Isatindigotindolosides, indoles containing a 1-S-β-glucopyranosyl unit at position C2, show promising bioactivity. Here, we report a copper-catalyzed C2- or C3-thioglycosylation of indoles with N-(thioglycosides)succinimides to construct indole alkaloid glucosides. This reaction is widely tolerant of functional groups, as various indoles and thioglycosides are suitable. It also provides a reliable method for performing late-stage modifications of natural products, such as gramine and melatonin. Total syntheses of isatindigotindolosides I and II were successfully accomplished using the C2-thiog

Endophytic fungi are striking resources rich in bioactive structures with agrochemical significance. In order to maximize the opportunity of search for bioactive compounds, chemical epigenetic manipulation was introduced to enhance the structural diversity of the fungal products, and an UPLC-ESIMS and bioassay-guided separation was used to detect novel bioactive metabolites. Consequently, four previously undescribed compounds including two cyclopentenones (globosporins A and B) and two monoterpenoid indole alkaloids (globosporines C and D), as well as three known compounds, were isolated from

Claviceps paspali is a fungus that mainly parasitises Paspalum dilatatum, generating a structure denominated sclerotium, in which indole-diterpenoid alkaloids are isolated. Its action mechanism is related to the inhibition of the gamma-aminobutyric acid receptor. It basically affects bovines, triggering the tremorgenic syndrome, the prevalence of this intoxication being relatively low in Europe. This work describes a clinical case on a cattle farm in Seville (southern Spain), composed of 91 bovines, 60% of which were principally affected with nervous clinical signs. The diagnosis was based on

Bisnicalaterine alkaloids are structurally complex and biologically significant bisindole natural products. Herein, we report the first enantioselective total syntheses of bisnicalaterines B, C, and their N4' epimers using an eight-step longest linear sequence. Central to this strategy is a site- and stereoselective bioinspired Friedel-Crafts-type coupling between the eburnamine and geissoschizol fragment, enabling a highly selective formation of the bisindole core. The geissoschizol framework is efficiently assembled using a stereocontrolled Bischler-Napieralski cyclization/reduction sequence

Human respiratory syncytial virus (RSV) is highly contagious and the leading cause of severe respiratory tract illness in infants, elderly, and immunocompromised individuals. Toll-like receptor 7 (TLR7), a pattern recognition receptor recognising the ssRNA of RSV, activates proinflammatory pathways and triggers secretion of interferons (IFNs). On the one hand, the inflammatory responses help clear out virus. On the other hand, they lead to severe lung damage. Banlangen is a traditional Chinese herbal medicine commonly prescribed for respiratory virus infection treatment, but the mechanisms of

Streptoindoles A and B are enantiomeric alkaloids featuring a novel methylene-bridged dimeric framework, produced by a marine-derived Streptomyces species. Here, we demonstrate their assembly via a nonenzymatic cascade comprising oxidation, intramolecular condensation, and Mannich reaction from N-acetyltryptamine and formaldehyde under mild conditions. Furthermore, we uncover a butane-2,3-dione-promoted reaction manifold that converts either tryptamine or N-acetyltryptamine into diverse alkaloid derivatives. These findings significantly expand the accessible chemical space of tryptamine-derive

The branches and leaves of Tabernaemontana catharinensis were extracted with supercritical fluid using a mixture of CO(2) plus ethanol (SFE), and the indole alkaloid enriched fraction (AF3) was selected for anti-Leishmania activity studies. We found that AF3 exhibits a potent effect against intracellular amastigotes of Leishmania amazonensis, a causative agent of New World cutaneous leishmaniasis. AF3 inhibits Leishmania survival in a dose-dependent manner, and reached 88% inhibition of amastigote growth at 100 microg/mL. The anti-parasite effect was independent of nitric oxide (NO), since AF3

Catharanthus roseus is a medicinal plant widely known for producing monoterpenoid indole alkaloids (MIAs), including therapeutic compounds such as vinblastine and vincristine, which are crucial for cancer treatment. However, the naturally low concentration of these alkaloids in plant tissues poses a significant challenge for large-scale production. This study explores the application of siderophore-producing bacteria for seed bacterization of Catharanthus roseus to enhance the production of MIAs, including vindoline, catharanthine, and vinblastine. Alkaloids are natural compounds useful as scaffolds for discovering new bioactive molecules. This study utilized alkaloid gramine to synthesize two groups of C3-substituted indole derivatives, which were either functionalized at N1 or not. The compounds were characterized by spectroscopic methods. The protective effects of the new compounds against in vitro oxidative hemolysis induced by standard oxidant 2,2'-azobis(2-amidinopropane dihydro chloride (AAPH) on human erythrocytes as a cell model were investigated. Additionally, the compounds were screened for antimicrobial activity. The results

The molecular network-guided exploration of the alkaloid extract of Callichilia inaequalis stems revealed a cluster attributed tentatively to dimeric monoterpene indole alkaloids of the rare criophylline subtype, initiating the dual study reported herein. A patrimonial-themed portion of this work was aimed at performing a spectroscopic reassessment of criophylline (1), a monoterpene bisindole alkaloid for which the nature of the inter-monomeric connectivity and configurational assignments have remained dubious. A targeted isolation of the entity annotated as criophylline (1) was undertaken to

On the occasion of receiving the Pharmaceutical Society of Japan Award 2020, I explained our research activities on the total syntheses of polycyclic alkaloids as an invited review. The structure of lundurine B, which has an unstable cyclopropane fused indoline skeleton, was proved firstly by the total synthesis. I also describe the total syntheses of optically active lundurine B and rapidilectine B utilizing asymmetric desymmetrization of the spiro intermediate. We developed an intramolecular bond formation reaction between the 2-position of the furan ring to the iminium cation (furane-iminiu

Indoles are chemical compounds that naturally occur in Prokaryotes and Eukaryotes. For example, indole alkaloids mainly occur in plant families such as Nyssaceae, Rubiaceae, Apocynaceae, and Loganiaceae. The popularity of indoles has led to further research into their function and synthetic modification. Meanwhile, the Wnt (Wingless and Int-1) signaling pathway is responsible for cell regulatory processes, while its dysfunction often constitutes the molecular basis for the pathogenesis of neoplastic processes.
