mechanistically, PPARδ enhances CD8+ T ce

A hybrid nanoadjuvant cascading activation of the cGAS-STING-IFN-Ⅰ pathway to enhance radio-immunotherapy.

The cGAS-STING signaling pathway is crucial for radiotherapy (RT)-induced immune modulation. However, under hypoxic conditions, reduced DNA damage and enhanced DNA damage repair (DDR) lead to lower cytosolic double-stranded DNA (dsDNA) levels, making standard RT doses inadequate for sustained cGAS-STING activation, resulting in transient immune responses. In this study, we developed the As-Mn@MnO2@Alb nanosystem as a potent radiosensitizer and cGAS-STING pathway amplifier. Arsenic trioxide (ATO)-mediated radiosensitization suppresses DDR, enhances immunogenic cell death, and increases tumor-as

T-cell activity feedback-guided photothermal immunotherapy enabled by immune-responsive redox nanodots.

Immunogenic cell death (ICD)-based tumor therapies are fundamentally limited by the absence of real-time immune activity readouts and by dysregulated oxidative stress during treatment, which together undermine immune preservation and therapeutic consistency. To address these challenges, an ultrasmall polychlorogenic acid nanodot platform, termed PCGA-ND, was developed via Fe3+ and TEMPO catalyzed oxidative polymerization of chlorogenic acid. The resulting PCGA-NDs exhibited a uniform size of approximately 5 nm. Compared with conventional polyphenol-based materials, PCGA-NDs exhibited significa

HIFU postoperative hypoxia-activated metal-organic frameworks modulate the tumor microenvironment to augment immunotherapy.

Limited intensity and duration of high-intensity focused ultrasound (HIFU)-induced immune response largely hinder postoperative immunotherapy due to low immunogenicity and immunosuppression of tumor microenvironment (TME). In this study, effect-specific metal-organic frameworks (MOFs) were designed based on the severe hypoxia of postoperative tumors through regulating TME to enhance body's antitumor immune response. The combination of iron ions, hypoxic-activated prodrug banoxantrone, and indoleamine 2,3-dioxygenase (IDO) signaling pathway inhibitor NLG919 is utilized to construct MOFs loaded

A Programmable Nanoreactor Orchestrates Cascade of DNA Sensing to Amplify cGAS-STING Activation for Cancer Immunotherapy.

The cGAS-STING pathway, a critical cytosolic DNA-sensing mechanism in innate immunity, holds significant promise for cancer immunotherapy. However, conventional DNA-damaging therapies lack tumor specificity and cause damage to normal tissue. Furthermore, dendritic cells (DCs), central to the STING-mediated immune response, exhibit extrinsic immunosuppression via inhibitory receptors such as T-cell immunoglobulin and mucin-domain containing-3 (TIM-3), which impairs DNA internalization and subsequent pathway activation. Herein, we engineered a telomere stress-induced nanoreactor composed of a pH

Self-assembled supramolecular nanoparticles of α-mangostin for targeted STING activation and enhanced tumor immunotherapy.

Targeted activation of the stimulator of interferon genes (STING) signaling pathway represents a promising strategy to counteract immunosuppressive tumor microenvironments (TME) and elicit anti-tumor immunity. However, clinical translation of STING agonists has been strongly hindered by the challenges in achieving tumor-targeted STING activation. Here, we report a facile and efficient supramolecular self-assembly strategy for tumor-targeted STING activation of α-mangostin (MGT), a natural non-cyclic dinucleotide STING agonist. MGT can self-assemble into uniform supramolecular nanoparticles (MG

A locoregional program of photothermal debulking, stemness suppression, and dendritic cell activation for durable glioblastoma immunotherapy.

Glioblastoma (GBM), especially in large or unresectable tumors, remains difficult to treat because of the lack of effective debulking, the persistence of therapy-resistant glioma stem cells (GSCs), and a profoundly immunosuppressive tumor microenvironment. Here, a locoregional hydrogel platform is developed to integrate photothermal debulking, post-ablation GSC suppression, and dendritic-cell-mediated immune activation for GBM therapy. The brain-compliant injectable hydrogel (MIN-PPIC@iGel) co-encapsulates indocyanine green/napabucasin-loaded micelles and poly(I:C)-loaded polymersomes, enablin

Bioinspired Nanoplatform Potentiates Sonodynamic Immunotherapy by Remodeling the Antioxidant Tumor Microenvironment and Activating STING pathway.

BACKGROUND: Sonodynamic therapy (SDT) has emerged as a promising modality for treating deep-seated tumors. It has been demonstrated that SDT effectively induces immunogenic cell death (ICD), thereby initiating a systemic anti-tumor immune response - a process known as sonodynamic immunotherapy. However, its efficacy is severely limited by the hypoxic tumor microenvironment (TME) and elevated glutathione (GSH) levels, which together scavenge reactive oxygen species (ROS) and create a potent antioxidant barrier. METHODS: Ultra-small Mn3O4 nanoparticles with multi-enzyme mimicking activity were

LDH and glycolytic activity as predictors of immunotherapy response in gastric cancer: a systematic review and meta-analysis.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have transformed gastric cancer (GC) treatment, but response heterogeneity necessitates reliable prognostic biomarkers. This meta-analysis investigates the predictive value of metabolic (LDH, glycolytic activity) and inflammatory markers (NLR, PLR, LMR) in GC patients receiving ICIs. METHODS: We systematically analyzed 17 studies (n=3,842) from major databases through March 2024. Pooled hazard ratios (HRs) for overall survival (OS) and progression-free survival (PFS) were calculated using random-effects models, with subgroup analyses by treatmen

Checkpoint immunotherapy is associated with preferential activation of tumor antigen-specific CD4+ T cells in MDS.

A growing body of literature suggests that the efficacy of DNA hypomethylating agents are mediated via activation of antitumor immune mechanisms. Based upon this hypothesis, early phase trials combining immune checkpoint inhibitors (ICIs) with azacitidine in patients with myelodysplastic syndrome (MDS)/acute myeloid leukemia (AML) were undertaken, but clinical and immunologic efficacy have proven disappointing. In these studies, the lack of antigen specificity made systematic assessment of the anti-MDS immune response challenging. We hypothesized that combining vaccination against the New York

Mitochondria-targeting polymer cLipG/CuET activates the cGAS/STING pathway to enhance cholangiocarcinoma immunotherapy.

BACKGROUND: The remodeling of extracellular matrix often fails chemotherapeutic agents to fight intrahepatic cholangiocarcinoma (ICC). The cGAS/STING pathway can trigger the innate immune response to obtain better immunotherapeutic outcomes. RESULTS: A cLipG/CuET nanocomposite was designed and synthesized based on glycyrrhizic acid (GA) and copper diethyldithiocarbamate (CuET). GA can open mitochondrial permeability transition pores (MPTPs), allowing Cu (II) to interrupt mitochondrial function through copper toxicity. By activating the cGAS-STING signaling pathway in macrophages, this mitocho

Hypoxia-activated paclitaxel prodrugs enable PD-L1 degradation to potentiate cancer chemo-immunotherapy.

The combination of chemotherapy and immune checkpoint blockade therapy shows great potential in tumor treatment, but their integration remains a great challenge. Herein, hypoxia-responsive prodrugs integrating paclitaxel (PTX) and bromodomain-containing protein 4 inhibitor JQ1 were designed and fabricated into nanoparticles (DJNP NPs) in the presence of DSPE-PEG2000. After accumulation at the tumor site, DJNP NPs can respond to tumor hypoxic condition and trigger the release of PTX and JQ1. The released PTX can result in cell apoptosis, which further evokes immunogenic cell death to boost the
